Caffeine-induced calcium oscillations in heavy-sarcoplasmic-reticulum vesicles from rabbit skeletal muscle.

Heavy-sarcoplasmic-reticulum vesicles from rabbit skeletal muscle show not only caffeine-induced calcium release in a medium allowing active calcium loading, but also oscillations in calcium concentration under appropriate conditions. The xanthine derivatives 7-isobutyl-1-methylxanthine and theophylline also induce oscillations under the same conditions. Calcium-releasing substances with other chemical structures such as adenosine nucleotides or calmodulin antagonists do not induce this effect. With the help of specific inhibitors such as ruthenium red, neomycin or magnesium it was demonstrate…

Inhibition of P-type and N-type calcium channels by dopamine receptor antagonists.

P-type Ca2+ channels in cerebellar Purkinje neurons and N-type Ca2+ channels in sympathetic neurons were found to be inhibited by D2 dopamine receptor antagonists with diverse structures, including phenothiazines (chlorpromazine and thioridazine), diphenylbutylpiperidines (fluspirilene and pimozide), butyrophenones (haloperidol and spiperone), and a piperazine (fluphenazine). Dopamine and quinpirole had no effect on P-type Ca2+ channels. In all cases, inhibition was characterized by slow onset and offset. The effects of P-type and N-type channels were very similar. Fluspirilene was the most po

Calcium, phosphorus and magnesium intakes correlate with bone mineral content in postmenopausal women.

Qualitative and quantitative differences in the dietary habits of postmenopausal women were studied to assess their influence on bone health and osteoporosis. A total of 194 postmenopausal women were studied with forearm DEXA densitometry. 70 were osteoporotic and 124 served as controls. Women had been menopausal for 5-7 years, and had never been treated with hormone replacement or drug therapy. A 3-day dietary recall was completed on Sunday, Monday and Tuesday after the examination: the results were processed by computer and daily calcium, phosphorus and magnesium intakes were related to bone…

Torsade de pointes and elevated magnesium and calcium requirements associated with intravenous pentamidine.

We present a report of markedly elevated calcium and magnesium requirements in an AIDS patient receiving central venous nutrition. These increased requirements were attributed to urinary magnesium losses associated with intravenous pentamidine therapy. The combination of intravenous pentamidine and electrolyte abnormalities resulted in the development of the dysrhythmia, torsade de pointes, in the patient. A review of literature presenting similar cases is included. Nutrition practitioners should be aware of the often prolonged effect of intravenous pentamidine on magnesium and calcium homeost…

Decreased melatonin biosynthesis, calcium flux, pineal gland calcification and aging: a hypothetical framework.

Increased pineal calcifications and decreased pineal melatonin biosynthesis, both age related, support the notion of a pineal bio-organic timing mechanism. Decreased calcium ion availability is the single common denominator of diminished beta-postreceptor- and alpha-receptor-stimulating functions in beta-receptor potentiation, which is necessary for nocturnal peak melatonin production. A comprehensive framework for the interaction of aging pineal cell mechanisms, calcium flux and melatonin biosynthesis is presented.

Epstein-Barr virus latent membrane protein 2A blocks calcium mobilization in B lymphocytes.

LMP2A is expressed in latent Epstein-Barr virus (EBV) infection and interacts with LMP1 and members of the src tyrosine kinase family in the plasma membrane. Since tyrosine kinase mediate receptor-induced changes in intracellular free calcium, the effect of LMP2A on receptor-mediated intracellular calcium mobilization was evaluated by stably expressing LMP2A in an EBV-negative Burkitt tumor cell line (BJAB) or in LMP1-converted BJAB cells. LMP2A significantly blocked calcium mobilization following class II, CD19, or immunoglobulin M cross-linking. LMP2A effects were partially reversed in LMP1-…

Calcium-dependent regulation of cation transport in cultured human nonpigmented ciliary epithelial cells.

