Evaluation of drug-drug interactions of traditional Japanese medicines (Hochuekkito, Goreisan, and Ninjin'yoeito) in CYP3A-humanised mice.

1. Traditional Japanese medicines (Kampo medicines) are increasingly used concurrently with other medications, but evidence regarding their potential for drug-drug interactions (DDIs) is insufficient. Given the important role of cytochrome P450 3 A (CYP3A) in DDIs, this study assessed the CYP3A-mediated DDI risk of three Kampo medicines (Hochuekkito, Goreisan, and Ninjin'yoeito).2. We utilised a CYP3A-humanised mouse model to comprehensively evaluate the effects of the three Kampo medicines on CYP3A activity, including an analysis of the area under the plasma concentration-time curve (AUC) and

Schistosomiasis Morbidity Hotspots: Roles of the Human Host, the Parasite and Their Interface in the Development of Severe Morbidity.

Schistosomiasis is the second most important human parasitic disease in terms of socioeconomic impact, causing great morbidity and mortality, predominantly across the African continent. For intestinal schistosomiasis, severe morbidity manifests as periportal fibrosis (PPF) in which large tracts of macro-fibrosis of the liver, visible by ultrasound, can occlude the main portal vein leading to portal hypertension (PHT), sequelae such as ascites and collateral vasculature, and ultimately fatalities. For urogenital schistosomiasis, severe morbidity manifests as pathology throughout the urinary sys

MicroRNA-33a-5p Modulates Japanese Encephalitis Virus Replication by Targeting Eukaryotic Translation Elongation Factor 1A1.

UNLABELLED: Japanese encephalitis virus (JEV) is a typical mosquito-borne flavivirus responsible for acute encephalitis and meningitis in humans. However, the molecular mechanism for JEV pathogenesis is still unclear. MicroRNAs (miRNAs) are small noncoding RNAs that act as gene regulators. They are directly or indirectly involved in many cellular functions owing to their ability to target mRNAs for degradation or translational repression. However, how cellular miRNAs are regulated and their functions during JEV infection are largely unknown. In the present study, we found that JEV infection do

Identification of candidate host proteins that interact with LipL32, the major outer membrane protein of pathogenic Leptospira, by random phage display peptide library.

Leptospirosis is a worldwide zoonotic disease caused by pathogenic Leptospira spp. Rodent species are the major reservoir hosts that can excrete leptospires in their urine leading to environmental contamination. After gaining entry into the host via skin breaks and mucosa, leptospires disseminate through the bloodstream to target organs causing a wide range of disease manifestations in susceptible mammalian hosts. The crucial step of infection requires host-pathogen interactions. LipL32, the major outer membrane protein (OMP) of pathogenic Leptospira, is conserved among pathogenic leptospires,

Demystifying the impact of prenatal tobacco exposure on the placental immune microenvironment: Avoiding the tragedy of mending the fold after death.

Prenatal tobacco exposure (PTE) correlates significantly with a surge in adverse pregnancy outcomes, yet its pathological mechanisms remain partially unexplored. This study aims to meticulously examine the repercussions of PTE on placental immune landscapes, employing a coordinated research methodology encompassing bioinformatics, machine learning and animal studies. Concurrently, it aims to screen biomarkers and potential compounds that could sensitively indicate and mitigate placental immune disorders. In the course of this research, two gene expression omnibus (GEO) microarrays, namely GSE2

The effects of metals as endocrine disruptors.

This review reports current knowledge regarding the roles that cadmium (Cd), mercury (Hg), arsenic (As), lead (PB), manganese (Mn), and zinc (Zn) play as endocrine-disrupting chemicals (EDCs). The influence of these metals on the endocrine system, possible mechanisms of action, and consequent health effects were correlated between experimental animals and humans. Analysis of the studies prompted us to identify some critical issues related to this area and showed the need for more rigorous and innovative studies. Consequently, it was recommended that future studies need to: (1) identify the mec

Environmental changes and risk of plague epidemics in Indonesia.

BACKGROUND: Historical epidemiological data indicate that plague epidemics caused thousands of deaths in Indonesia between 1911 and 1956. During this period, silent phases of the disease were observed, followed by re-emergences several years or even decades later in certain regions. The Indonesian government, both at the regional and central levels (notably the Ministry of Health of the Republic of Indonesia), has undertaken decades of epidemic management efforts, including plague surveillance, medical treatments, vector control, and improvements in individual and environmental sanitation. The

MIDAS: rapid, multiplexed molecular profiling for integrated host-pathogen analysis.

Addressing the need for integrated molecular analysis in complex infectious conditions requires technologies that rapidly quantify both pathogens and host responses. This is particularly relevant in sepsis research, where existing tools typically measure either microbial or host biomarkers. Here, we present MIDAS (Multiplexed Intelligent Diffraction Analysis System), a proof-of-concept platform that integrates shape-encoded hydrogel particles with lens-free diffraction imaging and deep learning-based analysis to enable simultaneous quantification of bacterial RNA and inflammatory proteins in a

Modeling Toxoplasma gondii-gut early interactions using a human microphysiological system.

