Pharmacological inhibition of both DGKα and DGKζ is required for optimal T cell activation.

Preclinical studies indicate that blocking diacylglycerol kinase (DGK) family members DGKα or DGKζ can improve antitumor immunity, prompting the development of clinical-stage, potent and selective small molecule inhibitors of DGKα and/or DGKζ. DGKα and DGKζ are the most widely expressed DGK family members by immune cells, and both enzymes convert the signaling lipid diacylglycerol (DAG) to phosphatidic acid. DAG is a critical second messenger downstream of T cell receptor (TCR) stimulation that promotes activation and effector function. Blocking DGKα or DGKζ activity enhances DAG-mediated sign

Generation and Characterization of CAR-T Cells.

CARs are synthetic receptors that link antigen binding to T-cell activation. Most CARs used in the clinic for treating cancer are second generation (2G) and comprise (i) a single chain variable fragment (scFv) that binds the target tumor antigen, (ii) a linker/hinge region, (iii) a transmembrane domain, (iv) a costimulatory endodomain, and (v) the endodomain of CD3 zeta. Our lab is focused on the development of function and safety-enhanced, next-generation CAR-T cells for the treatment of solid tumors. For example, we have designed switchable CARs that can be remotely turned on or off upon sma

Bioinformatic identification of CD8+ T cell activation mediated by key genes in fecal microbiota transplantation for irritable bowel syndrome.

BACKGROUND: The effect of fecal microbiota transplantation (FMT) in treating irritable bowel syndrome (IBS) may be attributed to the modulation of CD8 + T cells. This study aims to identify FMT-mediated key genes to explore the underlying mechanism. METHODS: Transcriptomic datasets GSE138297 (colonic biopsies from 8 IBS patients pre- and post-FMT) and GSE134649 (single-cell data from 3 healthy colon tissues) were obtained from GEO during December 2023-December 2024. Key genes were identified by intersecting differentially expressed genes (DEGs) and the most relevant co-expression module deriv

Super-resolution imaging reveals altered actin architecture at CAR-T cell immune synapses.

The actin cytoskeleton plays a key integrative role in immunological synapse (IS) formation during T cell activation, but how these dynamics are altered in chimeric antigen receptor (CAR)-T cells remains unclear. Here, we used stimulated emission depletion (STED) microscopy to perform the first super-resolution analysis of actin remodeling at the IS in single- and dual (CD19/CD22) CAR-T cells, activated on supported lipid bilayers across different time points. Quantitative imaging reveals that CAR-T cells form structurally distinct synapses to untransduced cells, characterized by reduced actin

Soluble PD-L1-secreting umbilical cord-derived mesenchymal stromal cells transiently attenuate established xenogeneic T cell activation in vivo.

Immune overactivation damages tissues in T cell-mediated disorders, and cell-based therapies are being developed to suppress T cell responses. Mesenchymal stromal cells (MSCs) modulate immunity; however, their suppressive effects depend on inflammatory cytokine priming. Umbilical cord-derived MSCs are attractive for non-invasive collection and expansion, but their effects diminish after intravenous infusion. PD-1/PD-L1 signaling lowers T cell activation; therefore, soluble PD-L1 (sPD-L1)-secreting MSCs may provide contact-independent checkpoint engagement and we tested whether sPD-L1-MSCs can

Intracellular CD3 and surface CD8β distinguish activated human T cells from natural killer cells after αCD3/αCD28 stimulation.

Many studies of human T cell function involve activating primary T cells in vitro to assess functional outcomes. A common model of human T cell activation is polyclonal stimulation with αCD3/αCD28 antibodies. However, subsequent identification of these stimulated T cells in a mixed population is complicated by surface CD3 loss. In particular, both cytotoxic T cells and natural killer (NK) cells can appear CD3-CD8α+, leading to misidentification of NK cells as cytotoxic T cells. To address this limitation, we investigated incorporating intracellular CD3 and surface CD8β staining following αCD3/

Butyrophilin 3A1 engages the CDR2δ loop of the Vγ9Vδ2+ T cell receptor.

