The Role of Elements in Anxiety.

Elements (bioelements) are necessary factors required for the physiological function of organisms. They are critically involved in fundamental processes of life. Extra- and intracellular message and metabolic pathway factors as well as structural components include one or many elements in their functional structure. Recent years have seen an intensification in terms of knowledge gained about the roles of elements in anxiety disorders. In this chapter we present a review of the most important current data concerning the involvement of zinc, magnesium, copper, lithium, iron, and manganese, and t

Multi-omics analysis reveals the critical role of gut microbiota related tryptophan and glutathione metabolism in sepsis-associated encephalopathy.

BACKGROUND: Recent studies highlight the critical role of gut microbiota in sepsis pathogenesis and its potential link to neurological disorders, particularly sepsis-associated encephalopathy (SAE). However, the exact relationship between gut microbiota, their metabolites, and SAE’s etiology and progression remains enigmatic. We aimed to elucidate how gut bacteria, fungi, and their metabolites contribute to the development and progression of SAE. METHODS: This study was a prospective cohort study. Patients who met the criteria for sepsis 3.0 were included and were divided into SAE and non-SAE

Within-host evolution drives the emergence of ceftazidime-avibactam resistance mediated by IncN plasmid-encoded blaNDM-1 and blaKPC-33 in ST11-KL64 hypervirulent Klebsiella pneumoniae.

UNLABELLED: Ceftazidime-avibactam (CZA) is considered one of the most effective antimicrobial agents for treating infections caused by Klebsiella pneumoniae carbapenemase-producing Klebsiella pneumoniae. However, rising CZA resistance poses a critical clinical challenge, primarily driven by blaKPC mutations. From a single CZA-treated patient, we isolated two CZA-susceptible and three CZA-resistant K. pneumoniae strains. Whole-genome sequencing of CZA-resistant strains CYH4028 and CYH1535 revealed two key resistance pathways: (i) acquisition of a conjugative IncN plasmid harboring blaNDM-1 and

Impacts of climate change on mental health and its underlying mechanisms: an umberella review.

BACKGROUND: Climate change represents a major global health challenge with potential implications for mental health. Exposure to climate-related stressors is associated with an elevated risk of psychiatric disorders, including trauma- and stressor-related disorders (e.g., PTSD), depressive disorders, and anxiety disorders. Vulnerable populations-including children, women, older adults, individuals with pre-existing mental health conditions, and communities in low-income or disaster-prone regions-may be disproportionately affected. This umbrella review synthesizes current evidence on the mental

Decoding epithelial regeneration in the cornea: multi-omic analysis reveals cellular plasticity as central mechanism.

BACKGROUND: Rapid and efficient epithelial regeneration is fundamental for tissue homeostasis and proper function. As the outermost ocular structure, the cornea is transparent, multilayered, and vital for clear vision. Due to its exposed position, the cornea frequently undergoes various forms of injury affecting either the epithelium itself or its surrounding microenvironment, including corneal innervation and the tear film. Corneal abrasion, occurring commonly through trauma or as part of refractive surgical procedures, is typically viewed as a minor event since it usually resolves rapidly. C

Transcriptome profiling of human dermal MDPL fibroblasts reveals a characteristic molecular signature providing insights into pathogenic mechanisms.

The emerging perception that the mammalian dermis encloses fibroblasts with differing functional identities has profound implications for understanding a wide range of genetic pathological states, including aging. MDPL syndrome (mandibular hypoplasia, deafness, progeroid characteristics, and lipodystrophy; MIM #615381) is an extremely rare, genetic progeroid disorder. Patients reported variants in the POLD1 gene (NM_002691.3), encoding for the evolutionarily conserved catalytic subunit of DNA polymerase delta (Polδ). The protein is a critical enzyme reliable for synthesizing nascent DNA strand

Flavonoids from Rhododendron nivale Hook. f ameliorate alcohol-associated liver disease via activating the PPARα signaling pathway.

BACKGROUND: Flavonoids are increasingly recognized for their potent antioxidant properties and potential therapeutic roles in the management of alcohol-associated liver disease (ALD). Extracts derived from Rhododendron nivale Hook. f. (FRN) have been shown to influence glutathione metabolism in aging animal models, exhibiting notable antioxidant effects. However, the specific impact of FRN on ALD remains insufficiently explored. HYPOTHESIS/PURPOSE: This study seeks to elucidate the efficacy of FRN in alleviating the pathology associated with ALD, delving into the underlying molecular mechanis

Lentinula edodes Cultured Extract and Rouxiella badensis subsp. acadiensis (Canan SV-53) Intake Alleviates Immune Deregulation and Inflammation by Modulating Signaling Pathways and Epigenetic Mechanisms.

Puberty is a critical developmental period of life characterized by marked physiological changes, including changes in the immune system and gut microbiota development. Exposure to inflammation induced by immune stressors during puberty has been found to stimulate central inflammation and lead to immune disturbance at distant sites from the gut; however, its enduring effects on gut immunity are not well explored. Therefore, in this study, we used a pubertal lipopolysaccharides (LPS)-induced inflammation mouse model to mimic pubertal exposure to inflammation and dysbiosis. We hypothesized that

Type I Interferon Signaling Is a Common Factor Driving Streptococcus pneumoniae and Influenza A Virus Shedding and Transmission.

