Williams syndrome: a relationship between genetics, brain morphology and behaviour.

BACKGROUND: Genetically Williams syndrome (WS) promises to provide essential insight into the pathophysiology of cortical development because its ∼28 deleted genes are crucial for cortical neuronal migration and maturation. Phenotypically, WS is one of the most puzzling childhood neurodevelopmental disorders affecting most intellectual deficiencies (i.e. low-moderate intelligence quotient, visuospatial deficits) yet relatively preserving what is uniquely human (i.e. language and social-emotional cognition). Therefore, WS provides a privileged setting for investigating the relationship between

Ictal SPECT in neocortical epilepsies: clinical usefulness and factors affecting the pattern of hyperperfusion.

INTRODUCTION: The aims of this analysis were to: (1) determine the value of ictal SPECT in the localization of neocortical epileptogenic foci, (2) evaluate the relationships between the results of ictal SPECT and other potential affecting factors, and (3) compare traditional visual analysis and the subtraction method. METHODS: We retrospectively analyzed 81 consecutive patients with neocortical epilepsy who underwent epilepsy surgery and achieved a favourable surgical outcome, including 36 patients with normal MRI. Side-by-side visual analysis and subtraction images were classified as correct

Integration of diverse information in working memory within the frontal lobe.

Ability to integrate diverse forms of information in current thought, or working memory, is essential for human reasoning and problem solving. We used functional imaging to identify brain regions preferentially involved in maintaining integrated versus unintegrated information in working memory. For equal amounts of verbal and spatial information, activation of prefrontal cortex was greater for maintaining integrated rather than unintegrated representations. Posterior brain regions showed the opposite pattern. These results demonstrate frontal-lobe specialization in maintaining working-memory

Brain serotonin2 receptors in major depression: a positron emission tomography study.

BACKGROUND: Postmortem and brain imaging studies that measured brain serotinin(2) (5-HT(2)) receptors in major depression reported an increase, decrease, and no change compared with controls. In this study, we assessed brain 5-HT(2) receptors in 20 depressed patients (mean +/- SD age, 40.1 +/- 9.5 years; range, 22-60 years) and 20 healthy controls similar in age (37.2 +/- 12.6 years; range, 19-59 years) using positron emission tomography and setoperone labeled with fluorine 18 ([(18)F]setoperone). METHODS: Patients with DSM-IV major depression and healthy controls underwent scanning with [(18

Brain metastasis in renal cell carcinoma: clinical data and neuropathological differential diagnoses.

Beside lung and skeleton, renal cell carcinoma (RCC) can also metastasize to the brain. In order to collect clinical and histopathological data on these tumors, we studied the local distribution of 50 metastases of RCC in different brain regions (frontal, parietal, temporal occipital lobe and brain stem/cerebellum) and found frequencies ranging from 18.4% to 22.4% in each region. This indicates that there is no preferable site of renal cell carcinoma metastases in the brain. In 46 (92%) of the cases the primary tumor or metastases in other organs had already been diagnosed before operation of

Brain insulin and insulin receptors in aging and sporadic Alzheimer's disease.

The search for the causes of neurodegenerative disorders is a major theme in brain research. Acquired disturbances of several aspects of cellular metabolism appear pathologically important in sporadic Alzheimer's disease (SDAT). Among these brain glucose utilisation is reduced in the early stages of the disease and the regulatory enzymes important for glucose metabolism are reduced. In the brain, insulin, insulin-like growth factors and their receptors regulate glucose metabolism and promote neuronal growth. To detect changes in the functional activity of the brain insulin neuromodulatory syst

Normal neuroanatomical variation in the human brain: an MRI-volumetric study.

Normative data on the in vivo size of the human brain and its major anatomically defined subdivisions are not readily available. In this study, high-resolution magnetic resonance imaging was used to measure regional brain volumes in 46 normal, right-handed adults (23 men, 23 women) between the ages of 22-49 years. Parcellation of the brain was based on neuroanatomical landmarks. The following brain regions were measured: the cerebral hemispheres, frontal lobe, temporal lobe, parietal lobe, occipital lobe, cingulate gyrus, insula, cerebellum, corpus callosum, and lateral ventricles. Males tend …

Contextual Updating in Attentional Orienting Relies on the Right Temporoparietal Junction: Evidence From rTMS.

The right temporoparietal junction (rTPJ) has been associated with multiple cognitive functions. Particularly, its involvement in attentional processes has been proposed, representing a key node in shifting and reorienting visuospatial attention toward unexpected stimuli. However, more recent evidence has demonstrated a more postperceptual function, suggesting a crucial engagement of rTPJ in contextual updating mechanisms. Therefore, considering the lack of consensus, the aim of the current study was to elucidate rTPJ contribution to attentional processes by applying repetitive transcranial ma

Tactile stimulus discrimination in adolescents with anorexia nervosa: a behavioral and neurophysiological study.

Body image disturbance (BID) is a key symptom of anorexia nervosa (AN) and involves body size overestimation. Although this overestimation might be related to perceptual deficits, little is known about tactile perception in AN. To clarify the role of putative tactile deficits in BID, the present study investigated 36 adolescent female AN patients and 41 matched healthy controls (HC) using a tactile oddball paradigm during parallel EEG and MEG (EMEG) measurement. Tactile perception was behaviorally tested via the Touch Test (tactile perception threshold), a deviant count task, and a tactile sti

While category-exclusive increases are frequently encountered especially in the anterior temporal lobe, category-exclusive decreases are rare at the neural population level. Category-selective decreases are largely concurrent with increases, yet emerging slightly later and of shorter duration, with population-level increases arguably playing a more s …

Neurostimulation improves reading and alters communication within reading networks in dyslexia.

