In a prospective study of 225 consecutive pediatric patients who required sedation for CT imaging, we monitored oral and nasal air flow, transcutaneous oxygen saturation, and cardiac rate and rhythm before and after the administration of IV Nembutal. In addition, the first 50 patients in this series had blood pressures mechanically monitored at 1-min intervals. Intravenous injection of the alpha-2 adrenergic agonists, clonidine and xylazine, have been previously shown to produce hypoxemia in sheep. To characterize this effect further, clonidine and ST-91, a clonidine analog that does not cross the blood-brain barrier, were injected i.v. in 10 conscious ewes. Although both clonidine (3-15 micrograms/kg) and ST-91 (3-30 micrograms/kg) produced arterial hypoxemia, clonidine was more effective (arterial PO2 was 91 +/- 4 mm Hg after saline, 30 +/- 3 mm Hg after 15 micrograms/kg of clonidine and 43 +/- 6 mm Hg after 30 micrograms/kg of ST-91; mean +/- S.E.

Authors report clinical and laboratory findings, treatment and evolution of six girls and three boys aged between 20 months and 13 years, diagnosed of juvenile polymyositis-dermatomyositis in the last seven years. Presenting symptoms were asthenia and proximal muscle weakness; in 3 cases characteristic skin lesions were associated. All were treated initially with prednisone p.o. (1-2 mg/kg/day) response being favourable in seven. Two patients with chronic evolution were treated with methotrexate and IV bolus of methylprednisolone.

Dercum's disease (adiposis dolorosa) is associated with very painful subcutaneous fatty deposits normally localized to the lower extremities in which pain treatment is usually unsuccessful. The gross and microscopic changes in swine following a single intravascular (iv) dose of T-2 toxin are described and evaluated quantitatively. T-2 toxin, in 70% ethanol, was given iv at 0 (5 pigs), 0.6 (5 pigs), 1.2 (1 pig), 4.8 (5 pigs), or 5.4 (2 pigs) mg/kg to 40 to 60 kg female crossbred pigs. 15 patients receiving cytotoxic drugs (other than cisplatin) that had previously produced nausea and vomiting refractory to first-line antiemetics were given a selective 5-HT3 receptor antagonist (GR38032F), 4 mg intravenously and 4 mg orally, immediately before chemotherapy, the oral dose being repeated 5 and 10 h later. Metronidazole resistance is an uncommon cause of treatment failure of Trichomonas vaginalis. High-dose intravenous metronidazole was used successfully to treat a patient in whom repeated oral therapy with this agent had failed. Metronidazole resistance was documented by aerobic cultures for susceptibility; anaerobic cultures failed to indicate resistance. Metronidazole serum levels were documented. With failure of repeated oral therapy, intravenous metronidazole may be indicated if there is no evidence of other causes for treatment failure, such as patient noncompliance or reinfection.

In chronic experiments on rabbits, i.v. administration of the pentagastrin (0.01-1.0 micrograms/kg) exerted inhibitory effects on the myoelectrical activity of the pylorus and stomach, whereas the effect of pentagastrin on the duodenum was rather activating. The threshold dose of pentagastrin for pylorus was 5 times smaller than for the duodenum and 10 times smaller than for the stomach. Possible mechanisms of the pentagastrin effect on the contractile activity of the pylorus, stomach and duodenum smooth muscles, are discussed.

The effect of sodium salicylate and anaesthesia on the rectal absorption of theophylline was evaluated in rats. Theophylline proved to be slowly but completely absorbed on rectal infusion in conscious rats, compared with intravenous infusion. Pentobarbital anaesthesia did not influence absorption. In contrast to literature data, the results with salicylate showed that rectal absorption of theophylline was not enhanced.

Encainide is a class IC agent possessing a broad spectrum of antiarrhythmic actions in a variety of animal models. It increases the ventricular fibrillation threshold of the perfused rabbit heart and in situ dog myocardium.