Oral transmission of parasites via environmentally resistant cyst stages in contaminated food or water is a common route of human infection, but there are no effective vaccines available for any enteric parasitic infection. Our knowledge of parasite cyst stage conversion and interaction with the intestinal tract is limited. Here, we investigate infection dynamics of Toxoplasma gondii cyst-stage in murine jejunum and human intestinal microphysiological systems. We focus on parasite ingress, replication, and conversion of the cyst stage to the rapidly replicating dissemination stage. In vivo bio

E. coli Meningitis.

Escherichia coli (E. coli) is a gram-negative organism that commonly colonizes the gastrointestinal tract and has the ability to travel hematogenously to the central nervous system, resulting in inflammation of the brain and spinal cord membranes. E. coli meningitis is a significant cause of mortality and morbidity in neonates and infants throughout the world and is less common in adults. E. coli meningitis has a high degree of bacteremia and invasion of the blood-brain barrier with specific microbial and host factors as well as microbe- and host specific signaling molecules. Of dogs and hookworms: man's best friend and his parasites as a model for translational biomedical research.

We present evidence that the dog hookworm (Ancylostoma caninum) is underutilised in the study of host-parasite interactions, particularly as a proxy for the human-hookworm relationship. The inability to passage hookworms through all life stages in vitro means that adult stage hookworms have to be harvested from the gut of their definitive hosts for ex vivo research. This makes study of the human-hookworm interface difficult for technical and ethical reasons. The historical association of humans, dogs and hookworms presents a unique triad of positive evolutionary pressure to drive the A. caninu

The role of toll-like receptors in the induction of immune responses during rabies virus infection.

The host response to infection generally begins with interactions between pathogen-associated molecular patterns common to a variety of infectious agents and reciprocal pattern-recognition receptors (PRRs) expressed by cells of the innate immune system. The innate responses triggered by these interactions contribute to the early, innate control of infection as well as the induction of pathogen-specific adaptive immunity. The outcome of infection with wild-type rabies virus is particularly dependent upon the rapid induction of innate and adaptive immune mechanisms that can prevent the virus fro

Trichinella inflammatory myopathy: host or parasite strategy?

The parasitic nematode Trichinella has a special relation with muscle, because of its unique intracellular localization in the skeletal muscle cell, completely devoted in morphology and biochemistry to become the parasite protective niche, otherwise called the nurse cell. The long-lasting muscle infection of Trichinella exhibits a strong interplay with the host immune response, mainly characterized by a Th2 phenotype. The aim of this review is to illustrate the role of the Th2 host immune response at the muscle level during trichinellosis in different experimental models, such as knock-out or

Animals as reservoir hosts of human trypanosomes.

Present knowledge on reservoir hosts of Trypanosoma rhodesiense, T. gambiense and T. brucei in Africa and T. cruzi and T. rangeli in America and experimental transmission studies of T. cruzi in mammalian hosts and in lizards is discussed. The difficulty in differentiating the African species of human trypanosomes, which appear not to be host specific, is a major obstacle to epizootiological studies.

Human endothelial cells bioactivate organic nitrates to nitric oxide: implications for the reinforcement of endothelial defence mechanisms.

Although in therapeutic use for more than a century, the mode of cellular action of organic nitrates remains incompletely understood. Despite ample experimental evidence from animal studies to show that nitrates are metabolized to NO in the vascular smooth muscle, direct demonstration of such an activity in human vascular cells is still lacking. Moreover, the role of the endothelium in modulating the pharmacodynamic action of nitrates is far from clear. We therefore aimed to investigate whether or not human endothelial cells are capable of bioactivating these drugs to NO and whether the amount

Infection-driven bladder carcinogenesis: Molecular mechanisms of pathogen-urothelial cell interactions.

Bladder cancer (BC) is one of the most common malignancies affecting the genitourinary tract. Although infectious agents are recognized as etiological factors in various cancers, their involvement in BC has traditionally been underestimated. Accumulating evidence implicates several pathogens-notably Schistosoma haematobium, Human papillomavirus (HPV), uropathogenic Escherichia coli (UPEC), and BK virus-in the initiation and progression of BC. Schistosoma haematobium promotes squamous cell carcinoma (SCC) through inducible nitric oxide synthase (iNOS)-mediated DNA damage; HPV contributes to onc

Insight into the Life Cycle of Enterovirus-A71.

Human enterovirus 71 (EV-A71), a member of the Picornaviridae family, is predominantly associated with hand, foot, and mouth disease in infants and young children. Additionally, EV-A71 can cause severe neurological complications, including aseptic meningitis, brainstem encephalitis, and fatalities. The molecular mechanisms underlying these symptoms are complex and involve the viral tissue tropism, evasion from the host immune responses, induction of the programmed cell death, and cytokine storms. This review article delves into the EV-A71 life cycle, with a particular emphasis on recent advanc

People, pigs and parasites in New Guinea: relational contexts and epidemiological possibilities.

Within Papua New Guinea the relationship people have with their pigs varies between societies. These differences arise in the earliest phase of rearing piglets and result in domestic animals whose primary attachments are to other pigs, to places or to people. For Papua New Guineans, different pig management regimes fulfill ecological and social needs. In addition, however, the ways in which pigs are raised and managed, and the presence or absence of a local population of wild pigs, have consequences for the exposure of both domestic pigs and people to parasites that they may host. Effective co