The dynamics underlying respiratory contagion (the transmission of infectious agents from the airways) are poorly understood. We investigated host factors involved in the transmission of the leading respiratory pathogen Streptococcus pneumoniae Using an infant mouse model, we examined whether S. pneumoniae triggers inflammatory pathways shared by influenza A virus (IAV) to promote nasal secretions and shedding from the upper respiratory tract to facilitate transit to new hosts. Here, we show that amplification of the type I interferon (IFN-I) response is a critical host factor in this process,

Dramatic neurological and biological effects by botulinum neurotoxin type A on SH-SY5Y neuroblastoma cells, beyond the blockade of neurotransmitter release.

BACKGROUND: Gene expression profile analysis on mammalian cell lines and animal models after exposure to botulinum neurotoxin (BoNT) has been investigated in several studies in recent years. Microarray analysis provides a powerful tool for identifying critical signaling pathways involved in the biological and inflammatory responses to BoNT and helps determine the mechanism of the function of botulinum toxins. One of the pivotal clinical characteristics of BoNT is its prolonged on-site effects. The role of BoNT on the blockage of neurotransmitter acetylcholine release in the neuromuscular junct

Global Network Analysis of Neisseria gonorrhoeae Identifies Coordination between Pathways, Processes, and Regulators Expressed during Human Infection.

Neisseria gonorrhoeae is a Gram-negative diplococcus that is responsible for the sexually transmitted infection gonorrhea, a high-morbidity disease in the United States and worldwide. Over the past several years, N. gonorrhoeae strains resistant to antibiotics used to treat this infection have begun to emerge across the globe. Thus, new treatment strategies are needed to combat this organism. Here, we utilized N. gonorrhoeae transcriptomic data sets, including those obtained from natural infection of the human genital tract, to infer the first global gene coexpression network of this pathogen.

Varicella-Zoster Virus ORF63 Protects Human Neuronal and Keratinocyte Cell Lines from Apoptosis and Changes Its Localization upon Apoptosis Induction.

There are many facets of varicella-zoster virus (VZV) pathogenesis that are not fully understood, such as the mechanisms involved in the establishment of lifelong latency, reactivation, and development of serious conditions like postherpetic neuralgia (PHN). Virus-encoded modulation of apoptosis has been suggested to play an important role in these processes. VZV open reading frame 63 (ORF63) has been shown to modulate apoptosis in a cell-type-specific manner, but the impact of ORF63 on cell death pathways has not been examined in isolation in the context of human cells. We sought to elucidate

Fractionated radiosurgery for 9L gliosarcoma in the rat brain.

PURPOSE: Fractionated radiosurgery is being carried out in the clinic to improve the therapeutic ratio of single-dose radiosurgery using various fractionation schemes. Because there is a paucity of experimental radiobiological data in the literature on the tumor response and late-responding normal tissue of critical intracranial structures to radiosurgery, the present animal study was designed to compare the response following a single high dose of radiation with that obtained from calculated fractionated doses of radiosurgery. METHODS AND MATERIALS: Male Fischer rats with 9L gliosarcoma grow

Transcription factor Fra-1 reprograms androgen receptor chromatin binding and super-enhancers in osteoblast differentiation.

Transcriptional regulation by coregulators and other proteins is essential for osteoblast differentiation. Bone density maintenance requires tissue-specific androgen and androgen receptor (AR)-regulated signaling in osteoblasts. However, the transcriptional mechanisms through which androgens exert specific effects on osteoblasts remain unclear. Here, to explore androgen signaling in osteoblastic cells, we investigated comprehensive AR-binding sites by using chromatin immunoprecipitation and sequencing (ChIP-seq) analysis. We observed that the transcription factor (TF) Fra-1, an important playe

Long-term spatiotemporal dynamics of β-lactamase resistome in the Han River, South Korea.

The global dissemination of antimicrobial resistance, particularly resistance to widely prescribed β-lactam antibiotics, is a critical public health threat worldwide. Aquatic ecosystems act as key reservoirs where antibiotic resistance genes accumulate and are transmitted between environmental bacteria and human pathogens within a One Health framework. To move beyond short-term observations and better understand these cross-sectoral dynamics, we conducted 5-year longitudinal surveillance using shotgun metagenomics across different seasons along the Han River, South Korea. Our analysis revealed

The interplay between mental health, ecological factors, and problematic smartphone use: a network analysis in adolescents with major depressive disorder.

Adolescence is a critical period for the onset of major depressive disorder (MDD), during which emotional symptoms frequently co-occur with environmental adversity and maladaptive behavioral coping. In China, these processes may be further shaped by left-behind experience (LBE) - a prevalent structural vulnerability in which children grow up separated from migrating parents. To clarify these relationships, we applied network analysis in a clinical sample of 2341 adolescents (Mage = 14.99 years,77.92% female) with MDD. Participants were recruited from 14 psychiatric outpatient clinics across Ch
