Ecological Niche Modeling for the Prediction of the Geographic Distribution of Cutaneous Leishmaniasis in Tunisia.

Cutaneous leishmaniasis is a very complex disease involving multiple factors that limit its emergence and spatial distribution. Prediction of cutaneous leishmaniasis epidemics in Tunisia remains difficult because most of the epidemiological tools used so far are descriptive in nature and mainly focus on a time dimension. The purpose of this work is to predict the potential geographic distribution of Phlebotomus papatasi and zoonotic cutaneous leishmaniasis caused by Leishmania major in Tunisia using Grinnellian ecological niche modeling. We attempted to assess the importance of environmental f

Genotypic and phenotypic predictors of inflammation in patients with chronic kidney disease.

BACKGROUND: In complex diseases such as chronic kidney disease (CKD), the risk of clinical complications is determined by interactions between phenotypic and genotypic factors. However, clinical epidemiological studies rarely attempt to analyse the combined effect of large numbers of phenotype and genotype features. We have recently shown that the relaxed linear separability (RLS) model of feature selection can address such complex issues. Here, it is applied to identify risk factors for inflammation in CKD. METHODS: The RLS model was applied in 225 CKD stage 5 patients sampled in conjunction

Review and evaluation of penalised regression methods for risk prediction in low-dimensional data with few events.

Risk prediction models are used to predict a clinical outcome for patients using a set of predictors. We focus on predicting low-dimensional binary outcomes typically arising in epidemiology, health services and public health research where logistic regression is commonly used. When the number of events is small compared with the number of regression coefficients, model overfitting can be a serious problem. An overfitted model tends to demonstrate poor predictive accuracy when applied to new data. We review frequentist and Bayesian shrinkage methods that may alleviate overfitting by shrinking

Synopsis and meta-analysis of genetic association studies in osteoporosis for the focal adhesion family genes: the CUMAGAS-OSTEOporosis information system.

BACKGROUND: Focal adhesion (FA) family genes have been studied as candidate genes for osteoporosis, but the results of genetic association studies (GASs) are controversial. To clarify these data, a systematic assessment of GASs for FA genes in osteoporosis was conducted. METHODS: We developed Cumulative Meta-Analysis of GAS-OSTEOporosis (CUMAGAS-OSTEOporosis), a web-based information system that allows the retrieval, analysis and meta-analysis (for allele contrast, recessive, dominant, additive and codominant models) of data from GASs on osteoporosis with the capability of update. GASs were i

Cost-effectiveness analysis of pneumococcal conjugate vaccines, PCV21 vs PCV20, in adults aged  ≥60 years in Germany.

BACKGROUND: Higher-valency pneumococcal conjugate vaccines (PCVs) may reduce the burden of adult pneumococcal disease (PD), yet it is unclear whether higher acquisition costs are offset by improved health outcomes and cost savings. RESEARCH DESIGN AND METHODS: A published state-transition Markov model simulated vaccine-naïve adults aged ≥60 years stratified by risk group and followed until death or a maximum age of 100 years was adapted to compare PCV21 versus PCV20. Published epidemiological and cost data were used to populate the model. Costs and quality-adjusted life years were discounted

A selective machine learning algorithm for severe periodontitis labeling from questionnaire data.

Epidemiological cohorts often collect self-reported oral health (SROH) questionnaires but lack clinical periodontal measurements. We developed a selective, explainable machine learning (ML) pipeline that can assign labels for severe periodontitis (SP) or no periodontitis (NP). Three datasets (n = 498) with SROH questionnaires, demographics, and Community Periodontal Index of Treatment Needs (CPITN) scores were used to derive NP, moderate periodontitis (MP), and SP categories. MP cases were excluded from model development. After cleaning and feature engineering, non-similar label duplicates wer

Community-level wastewater influenza concentrations and school attendance in Jefferson County, Kentucky (2022-2025).

Community-transmitted seasonal infectious diseases pose a significant challenge to school systems across the nation. School attendance is central to student learning progress and is a key public health indicator. We aimed to evaluate the incremental explanatory value of community-level seasonal influenza wastewater concentrations for estimating school absenteeism within the corresponding geographic sewershed in Louisville, Kentucky (USA). We used community wastewater seasonal influenza concentrations from a catchment area that geographically encompassed 51 public elementary, middle, and high s

Associated-Factor Studies in Biomedical Research: Foundations, Limitations, and Criteria for Appropriate Use.

Associated-factor studies constitute a substantial portion of the observational epidemiology literature. However, their proliferation has raised important methodological concerns. Many are published without a clear research question, repeat well-established associations, or use causal language to interpret coefficients from exploratory models. This narrative methodological review and critical reflection provides guidance for determining when associated-factor studies add genuine scientific value, how to plan sample size using events per variable, and how to interpret and report results appropr

Beyond time-series counts: reframing the short-term association between exposure to air pollution and mortality using the novel repeated acute exposure (RAE) design.