Phosphoantigen (pAg) recognition by Vγ9Vδ2+ T cells plays a critical role in immunity to pathogens and cancer. The butyrophilin (BTN) family of molecules have emerged as key regulators of γδ T cells; however, the underlying mechanisms remain unclear. Here, we demonstrate an interaction between BTN3A1 and Vγ9Vδ2+ T cell receptor (TCR) in a cell-free assay, confirming that BTN3A1 is a direct ligand for the Vγ9Vδ2+ TCR. Furthermore, immobilized recombinant BTN2A1 plus BTN3A1 extracellular domains are sufficient to activate Vδ2+ T cells. Finally, we show that intracellular pAg accumulation can mod

Vγ9Vδ2 T cells in tuberculosis: protective immunity and translational perspectives.

BACKGROUND: Vγ9Vδ2 T cells constitute the predominant human circulating γδ T cell subset and serve as key mediators of anti-mycobacterial immunity through major histocompatibility complex (MHC)-unrestricted phosphoantigen recognition and rapid effector activation. SUMMARY: This review evaluates Vγ9Vδ2 T cell biology in the context of tuberculosis (TB). We delineate the three-stage developmental pathway of Vγ9Vδ2 T cells in the postnatal thymus and the butyrophilin 3A1/2A1 (BTN3A1/BTN2A1)-mediated "inside-out" antigen sensing mechanism, noting the limitations of extrapolating in vitro findings

In vivo genome-wide CRISPR screens identify FOXR1 as a suppressor of CD8+ T cell antitumor immunity.

T cell dysfunction critically limits the efficacy of T cell-based immunotherapies in solid tumors, yet the intrinsic regulators of T cell dysfunction remain incompletely understood. Through an in vivo genome-wide CRISPR screen in tumor-infiltrating CD8+ T cells, we identified Forkhead Box R1 (FOXR1) as a potent transcriptional suppressor of CD8+ T cell effector functions. Genetic ablation of FOXR1 significantly enhanced cytokine production and cytotoxic capacity in both murine and human CD8+ T cells, whereas its overexpression impaired T cell activation and effector molecule expression. Human LY9 governs CD4+ T cell IFN-γ immunity to Mycobacterium tuberculosis.

CD4+ T cells are indispensable for optimal immunity to Mycobacterium tuberculosis (M.tb), a pathogen that triggers tuberculosis (TB) in humans. M.tb-specific human CD4+ T cells are known to polarize toward an interferon-γ (IFN-γ)-producing, CCR4-CCR6+CXCR3+T-bet+RORγT+ T helper 1* cell (TH1*cell) memory phenotype. We report that autosomal recessive deficiency of the human lymphocytic surface receptor LY9 (SLAMF3 and CD229), which is found in less than 10-5 individuals in the general population, underlies TB in three unrelated patients due to selective impairment in IFN-γ production by TH1* cel

The Human Neonatal Skin Fibroblast, an Available Cell Source for Tissue Production and Transplantation, Exhibits Low Risk of Immunogenicity In Vitro.

The immunogenicity of allogeneic skin fibroblasts in transplantation has been controversial. Whether this controversy comes from a natural heterogeneity among fibroblast subsets or species-specific differences between human and mouse remains to be addressed. In this study, we sought to investigate whether fibroblasts derived from either adult or neonatal human skin tissues could induce different immune responses toward phagocytosis and T cell activation using in vitro co-culture models. Our results indicate that both phagocytosis and T cell proliferation are reduced in the presence of neonatal

Epitope-dependent effect of long-term cART on maintenance and recovery of HIV-1-specific CD8+ T cells.

HIV-1-specific CD8+ T cells are anticipated to become effector cells for curative treatment using the "shock and kill" approach in people living with HIV-1 (PLWH) under combined antiretroviral therapy (cART). Previous studies demonstrated that the frequency of HIV-1-specific CD8+ T cells is reduced under cART and their functional ability remains impaired. These studies analyzed T-cell responses to a small number of HIV-1 epitopes or overlapping HIV-1 peptides. Therefore, the features of CD8+ T cells specific for HIV-1 epitopes under cART remain only partially clarified. Here, we analyzed CD8+

Eribulin mesylate exerts antitumor effects via CD103.

Eribulin mesylate (ERB) is a synthetic analog of halichondrin B, inhibiting tumor cell growth by disrupting microtubule function. Recently, anticancer drugs have been shown to not only act directly on tumor cells but also to exert antitumor effects by modifying the tumor environment. Although ERB has also been speculated to modify the tumor microenvironment including the immune response to tumors, the precise mechanism remains unclear. In our study, ERB suppressed the tumor growth of MC38 colon cancer in wildtype mice, whereas ERB failed to inhibit the tumor growth in Rag1-deficient mice which

Negative times negative equals positive, THEMIS sets the rule on thymic selection and peripheral T cell responses.

