[Methicillin resistant Staphylococcus aureus: phenotypic and genotypic studies].

BACKGROUND: Staphylococcus aureus is a human opportunistic pathogen. Its important pathogenicity and the increasingly rate of resistance to methicillin are the main causes of morbidity and mortality. AIM: In order to evaluate the epidemiologic situation of Methicillin Resistant S. aureus (MRSA) at Charles Nicolle hospital. METHODS: A four years retrospective study (January 1999-December 2002) was conducted. RESULTS: 65 non redundant MRSA isolates were collected. Identification was based on morphology, culture and biochemical characters. Antibiotic susceptibility was determined by disk diffu

Variable phenotypic manifestations of a K44N mutation in the TGIF gene.

The etiologies and clinical spectra of HPE are extremely heterogeneous. Here, we report a Brazilian boy with lobar holoprosencephaly who was ascertained in a sample of 60 patients with HPE and HPE-like phenotypes and screened for molecular analysis of the major HPE causative genes: SHH, PTCH, SIX3, GLI2, and TGIF. This boy presented a p.K44N (c.132G>T) mutation in exon 2 of the TGIF gene which was inherited from his phenotypically normal mother. This mutation leads to lysine to arginine amino acid change and is predicted to be a damaging mutation. Clinical aspects involving variable phenotypic…

Pigmented purpuric dermatosis: classification by phenotypic and molecular profiles.

The categorization of pigmented purpuric dermatosis (PPD) as a form of cutaneous lymphoid dyscrasia has been suggested. Phenotypic and molecular studies were done on 43 patients with PPD. The molecular studies used a capillary gel electrophoresis T-cell receptor beta multiplex polymerase chain reaction assay. There were 2 principal categories: polyclonal PPD represented by 22 cases and monoclonal variants comprising 21 cases. Monoclonal cases had extensive skin lesions. An identical restricted T-cell repertoire independent of time and location was observed. Approximately 40% of the monoclonal

Streptococcus agalactiae in pregnant women: phenotypic and genotypic characters.

OBJECTIVE: Streptococcus agalactiae (GBS) is considered a leading cause of neonatal sepsis. We evaluated the phenotypic and genotypic characters of 73 S. agalactiae strains isolated from different women at the 35-37 weeks of pregnancy. METHODS: Isolates were characterized by serotyping (direct agglutination) and by pulsed-field-gel-electrophoresis (PFGE). Resistance to antimicrobials (penicillin, macrolides, lincosamides, quinolones and tetracyclines) was assessed. RESULTS: All isolates were serologically typeable and ascribable to one of the six tested serotypes (Ia, Ib, II, III, IV, and V)

Phenotypic characterization of isolated valvular interstitial cell subpopulations.

BACKGROUND AND AIM OF THE STUDY: Valvular interstitial cells (VICs) demonstrate a heterogeneous range of phenotypes such as variable expression of smooth muscle alpha-actin (SMalphaA). Myofibroblast-like VICs, expressing high levels of SMalphaA, are thought to be involved in myxomatous degeneration of mitral valves. The inability to isolate specific cell types has restricted potential investigations of valvular disease mechanisms. Thus, investigations were conducted into methods of isolating different cell subpopulations from primary VICs as a preparatory step for cell type-specific evaluation…

Phenotypic switching and its implications for the pathogenesis of Cryptococcus neoformans.

Phenotypic switching has been described in several strains of Cryptococcus neoformans. It occurs in vivo during chronic infection and is associated with differential gene expression and changes in virulence. The switch involves changes in the polysaccharide capsule and cell wall that affect the yeast's ability to resist phagocytosis. In addition, the phenotypic switch variants elicit qualitatively different inflammatory responses in the host. The host's immune response ultimately affects selection of the switch variants in animal models of chronic cryptococcosis. The biological relevance of ph

A comparison of vestibular and auditory phenotypes in inbred mouse strains.

The purposes of this research were to quantify gravity receptor function in inbred mouse strains and compare vestibular and auditory function for strain- and age-matched animals. Vestibular evoked potentials (VsEPs) were collected for 19 inbred strains at ages from 35 to 389 days old. On average, C57BL/6J (35 to 190 days), BALB/cByJ, C3H/HeSnJ, CBA/J, and young LP/J mice had VsEP thresholds comparable to normal. Elevated VsEP thresholds were found for elderly C57BL/6J, NOD.NONH2(kb), BUB/BnJ, A/J, DBA/2J, NOD/LtJ, A/WySnJ, MRL/MpJ, A/HeJ, CAST/Ei, SJL/J, elderly LP/J, and CE/J. These results s

Characteristics of COPD phenotypes classified according to the findings of HRCT.

The present study was performed to clarify the clinical characteristics of chronic obstructive pulmonary disease (COPD) patients classified into phenotypes according to the dominancy of emphysema and the presence of bronchial wall thickening evaluated by chest high-resolution computed tomography (HRCT). Eighty-five patients with stable COPD (FEV1 < or = 80%) were examined by chest HRCT. Emphysematous changes and bronchial wall thickening were evaluated visually, and COPD patients were classified into three phenotypes: absence of emphysema, with little emphysema with or without bronchial wall t…

Phenotypic characterization of autoreactive T cells in multiple sclerosis.

MS has been hypothesized to result from autoreactive T cell responses against myelin antigens. In this report, we examined myelin-specific CD8 and CD4 T cells for two markers differentially expressed on naïve, memory and chronically stimulated T cells, CD28 and CD57. We observed differential expression on CD8 T cells in response to myelin antigens, but not in response to the recall antigen mumps. We demonstrate these cells display reduced proliferation and this may explain why therapies that limit the proliferation of T cells have had little effect on the course of MS, particularly later in th

Genetic and phenotypic characterization of Listeria monocytogenes lineage III.

Listeria monocytogenes has been previously grouped into three evolutionary groups, termed lineages I, II and III. While lineages I and II are commonly isolated from various sources, lineage III isolates are rare and have several atypical and unique phenotypic characteristics. Relative to their prevalence in other sources, lineage III strains are overrepresented among isolates from food-production animals, and underrepresented among isolates from human clinical cases and foods. This work describes an extensive genotypic and phenotypic characterization of 46 lineage III isolates. Phylogenetic an

Divergent phenotypes in Gaucher disease implicate the role of modifiers.

BACKGROUND: Gaucher disease is classified into neuronopathic and non-neuronopathic forms with wide phenotypic variation among patients sharing the same genotype. While homozygosity for the common L444P allele usually correlates with the neuronopathic forms, how a defined genotype leads to a phenotype remains unknown. METHODS: The genetic and epigenetic factors causing phenotypic differences were approached by a clinical association study in 32 children homozygous for the point mutation L444P. Direct sequencing and Southern blots were utilised to establish the genotype and exclude recombinant …
