Metabolomics and network pharmacology revealing the transdermal moxibustion compounds and their therapeutic mechanisms for rheumatoid arthritis.

BACKGROUND/PURPOSE: Moxibustion, a traditional therapy employing the combustion of Artemisia argyi, exerts notable clinical benefits in rheumatoid arthritis (RA). However, the identity of the transdermal moxibustion compounds (TMOCs) and their molecular mechanisms of action remain poorly understood. This study sought to define the core TMOCs and delineate their therapeutic mechanisms in RA. STUDY DESIGN/METHOD: GC-MS was used to extract the relative concentrations of TMOCs in the subcutaneous tissue, which were incorporated into the network pharmacology to identify the center targets of prima

Subcutaneous methadone is not different than transdermal fentanyl for postoperative analgesia in dogs with thoracolumbar disc disease, a prospective, randomised, blinded clinical study.

BACKGROUND: Thoracolumbar disc disease is a common neurological condition in dogs, which incorporates different pain components. Multimodal analgesic treatments, especially postoperatively, are often based on opioids and require an intravenous catheter for drug application. This might impede early physiotherapy and mobilisation. Different composite pain scales and sensory testing devices exist to evaluate postoperative pain behaviour in dogs. At present, no data are available to clearly recommend one tool or technique after spinal surgery over others. Therefore, the primary aim of this study w

Oxidation-dependent effects of alpha-1 antitrypsin on wound healing and inflammation.

Wound healing requires a delicate balance between cellular and molecular factors, all affected by reactive oxygen species (ROS). While ROS decontaminate, they also might lead to impaired wound healing, as evident in radiation-exposed skin and in venous insufficiency. Human alpha-1 antitrypsin (hAAT) is a circulating antiprotease that is anti-inflammatory and tissue-protective. Accordingly, tissue repair is enhanced in hAAT-rich conditions. hAAT undergoes oxidative modification in high-ROS environments, which alters its functional properties. While its antiprotease function is lost, the consequ

Hispidulin-rich fraction of Clerodendrum fragrans Wild. (Sesewanua) dissolving microneedle as antithrombosis candidate: A proof of concept study.

Existing conventional antithrombosis drugs have caused many side effects, opening up opportunities for the development of new thrombotic drugs. There is potential to use the hispidulin-rich fraction of sesewanua (HRFS) as a new antithrombotic. The oral route limitation of hispidulin, as a low water solubility and non-polar compound, can be addressed. This study explores the potential of HRFS in the form of dissolving microneedles (DMN). The formula was created using polymers such as polyvinyl alcohol (PVA), polyvinyl pyrrolidone K-30 (PVP), and non-ionic surfactant. Ex vivo permeation studies