The activity of T cells is finely controlled by a set of negative regulators of T-cell antigen receptor (TCR)-mediated signaling. However, how those negative regulators are themselves controlled to prevent ineffective TCR-mediated responses remain poorly understood. Thymocyte-expressed molecule involved in selection (THEMIS) has been characterized over a decade ago as an important player of T cell development. Although the molecular function of THEMIS has long remained puzzling and subject to controversies, latest investigations suggest that THEMIS stimulates TCR-mediated signaling by repressi

Cyclophilin A associates with and regulates the activity of ZAP70 in TCR/CD3-stimulated T cells.

The ZAP70 protein tyrosine kinase (PTK) couples stimulated T cell antigen receptors (TCRs) to their downstream signal transduction pathways and is sine qua non for T cell activation and differentiation. TCR engagement leads to activation-induced post-translational modifications of ZAP70, predominantly by kinases, which modulate its conformation, leading to activation of its catalytic domain. Here, we demonstrate that ZAP70 in TCR/CD3-activated mouse spleen and thymus cells, as well as human Jurkat T cells, is regulated by the peptidyl-prolyl cis-trans isomerase (PPIase), cyclophilin A (CypA) a

A PD-L1-targeting chimeric switch receptor enhances efficacy of CAR-T cell for pleural and peritoneal metastasis.

Pleural and peritoneal metastasis accompanied by malignant pleural effusion (MPE) or malignant ascites (MA) is frequent in patients with advanced solid tumors that originate from the lung, breast, gastrointestinal tract and ovary. Regional delivery of CAR-T cells represents a new strategy to control tumor dissemination in serous cavities. However, malignant effusions constitute an immune-suppressive environment that potentially induces CAR-T cell dysfunction. Here, we demonstrated that the anti-tumor cytotoxicity of conventional 2nd-generation CAR-T cells was significantly inhibited by both th

Can the host immune response against SARS-CoV2 also cause an anticancer effect?

During the COVID-19 pandemic, it is important to assure the safety and management of cancer patients. Despite preliminary studies revealed that patients with cancer are more susceptible to infection and have poorer prognosis than other infected patients without cancer, mortality from COVID-19 in cancer patients appears to be principally driven by age, gender, and comorbidities. So, we have some comments about the pathogenesis attributed to the COVID-19 disease and cancer relationship and determination of subgroups in this and oncoming studies. Variable effects of anticancer treatments on the p

Filgotinib suppresses HIV-1-driven gene transcription by inhibiting HIV-1 splicing and T cell activation.

Despite effective antiretroviral therapy, HIV-1-infected cells continue to produce viral antigens and induce chronic immune exhaustion. We propose to identify HIV-1-suppressing agents that can inhibit HIV-1 reactivation and reduce HIV-1-induced immune activation. Using a newly developed dual-reporter system and a high-throughput drug screen, we identified FDA-approved drugs that can suppress HIV-1 reactivation in both cell line models and CD4+ T cells from virally suppressed HIV-1-infected individuals. We identified 11 cellular pathways required for HIV-1 reactivation as druggable targets. Usi

Kawasaki syndrome: role of superantigens revisited.

Kawasaki syndrome (KS) is an acute vasculitis in children complicated by the development of heart disease. Despite its description over 50 years ago, the etiology of coronary artery disease in KS is unknown. High dose intravenous immunoglobulin is the most effective approach to reduce cardiovascular complications. It remains unclear why patients with KS develop coronary artery aneurysms. A subset of patients is resistant to immunoglobulin therapy. Given the heterogeneity of clinical features, variability of history, and therapeutic response, KS may be a cluster of phenotypes triggered by multi

Recombinant Factor VIII Fc Inhibits B Cell Activation via Engagement of the FcγRIIB Receptor.

The development of neutralizing antibodies (inhibitors) against factor VIII (FVIII) is a major complication of hemophilia A treatment. The sole clinical therapy to restore FVIII tolerance in patients with inhibitors remains immune tolerance induction (ITI) which is expensive, difficult to administer and not always successful. Although not fully understood, the mechanism of ITI is thought to rely on inhibition of FVIII-specific B cells (1). Its efficacy might therefore be improved through more aggressive B cell suppression. FcγRIIB is an inhibitory Fc receptor that down-regulates B cell signali
