Cathodal Transcranial Direct Current Stimulation Does Not Change Implicit Associations Against Alcohol in Alcohol Use Disorder: A Preregistered Clinical Trial.

Addictive behaviour is shaped by the dynamic interaction of implicit, bottom-up and explicit, top-down cognitive processes. In alcohol use disorder (AUD), implicit alcohol-related associations have been shown to predict increased subsequent alcohol consumption and are linked to the risk of relapse. Explicit cognitive processes, exerting prefrontal top-down control, are particularly significant during the critical period following the decision to abstain. This study aims to map implicit and explicit cognitive processes in recently abstinent individuals with AUD and to explore the effect of cath

"Drink like a man!" masculine drinking norms, alcohol protective behavioral strategies, and severity of hazardous alcohol use among college men.

Learned alcohol-specific gender norms can impact drinking behaviors in nuanced ways. Studies have shown that endorsing the masculine drinking norm (MDN) of excess (e.g., "A real man can drink a lot") is positively associated with alcohol use, while the inverse was found for endorsing the MDN of control (e.g., "A real man knows when he's had enough to drink and is able to stop"). Studies have also indicated that using alcohol protective behavioral strategies (PBS) can reduce the risk for heavy drinking and that men tend to use alcohol PBS less often than women. Thus, there is a need to examine

Alcohol demand and alcohol use disorder among adolescents and young adults who drink heavily: An exploratory analysis including sex differences.

Behavioral economic alcohol demand, or the reward value of alcohol consumption, consistently shows associations between alcohol use and alcohol use disorder (AUD) in adults, yet research within clinical youth populations (considered to be ages 15-24) remains limited. It is critical to better understand how demand functions in clinical youth populations to predict alcohol related outcomes. Moreover, given the narrowing gap between male and female youth in alcohol use prevalence in recent years, exploring sex differences in alcohol demand and its role in the relationship between demand, alcohol

"I just saw the alcohol brand, I never really thought of the zeros": Young people's views of NoLo and alibi alcohol sponsorship.

Alcohol companies frequently use sports sponsorship to maintain high visibility, even in jurisdictions with advertising restrictions. Strategies such as alibi marketing - using brand-associated slogans, colours, or fonts in place of explicit brand names, and NoLo marketing - promoting zero-alcohol variants of core brands, enable continued brand exposure. This study investigates young people's awareness of alcohol sponsorship and their perceptions of alibi and NoLo marketing in sports contexts. In December 2024, ten online focus groups (N = 44) were conducted with participants aged 11-17, strat

Functional interaction between orexin/dynorphin transmission in the posterior paraventricular nucleus of the thalamus following alcohol dependence: mediation of alcohol-seeking behavior.

Stress is a major contributor to the chronic nature of alcohol use disorder (AUD). Orexin (OX) neurons project to the paraventricular nucleus of the thalamus (PVT)-particularly the posterior part (pPVT)- a structure that plays a key role in stress regulation. The blockade of OX receptors in the pPVT was shown to prevent the stress-induced reinstatement of alcohol seeking in alcohol-dependent rats. Accumulating evidence indicates interactions among OX and dynorphin (DYN) in the pPVT, but unclear is the role of OX and DYN transmission in the pPVT in the stress-induced alcohol seeking during alco

Chronic alcohol intake disrupts cytochrome P450 enzyme activity in alcoholic fatty liver disease: insights into metabolic alterations and therapeutic targets.

INTRODUCTION: Alcoholic fatty liver disease (AFLD) is a common consequence of chronic alcohol consumption, characterized by lipid accumulation and oxidative stress in the liver. Cytochrome P450 (CYP450) enzymes play essential roles in metabolizing alcohol and other compounds. However, the specific long-term effects of alcohol on these enzymes remain unclear. METHODS: This study the examines influence of prolonged ethanol exposure on CYP450 activity and expression in AFLD using a rat model. Key enzymes such as CYP2E1, CYP2D6, and CYP3A1 were assessed in relation to lipid accumulation and oxida

Ussurian pear extract alleviates acute alcoholic liver injury by enhancing alcohol metabolism and suppressing hepatic oxidative stress and inflammation in rats.

Acute alcoholic liver injury (AALI) is liver damage caused by excessive short-term alcohol intake and is a key manifestation of acute alcohol intoxication. The Ussurian pear (Pyrus ussuriensis) is a commonly consumed fruit believed to help alleviate the alcohol-related physical burden in Asian areas. In this study, we investigated the effects of the ethanol extract of Ussurian pear (EUP) on the regulation of ethanol-metabolizing enzymes and antioxidative capacity in ethanol-treated HepG2 cells, as well as its anti-inflammatory activity in LPS-stimulated rat peritoneal macrophages. Additionally

A randomized controlled trial to assess whether a telehealth-based contingency management intervention reduces alcohol use for individuals with alcohol use disorder.

