Calmodulin inhibitors chlorpromazine and calmidazolium cause calcium overload and oxidative stress despite reducing peak density of CaV1.2 current in ventricular myocytes.

Calmodulin (CaM) plays crucial roles in the regulation of the voltage-dependent L-type calcium (Ca2+) channel α1C subunit (CaV1.2) channel opening and closing, which is closely associated with the occurrence and progression of cardiac diseases. However, the specific mechanisms by which CaM regulates CaV1.2 channel Ca2+ currents have not been well established experimentally, and whether CaM can be a therapeutic target for cardiovascular diseases deserves further study. In this study, we investigated the effects of CaM inhibitors chlorpromazine (CPZ) and calmidazolium (CMZ) on CaV1.2 channel fun

Short QT Syndromes.

The short QT syndrome is a recently described genetic arrhythmogenic disorder, characterized by abnormally short QT intervals and a high incidence of sudden death and atrial fibrillation. Clinical manifestations may also be present in infants; a family history of cardiac sudden death is often present. Gain-of-function mutations in 3 genes encoding potassium channels and loss-of-function mutations in 2 genes encoding the cardiac L-type calcium channel have been identified. Today, the first choice therapy is implantable cardioverter-defibrillator implantation; however, pharmacologic treatment wi

Myasthenia gravis and myasthenic syndrome.

Myasthenia gravis (MG) and Lambert-Eaton myasthenic syndrome (LEMS) are autoimmune ion channel disorders mediated by antibodies to postsynaptic acetylcholine receptors and presynaptic voltage-operated calcium channels, respectively. MG and LEMS are currently treated with corticosteroids and immunosuppressive drugs with a positive clinical outcome. Since both diseases are mediated by circulating autoantibodies, they are good candidates for immunomodulation by intravenous immunoglobulins (IVIG). Standardized controlled studies are lacking in this field, but there is general agreement that IVIG a…

Synthesis and biological evaluation of an iodinated iberiotoxin analogue, [mono-iodo-Tyr5, Phe36]-iberiotoxin.

The synthesis and iodination of a structural analogue of the specific large-conductance calcium-activated potassium (BK) channel blocker, iberiotoxin (IbTX), a 37-amino acid scorpion neurotoxin, is reported. The synthesis of this analogue, [Tyr5, Phe36]-IbTX, was accomplished using standard solid-phase Fmoc (9-fluorenylmethoxycarbonyl) chemistry protocols. The linear peptide was cyclized via the formation of three intramolecular disulfide bridges and subsequently iodinated at the Tyr5 position. Upon purification, the iodinated analogue, [mono-iodo-Tyr5, Phe36]-IbTX, exhibited comparable biolog

[Discovery of heat-induced calcium release via ryanodine receptors and a novel mechanism of thermal runaway].

Regulation of thermogenesis in mammals is essential for maintaining body temperature homeostasis under fluctuating environmental temperature. Impairments in this regulation can lead to severe conditions, including fever or heatstroke. This review focuses on malignant hyperthermia (MH), a pathological escalation of thermogenesis in skeletal muscle. It highlights the role of type 1 ryanodine receptor (RYR1), a Ca2+ release channel, based on our recent studies. Previous studies have revealed that genetic mutations in RYR1 are associated with muscle disorders including MH, which are characterized

The mechanism of LQTS related CaM mutation E141G interfering with CaV1.2 channels function through its C-lobe.

Mutations in the CALM1-3 genes, which encode calmodulin (CaM), have been reported in clinical cases of long QT syndrome (LQTS). Specifically, the CaM mutant E141G (CaME141G) in the variant CALM1 gene has been identified as a causative factor in LQTS. This mutation disrupts the normal Ca2+-dependent inactivation (CDI) function of CaV1.2 channels. However, it is still unclear how CaME141G interferes with the regulatory role of wild-type (WT) CaM on CaV1.2 channels and leads to abnormal CDI. A CaM molecule contains two lobes with similar structure, the N-lobe and the C-lobe. In this study, a CaM-

Feedback contributions to excitation-contraction coupling in native functioning striated muscle.

Skeletal and cardiac muscle excitation-contraction coupling commences with Nav1.4/Nav1.5-mediated, surface and transverse (T-) tubular, action potential generation. This initiates feedforward, allosteric or Ca2+-mediated, T-sarcoplasmic reticular (SR) junctional, voltage sensor-Cav1.1/Cav1.2 and ryanodine receptor-RyR1/RyR2 interaction. We review recent structural, physiological and translational studies on possible feedback actions of the resulting SR Ca2+ release on Nav1.4/Nav1.5 function in native muscle. Finite-element modelling predicted potentially regulatory T-SR junctional [Ca2+]TSR do

Sex-Specific Differences in Endothelial Function Are Driven by Divergent Mitochondrial Ca2+ Handling.

