Evidence for and against the transmissibility of Alzheimer disease.

Nonhuman primates were inoculated intracerebrally with brain tissue from 52 patients with confirmed Alzheimer disease (AD) in order to investigate the possibility of an infectious etiology. Animals inoculated with brain tissue from two patients with familial AD developed a spongiform encephalopathy that was indistinguishable from Creutzfeldt-Jakob disease (CJD). Seventeen other cases of AD on test for more than 50 months failed to produce similar changes, and 33 cases have not been incubating for a sufficient period of time to ascertain the presence of a transmissible agent. The initial transm…

Tupaias (tree shrews)--a new animal model for gallstone research; first observations of gallstones.

In tupaias, rat-sized mammals with phylogenetic affinities to insectivores and primates, gallstones can be induced by diet. Twenty per cent butter, 20% sucrose, and 1% cholesterol, added to the standard diet, caused massive stone formation in 11 of 16 male tupaias. In the other five animals the bile contained cholesterol crystals or few stones, liquid crystals, and fat droplets. The stones were composed almost totally of cholesterol and closely resembled those described in squirrel monkeys, which had been fed a similar diet. The diet caused neither a change in body weight nor serious diarrhea …

Elemental analysis of a tumor from a nocturnal prosimian with special emphasis on bromine.

Few natrually occurring tumors have been reported in primates. A spherical mass was noticed on the medial aspect of the thigh and caudal abdomen of a 15-year old female Perodicticus potto. The mass was surgically removed and the recovery was uneventful. Histological examination provided the diagnosis of myeloliposarcoma. Thirty-two elements were detected by chemical analysis. These are Na, K, Rb, Cs, Li, Cu, Ag, Be, Mg, Ca, Sr, Zn, Cd, Hg, B, Al, Ga, Si, Sn, Ti, P, Bi, V, S, Se, Mo, F, Cl, Br, Fe, Co and Ni. It has been proposed by some that tumor tissue tends to be chemically similar to embry…

Viruses and cancer of the lower genital tract.

The importance of viruses as oncogenic agents in animals is well established. Recent work suggests that viruses may also be etiologically re … ng cervical neoplasia. Additional studies are also in progress on animal models, including subhuman primates, and efforts continue in the attempt to confirm the presence of viral genetic material or its expression in human cervical cancer cells. The possibility that human wart viruses have an oncogenic potential is suggested by clinicopathologic and electr …

Osseous coagulum: a histologic evaluation.

Chronic periodontitis can be successfully simulated in primates by the method employed in this study. Osseous defects can be created with a marked degree of similarity to one another and subsequently rendered into chronic lesions for healing-repair studies. The chronic periodontal ossious defects corrected by the osseous coagulum technique and by curettage in the rhesus monkey in this study were repaired by the regeneration of the architecture of the lost tissue. The use of the osseous coagulum in two- and three-walled periodontal osseous defects led to a more rapid osteogenesis in such defect…

Ex Vivo Therapeutics in Cardiac Xenotransplantation: A Scoping Review.

Genetically modified porcine hearts through xenotransplantation offer a promising solution for the current donor scarcity from human allotransplantation. Machine perfusion has successfully been used for graft preservation in two landmark in-human cardiac xenotransplantation cases using hypothermic oxygenated perfusion. Alternate machine perfusion approaches, including normothermic perfusion on a "beating heart" device such as the Transmedics Organ Care System (OCS), have not been fully evaluated. Although preclinical studies have shown that continuous machine perfusion enhances graft viability

Animal Models for Interventional Oncology Research: Overview and Considerations in the Era of New Approach Methodologies.

While translational animal models play a meaningful role in interventional oncology (IO) research, recent attention has focused on new approach methodologies (NAMs) prioritizing human biology-based paradigms. In this narrative review, animal models used in IO research, spanning mouse, rat, woodchuck, rabbit, pig, dog, and non-human primate platforms, are presented. In addition, "next generation" animal models, such as humanized models, genetically defined models, and comorbidity models, are summarized. Subsequently, NAM concepts, including organoids, organ-on-a-chip systems, ex vivo organ prep

Acute Cortical Blindness Associated with Occipital Lobe Compression from Simian T Cell Lymphoma Virus Lymphoma in a Baboon (Papio anubis).

Simian T cell leukemia virus (STLV) is associated with lymphoma in many captive and wild Old World nonhuman primate (NHP) species and is readily transmitted by bodily fluids. The disease is best characterized in baboons with a predisposition for aged female animals. Asymptomatic infections are common; however, clinical signs may include generalized lymphadenopathy, lethargy, and anorexia. Herein, we report a case of disseminated lymphoma in a middle-aged female STLV-positive olive baboon (Papio anubis) that presented with generalized lymphadenopathy and a rapid onset of decreased visual acuity

High-Throughput Screening for the Prevalence of Neutralizing Antibodies against Human Adenovirus Serotype 5.

Adenoviral vectors based on the human adenovirus species C serotype 5 (HAdV-C5) are commonly used for vector-based gene therapies and vaccines. In the preclinical stages of development, their safety and efficacy are often validated in suitable animal models. However, pre-existing neutralizing antibodies may severely influence study outcomes. Here, we generated a new HAdV-C5-based reporter vector and established a high-throughput screening assay for the multivalent detection of HAdV-C5-neutralizing antibodies in serum. We screened the sera of rhesus macaques at different primate centers, and of

Evolving dynamics of Aedes-borne diseases in Africa: a cause for concern.

