Molecular Identification of a Novel Species, Babesia panickeri sp. nov., from a Naturally Infected Domestic Cat of India and Its Comparison with Canine Babesia Isolates.

INTRODUCTION: Babesiosis is a tick-borne hemo-parasitic disease of domestic and wild animals. Parasites causing babesiosis are considered to infect only specific hosts but some sporadic reports in recent past are in strong disagreement with their host specificity. This is the first report of a domestic cat being naturally infected with a novel Babesia sp. in India. METHODS: Blood samples collected from dogs (n = 6) and a 3-month-old cat, with clinical symptoms of babesiosis, were submitted to two different laboratories for hematology analysis, light microscopical examination, and molecular co…

Pathophysiology 1. Mechanisms of Thrombosis in Cancer Patients.

Thrombosis is a major cause of morbidity and mortality in cancer patients. The pathogenesis of blood coagulation activation in oncological patients is complex and involves both clinical and biological factors. Abnormalities in one or more coagulation test are common in cancer patients, even without thrombotic manifestations, indicating an ongoing hypercoagulable condition. Moreover, venous thromboembolism (VTE) can be the first symptom of an occult malignancy in an otherwise healthy individual. The levels of laboratory markers of activation of blood coagulation parallel the development of mali

Rapid induction of inflammatory lipid mediators by the inflammasome in vivo.

Detection of microbial products by host inflammasomes is an important mechanism of innate immune surveillance. Inflammasomes activate the caspase-1 (CASP1) protease, which processes the cytokines interleukin (IL)-1β and IL-18, and initiates a lytic host cell death called pyroptosis. To identify novel CASP1 functions in vivo, we devised a strategy for cytosolic delivery of bacterial flagellin, a specific ligand for the NAIP5 (NLR family, apoptosis inhibitory protein 5)/NLRC4 (NLR family, CARD-domain-containing 4) inflammasome. Here we show that systemic inflammasome activation by flagellin lead

HIV-specific CD8+ T cells from HIV+ individuals receiving HAART can be expanded ex vivo to augment systemic and mucosal immunity in vivo.

Most HIV+ individuals require lifelong highly active antiretroviral therapy (HAART) to suppress HIV replication, but fail to eliminate the virus in part because of residual replication in gut-associated lymphoid tissues (GALT). Naturally elicited HIV-specific CD8+ T cells generated in the acute and chronic infectious phases exhibit antiviral activity, but decrease in number after HAART. Therapeutic vaccines represent a potential strategy to expand cellular responses, although previous efforts have been largely unsuccessful, conceivably because of a lack of responding HIV-specific central-memor

Rodent host cell/Lassa virus interactions: evolution and expression of α-Dystroglycan, LARGE-1 and LARGE-2 genes, with special emphasis on the Mastomys genus.

Arenaviruses are usually rodent-borne viruses that constitute a major threat for human health. Among them, Lassa Fever Virus (LFV) occurs in Western Africa where it infects hundreds of thousands of people annually. According to the most recent surveys, LFV is hosted by one of the multimammate rats, Mastomys natalensis, but has never been detected in its sibling and sometimes sympatric species Mastomys erythroleucus. This pattern suggests that intrinsic, i.e. genetic properties underlie such a drastic epidemiological difference (M. natalensis as a reservoir vs. M. erythroleucus as a non-reservo

Suppression of acute anti-friend virus CD8+ T-cell responses by coinfection with lactate dehydrogenase-elevating virus.

Friend virus (FV) and lactate dehydrogenase-elevating virus (LDV) are endemic mouse viruses that can cause long-term chronic infections in mice. We found that numerous mouse-passaged FV isolates also contained LDV and that coinfection with LDV delayed FV-specific CD8(+) T-cell responses during acute infection. While LDV did not alter the type of acute pathology induced by FV, which was severe splenomegaly caused by erythroproliferation, the immunosuppression mediated by LDV increased both the severity and the duration of FV infection. Compared to mice infected with FV alone, those coinfected w

Plasmodium falciparum variant surface antigen expression varies between isolates causing severe and nonsevere malaria and is modified by acquired immunity.

