Dcun1d3 is dispensable for spermatogenesis and male fertility in mice.

BACKGROUND: DCUN1D3, a member of the DCNL (defective in cullin neddylation-like) protein family, has been implicated in ultraviolet (UV) radiation-induced cell cycle checkpoints, cell growth, survival, and neddylation. However, its specific function in male germ cells and potential involvement in spermatogenesis remain poorly understood. METHODS: To investigate the role of Dcun1d3 in male reproduction, we generated Dcun1d3 knockout (KO) mice. Sperm parameters were evaluated using computer-assisted sperm analysis (CASA), while histological and immunohistochemical analyses were performed to ass

Dual-Modality Therapy: Synergistic Enhancement of Radio-Hyperthermia by Gold-Gold Sulfide Nanoparticles in MCF-7 Cells.

BACKGROUND: This study examines the synergistic impact of Gold-Gold Sulfide (GGS) nanoparticles combined with hyperthermia and radiotherapy on MCF-7 cancer cells. GGS nanoparticles, with strong near-infrared absorption and photothermal properties, enhance cellular sensitivity to radiotherapy. METHODS: MCF-7 cells were treated with varying GGS concentrations and exposed to radiation doses of 50, 100, and 200 cGy, alongside laser irradiation for 10, 40, and 80 s. The IC50 for GGS nanoparticles was approximately 350 μM. RESULTS: Results revealed a significant reduction in cell viability with th

Merkel cell carcinoma presenting as a nasal dorsum mass: A case report and literature review.

BACKGROUND: Merkel cell carcinoma (MCC) is a rare and aggressive skin cancer with high incidence in older and immunocompromised patients. Its occurrence in the nasal dorsum is extremely rare and poses significant diagnostic and therapeutic challenges. CASE SUMMARY: We report the case of a 65-year-old woman with diabetes mellitus and hypertension who presented with a dorsal nasal mass. The initial differential diagnosis favored hemangioma-based clinical examination on imaging. Histopathological examination after excision revealed MCC, necessitating a complex management strategy. A 3 cm × 2 cm

Unfolded protein response modulates Tyrosinase levels and melanin production during melanogenesis.

BACKGROUND: Melanocytes protect the body from ultraviolet radiation by synthesizing melanin. Tyrosinase, a key enzyme in melanin production, accumulates in the endoplasmic reticulum (ER) during melanin synthesis, potentially causing ER stress. However, regulating ER function for melanin synthesis has been less studied than controlling Tyrosinase activity. OBJECTIVE: This study investigates the regulatory mechanisms of melanin production, focusing on ER stress and the ER stress-induced response. METHODS: B16 mouse melanoma cells induced to undergo melanogenesis were treated with unfolded prot

Microbubbles at the bedside: intracavitary CEUS to confirm chest-drain position and loculations.

BACKGROUND: CEUS is increasingly used beyond hepatobiliary imaging; its pleuro-pulmonary applications remain limited, especially for device assessment. CASE DESCRIPTION: We describe a postoperative patient with moderate right pleural effusion and associated middle-basal atelectasis causing hypoxemic respiratory failure. After ultrasound-guided insertion of a 12-Fr pigtail drain, the catheter trajectory and tip could not be confidently visualized due to limited patient mobilization and small-bore caliber. A second-generation UCA (sulfur hexafluoride) diluted 1 mL in 10 mL saline was instilled

Hyaluronic acid-functionalized nanoarmor enhances the stable colonization ability of Paenibacillus polymyxa JF_P68 and boosts its biological control efficacy against pear anthracnose.

BACKGROUND: The application of biocontrol agents offers a promising strategy for managing pear anthracnose, yet their efficacy is often limited by unstable colonization on fruit surfaces. RESULTS: Here, we isolated Paenibacillus polymyxa JF_P68, which exhibits strong inhibitory activity against Colletotrichum fructicola. To enhance its environmental tolerance, we developed a hyaluronic acid (HA)-functionalized nanoarmor using a layer-by-layer coating technique. This nanoarmor significantly improved the JF_P68's resistance to ultraviolet radiation and oxidative stress. In vivo trials demonstra

Cardiovascular risks in men with prostate cancer: a pragmatic, clinician-oriented review of risk stratification and management strategies.

BACKGROUND/OBJECTIVES: Recent advances in prostate cancer management have improved overall survival and quality of life for those afflicted. However, these patients continue to suffer from a high burden of cardiovascular disease through a variety of mechanisms, including both the cancer itself as well as prostate cancer-specific treatments. SUBJECTS/METHODS: Cardiovascular disease is now the leading cause of death in this population, and current data suggests that many patients remain sub-optimally controlled from a risk factor standpoint. There is a lack of consensus and standardized guideli

Hemorrhagic astroblastoma: atypical presentation of a rare tumor. Illustrative case.

BACKGROUND: Astroblastoma is an extremely rare tumor of the central nervous system, and its origin and validity as a different entity are still being debated. Because of its rarity and similarities to other glial neoplasms, it is often misdiagnosed, impacting treatment and outcomes. OBSERVATIONS: Astroblastoma is very rare and mainly affects children and young adults. It does not have a definitive World Health Organization grade and is divided into low- and high-grade categories. The clinical presentation is consistent with signs of intracranial hypertension. Radiographic imaging shows a bubb