We performed 86Rb flux studies to examine Na-K-adenosinetriphosphatase (ATPase), Na-K-2Cl cotransporter, and potassium channel activity in an established cell line derived from human nonpigmented ciliary epithelium (ODM2). The elevation of intracellular calcium by A23187 (3 microM) or thapsigargin (200 nM) increased both ouabain-sensitive potassium (86Rb) uptake (Na-K-ATPase mediated) and ouabain-insensitive potassium (86Rb) uptake. The ouabain-insensitive component could be inhibited substantially by bumetanide (0.1 mM), suggesting the involvement of a Na-K-2Cl cotransporter. The increase of …

Calcium gating of H+ fluxes in chloroplasts affects acid-base-driven ATP formation.

In previous work, calcium ions, bound at the lumenal side of the CF0H+ channel, were suggested to keep a H+ flux gating site closed, favoring sequestered domain H+ ions flowing directly into the CF0-CF1 and driving ATP formation by a localized delta approximately mu H+ gradient. Treatments expected to displace Ca++ from binding sites had the effect of allowing H+ ions in the sequestered domains to equilibrate with the lumen, and energy coupling showed delocalized characteristics. The existence of such a gating function implies that a closed-gate configuration would block lumenal H+ ions from e…

[Growth and serial changes of bone calcium content after renal transplantation in childhood].

We evaluated metacarpal index (MCI) and bone mineral content (BMC) of right 2nd metacarpal bone X-ray films using the microdensitometer technique in 12 pediatric and 32 adult renal transplant (Tx) recipients. Grafts were well functioning for more than 1 year in all adults (serum creatinine < or = 2.0 mg/dl) and in 9 of the children (serum creatinine < or = 1.2 mg/dl). Immunosuppression consisted of cyclosporin (CyA), methylprednisolone (MPL), mizoribine and anti-lymphocyte globulin for all children. 18 of the adults were given CyA and 14 were given conventional immunosuppression. BMC was found

The calcium second messenger system in bipolar disorders: data supporting new research directions.

Studies of aberrations in second messenger function may help to elucidate some of the multiple complex neurobiological alterations in bipolar affective disorders. The phosphatidylinositol and calcium ion (Ca2+) second messengers are of particular interest because of evidence of hyperactivity of these signaling mechanisms in both mania and bipolar depression and of their normalization by lithium and other mood-stabilizing treatments. Because the intracellular Ca2+ signal has a biphasic action, a single aberration could explain diverse clinical manifestations of the same illness, and a single ac…

Molecular diversity of neuronal-type calcium channels identified in small cell lung carcinoma.

Using the polymerase chain reaction (PCR), we identified RNA transcripts for two distinct classes of neuronal-type voltage-gated Ca2+ channels (VGCC) in a prototypic small cell lung carcinoma (SCLC) cell line, SCC-9. Oligonucleotide primers were designed to encode amino acid sequences common to alpha 1-subunits of known neuronal VGCC classes. Sequencing of complementary DNA (cDNA) clones derived from two independent PCR products revealed that one corresponded to a brain class A VGCC fragment predicted to encode a P-type VGCC (insensitive to dihydropyridines and omega-conotoxin) characteristic

Root canal sealer cytotoxicity with human gingival Fibroblasts. III. Calcium hydroxide-based sealers.

The cytotoxicity of four different calcium hydroxide-based root canal sealers (Sealapex, Apexit, CRCS, and Endoflas FS) was tested using human gingiva fibroblasts. The fibroblasts were acquired from the attached gingiva of apparently healthy patients. The sealers were allowed to set for 24 and 48 h before being covered with a fibroblastic suspension. The estimation of the incorporation rate of L-[14C]leucine in the fibroblasts after being in contact with the sealers was utilized to designate the fibroblasts' protein synthesis potential. The research was conducted over a period of 21 days. Endo…

Moderation of myocardial ischemia reperfusion injury by calcium channel and calmodulin receptor inhibition.

