Neoplastic hormone-producing cells of the placenta produce and secrete parathyroid hormone-related protein. Studies by immunohistology, immunoassay, and polymerase chain reaction.

BACKGROUND: Parathyroid hormone-related protein (PTHrP) was originally discovered as a product of tumors associated with hypercalcemia. Subsequent studies have demonstrated the widespread production of PTHrP in both fetal and adult tissues, among them the placenta, where it has been identified in amniotic epithelial cells and trophoblasts. However, studies of PTHrP production by the neoplastic placenta have not been reported. EXPERIMENTAL DESIGN: We have studied placental cells and tissues for PTHrP by immunohistology, immunoassay, and the polymerase chain reaction. We studied normal placenta…

Immunohistochemical localization, messenger ribonucleic acid abundance, and activity of 15-hydroxyprostaglandin dehydrogenase in placenta and fetal membranes during term and preterm labor.

Type 1 15-hydroxyprostaglandin dehydrogenase (PGDH) is the main enzyme responsible for the metabolism of prostaglandin E2 (PGE2) and PGF2 alpha. To examine the possibility that a deficiency of PGDH might contribute to preterm labor, we measured localization of immunoreactive (IR-) PGDH, PGDH mRNA, and PGDH enzyme activity in chorio-decidua, placenta, and amnion in patients after term elective cesarean section (n = 9), after spontaneous vaginal term delivery (n = 10), and at idiopathic preterm labor (PTL) in the absence of infection (< 36 weeks gestation; n = 11). Localization of IR-PGDH was de…

Mouse placenta: hemodynamics in the main maternal vessel and histopathologic changes induced by 2-methoxyethanol and 2-methoxyacetic acid following maternal dosing.

The two main maternal vessels that are a major, if not the entire, source of maternal blood for the mouse placenta are unique in possessing intraluminal valvular projections. The morphologic configuration of these projections suggests their potential to converge, diverge, and rotate blood currents flowing under systolic pressure. The intravascular occurrence of circular fibrin bodies composed of concentric fibrin strands coagulated from the plasma and almost no blood cellular elements in these strands lends credence to this concept. Histopathologic changes in the extraembryonic and embryonic t…

cDNA cloning of cytochrome P-450 related to P-450p-2 from the cDNA library of human placenta. Gene structure and expression.

We have isolated and analyzed cDNA (designated P-450HP cDNA) clones from a human placenta cDNA library, using the cDNA for rabbit pulmonary cytochrome P-450p-2, a prostaglandin omega-hydroxylase, as a hybridization probe. The cDNA obtained encoded a polypeptide comprising 511 amino acids with a calculated molecular mass of 58987 Da, and the amino acid sequence similarity with P-450p-2 and rat liver laurate omega-hydroxylase (P-450LA omega) was only about 50%. RNA blot analysis showed that the mRNA hybridizable with the human P-450HP cDNA was inducibly expressed 3-5-fold in rabbit small intesti…

Possible synthesis of pregnancy-'specific' beta-1-glycoprotein (sp1) and placenta-'specific' tissue proteins (PP10,PP12) by human and cynomolgus monkey leukocytes.

Using an enzyme-bridge immunoperoxidase (PAP) technique, it was revealed that pregnancy- and/or placenta-specific proteins, SP1, PP10 and PP12 were localized in human and cynomolgus monkey (Macaca fascicularis) polymorphonuclear neutrophils, and PP10, in mononuclear phagocytes. There was no difference in staining results, irrespective of whether the specimens were from human or monkey males, or pregnant or nonpregnant females. The results suggest that SP1, PP10 and PP12 may be synthesized by polymorphonuclear neutrophils and/or mononuclear phagocytes and could represent markers not only for tr…

Detection of highly pathogenic avian influenza A(H5N1) virus, clade 2.3.4.4b, genotype B3.13 in placentas from aborted bovine fetuses.

Infection by highly pathogenic avian influenza (HPAI) A(H5N1) virus (family Orthomyxoviridae, taxon species Alphainfluenzavirus influenzae), clade 2.3.4.4b, genotype B3.13, was first diagnosed in dairy cattle in the United States in March 2024. Here, we present a cluster of bovine abortions in whose placentas HPAI A(H5N1) was detected. Only placentas were submitted in all 5 cases received. Grossly, 3 placentas had multifocal edema; no significant gross abnormalities were observed in the other 2 placentas. Microscopically, one placenta had random areas of mineralization and another placenta had…

Prenatal metal exposure moderates the effect of CRH-related DNA methylation in the placenta on infant communication outcomes at 2 months of age.

BACKGROUND/ AIM: Prenatal stress and environmental metal mixtures impact neurobehavioral development, yet their interplay and potential mechanisms via placental epigenetics remain unclear. We investigated whether prenatal metal mixtures moderate the epigenetic pathway linking maternal stress at delivery (corticotropin-releasing hormone, CRH) to infant neurobehavioral development. METHODS: In a prospective cohort of 120 mother-infant pairs, we measured maternal plasma CRH at delivery, placental DNA methylation (Infinium EPIC v2.0), and placental concentrations of 18 metals (ICP-MS). Infant neu…

Differential expression of VEGF, PlGF and their receptors in placentas from anti-Ro/SSA- and anti-La/SSB-positive pregnant women.