Assessing the robustness of epidemiological associations requires diverse methodological approaches. This study introduces the Repeated Acute Exposure (RAE) design, a survival-based framework utilising an extended Cox model with time-varying coefficients to evaluate the short-term association between PM10 and daily mortality in Valencia, Spain. By incorporating a time-varying coefficient to capture the dynamic intensity of the risk across a 7-day window, this approach conceptualises every exposure increment as contributory to the mortality risk. We adjusted for temperature, relative humidity,

Rethinking social support in depressive symptoms: A comprehensive analysis of support source, gender, and developmental stage.

The critical importance of social support in alleviating depressive symptoms is well-established across diverse theoretical and intervention frameworks. This study extends current understanding by examining how different sources of social support, gender, and developmental stage interact, moving beyond traditional models that often emphasize single moderating factors. Using a stratified sampling approach, data were collected from 4395 university students (mean age = 20 years), who completed the Center for Epidemiologic Studies Depression Scale, the Multidimensional Perceived Social Support Sca

Chronic traumatic encephalopathy: A devastating legacy of repetitive concussion.

Repetitive concussive and subconcussive traumatic brain injury (TBI) is increasingly linked to chronic traumatic encephalopathy (CTE), yet a central challenge remains in connecting exposure to long-term neurodegeneration through a coherent mechanistic framework. Here, we synthesize evidence across epidemiology, neuropathology, and clinical studies to define the continuum from repetitive injury to disease. Primary injury initiates secondary cascades, including mitochondrial dysfunction, metabolic stress, neuroinflammation, and axonal injury across neuronal, glial, and vascular compartments, whi

Determination of a cytotoxicity threshold for α-Fe2O3 nanoparticles based on an in vitro-in vivo dosimetry model in alveolar epithelial cells.

This study aimed to extrapolate a cytotoxicity threshold in air using a combined in vitro-in vivo dosimetry model to assess the toxicity of α-Fe2O3 nanoparticles (NPs), innovatively applying this approach to NPs risk assessment. Cultured A549 cells treated with α-Fe2O3 NPs in various concentrations over 24-h and 72-h. Cell survival and mitochondrial membrane permeability were assessed using the MTT and MMP assay, respectively. The dose-response results served for estimating the Benchmark Dose (BMD) and BMDL using the EPA BMD software. BMDLs were extrapolated into equivalent air concentrations

Nicotine Attenuates Pathogenesis of Parkinson's Disease via α7-nAChR-Mediated Lipid Metabolic Reprogramming and Anti-inflammatory Signaling.

Nicotine-an abundant tobacco alkaloid-has been epidemiologically linked to a lower incidence of Parkinson's disease (PD). In a 6‑hydroxydopamine (6‑OHDA) rat model, we found that chronic nicotine (1.5 mg kg-1 day-1) was associated with better motor performance and a smaller drop in striatal brain‑derived neurotrophic factor (BDNF) (p < 0.05). Untargeted lipidomics indicated partial normalization of 6‑OHDA‑induced changes in sphingolipids, phospholipids, and neutral lipids, alongside reduced lipid‑peroxidation markers. In SH‑SY5Y cells, nicotine attenuated 6‑OHDA‑related cytotoxicity, mitochond

Machine Learning-Assisted 3D SERS Chip with Acoustic Enrichment for High-Accuracy Diagnosis of Respiratory Viruses and Emerging Pathogens.

Outbreaks of SARS-CoV-2, first investigated as an unknown pathogen, have reflected the severe threat that pathogen X poses to public health and social security. Early and precise diagnosis and classification of infectious respiratory diseases with similar symptoms are essential for the risk assessment of public health or epidemiological investigations. Current technologies are limited to detect known viruses, leading to false negatives for novel or mutated pathogens. Here, we propose an ML-assisted SERS strategy for screening various types of respiratory viruses and potential pathogen X in cas

A nonhuman primate model mirrors human congenital cytomegalovirus infection and reveals a spectrum of vertical transmission outcomes.

Congenital cytomegalovirus (cCMV) is the leading infectious cause of birth defects worldwide, yet immune determinants of protection to inform design of a maternal vaccine remain elusive. Here, we characterized the outcome of primary rhesus CMV (RhCMV) infection during pregnancy in an immune competent nonhuman primate (NHP) model. RhCMV DNA was detected in amniotic fluid and/or fetal tissues in six of 12 (50% placental transmission) CMV-naive rhesus macaque dams inoculated intravenously with RhCMV in early second trimester gestation. Widespread tissue dissemination dominated by one of two inocu
