The most commonly used oral antidiabetic drug, metformin, is a substrate of the hepatic uptake transporter OCT1 (gene name SLC22A1). However, OCT1 deficiency leads to more pronounced reductions of metformin concentrations in mouse than in human liver. Similarly, the effects of OCT1 deficiency on the pharmacokinetics of thiamine were reported to differ between human and mouse. Here, we compared the uptake characteristics of metformin and thiamine between human and mouse OCT1 using stably transfected human embryonic kidney 293 cells. The affinity for metformin was 4.9-fold lower in human than in

BACKGROUND: Metal implants can produce heat and damage adjacent tissues under microwave irradiation, which makes local metal implants in the body a contraindication for microwave therapy. However, with the wide application of titanium alloy implants which have low permeability and low conductivity, this concept has been challenged. Our team members have confirmed through previous research that continuous low-power microwave irradiation does not cause thermal damage to the surrounding tissues of the titanium alloy. Is there any other way to further increase the dose of microwave irradiation whi

It has been known for decades that pigs, as well as some other mammals, possess a peculiar lymph node architecture commonly referred to as "inverted". Although some direct consequences of this structure have been described, such as the altered circulation pattern of mature lymphocytes, the overall impact of this inversion on the induction and maturation of the adaptive immune response has never been explored. Here, we tentatively propose a series of hypotheses that could be experimentally tested. If confirmed, these hypotheses might prompt a reconsideration of conventional vaccination strategi

Several authors have explored the use of hypnosis in addressing female issues related to eating disorders, anorexia, and bulimia, and in particular those related to the specificity of the female body connected to bones, blood and hormones at different stages of life: from menarche to menopause, through pregnancy, childbirth, and breastfeeding, thus linked to the specificity of the female body as a reproductive function of the human species. This article goes beyond menopause and aims at exploring the contribution of hypnosis in addressing the theme of aging as a phase of life that, despite bri

Fusobacterium nucleatum is a Gram-negative, anaerobic bacterium that naturally colonizes the human oral cavity and is recognized as an etiological agent in both local and systemic diseases, including periodontitis, preterm birth, and colorectal cancer. To advance research into the systemic impact of oral-origin F. nucleatum-associated pathologies, it is crucial to optimize the use of animal models. In this study, we present a detailed protocol for establishing a mouse model of experimental periodontitis through oral inoculation of bacteria, including F. nucleatum. This model provides a valuabl

The detection of platelet-specific antibodies plays an important role in the clinical diagnosis of primary immune thrombocytopenia (ITP). MAIPA assay and PakAuto assay are the two main methods for this purpose. However, a comprehensive comparative evaluation of these techniques in clinical practice remains lacking. In this study, we collected 48 patients with thrombocytopenia. MAIPA and PakAuto are used to examine platelet-specific antibodies synchronously. Based on methodologies and analysis of experimental results, we conclude that PakAuto assay with a high sensitivity and accuracy can off

Ischemic heart disease (IHD) remains a leading contributor to cardiovascular disease (CVD) worldwide. Despite advances in diagnostic and therapeutic approaches, translational research demands robust large animal models to bridge the gap between experimental interventions and clinical application. Among these, porcine models have gained prominence due to their anatomical, physiological, immunological, and genomic similarities to humans. This review provides a comprehensive overview of current methodologies for establishing porcine IHD models, critically assesses emerging rehabilitative strategi

There is growing evidence that microbial dysbiosis is intimately related to carcinogenesis across several types of human cancer. Neisseria gonorrhoeae is best known for causing acute exudative genitourinary infection in males. N. gonorrhoeae can also cause chronic, asymptomatic infection of the female genitourinary tract along with the oropharynx and rectum of both sexes. Epidemiological studies suggest that N. gonorrhoeae is an independent risk factor for cancer of the anus, bladder, cervix, prostate, and oropharynx. It is not clear however if this association is causal. The purpose of this r

In this chapter, indirect enzyme-linked immunosorbent assays (ELISAs) to quantitatively measure autoantibodies directed to human natural and homocysteinylated alpha-1 antitrypsin (anti-AATA and anti-HAATA, respectively) in serum are described. The illustrated ELISA protocols are slightly different, since the two protein forms have different biochemical features and, consequently, different affinity for the matrix (polystyrene microplate wells), so that specific experimental conditions have to be performed for the quantification of the serum antibody recognition. These procedures can be carried

MicroRNA (miRNA) is a small, non-coding RNA molecule (~22 nucleotides) that acts as a post-transcriptional gene regulator, primarily by inhibiting the translation of target mRNA transcripts or affecting cell mRNA stability. Since miRNAs are comprehensively involved in gene regulation, their abnormalities are associated with various human diseases, including cardiovascular disease. Additionally, targeted inhibition of disease-related miRNAs and their targets should have therapeutic potential. Therefore, this chapter describes the experimental steps for targeted inhibition of specific miRNAs usi

