Antinuclear antibodies in patients with systemic lupus erythematosus: a comparison of counterimmunoelectrophoresis and immunoblotting.

Traditionally, the method used mostly to identify antinuclear antibody (ANA) specificities is the counterimmunoelectrophoresis technique (CIE), in which a salt extract of rabbit thymus powder (so-called extractable nuclear antigen or ENA) serves as the source of antigen. Recently, the immunoblotting technique (IBT) has been introduced in the serology of antinuclear antibodies. A nuclear extract of HeLa cells is generally used as antigen in this method. In this paper, we compared both methods using sera of patients with active systemic lupus erythematosus (SLE). Only anti-Sm, anti-RNP, and Anti

Cardiovascular abnormalities in multisystem inflammatory syndrome in children related to COVID-19.

INTRODUCTION: The COVID-19 pandemic began with the identification of SARS-CoV-2 in December 2019. Although children usually have milder acute symptoms, they can develop severe systemic symptoms termed pediatric multisystem inflammatory syndrome (MIS-C). This study reviews research in children and adolescents diagnosed with MIS-C, focusing on cardiovascular abnormalities. METHODOLOGY: This systematic review was conducted following PRISMA guidelines. The review protocol was prospectively registered in the Prospective Register of Systematic Reviews (PROSPERO; registration number: CDR420251232497

Anti-f with anti-c detected during pre-transfusion testing: A diagnostic challenge.

The Rh blood group system includes several compound antigens, among which the f antigen (ISBT 004006) is expressed when the c and e antigens occur in cis position on the same RhCE protein. Anti-f is a rare alloantibody but is clinically significant due to its potential to cause hemolytic transfusion reactions and hemolytic disease of the newborn. We report a rare case of anti-f with coexisting anti-c identified during pre-transfusion testing in a 40-year-old female admitted for elective hysterectomy. Her blood group was O Rh D positive and her antibody screen was positive with a negative autoc

Current knowledge and challenges of sepsis-associated encephalopathy.

Encephalopathy is a common complication of sepsis, occurring in up to 70% of patients admitted to the intensive care unit. It is primarily characterized by a deterioration in condition, ranging from delirium to coma, but also by electroencephalographic changes and seizures. Sepsis-associated encephalopathy (SAE) is linked to increased mortality, which rises in proportion to the severity of both clinical manifestations and electroencephalographic abnormalities, and is frequently followed by long-term cognitive impairment and functional disability. The pathophysiology of SAE is complex and inclu

Steroid hormone profiling reveals altered adrenal androgen production in endometriosis.

OBJECTIVE: Endometriosis is a chronic, hormone-dependent condition affecting an estimated 190 million women worldwide. Our understanding of hormonal contributions to endometriosis pathophysiology is incomplete, hindering the identification of diagnostic biomarkers and novel therapeutic targets. Although the role of estrogens is well established, research on androgens in endometriosis is limited and the contribution of adrenal-derived 11-oxygenated androgens remains largely unknown. METHODS: We performed steroid androgen profiling to measure androgen concentrations in serum from healthy contro

Speech glottal flow performance in early stages of Parkinson's disease and multiple systems atrophy.

The speech phonation subsystem has been widely studied for assessing phonatory impairments in the presence of numerous pathologies, especially neurodegenerative diseases. Perturbation or/and cepstral measures have been the traditional acoustic features for this purpose. However, these measures have the drawback of involving all the speech production subsystems. To isolate the phonatory component, analysis of the glottal flow generated by the vibration of the vocal folds is an appealing alternative. However, despite its significant pertinence, this approach has been relatively little studied. I

Septic Arthritis of the Left Ankle Caused by Brucella suis biovar 5: A Case Report.

We report a rare case of human brucellosis caused by Brucella suisbiovar 5 in a 75-year-old man from Nagano Prefecture, Japan. He had no history of immune disorders and presented with fatigue, left ankle pain, and swelling. Initial antimicrobial therapy for suspected cellulitis and tenosynovitis failed, leading to admission to our hospital. Imaging and synovial fluid analysis suggested septic arthritis with a periarticular abscess. Gram-negative rods were recovered from synovial fluid obtained via arthrocentesis of the left ankle joint. Although the isolate could not be identified using matrix

Trends in the Frequency and Antimicrobial Resistance of Common Gram-Negative Bacilli Isolated From Urine Cultures.

