A Review on Curcumin: Pharmacological Promises and Biomedical Activities.

Turmeric is a common spice obtained from the rhizomes of the Curcuma longa plant. It belongs to the Zingiberaceae (ginger family). The rhizome is a horizontally growing stem that sends roots and shoots from below ground. Turmeric has many uses, including spice, food preservation, flavor enhancement, and coloring. Also, it is used in conventional medicine as a home treatment for many diseases. Turmeric obtains its yellow color from curcuminoids, polyphenolic pigments that dissolve in fat. The primary curcuminoid form in turmeric is curcumin or diferuloylmethane (1,7-bis (4-hydroxy-3-methoxyphen

Epigallocatechin-3-Gallate: A potential amyloid Fibril Disaggregator of Serum amyloid A1.

Serum amyloid A1 (SAA1) is a 122-amino acid protein that, after cleavage, matures into a 104-amino acid form. Its N-terminus is responsible for binding high-density lipoprotein (HDL), while the C-terminus maintains its structural integrity. As an acute-phase protein, SAA1 is produced by the liver in response to acute inflammation. SAA1 is also a precursor to amyloid A (AA), and its accumulation can lead to AA amyloidosis-a condition secondary to chronic inflammation that causes tissue damage and organ dysfunction. Our study explores methods to disaggregate SAA1 fibrils isolated from the cat sp

The golden touch: a comprehensive network pharmacology-guided review of synergy between curcumin and PARP inhibitors.

Cancer remains the second leading cause of death globally. Poly (ADP-ribose) polymerase inhibitors (PARPi) are effective targeted therapies, but their use is largely limited to BRCA-deficient cancers and is often hampered by resistance. This review explores combining PARPi with curcumin, a natural polyphenol, as an adjunct therapy. We reviewed literature and employed a network pharmacology approach to investigate shared molecular mechanisms. Curcumin induces DNA double-strand breaks while simultaneously impairing multiple repair pathways, including homologous recombination and non-homologous e

UGT708 glycosyltransferases: Nature's architects of C-glycosides.

Uridine Diphosphate glycosyltransferases (UGTs) catalyze the transfer of glycosyl moieties from donors to acceptors, a modification critical for plant growth, development, and metabolic homeostasis. These enzymes are ubiquitous across all life domains, playing key roles in the biosynthesis of diverse glycosides. This review focuses on the plant UGT708 family, which is uniquely characterized by its ability to form stable C-glycosidic bonds on flavonoid backbones, enhancing metabolic stability and bioactivity. We conducted a comprehensive analysis of UGT distribution and functional evolution acr

The coordination chemistry of 2,2'-bis-p-tBu-calix[4]arene.

A review of the coordination chemistry of 2,2'-bis-p-tBu-calix[4]arene (H8bisTBC[4]) reveals thirteen published structures, seven of which are homometallic complexes of manganese (×5) and copper (×2), and six which are heterometallic 3d-4f (4 × Mn-Gd, 1 × Fe-Gd and 1 × Cu-Tb). In the majority of cases the disposition of the inverted, fully deprotnated bisTBC[4] ligand favours the formation of [M4] butterflies, with two metals housed in the polyphenolic TBC[4] pockets and two encapsulated in the space between the pockets. The dominant structure directing ability of the ligand is reflected in th

Mechanism of H2S in Inhibiting the Senescence and Browning of Fresh-Cut Potatoes.

The market for fresh-cut fruits and vegetables is gradually expanding and is popular among consumers, but fresh-cut fruits and vegetables are highly susceptible to browning, causing a decrease in their quality and nutrition. Although anti-browning reagents and cryopreservation methods are often used for fresh-cut fruits and vegetables, the effects are not satisfactory. In this paper, hydrogen sulfide (H2S) donor NaHS solution was used for fumigation of fresh-cut potatoes to explore the mechanism of H2S signaling on the browning of fresh-cut potatoes at the biochemical level. Fresh-cut potatoes

Self-assembled copper chlorogenic acid nanoparticles: Inducing pyroptosis and cuproptosis to activate antitumor immunity.

Currently, the tumor treatment faces many challenges. Although chemotherapy is the most effective and economical choice, most chemotherapy drugs lack tumor specificity, have low bioavailability, and face many obstacles in clinical application. Significantly, the tumor immunosuppressive microenvironment (TIME) and low immunogenicity in solid tumors also seriously affect the clinical effect of tumor therapy. Here, metal-phenolic network nanoparticles self-assembled by copper-coordinated natural drug chlorogenic acid (ChA) (ChA-Cu NPs) are designed to amplify cell immunogenic death mediated by cu

Mitigating Oxidative Browning in Litchi by Regulating Biochemical Markers and Targeted Gene expression via Exogenous Nitric Oxide.

