We report in this study that activation of the JNK by the growth factor, CSF-1 is critical for macrophage development, proliferation, and survival. Inhibition of JNK with two distinct classes of inhibitors, the pharmacological agent SP600125, or the peptide D-JNKI1 resulted in cell cycle inhibition with an arrest at the G(2)/M transition and subsequent apoptosis. JNK inhibition resulted in decreased expression of CSF-1R (c-fms) and Bcl-x(L) mRNA in mature macrophages and repressed CSF-1-dependent differentiation of bone marrow cells to macrophages. Macrophage sensitivity to JNK inhibitors may

Increasing evidence demonstrates that IL-6 has a protective role during liver injury. IL-6 activates intracellular pathways via the gp130 receptor. In order to identify IL-6-gp130 pathways involved in mediating liver protection, we analyzed hepatocyte-specific gp130 knockout mice in a concanavalin A-induced (Con A-induced) model of immune-mediated hepatitis. We demonstrated that IL-6-gp130-dependent pathways in hepatocytes alone are sufficient for triggering protection in Con A-induced hepatitis. gp130-STAT3 signaling in hepatocytes mediates the IL-6-triggered protective effect. This was demon

Fibroblast proliferation, differentiation, and migration contribute to the characteristic pulmonary vascular remodeling seen in primary pulmonary hypertension (PPH). The identification of mutations in the bone morphogenetic protein type II receptor (BMPRII) in PPH have led us to question what role BMPRII and its ligands play in pulmonary vascular remodeling. Thus, to further understand the functional significance of BMPRII in the pulmonary vasculature, we examined the expression of TGF-beta superfamily receptors in human fetal lung fibroblasts (HFL) and investigated the role of BMP4 on cell cy

In order to establish infection to host cells, viruses suppress or escape from the host immune response against microorganisms by various strategies. Interferon (IFN) system is an important contributor of innate immunity. IFN is induced by viral infection, and it promotes antiviral state through induction and/or activation of the effector molecules. Many viruses possess the suppression or inhibition mechanisms for the anti-viral effector molecules, whereas they also perform inhibition of IFN signaling pathway, JAK/STAT pathway. We consider that latter is a most effective strategy counteracting

Ovarian hormones are considered to be capable of regulating expression of beta-catenins. A possible role of beta-catenin in alteration of cell morphology has been proposed, but little is known about beta-catenin expression during changes in the tumor morphology of endometrial carcinomas induced by progesterone therapy. To clarify changes in expression of beta-catenin and their relation to morphological alteration, expression of hormone receptors and several cell kinetic markers, sequential biopsy and hysterectomy specimens of 23 endometrial carcinoma and 6 complex hyperplasia with atypia (atyp

During the immediate-early (IE) phase of reactivation from latency, the Kaposi's sarcoma-associated herpesvirus (KSHV) replication and transcription activator protein (RTA) (or ORF50) is thought to be the most critical trigger that upregulates expression of many downstream viral lytic cycle genes, including the delayed-early (DE) gene encoding the replication-associated protein (RAP) (or K8). RAP physically interacts with and stabilizes the cellular transcription factor CCAAT/enhancer-binding protein-alpha (C/EBPalpha), leading to upregulated expression of the cellular C/EBPalpha and p21(CIP-1

IL-4 and IL-13, cytokines with similar biological effects may influence growth and progression of B-cell tumors through regulation of key cell surface molecules important in intercellular communications. In this study, we demonstrate that IL-4 and IL-13 exhibited differential effects on CD23 and CD44 expression and binding to hyaluronan in BL30/B95-8, a Burkitt's lymphoma (BL), and MK3.31, an Epstein-Barr virus transformed normal human B cell line (B-LCL). Studies conducted to understand the molecular mechanisms underlying this differential effect show that IL-4 induced phosphorylation of JAK1

Initiation of the Epstein-Barr virus (EBV) lytic cycle is dependent on the transcription of the BZLF1 gene. The BZLF1 gene promoter (Zp) was activated by crosslinking of cell surface immunoglobulin (Ig) with anti-Ig antibody in B cells, even in the absence of other viral genes. We identified several anti-Ig response elements within Zp, which were originally defined as 12-O-tetradecanoylphorbol-13-acetate (TPA) response elements (ZI repeats and ZII, an AP-1-like domain). Since anti-Ig crosslinking leads to activation of protein kinase C (PKC) and an increase in intracellular calcium level, Zp w

Imine reductases (IREDs) represent a highly promising class of biocatalysts for the asymmetric synthesis of chiral amines bearing two stereogenic centers. Herein, we report an IRED-catalyzed asymmetric reductive amination strategy that converts racemic ketones into γ-piperidinamines with contiguous stereocenters and operates through a pathway consistent with a dynamic kinetic resolution-asymmetric reductive amination (DKR-ARA). The wild-type AcIRED afforded the 1-benzyl-N-cyclopropyl-3-methylpiperidin-4-amine (1a) in 60.8% conversion, but with only 78.5% enantiomeric excess (eecis) for (3S,4R)

Microplastic (MP; MPs) and nanoplastic (NP; NPs) pollution poses a growing threat to global health. Their small size and large surface area facilitate ingestion, accumulation, and interactions with biological systems, leading to toxicity through oxidative stress, inflammation, mitochondrial dysfunction, and cell death. These effects may occur directly or through the release of adsorbed pollutants and plastic additives. Resveratrol (3,5,4'-trihydroxy-trans-stilbene; RV), a natural polyphenol found in grapes, berries, and peanuts, has attracted interest as a potential protective agent because of