The sustained increase in antimicrobial resistance among Gram-negative bacilli (GNB) compromises the effectiveness of empirical treatment regimens. This study aims to describe the evolution of the frequency and antimicrobial resistance profiles of the most commonly isolated GNB in urine cultures in our area. A cross-sectional, descriptive, and retrospective study was conducted, analyzing urine cultures from patients with suspected UTIs between 2021 and 2025. Microorganism identification was performed using MALDI Biotyper or MicroScan systems, and antimicrobial susceptibility testing was carrie

Smarter biopsy decisions in thyroid nodules via dual-modal photoacoustic and ultrasound imaging.

Thyroid nodules are primarily diagnosed using ultrasound imaging (USI), but its low specificity leads to unnecessary fine-needle aspiration biopsies (FNABs). In particular, USI's limited ability to differentiate follicular neoplasms from benign nodules contributes to suboptimal biopsy decision-making. We propose a dual-modal imaging approach that combines multiparametric photoacoustic imaging (PAI) and USI to support smarter biopsy decisions. In 106 patients with 29 benign nodules, 45 papillary thyroid carcinomas, and 32 follicular neoplasms, three PAI-derived parameters-the photoacoustic spec

Immunophenotyping of conventional and non-conventional T cell populations by surface staining of peripheral blood mononuclear cells.

Here, we present a comprehensive and reproducible protocol for the isolation, cryopreservation, and immunophenotyping of human peripheral blood mononuclear cells (PBMCs) using a 28-color flow cytometry panel. This high-dimensional panel enables the simultaneous assessment of conventional and unconventional T cells and their activation or functional states in a single sample, making it a valuable tool for translational and clinical immunology research. This protocol outlines detailed procedures for blood processing, density gradient separation, and optimized washing steps to minimise platelet c

Chronic Myeloid Leukemia Presenting With Bilateral Optic Neuropathy and Sensorineural Hearing Loss as the First Clinical Presentation: A Case Report.

Chronic myeloid leukemia (CML) is a myeloproliferative disorder that commonly manifests in chronic, accelerated, or blast phase. Typically observed in individuals aged 60-65 years, CML is infrequently diagnosed in adolescents. The usual presentation in late adulthood involves nonspecific symptoms such as fever, fatigue, and weight loss, with rare reports of initial neurological involvement. A 17-year-old male presented with bilateral vision loss and profound hearing loss, alongside a medical history marked by fever, night sweats, and weight loss. A positive tuberculosis contact raised suspicio

Spontaneous Involution of a Thyroid Nodule: When Nodules Can't Be Trusted.

A thyroid nodule (TN) is an abnormal growth within the thyroid gland, with a large prevalence in adults. They can be classified as benign or malignant based on their characteristics. Most nodules are non-palpable and require ultrasound and histopathological analysis for identification. This study reports a rare case of TN involution despite initially exhibiting malignant ultrasound characteristics. The patient presented with a cystic TN of 27.43 x 13.31 x 35.84 mm, initially classified as Thyroid Imaging Reporting and Data System (TIRADS)-2 and Bethesda II, which later transformed into a solid

Primary Intraventricular Amelanotic Melanoma: Case Report and Literature Review.

Primary amelanotic melanoma of the central nervous system (CNS) represents an exceedingly rare neoplasm within the category of primary CNS melanocytic tumors. The absence of melanin pigmentation often complicates its identification, necessitating histopathological and immunohistochemical evaluation for accurate diagnosis. We describe the case of a 70-year-old man who presented with Parkinsonian features and was found to have a well-circumscribed intraventricular mass in the left atrium of the lateral ventricle (38×45×29 mm), hypointense on T1-weighted MRI with heterogeneous enhancement. Does Tilmanocept based lymphoscintigraphy measure up against colloid-based lymphoscintigraphy for sentinel node biopsy in melanoma and early oral squamous cell cancer?

