Congenital Hyperinsulinism of Infancy (CHI) is a genetically heterogeneous disorder characterized by profound hypoglycemia related to inappropriate insulin secretion. Two histopathologically and genetically distinct groups are recognized among patients with CHI due to ATP-sensitive potassium channel (KATP) defects: a diffuse type (Di-CHI), which involves the whole pancreas, and a focal form (Fo-CHI), which shows adenomatous islet-cell hyperplasia of a particular area within the normal pancreas. The beta-cell KATP channel consists of two essential subunits: Kir6.2 encoded by the KCNJ11 gene whi

INTRODUCTION: Abnormal control of the complement alternative pathway (CAP) (factor H, factor I and membrane cofactor protein (MCP) deficiencies) is a well established risk factor for the occurrence of haemolytic uraemic syndrome (HUS). In some instances, HUS may be associated with an unusual glomerulonephritis with isolated C3 deposits (glomerulonephritis C3). We determined whether HUS and glomerulonephritis C3 share common genetic susceptibility factors.
METHODS: We identified 19 patients with glomerulonephritis C3. We measured levels of circulating complement components, performed assays fo

Polymorphisms in DNA-repair genes could contribute to the interindividual differences in cancer susceptibility in smokers. By reducing DNA-repair capacity, these polymorphisms may influence the net level of smoking-induced genetic damage significantly, a critical step in the cascade of events leading to cancer. In this biomonitoring study, we examined the relationship between polymorphisms in the DNA-repair gene XPD/ERCC2 and genetic damage. We tested the hypothesis that coding polymorphisms in XPD/ERCC2 limit DNA-repair efficiency in humans leading to increased frequencies of chromosome aberr

PURPOSE: X-linked retinitis pigmentosa (XLRP) is a degenerative disease of the retina characterized in the early stages of disease by night blindness as a result of rod photoreceptor loss, progressing to severe disease with loss of central vision by the third decade in affected males. XLRP displays exceptional genetic heterogeneity, with five reported loci on the human X-chromosome. To investigate the level of heterogeneity for XLRP in the patient pool in the current study, extensive haplotype analysis, linkage analysis, and mutation screening were performed.
METHODS: Haplotype analysis of a

Identical anomalies produced by different causes such as aneuploidy, gene mutation, teratogenic chemicals, and certain surgical procedures are a clear indication that embryonic primordia respond as units in the production of developmental anomalies of anatomic structure. Hence, they must also act as units during normal ontogeny. The presence of identical malformations in different mammalian species identifies developmental and anatomic homology by virtue of descent from a common ancestor. These dys- and orthomorphogenetically reactive units are the equivalents of the classic experimental embry

The genetic basis of autoimmune haemolytic anaemia in NZB mice has been investigated using crosses and backcrosses with the NZC strain. These mating combinations reveal two dominant or co-dominant genes governing occurrence of the disorder. One of these, Aia-2, is common to both the NZB and the NZC strains. The other gene, Aia-1, is peculiar to the NZB strain and is linked to the b (black/brown) coat colour locus on chromosome 4. The cross-over value of 37+/-4% places this gene in the neighbourhood of the minor histocompatibility locus H-18, provided it is on the same side of the b locus. Prev

BACKGROUND: Variants in the ion channel gene KCNQ1 are the leading cause of congenital long QT syndrome (LQTS), a disorder characterized by QT prolongation and risk of fatal arrhythmias. The expanding use of genetic testing has led to the identification of numerous variants of uncertain significance, complicating clinical interpretation and risk stratification. Functional characterization of KCNQ1 variants is essential to define their role in long QT syndrome and advance precision medicine in cardiovascular care.
METHODS: We generated a nearly comprehensive variant library of 13 403 protein-c

BACKGROUND: The tight junction family of claudin proteins regulates paracellular ion transport in the nephron and across epithelial and endothelial barriers throughout the body. Rare recessive variants in CLDN16 and CLDN19 cause familial hypomagnesemia with hypercalciuria and nephrocalcinosis (FHHNC), while common variants in CLDN2, CLDN10, and CLDN14 have been associated with kidney stones. Despite evidence that most claudin proteins are expressed within the nephron, their genetic contribution to kidney stone susceptibility has not been systematically explored.
METHODS: We analyzed 415 237 U

Background/Objectives: Inflammatory and oxidative-stress-related processes contribute to post-myocardial infarction (MI) remodeling and may influence long-term cardiovascular outcomes. Recent findings have highlighted the potential role of non-coding RNAs in regulating these processes. LncRNA MALAT1 acts as a ceRNA that "sponges" miR-146a, reducing its ability to repress downstream targets such as COX-2. The aim of this study was to assess MALAT1 and miR-146a expression in PBMCs and plasma COX-2 in controls and patients six months post-MI. In addition, we investigated whether MALAT1 rs3200401