Brucellosis is a highly contagious bacterial disease affecting a wide range of animals, as well as humans. The existence of the clinically diagnosed brucellosis in avian species is controversially discussed. In the current study, we set to summarize the current knowledge on the presence of brucellae in avian species. Anti-Brucella antibodies were monitored in different avian species using classical diagnostic tools. Experimental infection of chicken embryos induced the disease and resulted in the development of specific lesions. Few empirical studies have been performed in adult poultry. Howev

Toxoplasma gondii is an obligate intracellular parasite belonging to the phylum Apicomplexa that infects all warm-blooded animals, including humans. T. gondii can replicate in every nucleated host cell by orchestrating metabolic interactions to derive crucial nutrients. In this review, we summarize the current status of known metabolic interactions of T. gondii with its host cell and discuss open questions and promising experimental approaches that will allow further dissection of the host-parasite interface and discovery of ways to efficiently target both tachyzoite and bradyzoite forms of T.

Gelsolin is the fourth most abundant protein in the body and its depletion in the blood has been found in multiple sclerosis (MS) patients. How gelsolin affects the MS brain has not been studied. We found that while the secreted form of gelsolin (pGSN) decreased in the blood of experimental autoimmune encephalomyelitis (EAE) mice, pGSN concentration increased in the EAE brain. Recombinant human pGSN (rhp-GSN) decreased extracellular actin and myeloperoxidase activity in the brain, resulting in reduced disease activity and less severe clinical disease, suggesting that gelsolin could be a potent

Toll-like receptor (TLR) signalling is involved in first-line defence against Leishmania parasites by triggering NF-κB activation and downstream production of proinflammatory cytokines. Experimental models of visceral leishmaniasis (VL) support a protective role for TLRs 2, 4 and 9 in host immune responses to Leishmania infection. There are limited data available on expression of these TLRs in human VL, particularly in sites of infection, such as the spleen. This study aimed to determine whether the expression of mRNA encoding the expression of TLRs 2, 4 and 9 was altered in VL and compare exp

Urogenital schistosomiasis is one of the greatest single infectious sources of human morbidity and mortality known. Through a complex cycle of infection, migration and eventual maturation and mating, S. haematobium (the aetiological agent of urogenital schistosomiasis) deposits highly immunogenic eggs within the bladder and other pelvic organs, activating a wide range of immune programs that determine both infection outcome as well as downstream immunopathology. In this review, we discuss the experimental and observational bases for our current understanding of these immune programs, focusing

Acute respiratory distress syndrome (ARDS) is a common critical pulmonary complication after esophagectomy and other thoracic surgeries (e.g., lung transplantation, pulmonary thromboendarterectomy). Direct pulmonary ischemia-reperfusion injury (PIRI) is known to play the main role in induction of ARDS in these cases. Large animal models are an appropriate choice for ARDS as well as PIRI study because of their physiological and anatomic similarities to the human body. With regard to large animal models, we reviewed different methods of inducing in situ direct PIRI and the commonly applied metho

A hyperspectral imaging system to measure and analyze the reflectance spectra of the human tongue with high spatial resolution is proposed for tongue tumor detection. To achieve fast and accurate performance for detecting tongue tumors, reflectance data were collected using spectral acousto-optic tunable filters and a spectral adapter, and sparse representation was used for the data analysis algorithm. Based on the tumor image database, a recognition rate of 96.5% was achieved. The experimental results show that hyperspectral imaging for tongue tumor diagnosis, together with the spectroscopic

Animal models provide the opportunity for in-depth and experimental investigation into the anatomical and physiological underpinnings of human neurological disorders. Rodent models of thiamine deficiency have yielded significant insight into the structural, neurochemical and cognitive deficits associated with thiamine deficiency as well as proven useful toward greater understanding of memory function in the intact brain. In this review, we discuss the anatomical, neurochemical and behavioral changes that occur during the acute and chronic phases of thiamine deficiency and describe how rodent m

The failure of closure of the neural tube during development leads to malformations called neural tube defects (NTDs). The most common neural malformations in humans include anencephaly, encephalocele, exencephaly, craniorachischisis spina bifida with or without myelomeningocele, lipomyeloschisis, lipomyelomeningocele, meningocele and myelocystocele. Current preventive strategies are mainly based on pharmacologic/folic acid supplementation. However, stem cell-based and other combination approaches may emerge as potential treatment options for NTDs. This review provides an account of experiment