In areas of endemic parasite transmission, protective immunity to Plasmodium falciparum malaria is acquired over several years with numerous disease episodes. Acquisition of Abs to parasite-encoded variant surface Ags (VSA) on the infected erythrocyte membrane is important in the development of immunity, as disease-causing parasites appear to be those not controlled by preexisting VSA-specific Abs. In this work we report that VSA expressed by parasites from young Ghanaian children with P. falciparum malaria were commonly and strongly recognized by plasma Abs from healthy children in the same a

Enhanced antigen-specific delayed-type hypersensitivity and immunoglobulin G2b responses after oral administration of viable Lactobacillus casei YIT9029 in Wistar and Brown Norway rats.

In this study, the effects of orally administered viable Lactobacillus casei Shirota strain YIT9029 on the immunity parameters of Wistar and Brown Norway rats were examined. For this purpose, we used the Trichinella spiralis host resistance model. Two weeks before and during T. spiralis infection, rats were fed 10(9) viable L. casei bacteria 5 days per week. The T. spiralis-specific delayed-type hypersensitivity (DTH) response was significantly enhanced in both Wistar and Brown Norway rats given L. casei. In both rat strains fed L. casei, serum T. spiralis-specific immunoglobulin G2b (IgG2b) c

Localized expression of mRNA for phagocyte-specific chemotactic cytokines in human periodontal infections.

In bacterial infections, mononuclear and polymorphonuclear phagocytes are key components of host defenses. Recent investigations have indicated that chemokines are able to recruit and activate phagocytes. In particular, interleukin-8 (IL-8) attracts polymorphonuclear leukocytes (PMNs), while monocyte chemoattractant protein-1 (MCP-1) is selective for cells of the monocyte/macrophage lineage. In this investigation, we analyzed the in situ expression of IL-8 and MCP-1 mRNAs in human periodontal infections. Specific mRNA was detected by in situ hybridization using 35S-labeled riboprobes in frozen

Cell Therapy Transplant Canada (CTTC) Consensus-Based Review for the Use of Supportive Care and Topical Therapies for Chronic Graft-Versus-Host Disease.

This is a consensus-based Canadian review with the primary purpose of expanding knowledge regarding supportive and topical therapies for chronic graft vs. host disease (cGvHD). There is often a lack of confidence in treating certain organ specific manifestations, and access to specialists with organ specific expertise varies across the country. This paper covers information gained from a literature review and Canadian transplanter expertise and will serve as a guide for hematopoietic cell transplant healthcare providers to topical cGvHD therapies accessible in Canada. We will review common sym

T cell responses in hepatitis C virus infection: historical overview and goals for future research.

Hepatitis C virus (HCV)-specific T cells are key factors in the outcome of acute HCV infection and in protective immunity. This review recapitulates the steps that immunologists have taken in the past 25years to dissect the role of T cell responses in HCV infection. It describes technical as well as disease-specific challenges that were caused by the inapparent onset of acute HCV infection, the difficulty to identify subjects who spontaneously clear HCV infection, the low frequency of HCV-specific T cells in the blood of chronically infected patients, and the lack of small animal models with i

Heterotopic transplantation as a model to study the regulation of spermatogenesis; some histomorphological considerations about sperm decline in man.

A novel animal model (syngeneic neonatal testicular graft transplanted under the skin of the outer ear in adult inbred Fischer rats that had been castrated, hypophysectomised, and/or subjected to hormonal replacement therapy) was developed to study regulation of spermatogenesis and Sertoli cell number. Given that Sertoli cell number and testicular size are important in determining spermatozoan production rates, this model was first used to study Sertoli cell proliferation, testicular size, and establishment of germ cells. The specific objectives were to determine the developmental pattern of S

Selective autophagy: xenophagy.

Xenophagy is an autophagic phenomenon that specifically involves pathogens and other non-host entities. Although the understanding of the relationship between autophagosomes and invading organisms has grown significantly in the past decade, the exact steps to confirm xenophagy has been not been thoroughly defined. Here we describe a methodical approach to confirming autophagy, its interaction with bacterial invasion, as well as the specific type of autophagic formation (i.e. autophagosome, autolysosome, phagolysosome). Further, we argue that xenophagy is not limited to pathogen interaction wit

Rosacea: current thoughts on origin.