BACKGROUND: Contingency management (CM) is an intervention for alcohol use disorder (AUD) that reinforces abstinence, as confirmed by alcohol biomarkers. CM is usually brief (12-16 weeks) despite evidence that longer interventions have better long-term outcomes. Most CM models are in-person which can also be a barrier for treatment. Studies of longer duration telehealth-based CM models are needed. AIMS: To determine if a telehealth-based CM model that utilizes phosphatidylethanol (PEth) to confirm abstinence is effective at reducing alcohol use during a 26-week intervention and 12-month follo

Alcohol approach-avoidance task behavior and brain potentials differentially predict ecologically assessed alcohol craving and consumption in early emerging adulthood.

AIMS: The current study measured the extent to which different neurobehavioral indices of incentive-motivational salience attribution to alcohol cues predict alcohol craving and consumption in the natural environment. DESIGN, SETTING, AND PARTICIPANTS: Laboratory study at a university in Missouri, USA, followed by a smartphone-based 21-day ecological momentary assessment (EMA) protocol. Participants were emerging adults (N = 218-268 [52-56% female], age 18-20). MEASUREMENTS: Participants completed an alcohol cue approach-avoidance task while their electroencephalogram (EEG) was recorded. Beh

Cannabidiol modulates brain molecular alterations, gut microbiota dysbiosis and alcohol self-administration in a mouse model of fetal alcohol spectrum disorder.

Fetal Alcohol Spectrum Disorder (FASD) is a range of neurodevelopmental abnormalities caused by Perinatal Alcohol Exposure (PAE), leading to profound behavioral and molecular disturbances in the offspring. Unraveling the central and peripheral mechanisms involved, including the microbiota-gut-brain axis, is crucial to improving our understanding of the disease and developing new treatment strategies from a sex perspective. In this study, we investigated the impact of PAE on emotional behavior, brain biomarkers, and gut microbiota composition and diversity in a preclinical C57BL/6 J mouse model

Bioprotective Role of Phytocompounds Against the Pathogenesis of Non-alcoholic Fatty Liver Disease to Non-alcoholic Steatohepatitis: Unravelling Underlying Molecular Mechanisms.

Non-alcoholic fatty liver disease (NAFLD), with a global prevalence of 25%, continues to escalate, creating noteworthy concerns towards the global health burden. NAFLD causes triglycerides and free fatty acids to build up in the liver. The excessive fat build-up causes inflammation and damages the healthy hepatocytes, leading to non-alcoholic steatohepatitis (NASH). Dietary habits, obesity, insulin resistance, type 2 diabetes, and dyslipidemia influence NAFLD progression. The disease burden is complicated due to the paucity of therapeutic interventions. Obeticholic acid is the only approved th

Recent Advances in Mesenchymal Stem/Stromal Cell-Based Therapy for Alcohol-Associated Liver Disease and Non-alcoholic Fatty Liver Disease.

Alcohol-associated liver disease (ALD) and non-alcoholic fatty liver disease (NAFLD) represent pathological conditions that include many distinct stages, potentially leading to the final stage of cirrhotic liver. To date, liver transplantation is the sole successful treatment with concomitant limitations related to donor organ shortage and the need of life-long immunosuppressive therapy. Recently, cell-based therapies for ALD and NAFLD have been proposed with mesenchymal stem/stromal cells (MSCs) as promising effectors. MSC therapeutic applications offer hepatoprotection, regulation of the inf

Event-level effects of alcohol, cannabis, and simultaneous alcohol and cannabis use on bystander intentions in response to hypothetical situations among college women.

Bystanders can play an important role in preventing alcohol-related harm (e.g., unintentional injury) or sexual aggression. While the impact of one's own acute alcohol intoxication on sexual aggression bystander intervention has been explored, less is known about how alcohol impacts the ability to intervene in alcohol-related harm. Further, scant research has examined one's own cannabis or simultaneous alcohol and cannabis use (i.e., using alcohol and cannabis at the same time so that their effects overlap) on bystander intervention. The present study employed an ecological momentary assessmen

Testing Alcohol Container Warning Labels Among Alcohol Consumers in the Field Over a 4-Week Period: A Protocol for a Randomized Field Trial.

OBJECTIVE: Online and lab-based experiments examining the impact of alcohol labels typically test a one-time exposure to labels and assess short-term, nonbehavioral outcomes. These studies do not simulate a real-world label dose or assess actual alcohol use. This pilot aimed to develop a new protocol for testing alcohol labels that better reflects real-world exposure by presenting labels on consumers' own alcohol products over time and assessing effects on several outcomes, including alcohol use. METHOD: Forty alcohol consumers in Canada completed an online baseline survey, were randomized to
