Advances in Odontogenic Regeneration: From Fundamental Research to Clinical Application.

Dentin is the primary mineralized tissue of the tooth, essential for pulp protection and enamel support. Its regenerative capacity is critical for dental health. This review summarizes recent progress in odontogenic regeneration, covering fundamental biological mechanisms and translational applications. A literature search of peer-reviewed studies on odontogenic regeneration, dental tissue engineering, and biomaterial-based strategies was conducted. Key advances include signaling pathway-guided stem cell differentiation into functional odontoblasts, which promotes reparative dentin matrix form…

Simultaneous regeneration of skin and bone in full-thickness cranial composite defects.

Traumatic cranial defects often involve concurrent loss of soft and hard tissues and can progress to chronic defects due to delayed healing associated with infection or other co-morbidities. Despite autologous reconstruction remaining the clinical standard, it requires staged procedures using heterogeneous tissues, increasing operative time, costs, and surgical risks. Moreover, current tissue engineering approaches focus on single tissues or acute tissue defect models, limiting their clinical applications. Herein, we describe an acellular, material-driven 3D-printed composite scaffold designed…

In vivo delivery strategies for therapeutic CRISPR genome editing.

CRISPR-based genome and epigenome editing technologies have rapidly evolved from programmable nucleases into a diverse therapeutic toolbox encompassing conventional CRISPR systems, base editing, prime editing, RNA targeting, and epigenetic modulation. While early clinical successes relied on ex vivo manipulation of patient-derived cells, recent advances in delivery chemistry and vector engineering are enabling direct in vivo editing across multiple organs. Here, we provide a comprehensive review of delivery modalities of CRISPR systems solely in vivo that underpin their therapeutic translation

Human Multi-Organ-on-a-Chip Platforms for Next-Generation Drug Delivery Strategies.

Human multi-organ-on-a-chip (MOoC) platforms have rapidly emerged as next-generation preclinical models for evaluating therapeutic delivery and efficacy. By integrating dynamic perfusion, physiologically relevant microenvironments, and systemic tissue-tissue crosstalk, MOoCs provide unprecedented opportunities to study drug absorption, distribution, metabolism, and toxicity under human-relevant conditions. In this review, we summarize recent progress in MOoC technologies and their application to diverse drug delivery strategies, including stem cell-based carriers, gene and nucleic acid therapi

Sacrificial oil shell method for the generation of alginate microbeads adapted to multicellular spheroid culture.

Three-dimensional cell culture provides a powerful framework for studying the growth of tissue in vitro. To account for biological variability, it is important to generate a large number of model tissues within well-controlled environments. In this context, droplet microfluidics has emerged as a promising tool for encapsulating cells in extracellular matrices with well-defined mechanical properties. However, its use in biology laboratories remains limited due to technical and biological challenges. In this work, we present a simple method for encapsulating mammalian cells in alginate gel micro

Advances in 3D printing for the repair of tympanic membrane perforation: a comprehensive review.

Tympanic membrane perforation (TMP) is one of the most common conditions in otolaryngology worldwide, and hearing damage caused by inadequate or prolonged healing can be distressing for patients. This article examines the rationale for utilizing three-dimensional (3D) printing to produce scaffolds for repairing TMP, compares the advantages and disadvantages of 3D printed and bioprinted grafts with traditional autologous materials and other tissue engineering materials in TMP repair, and highlights the practical and clinical significance of 3D printing in TMP repair while discussing the current…

Kartogenin-loaded hydrogel promotes intervertebral disc repair via protecting MSCs against reactive oxygen species microenvironment by Nrf2/TXNIP/NLRP3 axis.

Intervertebral disc (IVD) degeneration (IDD) and the consequent low back pain present a major medical challenge. Stem cell-based tissue engineering is promising for the treatment of IDD. However, stem cell-based treatment is severely impaired by the increased generation of reactive oxygen species (ROS) in degenerative disc, which can lead to a high level of cell dysfunction and even death. In this study, a kartogenin (KGN)@PLGA-GelMA/PRP composite hydrogel was designed and used as a carrier of ADSCs-based therapies in disc repair. Injectable composite hydrogel act as a carrier for controlled r…

Intravascular Fluorescence Molecular Imaging of Atherosclerosis.

Optical molecular imaging using near-infrared fluorescence (NIRF) light is an emerging high-resolution imaging approach to image a wide range of molecular and cellular species in vivo. Imaging using NIR wavelengths (650-900 nm) enables deeper photon penetration into tissue and reduced tissue autofluorescence, resulting in higher sensitivity to detect exogenously administered NIR fluorophores (injectable molecular imaging agents). Greater imaging depth of several centimeters is further achievable in the NIR window as blood absorption is as an order of magnitude lower than in the visible range.

Positional Memory in Vertebrate Regeneration: A Century's Insights from the Salamander Limb.

Salamanders, such as axolotls and newts, can regenerate complex tissues including entire limbs. But what mechanisms ensure that an amputated limb regenerates a limb, and not a tail or unpatterned tissue? An important concept in regeneration is positional memory-the notion that adult cells "remember" spatial identities assigned to them during embryogenesis (e.g., "head" or "hand") and use this information to restore the correct body parts after injury. Although positional memory is well documented at a phenomenological level, the underlying cellular and molecular bases are just beginning to be

Self-assembled insulin-like growth factor 1 peptides induce adipose stem cell differentiation to repair cartilage injury.

BACKGROUND: Tissue engineering using adipose-derived mesenchymal stem cells (ADSCs) promotes the regeneration of articular cartilage. However, insulin-like growth factor 1 (IGF-1), which is used to induce the differentiation of ADSCs into chondrocytes during treatment, is prone to instability and short tissue retention. METHODS: Nap-FFG-GYGSSSRRAPQT was used as an IGF-1 mimicking molecule. MTT and CCK-8 assays were performed to evaluate the proliferation ability of ADSCs. QRT-PCR and Western blot assays were used to assess the expression of cartilage-related genes. International Cartilage Reg…

Potential of Bone-Marrow-Derived Mesenchymal Stem Cells for Maxillofacial and Periodontal Regeneration: A Narrative Review.

Bone-marrow-derived mesenchymal stem cells (BM-MSCs) are one of the most widely studied postnatal stem cell populations and are considered to utilize more frequently in cell-based therapy and cancer. These types of stem cells can undergo multilineage differentiation including blood cells, cardiac cells, and osteogenic cells differentiation, thus providing an alternative source of mesenchymal stem cells (MSCs) for tissue engineering and personalized medicine. Despite the ability to reprogram human adult somatic cells to induced pluripotent stem cells (iPSCs) in culture which provided a great op…

A sterilizable, biocompatible, tropoelastin surface coating immobilized by energetic ion activation.

Biomimetic materials which integrate with surrounding tissues and regulate new tissue formation are attractive for tissue engineering and regenerative medicine. Plasma immersion ion-implanted (PIII) polyethersulfone (PES) provides an excellent platform for the irreversible immobilization of bioactive proteins and peptides. PIII treatment significantly improves PES wettability and results in the formation of acidic groups on the PES surface, with the highest concentration observed at 40-80 s of PIII treatment. The elastomeric protein tropoelastin can be stably adhered to PIII-treated PES in a c…
