An O-acetylated glucomannan from Bletilla striata: Structural characterization and ADP-dependent platelet-activating hemostatic activity.

Previous work demonstrated that crude Bletilla striata polysaccharides exhibited hemostatic effects against gastric bleeding through the activation of ADP pathway, whereas their key homogeneous active constituent remains structurally undefined. Herein, bioactivity-guided fractionation afforded a novel homogeneous polysaccharide BSP-60 (6271 Da), identified as the core hemostatic component. Differing markedly from all reported B. striata polysaccharide, BSP-60 consists of glucose (Glcp) and mannose (Manp) at a molar ratio of 1:1.88, featuring a linear unbranched chain of regularly alternating →

Translating mechanobiology in osteoarthritis to biomaterial-informed therapeutic strategies.

Osteoarthritis (OA) is the most prevalent degenerative joint disease worldwide, affecting approximately 250 million individuals. Mechanical imbalance serves as both its primary driving force and pathological hallmark. Mechanobiology-based interventions remain constrained by an insufficient understanding of how chondrocytes perceive and adapt to aberrant mechanical environments. Here, we review the cascading mechanisms linking mechanical imbalance to cellular dysfunction in OA and emerging therapeutic strategies targeting these mechanobiological processes. At the biomechanical level, joint mech

Cancer-associated fibroblasts in therapeutic resistance of hepatocellular carcinoma: evidence, mechanisms, and translational implications.

Hepatocellular carcinoma (HCC) remains difficult to treat because responses to tyrosine kinase inhibitors (TKIs) and immune checkpoint inhibitors (ICIs) are limited and heterogeneous. Cancer-associated fibroblasts (CAFs) can influence therapeutic responses through paracrine signaling, immune regulation, and extracellular matrix remodeling, but CAF-driven tumor progression should not be equated with therapeutic resistance. This review critically evaluates HCC-specific evidence according to defined drug exposure, treatment-related outcomes, and functional rescue. The most mechanistically resolve

Alveolar stem cells transdifferentiate to drive bronchiolar regeneration.

The lung consists of two anatomically distinct compartments: the airways and the alveoli. Although airway epithelial stem cells are known to mobilize for alveolar regeneration, whether alveolar cells can reciprocally traverse these anatomical boundaries and contribute to bronchial repair remains unclear1-4. Here we developed dual-recombinase-mediated lineage-tracing techniques in mice to demonstrate that alveolar type 2 (AT2) cells migrate into injured bronchioles and transdifferentiate into club cells and ciliated cells, thereby actively contributing to airway regeneration. Mechanistically, a

Synthetic Surrogates of Collagen-Rich Microenvironments: Integrating Modular Bioactive Fibrillar Structure and Tunable Viscoelasticity via Multifunctional Assembling Peptides.

Collagen imparts structure, viscoelasticity, and bioactivity to the extracellular matrix (ECM) with variance between organs and between healthy and diseased states. Synthetic mimics of collagen-rich tissues remain a need in applications, from biological studies to regenerative medicine, for parsing and controlling these properties. We designed multifunctional collagen mimetic peptides (mfCMPs) that self-assemble into triple helices and fibrils and contain integrin binding motifs: GFOGER, a binding site within intact collagen I, and RGD, a cryptic binding site available within denatured collage

The involvement of circular RNAs in breast cancer metastasis: Insight into EMT-related signaling pathways.

Breast cancer, a complex and heterogeneous disease marked by uncontrolled cell growth in breast tissue, presents a significant global health challenge due to its high mortality rate. Metastasis, a major driver of cancer progression and mortality, poses a significant challenge in clinical oncology. This complex process involves profound changes in cytoskeletal dynamics and interactions with the extracellular matrix, including integrin-mediated adhesion and the activity of matrix metalloproteinases (MMPs). Circular RNAs (circRNAs) are a type of endogenous non-coding RNA with a covalently closed-

Dye-Combination Micelles for Two-Photon Phototherapy.

Two-photon excitation (TPE) phototherapy provides high spatial resolution and deep-tissue penetration with minimal invasiveness. In this study, we introduce a modular and scalable approach to transform a traditional TPE imaging dye into a highly effective type-I photosensitizer (PS) through minimal chemical modification. The newly developed selenium-bridged dye demonstrates pronounced two-photon absorption, efficient ROS generation upon TPE, and strong antitumor activity both in vitro and in hypoxic in vivo tumor environments. For subcellular targeting, we conjugated organelle-specific functio

Time-dependent imprinting of gut homing receptors on lymphocytes by migratory antigen presenting cells.