Aedes-borne viruses, yellow fever (YF), dengue, Chikungunya and Zika are taking a huge toll on global health as Africa faces re-emergence with potential for massive human catastrophe. Transmission driven by diverse vectors in ecological settings that range from urban to rural and sylvatic habitats with human and nonhuman primate/reservoir activities across such habitats has facilitated virus movement and spillover to susceptible human populations. Approved vaccine exists for YF, although availability for routine and mass vaccination is often constrained. Integrating vector surveillance, unders

Pulmonary-Resident Memory Lymphocytes: Pivotal Orchestrators of Local Immunity Against Respiratory Infections.

There is increasing evidence that lung-resident memory T and B cells play a critical role in protecting against respiratory reinfection. With a unique transcriptional and phenotypic profile, resident memory lymphocytes are maintained in a quiescent state, constantly surveying the lung for microbial intruders. Upon reactivation with cognate antigen, these cells provide rapid effector function to enhance immunity and prevent infection. Immunization strategies designed to induce their formation, alongside novel techniques enabling their detection, have the potential to accelerate and transform va

Idiopathic Chronic Diarrhea in Rhesus Macaques Is Not Associated with Enteric Viral Infections.

While recent changes in treatment have reduced the lethality of idiopathic chronic diarrhea (ICD), this condition remains one of the most common causes of rhesus macaque deaths in non-human primate research centers. We compared the viromes in fecal swabs from 52 animals with late stage ICD and 41 healthy animals. Viral metagenomics targeting virus-like particles was used to identify viruses fecally shed by each animal. Five viruses belonging to the Picornaviridae, one to the Caliciviridae, one to the Parvoviridae, and one to the Adenoviridae families were identified. The fraction of reads matc

Pathology and One Health implications of fatal Leptospira interrogans infection in an urbanized, free-ranging, black-tufted marmoset (Callithrix penicillata) in Brazil.

Leptospirosis is a zoonotic neglected disease of worldwide public health concern. Leptospira species can infect a wide range of wild and domestic mammals and lead to a spectrum of disease, including severe and fatal forms. Herein, we report for the first time a fatal Leptospira interrogans infection in a free-ranging nonhuman primate (NHP), a black-tufted marmoset. Icterus, pulmonary haemorrhage, interstitial nephritis, and hepatocellular dissociation were the main findings raising the suspicion of leptospirosis. Diagnostic confirmation was based on specific immunohistochemical and PCR assays

Parietal Cortex Regulates Visual Salience and Salience-Driven Behavior.

Unique stimuli stand out. Despite an abundance of competing sensory stimuli, the detection of the most salient ones occurs without effort, and that detection contributes to the guidance of adaptive behavior. Neurons sensitive to the salience of visual stimuli are widespread throughout the primate visual system and are thought to shape the selection of visual targets. However, a neural source of salience remains elusive. In an attempt to identify a source of visual salience, we reversibly inactivated parietal cortex and simultaneously recorded salience signals in prefrontal cortex. Inactivation

Spatiotemporal expansion of primary progenitor zones in the developing human cerebellum.

We present histological and molecular analyses of the developing human cerebellum from 30 days after conception to 9 months after birth. Differences in developmental patterns between humans and mice include spatiotemporal expansion of both ventricular and rhombic lip primary progenitor zones to include subventricular zones containing basal progenitors. The human rhombic lip persists longer through cerebellar development than in the mouse and undergoes morphological changes to form a progenitor pool in the posterior lobule, which is not seen in other organisms, not even in the nonhuman primate

Apolipoprotein L1 and Kidney Disease in African Americans.

Genetic variants in the Apolipoprotein L1 (APOL1) gene cause high rates of kidney disease in African Americans. These variants, found only in individuals with recent African ancestry, confer enhanced innate immunity against African trypanosomes. Although they are among the most powerful disease-causing common variants discovered to date, we are just beginning to understand how they promote kidney injury. Since APOL1 is present in only a few primate species, much of our current knowledge has come from natural experiments in humans and in vitro studies while awaiting the development of transgeni

Human speech- and reading-related genes display partially overlapping expression patterns in the marmoset brain.

Language is a characteristic feature of human communication. Several familial language impairments have been identified, and candidate genes for language impairments already isolated. Studies comparing expression patterns of these genes in human brain are necessary to further understanding of these genes. However, it is difficult to examine gene expression in human brain. In this study, we used a non-human primate (common marmoset; Callithrix jacchus) as a biological model of the human brain to investigate expression patterns of human speech- and reading-related genes. Expression patterns of s

Sebaceous gland metaplasia in the oesophagus of a cynomolgus monkey (Macaca fascicularis).

Microscopical evaluation of tissues from a mature female cynomolgus monkey assigned to a 13-week oral toxicity study revealed nests of sebaceous cells within the mucosa of the distal oesophagus. The sebaceous cell nests showed normal differentiation from basally-oriented seboblasts to centrally-orientated mature sebocytes. No excretory ducts were found, but solitary or small groups of vacuolated sebocytes were present in the overlying normal, non-keratinized squamous epithelium. The finding of sebaceous cells, of presumed ectodermal origin, in the oesophageal mucosa, which is of endodermal ori
