Mimicking oxidative damage in γS-crystallin with site-specific incorporation of 5-hydroxytryptophan.

The human eye lens plays an essential role in vision by focusing light onto the retina. This transparent tissue consists of densely packed crystallin proteins that exhibit remarkable solubility despite minimal protein turnover. Post-translational modifications that accumulate over a lifetime can reduce crystallin solubility, resulting in the precipitation or phase separation of protein aggregates. Oxidation is a common type of modification that can cause such opacification of the lens, particularly in age-related cataract. Here, we study the oxidation of W163 in γS-crystallin, a structural len

DNA methylation profiling predicts postsurgical regrowth in SF1-lineage nonfunctioning pituitary neuroendocrine tumors.

BACKGROUND: Nonfunctioning pituitary neuroendocrine tumors (NFPitNETs) account for ∼30-35% of PitNETs; ∼75% arise from the SF1 lineage. Recurrence remains common despite resection (∼30% in 10 years), and routine histopathology/IHC has limited value in predicting recurrence risk. This study evaluated whether DNA methylation profiling improves recurrence risk stratification. MATERIALS AND METHODS: Genome-wide tissue methylation (Illumina EPIC v1, 850K) was analyzed in 117 retrospective NFPitNETs with clinical and imaging follow-up. Unsupervised consensus clustering defined methylation-based sub

Dacron ingrowth during penile prosthesis revision: presentation, outcomes, and surgical management.

The Boston Scientific American Medical Systems 700™ (AMS 700™, Boston Scientific Corporation, United States of America) inflatable penile prostheses feature an inner Dacron layer that facilitates controlled expansion. Over time, outer silicone rupture exposes the Dacron, allowing for tissue ingrowth and complicating explantation. We herein describe our experience managing this situation in a retrospective, single-surgeon case series. Operative reports of all AMS 700™ inflatable penile prostheses explants from 2011 to 2025 (CPT 54406, 54410, 54411, 54416, 54417) of a single surgeon at one insti

Human In Vivo Validation of Frequency-Dependent QTI.

PURPOSE: The aim of this work is to investigate if q-space trajectory imaging (QTI) waveforms can be designed to probe QTI metrics at a single centroid frequency under realistic experimental conditions for in vivo human brain imaging. METHODS: Realistic diffusion encoding waveforms based on double-rotation gradient waveform and magic-angle spinning of the q-vector with varying encoding bandwidth, alignment of encoding spectra across encodings (tuning) and across axes (spectral isotropy) were designed to investigate the accuracy of the centroid frequency approximation. QTI metrics were compute

From the perspective of prolactin: a view on obesity.

The physiological roles of prolactin extend beyond its classical functions in reproductive regulation. Emerging evidence indicates that prol … e associated with increased circulating prolactin levels, which are potentially mediated by adipose tissue expansion, enhanced aromatase-dependent estrogen production, and chronic low-grade systemic inflammation. This review aimed to provide a systematic synthesis of current evidence regarding the mechanistic links between hyperprolactinemia and obesity, with an e …

Non-invasive in vivo monitoring of transplanted human intestinal organoids using bioluminescence.

BACKGROUND: Xenotransplantation of pluripotent stem cell-derived human intestinal organoids (HIOs) into animal hosts models early human small intestinal development. However, traditional methods to assess HIO engraftment and expansion involve invasive, non-survival procedures. We developed a novel HIO platform featuring stable intestinal epithelial expression of a red-shifted luciferase, enabling sensitive, longitudinal in vivo tracking of HIO xenografts. METHODS: Human embryonic stem cell (hESC) line H9 was lentivirus-transduced with a plasmid encoding red-shifted luciferase under a ubiquiti

Toward precision hernia surgery: integrating biomarkers, genomics, quantitative imaging, and artificial intelligence in abdominal wall repair.

PURPOSE: Despite technical advances, the expansion of minimally invasive approaches and the development of novel biomaterials, recurrence and complications following abdominal wall hernia (AWH) repair remain frequent. This variability likely reflects anatomical and technical factors and host biological heterogeneity. Precision herniology is emerging as a translational paradigm integrating biomarkers, genetics, quantitative imaging, and artificial intelligence to refine risk stratification and therapeutic planning. METHODS: We conducted a critical narrative review with translational scope. Pub

Endurance exercise remodels pulmonary vein sleeve myocytes and promotes a proarrhythmic atrial substrate.