The left temporo-parietal cortex (TPC) is critical for phonological decoding during reading and appears hypoactive in dyslexia. Therefore, a promising approach to alleviating phonological deficits in dyslexia is to modulate left TPC functioning. However, it is unclear how neurostimulation alters activity and network interactions in dyslexia. To address this gap, we combined facilitatory transcranial magnetic stimulation (TMS) to the left TPC in adults with dyslexia with an overt word and pseudoword reading task during functional neuroimaging. We found TMS-induced improvements in pseudoword rea

Multiscale chemogenetic dissection of fronto-temporal top-down regulation for object memory in primates.

Visual object memory is a fundamental element of various cognitive abilities, and the underlying neural mechanisms have been extensively examined especially in the anterior temporal cortex of primates. However, both macroscopic large-scale functional network in which this region is embedded and microscopic neuron-level dynamics of top-down regulation it receives for object memory remains elusive. Here, we identified the orbitofrontal node as a critical partner of the anterior temporal node for object memory by combining whole-brain functional imaging during rest and a short-term object memory

Temporoparietal structural-functional coupling abnormalities in drug-naïve first-episode major depressive disorder.

INTRODUCTION: Major depressive disorder (MDD) is a debilitating and heterogeneous disease. Many MDD patients experience concurrent anxiety symptoms, often referred to as anxious depression (MDD-ANX). The relationships between network alterations in structural connectivity (SC) and functional connectivity (FC) in MDD and its anxiety-related subtype remain areas that require further investigation. METHODS: We investigated SC-FC coupling at the system and regional levels in 80 never-treated first-episode MDD patients and 80 healthy control (HC) subjects. For brain systems and regions showing sig

Neural mechanisms underlying improved new-word learning with high-density transcranial direct current stimulation.

Neurobehavioral studies have provided evidence for the effectiveness of anodal tDCS on language production, by stimulation of the left Inferior Frontal Gyrus (IFG) or of left Temporo-Parietal Junction (TPJ). However, tDCS is currently not used in clinical practice outside of trials, because behavioral effects have been inconsistent and underlying neural effects unclear. Here, we propose to elucidate the neural correlates of verb and noun learning and to determine if they can be modulated with anodal high-definition (HD) tDCS stimulation. Thirty-six neurotypical participants were randomly alloc

The Role of Occipitotemporal Network for Speed-Reading: An fMRI Study.

The activity of occipitotemporal regions involved in linguistic reading processes, such as the ventral occipitotemporal cortex (vOT), is believed to exhibit strong interactions during higher-order language processing, specifically in the connectivity between the occipital gyrus and the temporal gyrus. In this study, we utilized functional magnetic resonance imaging (fMRI) with psychophysiological interaction (PPI) and dynamic causal modeling (DCM) to investigate the functional and effective connectivity in the occipitotemporal network during speed reading. We conducted the experiment with nati

Atypical functional connectivity of temporal cortex with precuneus and visual regions may be an early-age signature of ASD.

BACKGROUND: Social and language abilities are closely intertwined during early typical development. In autism spectrum disorder (ASD), however, deficits in social and language development are early-age core symptoms. We previously reported that superior temporal cortex, a well-established social and language region, shows reduced activation to social affective speech in ASD toddlers; however, the atypical cortical connectivity that accompanies this deviance remains unknown. METHODS: We collected clinical, eye tracking, and resting-state fMRI data from 86 ASD and non-ASD subjects (mean age 2.3

Coupling between slow waves and sharp-wave ripples engages distributed neural activity during sleep in humans.

Hippocampal-dependent memory consolidation during sleep is hypothesized to depend on the synchronization of distributed neuronal ensembles, organized by the hippocampal sharp-wave ripples (SWRs, 80 to 150 Hz), subcortical/cortical slow-wave activity (SWA, 0.5 to 4 Hz), and sleep spindles (SP, 7 to 15 Hz). However, the precise role of these interactions in synchronizing subcortical/cortical neuronal activity is unclear. Here, we leverage intracranial electrophysiological recordings from the human hippocampus, amygdala, and temporal and frontal cortices to examine activity modulation and cross-r

The electroclinical features and surgical outcomes of inferior perisylvian epilepsy.

OBJECTIVE: To summarize the clinical and electrophysiological observations of epilepsy originating from the inferior perisylvian cortex, and … ION: Inferior perisylvian epilepsy possesses semiological manifestations similar to those of mesial temporal lobe epilepsy or rolandic opercular epilepsy, hence these conditions should be carefully differentiated. Performing lesionectomy or cortectomy, sparing the mesial temporal structures, was found to be an effective and safe treatment modality for IPE.

Spontaneous cortical MEG activity undergoes unique age- and sex-related changes during the transition to adolescence.

BACKGROUND: While numerous studies have examined the developmental trajectory of task-based neural oscillations during childhood and adolescence, far less is known about the evolution of spontaneous cortical activity during this time period. Likewise, many studies have shown robust sex differences in task-based oscillations during this developmental period, but whether such sex differences extend to spontaneous activity is not understood. METHODS: Herein, we examined spontaneous cortical activity in 111 typically-developing youth (ages 9-15 years; 55 male). Participants completed a resting st

Beyond the average brain: individual differences in social brain development are associated with friendship quality.

We tested whether adolescents differ from each other in the structural development of the social brain and whether individual differences in social brain development predicted variability in friendship quality development. Adolescents (N = 299, Mage T1 = 13.98 years) were followed across three biannual waves. We analysed self-reported friendship quality with the best friend at T1 and T3, and bilateral measures of surface area and cortical thickness of the medial prefrontal cortex (mPFC), posterior superior temporal sulcus (pSTS), temporoparietal junction (TPJ) and precuneus across all waves.