Gram-negative bacteria are surrounded by a multilayered cell envelope with a mostly impermeable outer membrane that provides intrinsic resistance to many antibiotics1-5. The outer membrane is an asymmetrical bilayer with phospholipids in the inner leaflet and lipopolysaccharide (LPS) in the outer leaflet6,7. An LPS transport (Lpt) machine called LptB2FGCADE moves LPS across a protein bridge from the inner membrane to the outer membrane8,9. LPS biosynthesis is regulated to prevent toxic accumulation of LPS molecules in the inner membrane during growth10-12. Whether LPS transport across the Lpt

Immune-mediated inflammatory diseases (IMIDs) refer to a heterogeneous group of conditions driven by dysregulated immune responses that affect multiple organs and systems, leading to substantial morbidity and premature mortality. Accumulating evidence indicates that excessive interleukin-6 (IL-6) production and aberrant activation of downstream signaling pathways play central roles in sustaining chronic inflammation and acute hyperinflammatory states. This review aims to summarize the current understanding of IL-6 biology and signaling modes, and critically evaluate therapeutic strategies targ

Rapid and ultrasensitive detection of Vibrio parahaemolyticus (V. parahaemolyticus, Vp) is of great significance for the early prevention of foodborne disease. Traditional methods for detecting Vp are time-consuming, exhibiting low sensitivity and specificity. In this study, CRISPR/Cas12a system is integrated with electrochemical sensing and polymerase chain reaction (PCR) to establish a PCR-based E-CRISPR biosensor for Vp detection. The target DNA extracted from Vp is amplified by PCR, then activate CRISPR/Cas12a system to cleave methylene blue (MB)-labeled hairpin DNA probes on electrode, re

Mitogen-activated protein kinase kinase (MEK) is a component of an important signaling pathway involved in the development and progression of pancreatic ductal adenocarcinoma (PDAC). However, MEK-targeted therapeutics are not effective, and therefore not indicated, for patients with PDAC. We have found that annexin A8 (ANXA8) is involved in resistance to MEK inhibitor therapy in PDAC. Expression of ANXA8 was induced at both the mRNA and protein levels early by MEK inhibitor treatment in PDAC cells, and the level of ANXA8 mRNA expression was inversely correlated with sensitivity to the MEK inhi

Antibody-drug conjugates (ADCs) have significantly advanced cancer therapy by enabling the selective delivery of cytotoxic agents to tumour cells. However, ADC efficacy remains constrained by its dependence on a single target antigen, which limits tumour targeting and promotes resistance in heterogeneous tumours with variable and low antigen expression1-6. Here we introduce an in vivo bioorthogonal ligation strategy that generates functional antibody-ADC click constructs following systemic administration. This platform provides a modular and translatable approach for enhanced targeted drug del

This study compares the ability of the cytokinin (CK) trans-zeatin (tZ) and the CK sugar conjugate 6-(3-methoxybenzylamino)purine-9-arabinoside (BAPA) to induce resistance against the bacterial pathogen Pseudomonas syringae in Arabidopsis thaliana. Treatment with either tZ or BAPA significantly reduced bacterial growth after a later infection. This chemically induced resistance (IR) required the CK receptor AHK3, highlighting its critical role in mediating resistance by tZ and BAPA. This is remarkable as these compounds show either high or no affinity for this CK receptor, respectively. Surpri

Rapid and ultrasensitive nucleic acid detection is essential for environmental monitoring and biomedical diagnostics. Here, we report a modular one-pot isothermal platform that integrates catalytic hairpin assembly (CHA), rolling circle amplification (RCA), and CRISPR-Cas12a to construct a self-reinforcing multilayer DNA circuit (CRC). In this system, Cas12a cis-cleavage generates short DNA fragments that recursively activate CHA and RCA, forming a self-sustained cascade amplification loop, while trans-cleavage enables real-time fluorescence signal readout. Using this one-pot platform, ultralo

CD169/Siglec-1 is a type-I transmembrane lectin receptor expressed on subcapsular sinus macrophages and activated monocytes, which plays a key role in innate immune surveillance and orchestration of adaptive immunity. As a sialic acid-binding immunoglobulin-like lectin (Siglec), CD169 recognizes sialylated gangliosides and glycoconjugates present on surface of several viral envelopes, including HIV-1, SARS-CoV-2, and Ebola virus as well as sialylated bacterial capsules such as those of Neisseria meningitidis and Campylobacter jejuni. Moreover, different viruses were shown to be captured by CD1

Various Ni-based metalloenzymes employ synergistic heterometallic cooperativity where a Ni center and a second metal ion cofacilitate small-molecule conversion. Inspired by these biological systems, synthetic model complexes have been developed that exploit bimetallic cooperativity to achieve analogous reactivity. In this work, we report a bimetallic Ni-Zn complex comprising a pincer-type diamido-diaminodiphosphine ligand, in which a weakly Lewis acidic Zn(II) center stabilizes a T-shaped Ni(0) site. Subsequent reactivity with H2 led to the isolation of a rare Ni dihydride complex, which dynam

To meet the urgent need for sensitive detection of foodborne pathogens and antibiotic residues-key to preventing outbreaks and curbing antimicrobial resistance-we developed a stable fluorescent probe, H4TCPE@UiO-66 (H@U). The probe retains strong fluorescence under harsh conditions (18.70% intensity change at pH 3, 11.36% at 80 °C) and exhibits a 6.90-fold signal amplification over H4TCPE. In addition, H@U is able to be a sensitive probe integrated with millimetre-scale polystyrene microspheres (mPS) and realize rapid detection of chloramphenicol within 20 min with a detection limit of 33.52 p
