Allelic Diversity, De Novo CAG Expansions and Intergenerational Instability at the HTT Locus in a Sample from India.

BACKGROUND: Huntington's disease (HD) is an inherited, neurodegenerative disorder caused by the expansion of an unstable CAG repeat sequence in the Huntingtin (HTT) gene. The prevalence of HD, allelic diversity, rate of novel expansions and the clinical correlates vary across populations. OBJECTIVE: We analyzed the diversity of alleles and their clinical correlates and examined the inheritance patterns and the pattern of instability of CAG repeats in a few families. METHODS: Clinical history and pedigree structure were collected from records or through interviews between 2016 and 2019. Genet

Prevalence of MUTYH Monoallelic Variants in Patients With Hereditary Cancer Using Multigene Panel Testing.

BACKGROUND: The MUTYH gene is involved in DNA repair and is known for MAP (MUTYH-associated polyposis), an autosomal recessive disorder that predisposes individuals to colorectal cancer (CRC), with a lifetime risk ranging from 40% to 90%. Homozygosity or double heterozygosity (DH) for pathogenic variants (PVs) in MUTYH causes MAP, but several studies suggest that monoallelic PVs may also increase cancer risk, mainly CRC and breast cancer (BC). METHODS: We analyzed MUTYH status in a cohort of 130 patients referred to our familial cancer clinic for suspected hereditary cancer, describing their

Successful pre-implantation genetic testing for autosomal recessive cutis laxa: clinical utility of a multidisciplinary team approach.

PURPOSE: Pre-implantation genetic testing for monogenic disorders (PGT-M) enables couples at risk of transmitting serious genetic disorders to their offspring to give birth to healthy children. Here, we present an illustrative case of the use of PGT-M in a couple who were both carriers of autosomal recessive cutis laxa (ARCL) and describe the application of PGT-M in our reproductive center using a multidisciplinary team approach. METHODS: The couple experienced four adverse pregnancy outcomes, and both partners were subsequently identified as carriers of ARCL caused by PYCR1 gene variants dur

Association of Sperm DNA Fragmentation and Intracytoplasmic Sperm Injection Outcomes in an Age-adjusted Cohort: A Retrospective Cohort Study.

BACKGROUND: Sperm DNA fragmentation index (DFI) is increasingly recognised as a critical parameter in male infertility assessment. However, its prognostic utility in assisted reproduction, particularly in intracytoplasmic sperm injection (ICSI) cycles, remains uncertain. AIM: This study aims to evaluate the association between sperm DFI and clinical outcomes following ICSI and to examine whether this relationship is independent of maternal age. SETTINGS AND DESIGN: A multicentre retrospective cohort study across private fertility centres in India. MATERIALS AND METHODS: A total of 688 coupl

Decoding hybridization barriers: the molecular and genetic orchestration of the triploid block in Arabidopsis thaliana.

Plant evolution has been greatly influenced by polyploidization phenomena. Polyploid plants yield more and are more resistant to unfavorable environments than their diploid relatives. The triploid block, a postzygotic barrier that causes failure of endosperm development and thus seed arrest, often prevents polyploid breeding. Alterations in the parental dose in interploidy crosses alter endosperm development by changing the correct maternal: paternal ratio (2m:1p) that this tissue requires to properly fulfill its proliferation and cellularization. After many years of research, the study of epi

Long-Term Neurodevelopmental Outcomes of Late Preterm Children: A Pilot Study on the Role of Early Nutrition.

Background: Late preterm delivery accounts for approximately 5% of all births. Although there is a growing body of literature recognizing that children born late preterm are at risk for mild neurodevelopmental issues, these children are often excluded from follow-up programs. From a preventive perspective, it could be useful to promote protective factors for child development. Among these, human milk feeding is highly recommended due to its advantages for child health and development. However, there is limited research on the complex interactions between late preterm birth, human milk feeding,

Serum EMMPRIN/CD147 promotes the lung pre-metastatic niche in a D2A1 mammary carcinoma mouse model.

Several types of cancer, including breast cancer, metastasize to the lung. However, before the disseminating tumor cells (DTCs) arrive there, the lung must be prepared as a hospitable environment for them, forming the pre-metastatic niche (PMN). It is now accepted that the primary tumor can release soluble mediators or extracellular vesicles that activate the PMN resident cells, recruit immune cells, promote angiogenesis, and remodel the extracellular matrix (ECM), even before the arrival of DTCs to the niche. However, not all the components of the tumor secretome are known. Here we demonstrat

A lack of commensal microbiota influences the male reproductive tract intergenerationally in mice.

IN BRIEF: Germ-free mice display epididymal transcriptomic changes that were also evident in their conventionalized male offspring and mice lacking T and B cells. This paper demonstrates the role of microbiota and immune cells in the epididymis. ABSTRACT: The microbiome encompasses the array of microorganisms inhabiting various niches in the body and is necessary for numerous physiological processes, including normal metabolism and a functioning immune system. Not only does the absence of a microbiome in mice impact the exposed animals but also inherited phenotypes in successive generations o

From molecules to models: A holistic review of autism spectrum disorder mechanisms and research tools.

Autism Spectrum Disorder (ASD) is a complex neurodevelopmental condition characterized by persistent deficits in social communication and interaction, as well as restricted, repetitive patterns of behavior, interests, or activities. These features are associated with atypical early brain development and connectivity. While ASD has been traditionally associated with molecular genetic alterations, recent research highlights the significant contribution of various environmental factors to the pathophysiology of the disorder. Pathogenic genetic variations in key regulatory genes remain central to

Six cases of ectopic cutaneous ossification associated with GNAS gene variants.

The GNAS gene gives rise to stimulatory G protein Gs-alpha (Gsα), a pivotal transducer of hormonal signalling pathways. Variants inherited maternally lead to reduced Gsα activity in certain tissues, resulting in resistance to multiple hormones and manifesting clinically as pseudohypoparathyroidism (PHP). In contrast, paternal transmission is linked to pseudopseudohypoparathyroidism (PPHP), which is characterized by extensive ectopic cutaneous ossification but lacks endocrine resistance. Thus, the phenotypic spectrum arising from GNAS gene variants is profoundly influenced by the parent-of-orig