BACKGROUND: Chronic lung allograft dysfunction (CLAD) leads to declining respiratory function and high mortality, representing the main barrier to long-term survival in lung transplantation (LT). We performed the first genome-wide association study (GWAS) investigating donor's and recipient's genetic factors associated with CLAD.
METHOD: We genotyped 392 donor-recipient pairs from the multicentric Cohort in Lung Transplantation. We tested 4.5 million SNPs for association with CLAD using multivariable logistic regression models corrected for age, sex, initial disease and genetic ancestry. Thre

Germline and somatic cancer variants in tumor suppressor genes (TSGs) share loss of function mechanisms with studies of a few genes (DICER1, CEBPA) highlighting differences in variant consequence and location. To systematically assess whether TSGs display distinct mutational patterns we leveraged large public genetic databases and compared high-quality pathogenic/likely pathogenic (P/LP) germline variants in ClinVar, with oncogenic/likely oncogenic (O/LO) somatic tumor variants from cBioPortal across 40 TSGs. We obtained 32,941 P/LP germline and 12,907 O/LO somatic variants. Only 3,863 (9.2%)

CDHR1 is a recently identified cause of autosomal recessive retinal degeneration manifesting as three distinct clinical phenotypes: macular dystrophy, cone-rod dystrophy or retinitis pigmentosa. In this review, we summarise the discovery and characterisation of CDHR1, clinical phenotypes, natural history and therapeutic approaches including gene supplementation and CRISPR gene editing. CDHR1 is a non-classical cadherin that is highly expressed in cone and rod photoreceptors and is essential for the higher-order organisation of the functionally critical outer segments. Promising pre-clinical da

Disease outbreaks caused by Mycobacterium marinum pose a major challenge to marine aquaculture and threaten the sustainable production of Cynoglossus semilaevis. Increasing evidence suggests that host genetic variation and gut microbial communities may jointly influence disease-related phenotypes, yet their interaction under mycobacterial challenge remains poorly understood. In this study, we combined host whole-genome resequencing and gut 16S rRNA sequencing to explore host-microbiome associations in C. semilaevis following M. marinum challenge. Gut microbiota analysis revealed significant di

Sleepy cod (Oxyeleotris lineolata) is native to Australia and is now an economically valuable fish cultured in China and Southern Asian countries. Its growth rate exhibits as sexually dimorphic, with males generally growing more rapidly and attaining a larger body size compared to females. Thus, the effective development of sex control breeding can significantly contribute to increased yields and output value. Nevertheless, due to the lack of genomic and transcriptomic data, the molecular mechanisms underlying sex determination and gonadal differentiation in sleepy cod remain poorly understood

INTRODUCTION: The diagnosis of infertility generates a significant psychological impact on the couples who experience it. The loss of fertility triggers a psychological mourning process that affects self-esteem and identity. The aim of this study is the follow-up of couples 25 years after conceiving children with artificial insemination of donor semen, to explore from a mixed methods approach the relationship between the mode of elaboration of infertility grief, the communication strategies of the genetic origins of babies and the filiation experience.
METHOD: The macrostages of the process a

Depending on zygosity and the specific change, different variants in the GBA1 gene can cause Parkinson's disease (PD, PARK-GBA1) with reduced penetrance, act as genetic risk factors for PD or parkinsonism, and/or lead to Gaucher's disease (GD). This MDSGene systematic literature review covers 27,963 patients carrying GBA1 variants from 1082 publications with 794 variants, including 13,342 patients with PD or other forms of parkinsonism. It provides a comprehensive overview of demographic, clinical, and genetic findings from an ethnically diverse sample originating from 82 countries across five

The aim of this study was to investigate three unrelated Simmental calves with atypical white coat color, identify potential genetic causes using a trio-based whole-genome sequencing approach, and assess the prevalence of the identified variants in the breed. Several inherited alleles affecting coat color, ranging from fawn to red spotted and white-headed, have been described in Simmental cattle originating from Switzerland. However, no genetic variant has yet been associated with an almost completely white coat in this breed. Clinical examination revealed different syndromic disorders of whit

BACKGROUND: Burkholderia thailandensis is an environmental Gram-negative bacterium closely related to the highly pathogenic B. pseudomallei, the causative agent of melioidosis. Although B. thailandensis is generally considered avirulent, sporadic human infections and genomic features suggest potential for pathogenic adaptation. We report the whole-genome sequencing and comparative analysis of a B. thailandensis strain (BUR-TH_250524) isolated from soil in Kerala, India, to assess its taxonomic identity, genetic composition, and potential for antimicrobial resistance and virulence. Polycystic ovary syndrome (PCOS) affects 6-20% of reproductive-aged women. It is associated with increased risks of metabolic syndrome, Type 2 diabetes, cardiovascular diseases, mood disorders, endometrial cancer and non-alcoholic fatty liver disease. Although various susceptibility loci have been identified through genetic studies, they account for ∼10% of PCOS heritability. Therefore, the etiology of PCOS remains unclear. This review explores the role of epigenetic changes and modifications in circadian clock genes as potential contributors to PCOS pathogenesis. Epigenetic alterations, such