Members of the vascular endothelial growth factor (VEGF) family of growth factors and their receptors, including VEGF-A, placental growth factor (PlGF), VEGF receptor (VEGFR)-1, and VEGFR-2 with co-receptor neuropilin-1 (NRP-1), are known to play a crucial role in the normal development and maintenance of the morphofunctional features of the placenta, also contributing to the peculiar immune tolerance occurring during pregnancy. Indeed, an altered expression of VEGF family members and their receptors has been implicated in inadequate placentation and dysregulation of the immune tolerance in a …

Exploring the role of specialized pro-resolving mediators, fatty acid oxidation markers and transporters in Gestational Diabetes Mellitus (GDM) placentae.

BACKGROUND: The role of fatty acids is critical throughout pregnancy and can influence pregnancy outcome. Materno-fetal transfer of fatty acids depends on placental fatty acid metabolism which includes fatty acid specialized pro-resolving mediators (SPMs) production, activation of peroxisome proliferated activated receptors (PPARs) and fatty acid oxidation. Alterations in any of these mechanisms may influence fetal development. This study reports placental levels of SPMs (protectins and lipoxins), PPARα, fatty acid oxidation marker (CPT1A) and fatty acid transporters (FATP1 and FATP4) in GDM a…

Persistent Cord Presentation Due to Marginal Cord Insertion at the Lower Edge of a Low-Lying Placenta: A Case of Successful Vaginal Delivery.

The position of the umbilical cord within the uterus is influenced by its insertion site, with low insertion near the internal os raising concerns regarding the risk of cord prolapse and feasibility of vaginal delivery. This report describes a case of persistent cord presentation caused by marginal cord insertion at the lower edge of a low-lying placenta, further complicated by preterm premature rupture of membranes (pPROM) at 30 weeks. Given the fetal immaturity, expectant management was pursued despite the potential risk of cord prolapse. Although persistent amniotic fluid leakage occurred, …

Predicting Time to Delivery in Hypertensive Disorders: Assessing PlGF and sFlt-1 with the Novel Parameter 'Mtp-Multiples of a Normal Term Placenta'.

Background: Imbalanced angiogenesis is characteristic of normal placental maturation but it also signals placental dysfunction, underlying hypertensive disorders during pregnancy. This study aimed to investigate the relationship between angiogenic placental aging, measured by markers placental growth factor (PlGF) and soluble fms-like tyrosine kinase-1 (sFlt-1) using the new index "Multiples of a normal term placenta" (Mtp) and the duration of pregnancy. Methods: A retrospective observational study was conducted, including singleton pregnancies diagnosed or suspected of hypertensive disorders …

Expression of the mucin-like glycoprotein CD24 and its ligand siglec-10 in placentas with acute and post SARS-CoV-2 infection.

CD24 is a mucin-like glycoprotein expressed on trophoblast cells and endothelial tissue of first and third trimester placentas. As an immune suppressor, CD24 may contribute to maternal immune tolerance to the growing fetus. CD24 is known to interact with the sialic acid-binding immunoglobulin-type lectins (Siglecs), specifically siglec-10. The aim of this study was to investigate the expression of both, CD24 and siglec-10 on placental tissue slides from acute covid patients, patients who survived a covid-19 infection and normal term controls. For the evaluation of CD24 & siglec-10 we used a to…

Placental Transcriptomic Signatures of Prenatal Phthalate Exposure and Identification of Placenta-Brain Genes Associated with the Effects of Phthalate Exposure on Neurodevelopment in Children.

Prenatal exposure to phthalates may affect placental function and fetal development, but the underlying mechanisms are unclear. The aim of our study was to explore the alterations in the placental transcriptome associated with prenatal phthalate exposure and to further analyze whether the placental-brain axis (PBA) genes play a mediating role in the association between prenatal phthalate exposure and children's neurodevelopment. We included 172 participants from the Ma'anshan Birth Cohort and collected data on seven phthalate metabolites in urine during pregnancy, placental tissue RNA-seq, and…

Clinical evaluation of the effect for prophylactic balloon occlusion in pregnancies complicated with placenta accreta spectrum disorder: A systematic review and meta-analysis.

BACKGROUND: Placenta accreta spectrum (PAS) disorder is a critical and severe obstetric condition associated with high risk of intraoperative massive hemorrhage and cesarean hysterectomy. Severe obstetric hemorrhage is currently one of the leading causes of maternal death worldwide. Prophylactic balloon occlusions, including prophylactic balloon occlusion of the abdominal aorta (PBOAA) and prophylactic balloon occlusion of the internal iliac arteries (PBOIIA), are the most common means of controlling hemorrhage in patients with PAS disorder, but their effectiveness is still debated. OBJECTIVE…

A novel approach in the management of placenta accreta spectrum disorders: A single-center multidisciplinary surgical experience at Tu Du Hospital in Vietnam.

OBJECTIVE: Placenta accreta spectrum disorders (PASD) are the leading cause which results in highly maternal mortality during pregnancy. Although hysterectomy has been the gold standard for PASD, the recent study along with our experience has been demonstrated that the association between uterine myometrial resection and transverse B-Lynch suture in conservative management might be effective in the appropriate patients, thus we hereby attempted to determine this issue. MATERIALS AND METHODS: A retrospective observational study enrolled 65 patients at Tu Du Hospital in Vietnam between January …

Absence of SARS-CoV-2 Spike glycoprotein expression in placentas from individuals after mRNA SARS-CoV-2 vaccination.

Current public health initiatives to contain the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) global pandemic focus on expanding vaccination efforts to include vulnerable populations such as pregnant people. Vaccines using messenger ribonucleic acid (mRNA) technology rely on translation by immune cells, primarily at the injection site. Hesitancy remains among the general population regarding the safety of mRNA vaccines during gestation, and it remains unknown whether the SARS-CoV-2 Spike protein (the product of mRNA vaccines available) accumulates in the placenta after vaccinat…