Nitric oxide has been shown to influence oxidative metabolism in plants, enhancing their resilience to various biotic and abiotic stresses. Post-harvest oxidative stress is a key factor leading to quality deterioration in litchi (Litchi chinensis Sonn.) fruit, with visible symptoms that significantly reduce shelf life and consumer acceptability. Therefore, the effect of exogenous sodium nitroprusside (SNP; 1.0 mM and 2.0 mM) on litchi (cv. Purbi) fruit was examined during storage at 7 ± 1°C. Different biochemical changes related to post-harvest quality and pericarp browning of litchi were eval

Lignin-Derived Oligomers as Promising mTOR Inhibitors: Insights from Dynamics Simulations.

The mammalian target of rapamycin pathway, mTOR, is a crucial signaling pathway that regulates cell growth, proliferation, metabolism, and survival. Due to its dysregulation it is involved in several ailments such as cancer or age-related diseases. The discovery of mTOR and the understanding of its biological functions were greatly facilitated by the use of rapamycin, an antibiotic of natural origin, which allosterically inhibits mTORC1, effectively blocking its function. In this entirely computational study, we investigated mTOR's interaction with seven ligands: two clinically established inh

Characterization, digestive properties and glucose metabolism regulation of curcumin-loaded Pickering emulsion.

Curcumin, a polyphenolic compound with antioxidant and hypoglycemic properties, has limited applications due to low stability and poor oral bioavailability. This study aimed to overcome these challenges by developing stable oil-in-water Pickering emulsions with curcumin and evaluating their stability and digestive characteristics. Using a T2DM mouse model, we further examined the effects of curcumin-loaded Pickering emulsions on glucose metabolism. The emulsions demonstrated high curcumin encapsulation efficiency (96.72 % ± 2.15 %) and minimal average droplet size. Both emulsions showed stable

Ultrafast ROS Scavenging Activity of Amur Maple Tree Extracts Confers Robust Cardioprotection for Myocardial Ischemia/Reperfusion Injury.

Ginnalin A (GA), a polyphenolic compound derived from amur maple trees, has been identified as a powerful scavenger of reactive oxygen species (ROS). Recognizing the pivotal role of ROS in exacerbating secondary damage during myocardial ischemia-reperfusion injury (MIRI), we fractionated GA-enriched extracts from the leaves of the amur maple tree, Acer tataricum L. subsp. ginnala (Maxim.) Wesm., using common solvents of dichloromethane (DCM) and ethyl acetate (EA). When co-administered for 30 min, the DCM- and EA-fractioned extracts effectively protected cardiomyocytes from H2O2-induced damage

Curcumin: A Potential Detoxifier Against Chemical and Natural Toxicants.

The human body gets exposed to a variety of toxins intentionally or unintentionally on a regular basis from sources such as air, water, food, and soil. Certain toxins can be synthetic, while some are biological. The toxins affect the various parts of the body by activating numerous pro-inflammatory markers, like oxidative stresses, that tend to disturb the normal function of the organs ultimately. Nowadays, people use different types of herbal treatments, viz., herbal drinks that contain different spices for detoxification of their bodies. One such example is turmeric, the most commonly availa

Bioinspired Cascade-Catalytic Scaffold to Prevent Adverse Remodeling of Heart Valve under Pathological Conditions via Conversion of •O2- into NO.

In situ tissue-engineered heart valves (TEHVs) face significant challenges in patients with compromised endogenous repair capacity, particularly those with diabetic comorbidities. To address this issue, we developed a bimetallic nanozyme-functionalized scaffold capable of restoring •O2-/NO homeostasis through a bioinspired cascade catalytic reaction that modulates the adverse repair microenvironment. The nanozyme, composed of polyphenol-coordinated iridium (Ir)/ruthenium (Ru), exhibited tunable superoxide dismutase (SOD)- and endothelial nitric oxide synthase (eNOS)-like activities by adjustin

Curcumin as an Antioxidant Against Ziprasidone Induced Lipid Peroxidation in Human Plasma: Potential Relevance to Cortico Subcortical Circuit Function.

Oxidative stress observed in schizophrenia and other psychiatric disorders can induce neuronal damage and modulate intracellular signaling, ultimately leading to neuronal death by apoptosis or necrosis. The aim of this study was to estimate in vitro the possible antioxidant properties of curcumin, the natural polyphenolic antioxidant, and its protective effects against lipid peroxidation induced by the atypical antipsychotic Ziprasidone. Curcumin (5 µg/mL, 12.5 µg/mL, 25 µg/mL, 50 µg/mL) was added to human plasma and incubated for 1 and 24 h, alone and in the presence of Ziprasidone (40 ng/mL,

Curcumin-based benzothiazepane analogues exhibit selective anti-cancer activity in HCT-116 cells via precipitated particle formation and internalisation.

Despite extensive investigation into the anti-cancer activity of the natural polyphenol curcumin, its therapeutic application is restricted by its inherent physicochemical properties. Synthetic curcumin analogues, however, offer a promising strategy to improve the drug-like potential of curcumin. In this study, we evaluated three curcumin-based benzothiazepane analogues for their ability to selectively target colon cancer cells. Their cytotoxicity was assessed on intestinal cancerous HCT-116 and non-cancerous IPEC-J2 cells using cell viability assays and microscopic imaging. Two analogues, AT0