Circadian rhythms synchronize many physiological and immune processes, but their role in coordinating intestinal lymphocyte trafficking remains unclear. Here, we investigated the temporal regulation of gut homing receptors, α4β7 integrin and CCR9 chemokine receptor, on lymphocyte subsets and the involvement of migratory antigen presenting cells 5r4 in this process. Using wild-type and Period 2 (Per2) knockout mice, we analyzed receptor expression on CD4+, CD8+, and B lymphocytes from mesenteric lymph nodes (MLNs) at four Zeitgeber times (ZT5, ZT11, ZT17, ZT23). We observed circadian oscillatio

Deciphering the immunocellular regulatory network in inflammatory bowel disease: from susceptibility genes to cellular effectors and toward precision therapies.

Inflammatory bowel disease (IBD) is a chronic, immune-mediated intestinal disorder driven by dysregulated immune responses in genetically susceptible individuals. Despite recent advances in treatment, more than 30% of patients either fail to respond initially or lose response over time, underscoring the need for a deeper mechanistic understanding of immunogenetic pathways and the development of individualized therapeutic strategies. We first discuss how newly identified susceptibility genes (e.g., IL23R, NOD2, BDNF, SLC) and their polymorphisms influence immune cell function and epithelial bar

A Comparison of Biological Therapies vs Traditional Immunosuppressant in the Management of Inflammatory Bowel Diseases: A Narrative Review.

Inflammatory bowel disease (IBD), encompassing Crohn's disease and ulcerative colitis, is a chronic immune-mediated condition requiring long-term management. Traditional immunosuppressants, such as corticosteroids, thiopurines, and methotrexate, have been widely used but are associated with broad immunosuppressive effects and significant adverse events. The advent of biological therapies, including tumor necrosis factor (TNF) inhibitors, integrin inhibitors, and interleukin (IL)-12/23 inhibitors, has revolutionized IBD treatment by offering targeted immune modulation. Biologics have demonstrat

UC-associated autoantibodies to αvβ6 inhibit mucosal TGFβ activation and predispose to intestinal inflammation.

Ulcerative colitis (UC) is characterized by epithelial barrier dysfunction and dysregulated mucosal immune responses; however, the mechanisms driving disease onset remain poorly defined. Autoantibodies against the epithelial-restricted integrin αvβ6 are a highly specific biomarker of UC that can precede clinical diagnosis by up to 10 years. Because αvβ6 activates TGFβ at epithelial surfaces, we hypothesized that UC-associated αvβ6 autoantibodies inhibit mucosal TGFβ activation and disrupt epithelial homeostasis. We showed that αvβ6 autoantibodies were enriched in UC and that IgG from autoantib

The clinical pathological significance of ILK and FABP4 protein expression in EHCC.

This study investigates the clinical and pathological significance of integrin-linked kinase (ILK) and fatty acid-binding protein 4 (FABP4) expression in EHCC. We utilized EnVision immunohistochemistry technology to assess the expression levels of ILK and FABP4 in 100 cases of patients with EHCC. Data analysis was conducted using SPSS version 26.0 (Chicago). The proteins ILK and FABP4 exhibited elevated expression levels in extrahepatic cholangiocarcinoma (EHCC) tumors when compared to peritumoral tissues, adenomas, and normal biliary tract tissues, with statistical significance noted (P <.05

NKG2D as a molecular link between epithelial stress and immune activation in Crohn's disease: therapeutic implications.

Crohn's disease (CD) is a chronic inflammatory bowel disorder driven by dysregulated immune responses, epithelial barrier dysfunction, and progressive tissue damage. Despite advances in biologic therapies, treatment resistance and loss of response remain common. The activating receptor NKG2D, expressed on natural killer cells and T cell subsets, recognizes stress-induced ligands (MICA, MICB, ULBP) upregulated on intestinal epithelial cells under inflammatory conditions. In CD, sustained ligand expression promotes NKG2D-dependent cytotoxicity, pro-inflammatory cytokine release (IFN-γ, IL-17), a

2-Hydroxyisobutyrylation and Phosphorylation Crosstalk Guides Metastasis Prediction and Immunotherapy in Esophageal Squamous Cell Carcinoma.