Intracellular Ca2+ accumulation is implicated in the pathogenesis of myocardial reperfusion injury. To study approaches designed to modify Ca2+ uptake during coronary revascularization after acute infarction, a pig heart surgical infarct model (left anterior descending artery occlusion for 60 min) was subjected to 60 min hypothermic potassium cardioplegic arrest, followed by 60 min of global reperfusion. Four groups of six hearts each were studied in a randomized manner, i.e., cardioplegia alone (control), cardioplegia + 10 microM diltiazem (Ca2+ slow channel blocker), cardioplegia + 10 microM

Effect of delayed calcium hydroxide treatment on periodontal healing in contaminated replanted teeth.

The effect of delayed intracanal calcium hydroxide treatment on experimentally induced extensive inflammatory root resorption in monkeys was studied. A significant shift from inflammatory resorption to ankylosis was noted following calcium hydroxide treatment. Furthermore, calcium hydroxide treatment appeared to change the pattern of ankylosis over time, although the total ankylotic area remained the same. Ankylosis preceded by root resorption (replacement resorption) increased, while ankylosis not associated with root resorption decreased. It was concluded that intracanal calcium hydroxide tr…

In vitro magnesium- and calcium-induced sialidase activity in caprine intestinal mucosa cells.

Sialidase activity identical to 0.0196 mg per ml of liberated sialic acids (2.11 Units) was detected in small intestinal contents of goats. Mg++ and Ca++ induced in vitro detection in mucosa cells of both small and large intestines. Magnesium alone induced sialidase activities identical to 0.0128 mg per ml of liberated sialic acids (1.38 Units) and 0.0166 mg per ml (1.79 Units) in small and large intestinal mucosal cells, respectively. Mg++ and Ca++ together induced higher sialidase activities identical to 0.0191 mg per ml (2.06 Units) and 0.0194 mg per ml (2.09 Units) in small and large intes

Potentiation by aldosterone of vasopressin-induced calcium kinetics in vascular smooth muscle cells.

1. The present study was undertaken to examine the effect of aldosterone on arginine vasopressin (AVP)-induced Ca2+ kinetics in cultured rat vascular smooth muscle cells (VSMC). The pre-incubation of cells with 1 x 10(-6) mol/L aldosterone for 24 h did not affect the basal cytosolic free Ca2+ [( Ca2+]i) but enhanced the AVP-induced mobilization of [Ca2+]i (1 x 10(-8) mol/L AVP; 287 vs 401, 1 x 10(-6) mol/L AVP; 430 vs 714 nmol/L). 2. The pre-incubation of cells with 1 x 10(-7) mol/L aldosterone for 24 h did not show this enhancing effect on the AVP-induced mobilization of [Ca2+]i. Without the

Diproteverine (BRL 40015): a new type of calcium antagonist with potential antianginal properties.

The calcium channel-blocking activity and associated cardiovascular effects of diproteverine, a novel compound derived from papaverine, were investigated. Electrophysiological measurements in sheep Purkinje fibres showed diproteverine to reduce the amplitude of the slow action potential (IC30 = 2 microM) and to shorten the duration of the fast action potential at 50% repolarisation (IC30 = 2.5 microM). Higher concentrations (4-5 times) were required to block block the sodium channel, as assessed by a reduction in Vmax of the fast action potential. Papaverine was found to possess marginal membr…

Fluorescence digital image analysis of serotonin-induced calcium oscillations in single blood platelets.

The intracellular concentration of Ca2+ [( Ca2+]i) was monitored continuously in single rabbit blood platelets by digital imaging microscopy in conjunction with Fura-2, a specific Ca(2+)-indicator dye. Ionomycin as well as aluminium fluoride caused sustained increase in [Ca2+]i in the platelet, but oscillations of [Ca2+]i were not observed. Serotonin (5-HT) induced oscillatory increases in [Ca2+]i in the presence of 1 mM CaCl2; these had not been detectable in cell populations because the oscillations were not in synchrony. This effect of 5-HT was diminished when CaCl2 was omitted from the med
