Inflammation-targeted nanomedicine prevents tumor metastasis following photodynamic therapy by reversing epithelial-mesenchymal transition and ROS-mediated immunosuppression.

BACKGROUND: Prolonging the duration of photodynamic therapy (PDT) enhances the level of reactive oxygen species (ROS), thereby facilitating tumor ablation. However, our findings indicated that excessive ROS not only induces epithelial-mesenchymal transition (EMT) but also creates an immunosuppressive microenvironment in tumor, thereby triggering tumor metastasis. METHODS: We initially developed neutrophil membrane hybrid liposomes (NLs) that can specifically target inflamed tumor tissues following PDT. Then, we utilized NLs to encapsulate the antioxidant nanozyme FeGA and the antiplatelet dru

Photodynamic therapy reduced stemness and cell migration in triple-negative breast cancer cells through the PTEN/CD44 signaling pathway: A comparative study.

Triple-negative breast cancer (TNBC) is a highly aggressive, and the prognosis of affected patients is still unfavorable. Cancer stem cells have been highly implicated in tumor relapse and metastasis. Although zinc phthalocyanine (ZnPc) photodynamic therapy (PDT) has shown promising results in treating some cancers, the role of various PDT parameters on TNBC stemness remains unclear. In addition to performing in silico investigations, we evaluated the cytotoxic effects of ZnPc-PDT (8 and 24 J/cm2) on MDA-MB-468 cells using the MTT assay. We investigated its effects on apoptosis, cell cycle, an

Photo-immunomodulatory multifunctional nanoliposomes targeting myeloid derived suppressor cells augment antitumor immunity for colon cancer.

The therapeutic effect of cancer immunotherapy is limited by immune resistance caused by the immunosuppressive tumor microenvironment (TME). While photodynamic therapy (PDT) can enhance tumor immunogenicity and initiate anti-tumor immunity, its practical benefits are limited. To address this, we developed a TME-responsive nanoliposome for the co-delivery of the photosensitizer chlorin e6 (Ce6) and the immunomodulatory agent AMD3100. Our results demonstrated that Ce6-mediated PDT effectively induced immunogenic cell death (ICD) in colon cancer cells, as evidenced by the robust surface exposure

Unravelling the modes of phototoxicity of NIR absorbing chlorophyll derivative in cancer cells under normoxic and hypoxic conditions.

The efficacy of photodynamic treatment (PDT) against deep-seated tumor is hindered by low penetration depth of light as well as hypoxic conditions which prevails in tumor. To overcome this limitation, Near-infrared (NIR) absorbing photosensitizers have been investigated actively. In the present study we evaluated the PDT efficacy of an NIR absorbing chlorophyll derivative 'Cycloimide Purpurin-18 (CIPp-18)' in Human Breast carcinoma (MCF-7) and cervical adenocarcinoma (Hela) cells under normoxic and hypoxic conditions. PDT with CIPp-18 (2.0 µM, 3 h) and NIR light (700 ± 25 nm, 0.36-1.4 J /cm2)

Engineered microalgae for photo-sonodynamic synergistic therapy in breast cancer treatment.

Dynamic therapies such as photodynamic therapy (PDT) and sonodynamic therapy (SDT) have potential in cancer treatment. Microalgae have attracted increasing attention because of their high active mobility, flexibility in terms of functionality, and good biocompatibility. In this study, surface-engineered microalgae Chlorella vulgaris (Chl) modified with metal‒organic framework (MOF) nanoparticles (denoted Chl-MOF) are successfully developed for synergistic photo-sonodynamic therapy and immunotherapy. The resulting Chl-MOF can be used as an oxygenator for O2 generation through Chl-mediated photo

In vitro and in ovo photodynamic efficacy of nebulized curcumin-loaded tetraether lipid liposomes prepared by DC as stable drug delivery system.

Lung cancer is one of the most common causes of high mortality worldwide. Current treatment strategies, e.g., surgery, radiotherapy, chemotherapy, and immunotherapy, insufficiently affect the overall outcome. In this study, we used curcumin as a natural photosensitizer in photodynamic therapy and encapsulated it in liposomes consisting of stabilizing tetraether lipids aiming for a pulmonary drug delivery system against lung cancer. The liposomes with either hydrolyzed glycerol-dialkyl-glycerol tetraether (hGDGT) in different ratios or hydrolyzed glycerol-dialkyl-nonitol tetraether (hGDNT) were

Acidity-Activated Charge Conversion of 177Lu-Labeled Nanoagent for the Enhanced Photodynamic Radionuclide Therapy of Cancer.

Nanomaterials in combination with radionuclide therapy (RNT) provide new opportunities for cancer treatment. However, nanomaterials with efficient tumor accumulation have been less exploited for effective radionuclide-based therapy. Here, we report glycol chitosan-based nanoparticles (GCP-NPs) with acidic pH-dependent surface charge conversion for efficient radionuclide-based combination therapy. The nanoplatform can change the surface charge of nanoparticles from slight negative to positive in the acidic tumor microenvironment, which facilitates cellular internalization and penetration and th

The effect of high-risk HPV E6/E7 mRNA on the efficacy of topical photodynamic therapy with 5-aminolevulinic acid for cervical high-grade squamous intraepithelial lesions.

BACKGROUND: E6 and E7 high-risk human papillomavirus (HR-HPV) oncoproteins are closely associated with the initiation and progression of cervical cancer (CC) and pre-cancerous lesions. Cervical high-grade squamous intraepithelial lesions (HSIL), as pre-cancerous lesions, have a 5% chance of progressing to invasive cancer. Topical 5-aminolevulinic acid photodynamic therapy (ALA-PDT) is a novel non-invasive targeted therapy for intraepithelial lesions. Herein, we analyzed the effect of HR-HPV E6/E7 mRNA on ALA-PDT for cervical HSIL. METHODS: A retrospective analysis of 148 HR-HPV-positive patie

Anisotropic Truncated Octahedral Au with Pt Deposition on Arris for Localized Surface Plasmon Resonance-Enhanced Photothermal and Photodynamic Therapy of Osteosarcoma.

The multifunctional combined nanoplatform has a wide application prospect in the synergistic treatment of cancer. Nevertheless, the traditional treatment of phototherapy is limited by the catalytic nanomaterial itself, so the effect is not satisfactory. Here, the arris of the anisotropic truncated octahedral Au (TOh Au) was coated with noble metal Pt to form a spatial separation structure, which enhanced the local surface plasmonic resonance and thus boosted the photocatalytic effect. In this system, the highly efficient photocatalysis provides a strong guarantee for oncotherapy. On the one ha