Posttranslational modification (PTM) is pivotal in cancer progression. However, the mechanisms, biological function, and clinical significance underlying PTM crosstalk are unclear. Here, we performed systematic analyses of 2-hydroxyisobutyrylation (Khib), phosphoproteomic, proteomic, and transcriptomic profiles from 60 esophageal squamous cell carcinoma (ESCC) samples, comprising matched normal, primary tumor, and metastatic lymph node tissues. The integrative analysis identified 4499 proteins co-modified by Khib and phosphorylation on distinct residues. Interestingly, while the two PTMs targe

COL8A1-positive cancer-associated fibroblasts are drivers of 5-fluorouracil resistance in colorectal cancer.

Acquired resistance to 5-fluorouracil (5-FU) curtails the survival benefit of chemotherapy in colorectal cancer (CRC). We asked whether a discrete subset of cancer-associated fibroblasts (CAFs) drives this phenotype. Multi-omic profiling of 24 CRC cohorts identified 10 collagen genes linked to poor outcome and 5-FU resistance; single-cell RNA-seq localized them to specific CAFs. Cellular communication modelling, in vitro proliferation, migration, apoptosis, and drug-sensitivity assays, together with CRISPR/Cas9 and siRNA perturbations, xenografts, and immunochemical analyses, examined the func

The VP1 protein of foot-and-mouth disease virus: Bridging pathogenicity and control strategies.

The Foot-and-mouth disease virus (FMDV) is a highly contagious pathogen affecting cloven-hoofed animals. Its major structural protein, VP1, serves as a core target that governs viral pathogenicity and the host immune response. The distinctive structure and functions of VP1 are therefore crucial for developing vaccines and effective control strategies. This review systematically examines the multifaceted roles of VP1 in the FMDV life cycle, encompassing RGD motif-mediated viral adsorption, epitope-driven immunomodulation, and mechanisms underlying viral replication and immune evasion. [Life Science Research Using Small Membered Cyclic Unsaturated Compounds].

Small-membered unsaturated molecules possess unique molecular strain, electronic structures, and photochemical properties, making them attractive platforms for the development of functional molecules in life science research. This review summarizes our efforts to exploit cyclopropenones, cyclobutenediones, and deltic guanidines as photoreactive molecules and bioactive molecular scaffolds. Cyclopropenones can generate highly reactive alkynes upon photoirradiation, enabling light-controlled reactions such as carboxy group modification through photogenerated ynamines and amino group modification

Titanium Dioxide in Biomedical and Environmental Nanotechnology: From Photocatalytic Detoxification to Targeted Therapeutics.

Titanium dioxide (TiO2) has evolved from a conventional photocatalyst into a sophisticated nano-platform that bridges environmental sustainability and biomedicine. This paper proposes a unified interfacial redox design framework that links the electronic-structure engineering of the TiO2 with the spatial control of its reactive oxygen species (ROS). In the environmental sector, we highlight advances in photocatalytic detoxification, such as the cleavage of organophosphates via Ag-modified TiO2, driven by doping and metal-support interactions. In the biomedical domain, TiO2 is framed as an acti

CD44 restricts EGFR mobility to polarize cytoskeletal signalling modules driving bleb-based migration.

Cells under high confinement migrate efficiently in low-adhesion environments by forming stable, polarized, hydrostatic pressure-driven leader blebs. Here we investigated the basis of polarized bleb morphology in metastatic melanoma cells migrating under low-adhesive and highly confined microenvironments. Using high-resolution live imaging, molecular perturbations and biosensors, we show that EGF signalling through PI3K stabilizes and maintains polarized leader blebs. EGFR and PI3K activities form a gradient within leader blebs that decreases from rear to front, promoting phosphatidylinositol

CODNet models enable real-time assessment of colorectal cancer organoids and uncover the integral role of LTBP2 in the diffuse organoid subtype.

To achieve precise classification of colorectal cancer (CRC) at the patient-derived organoids (PDOs) level, we constructed a real-time CRC PDO assessment system through artificial intelligence (AI) assisted model using an optimized object detection algorithm. We constructed the CODNetV1 model for the identification, feature extraction and drug sensitivity detection of CRC PDOs (mAP of 88.05%). And we optimized CODNetV2 to classify four morphological subtypes of CRC PDOs (mAP of 85.65%). CODNetV1 and CODNetV2 achieved real-time, automated, multi-parameter detection of CRC PDOs, directly facilit