Rosacea is a clinical pattern beginning and evolving in the genetically susceptible individual in response to a host of exposures. It produces a variety of clinical presentations, which vary over time and with age. Recently, many specific mediators of rosacea development have been described. A primary genetic cause for rosacea is suggested as single genes often control such mediators: enzymes, neuroendocrine transmitters, and cytokines are found in pathways to rosacea signs and symptoms. Currently, neither a specific cause nor a laboratory indicator of rosacea has been suggested. However, broa

Exometabolome variation in the fungal pathogen of humans Candida albicans reveals specificities at the genetic clade and strain levels.

Candida albicans is a fungal pathobiont that thrives in different host niches and successfully confronts the host immune defenses. The success of C. albicans as a pathogen relies on a highly plastic metabolism, with examples of variations across isolates; however, detailed characterization of how C. albicans genetic diversity impacts the content of the metabolome is still missing. Here, we have implemented an NMR (nuclear magnetic resonance)-based high-throughput workflow to probe exometabolome variation in in vitro cultures of 96 clinical isolates spread across several genetic clusters (clade

Succession of the yellow tang (Zebrasoma flavescens) microbiome from hatch to settlement in a recirculating aquaculture system.

Ornamental fish aquaculture is receiving increasing scientific attention as a sustainable means of meeting demand for reef species from the aquarium trade. However, low juvenile recruitment in an aquaculture environment remains a key industry bottleneck for many high-demand pelagic-spawning species. Here, the microbiome of a yellow tang (Zebrasoma flavescens) larval cohort from hatch to the juvenile stage (76 days post-hatch) in a recirculating aquaculture system was investigated using a high throughput 16S rRNA gene amplicon sequencing approach. The taxonomic composition of microorganisms was

Exchanging the stem-loop A promoter of Zika virus with the corresponding region of tick-borne encephalitis virus generates a chimeric virus that replicates in vertebrate and mosquito cells.

The 5' end of the orthoflavivirus genome contains a stem-loop structure, known as stem-loop A (SLA), which interacts with the viral polymerase (the NS5 protein) and functions as the promoter during RNA replication. Previously, we replaced the SLA of Zika virus (ZIKV; a mosquito/vertebrate orthoflavivirus) with the corresponding region of Long Pine Key virus (a dual-host-associated insect-specific orthoflavivirus), producing a chimeric virus capable of replicating in both mosquito and vertebrate cells. Here, we investigated whether additional chimeric viruses could be created by replacing the S

Intravaginal Chlamydia trachomatis Challenge Infection Elicits TH1 and TH17 Immune Responses in Mice That Promote Pathogen Clearance and Genital Tract Damage.

While ascension of Chlamydia trachomatis into the upper genital tract of women can cause pelvic inflammatory disease and Fallopian tube damage, most infections elicit no symptoms or overt upper genital tract pathology. Consistent with this asymptomatic clinical presentation, genital C. trachomatis infection of women generates robust TH2 immunity. As an animal model that modeled this response would be invaluable for delineating bacterial pathogenesis and human host defenses, herein we explored if pathogen-specific TH2 immunity is similarly elicited by intravaginal (ivag) infection of mice with

Combining CD19 redirection and alloanergization to generate tumor-specific human T cells for allogeneic cell therapy of B-cell malignancies.

Allogeneic hematopoietic stem-cell transplantation can cure some patients with high-risk B-cell malignancies, but disease relapse following transplantation remains a significant problem. One approach that could be used to augment the donor T-cell-mediated antitumor effect is the infusion of allogeneic donor-derived T cells expressing a chimeric antibody receptor (CAR) specific to the B-cell antigen CD19. However, the use of such cells might result in toxicity in the form of graft-versus-host disease mediated by CD19-specific (CD19-CAR) T cells possessing alloreactive endogenous T-cell receptor

Candida albicans Oropharyngeal Infection Is an Exception to Iron-Based Nutritional Immunity.

Candida albicans is a commensal of the human gastrointestinal tract and a common cause of human fungal disease, including mucosal infections, such as oropharyngeal candidiasis and disseminated infections of the bloodstream and deep organs. We directly compared the in vivo transcriptional profile of C. albicans during oral infection and disseminated infection of the kidney to identify niche specific features. Overall, 97 genes were differentially expressed between the 2 infection sites. Virulence-associated genes, such as hyphae-specific transcripts, were expressed similarly in the 2 sites. Gen