Background Sex-specific differences in vasodilation are mediated in part by differences in cytosolic Ca2+ handling, but how variations in mitochondrial Ca2+ contributes to this effect remains unknown. Here, we investigated the extent to which mitochondrial Ca2+ entry via the MCU (mitochondrial Ca2+ uniporter) drives sex differences in vasoreactivity in resistance arteries. Methods and Results Enhanced vasodilation of mesenteric resistance arteries to acetylcholine (ACh) was reduced to larger extent in female compared with male mice in 2 genetic models of endothelial MCU ablation. Ex vivo Ca2+

Caffeine as a tool to investigate sarcoplasmic reticulum and intracellular calcium dynamics in human skeletal muscles.

Caffeine is worldwide used for its power to increase cognitive and physical performance. The ergogenic effects of caffeine, however, do not depend on a direct action on muscles. Actually, the actions of caffeine on skeletal muscles, take place at millimolar concentrations which are far above the micromolar level reached after a regular consumption of coffee or similar drinks, and close to a lethal concentration. At millimolar concentrations caffeine exerts a powerful effect on sarcoplasmic reticulum (SR) activating the release of calcium via ryanodine receptors and, possibly, inhibiting calciu

Diabetes and cancer: Debating the link through Ca2+/cAMP signalling.

The incidence of both cancer and diabetes is dramatically increasing in worldwide population, costing many millions from governments into expenditures related to medical health systems. Diabetes has been clinically linked to an increased risk for developing several types of cancer. The cellular mechanisms involved in this link are still under intensive debate in literature. In addition, a Ca2+ homeostasis dysregulation has been intensively debated as an issue involved in both cancer and diabetes. Calcium (Ca2+) channel blockers (CCBs), prescribed for treating hypertension, have also been showi

Intracellular emetic signaling evoked by the L-type Ca2+ channel agonist FPL64176 in the least shrew (Cryptotis parva).

Ca2+ plays a major role in maintaining cellular homeostasis and regulates processes including apoptotic cell death and side-effects of cancer chemotherapy including vomiting. Currently we explored the emetic mechanisms of FPL64176, an L-type Ca2+ channel (LTCC) agonist with maximal emetogenic effect at its 10 mg/kg dose. FPL64176 evoked c-Fos immunoreactivity in shrew brainstem sections containing the vomit-associated nuclei, nucleus tractus solitarius (NTS) and dorsal motor nucleus of the vagus. FPL64176 also increased phosphorylation of proteins ERK1/2, PKCα/βII and Akt in the brainstem. Mor

Nephrocalcinosis in siblings--familial hypomagnesemia, hypercalciuria with nephrocalcinosis (FHHNC syndrome).

Familial Hypomagnesemia, Hypercalciuria with Nephrocalcinosis is a rare autosomal recessive inherited disease associated with renal failure. Two girls born of consanguineous parentage aged 16 and 17 presented to us with renal failure, nephrocalcinosis and bone deformities. On evaluation they were found to have hypomagnesemia, hypercalciuria, increased fractional excretion of magnesium, hypocitraturia, renal failure and elevated PTH. Their parental screening was normal. There were no extra-renal features in them. One sibling had nephrolithiasis and the stone analysis revealed calcium phosphate

Lymphocytic intracellular calcium in a patient with complicated verapamil overdose.

Overdose with calcium channel blockers (CCBs) may lead to serious complications. CCBs act by blocking calcium entry into the cell, thus lowering intracellular calcium ([Ca2+]i). [Ca2+]i during CCB overdose has not yet been reported. We measured [Ca2+]i in lymphocytes of a patient with acute verapamil overdose with a complex clinical picture. A 59-year-old woman was admitted after a suicidal ingestion of 7200 mg of a sustained-release verapamil preparation. She presented with hypotension, complete atrioventricular block, stupor, hypokalemia, and hyperglycemia. Acute oliguric renal failure, acut

Rapid reversal of life-threatening diltiazem-induced tetany with calcium chloride.

We describe a patient who developed tetany with sudden respiratory arrest after the infusion of intravenous diltiazem. The administration of calcium chloride rapidly resolved the patient's tetany with prompt recovery of respiratory function, averting the need for more aggressive airway management and ventilatory support. The emergency physician should be aware that life-threatening tetany may accompany the administration of intravenous diltiazem and that calcium chloride may be a rapid and effective remedy.

Trifluoperazine and calcium antagonists accelerate cholinergic desensitization in Aplysia neurons.

The onset of desensitization of excitatory acetylcholine responses in isolated, voltage clamped Aplysia RB neurons to microperfused agonist was accelerated by trifluoperazine and the calcium antagonists SKF-525A and D-600. In the absence of these drugs the time course of desensitization is described as the sum of two exponentials plus a constant. All 3 compounds accelerated the fast exponential component of desensitization, but had opposite effects on the slow component: trifluoperazine and SKF-525A accelerated the slow component, whereas this phase was greatly slowed by D-600. These results p