BACKGROUND AND AIMS: The risk of atrial fibrillation (AF) is higher in endurance athletes. Pulmonary vein isolation (PVI) is effective in this group, implicating pulmonary vein (PV) remodelling, but underlying mechanisms are unclear. This study investigated if endurance training remodels PV sleeves and the PV-left atrial (LA) junction to promote PV triggers and a permissive peri-antral substrate for AF. METHODS: In canine and murine endurance-running models, in vivo PV-LA mapping, ex vivo PV electrophysiology, intracellular action potential (AP) profiling with machine learning classification,

Infants who develop bronchopulmonary dysplasia have an airway endotype defined by vimentin expression and ciliary loss.

RATIONALE: Bronchopulmonary dysplasia (BPD) arises from disrupted lung development after preterm birth and produces structural deficits at every level of the respiratory tree. Lower airway disease is emerging as a clinically significant BPD phenotype with increased mortality, yet the molecular mechanisms whereby preterm birth disrupts airway development remain poorly defined. OBJECTIVES: To develop a human model of lower airway disease following preterm birth and to define a molecular endotype of evolving BPD (eBPD) at baseline and in response to injury. METHODS: An ex vivo organotypic airwa

Intestinal epithelial peroxisome proliferator-activated receptor γ deficiency exacerbates insulin resistance in mice with metabolic dysfunction-associated steatohepatitis.

Peroxisome proliferator-activated receptor γ (PPARγ) is highly expressed in the intestinal epithelium and protects against intestinal inflammation. Intestinal inflammation and bacterial translocation contribute to the progression of metabolic dysfunction-associated steatohepatitis (MASH). We aimed to investigate the role of intestinal PPARγ in diet-induced MASH. Mice with intestinal epithelial cell-specific PPARγ deficiency (PPARγΔIEC) were generated using a villin-Cre transgene and floxed Pparg allele. Littermate mice carrying only the floxed Pparg allele (PPARγf/f), served as controls. MASH

Neurovascular Signaling in the Perivascular Space: Biophysical Principles and Disruption in Hypertension.

The brain's vasculature-from pial arteries through penetrating arterioles and the capillary bed to venous drainage-is enveloped by a narrow, compositionally elaborate perivascular space that provides the primary signaling interface between neural tissue and blood vessels. Here, we advance the principle that the geometry and composition of this space are not constant but change systematically along the vascular tree, and that perivascular space composition at these locations imposes biophysical constraints that dictate the fidelity and gain of neurovascular signaling. We argue that its narrow m

Cardiac drug potential: exploring individual and combined cardiac drugs to promote differentiation of adipose-derived stem cells into cardiomyocytes.

BACKGROUND: Stem cell–based cardiac regenerative medicine holds significant promise for regenerating damaged cardiomyocyte by cardiomyocyte differentiation. Among stem cell types, adipose-derived stem cells (ADSCs) are the most extensively studied due to their ease of isolation and expansion, yet current differentiation protocols remain inefficient and poorly reproducible. Given the well-established roles of certain cardiac drugs—such as Salbutamol, Ivabradine, and Entresto—their potential influence on the regulation of cardiomyocyte differentiation may provide a valuable foundation for advanc

Microbubble-Infused Hydrogel Scaffolds With Tunable Porosity for Regenerative-Medicine Applications.

Tissue-engineering scaffolds require interconnected porous networks to support cell infiltration, nutrient diffusion, and waste removal. Conventional methods to introduce porosity-such as particulate leaching, gas foaming, and freeze-drying-can leave cytotoxic residues. We propose a scalable, cytocompatible approach to tune hydrogel porosity using lipid-shelled gas microbubbles as a transient porogen. In this study, we demonstrate that lipid-shelled microbubbles can be incorporated into alginate, poly(ethylene glycol) diacrylate (PEGDA), or gelatin methacrylate (GelMA) precursors, and subseque

Effects of anti-RANKL, Zoledronate or combination therapy in a mouse model of fibrous dysplasia: a preclinical study.