OBJECTIVE: Sentinel lymph node biopsy (SLNB) is crucial in cutaneous melanoma and oral cavity squamous cell carcinoma (OCSCC) as it directs the need for systemic therapy. There has been growing interest in Technetium-99m-labeled Tilmanocept (TL) due to its specific binding capability. This study compares the efficacy of TL to 99mTc-antimony sulphide colloid (ASC), in these two biologically distinct cancers. DESIGN: In this prospective cross-sectional study, 40 patients were included in each radiotracer group. The primary outcomes measured SN identification rate on lymphoscintigraphy and surgi

Is Sounding the Uterus Prior to Intrauterine Device Placement Still Relevant?

BACKGROUND: Inherited kidney diseases (IKDs) in children pose unique diagnostic and therapeutic challenges. IKD significantly impact patient quality of life, morbidity, mortality, and cost to the healthcare system. With over 150 genetic abnormalities, they account for approximately 30% of cases requiring renal replacement therapy. There is an urgent need to advance both diagnosis and treatment strategies. In this review, we present recent advances in diagnosis and treatment for facilitating personalized treatment approaches. SUMMARY: The diagnostic landscape for IKDs have evolved significantl

The mouth, a site of control and pain: a patient-centered qualitative study on oral health experiences in anorexia nervosa.

INTRODUCTION: Anorexia nervosa (AN) is a severe eating disorder with profound psychosocial and physical impacts, including oral health consequences. Despite the documented oral health issues in AN, limited research explores patients' perceptions of their oral health, care and needs. METHODS: The DENT'ANOREXIE study employed a qualitative design, conducting semi-structured interviews with 12 female participants (ages 18-57) diagnosed with AN, at a single French hospital. Data were analyzed thematically using Braun and Clarke's six-step framework. RESULTS: Three key themes emerged: (1) symboli

Investigation on ABCC6-Deficient Human Hepatocytes Generated by CRISPR-Cas9 Genome Editing.

Patients affected by the rare disease pseudoxanthoma elasticum (PXE) exhibit the calcification of elastic fibers in ocular, dermal, and vascular tissues. These symptoms are triggered by mutations in the ATP-binding cassette transporter subfamily C member 6 (ABCC6), whose substrate remains unknown. Interestingly, ABCC6 is predominantly expressed in the liver tissue, leading to the hypothesis that PXE is a metabolic disorder. We developed a genome-editing system targeting ABCC6 in human immortalized hepatocytes (HepIms) for further investigations. The HepIms were transfected with an ABCC6-specif

Understanding Sex Differences in Autoimmune Diseases: Immunologic Mechanisms.

Autoimmune diseases such as systemic lupus erythematosus and Sjögren's syndrome show pronounced sex disparities in prevalence, severity, and clinical outcomes, with females disproportionately affected. Emerging evidence highlights sex-based differences in immune and inflammatory responses as key contributors to this bias. Genetic factors-including sex chromosomes, skewed X chromosome inactivation, and sex-biased microRNAs-as well as sex hormones and pregnancy modulate gene expression and immune cell function in a sex-specific manner. Additionally, sex hormone-dependent epigenetic modifications

A visual guide for emergency radiologists navigating the complexity of abdominal vasculitis: part 1 - true vasculitis.

Systemic vasculitides are rare, multi-organ inflammatory disorders involving blood vessels, classified by the size of the affected vessels. When abdominal arteries are involved, diagnosis becomes particularly challenging due to nonspecific symptoms such as fever, fatigue, and abdominal pain, which can mimic other acute conditions and hinder identification in emergency settings. Examples include Takayasu arteritis (large vessels), polyarteritis nodosa (medium vessels), and lupus-associated or drug-induced vasculitis (small vessels), each exhibiting distinct imaging features. Imaging techniques,

In vitro monitoring of Babesia microti infection dynamics in whole blood microenvironments.

Babesiosis - a globally emerging tick-borne infectious disease primarily caused by the intraerythrocytic piroplasm parasite, Babesia microti - has traditionally been studied using animal models such as mice. Compared to animal models, microfluidic-based models offer advantages, including direct analysis of human samples (such as patient blood), enhanced assay capacity (including physical/optical access, consistency, and throughput), low costs, and easy adoption. Here, we report an open microfluidic platform named "μ-Blood" for monitoring B. microti infection dynamics in vitro . Compared to oth