Bone fragility and pain are major clinical issues in fibrous dysplasia (FD) of bone, a genetic disorder characterized by increased bone resorption and lytic lesions. Both bisphosphonates (BPs) and denosumab are currently used to treat FD patients, although important concerns remain unsolved. BPs downregulate bone remodeling but their effects on FD lesions and pain are variable. Conversely, denosumab converts FD tissue into mineralized bone and prevents disease progression, but disease rebound occurs upon treatment withdrawal. The combination of these two drugs may represent an effective and sa

Two-step Xenogeneic Dermal Matrix-assisted Autologous Skin Grafting for Large Scalp Cutaneous Squamous Cell Carcinoma Ulcer.

Cutaneous squamous cell carcinoma (CSCC) of the scalp poses major reconstructive challenges, especially in older patients with poor surgical tolerance. Large postexcisional defects often require complex reconstruction, whereas free flaps and tissue expansion may be unsafe in high-risk individuals. We present a cost-effective, 2-stage approach using xenogeneic acellular dermal matrix (XADM) followed by autologous split-thickness skin grafting. The graft survived completely. Both scalp and donor site healed without infection or wound complications. Frozen sections confirmed negative margins, and…

Temporal and Spatial Gene Expression Dynamics in Neonatal HI Hippocampus with Focus on Arginase.

BACKGROUND: Hypoxic-ischemic (HI) brain injury triggers a dynamic, multi-phase response involving early microglial efferocytosis followed by extracellular matrix (ECM) deposition and scar formation. Arginase-1 (ARG1), a key enzyme in tissue repair, is implicated in both processes, yet its role in neonatal microglia remains poorly defined. We characterize ARG1-linked pathways in neonatal microglia, identifying distinct efferocytic and fibrotic phases post-HI. METHODS: HI was induced in P9 mice using the Vannucci model, and brains were collected at 24 h (D1) and 5 days (D5). Spatially resolved

Quantitative analysis of growth and diversification in venom data using database metrics.

Animal venomics is a growing field of research with evolutionary and biotechnological significance. Yet, fundamental questions regarding the origin, diversification, and bioactivity of venoms remain unresolved. Here, we analysed venom tissue-related data curated in Tox-Prot, currently the most comprehensive database for animal venoms, across three snapshots spanning two decades (2005, 2015, 2025). We assessed the taxonomic landscape related to Tox-Prot entries, sequence length distribution, protein family abundances, and habitat-specific venom patterns. Our results consistently show that snake

Decoding Low-Density Neutrophils in Breast and Lung Cancer: From Bystanders to Drivers.

Neutrophils, once viewed as short-lived and functionally uniform antimicrobial cells, are now recognized as highly plastic immune regulators with context-dependent roles in cancer. Among circulating neutrophil subsets, low-density neutrophils (LDNs) have emerged as a clinically relevant population in oncology due to their distinct phenotypic, potent immunosuppressive activity, and strong association with disease progression. LDNs are rare in healthy individuals but markedly expanded in patients with solid tumors. LDNs display enhanced activation, increased expression of immunoregulatory molecu

Quantification of SARM1 NADase Activity in Human Peripheral Blood Mononuclear Cells.

SARM1 (sterile α and TIR motif-containing protein-1) is an NADase enzyme that serves as the central executioner of Wallerian axon degeneration. Given this, SARM1 is of high interest as a candidate therapeutic target, and SARM1 inhibitors are currently in clinical trials for treatment of neurodegeneration. Beyond neuroscience, emerging studies reveal that SARM1 may also drive aspects of bone fragility, liver pathology, adipose expansion, and insulin resistance in metabolic disease. However, we lack methods to quantify SARM1 activation in humans to better define patients at high risk of SARM1-me

Intracellular Ca2+ balances the anti-inflammatory potential of IL-33-activated mast cells (MCs).

IL-33 is an alarmine released upon tissue injury. Mast cells (MCs) highly express the IL-33 receptor T1/ST2 (IL-33R) and are thus highly susceptible to released IL-33. Activation of the IL-33R on MCs induces the MyD88-dependent activation of the TAK1-IKK2-p65/RelA signaling pathway, ERK1/2 and p38 which together mediate the production of IL-6, IL-13 and IL-2. Whereas MC-derived IL-6 and IL-13 act as pro-inflammatory cytokines, MC-derived IL-2 acts as an anti-inflammatory cytokine by inducing Treg cell expansion in vitro and in vivo. The mechanism, which mediate this pleiotropic nature within I
