Limitation of Trypanosoma brucei parasitaemia results from density-dependent parasite differentiation and parasite killing by the host immune response.

In the bloodstream of its mammalian host, the "slender" form of Trypanosoma brucei replicates extracellularly, producing a parasitaemia. At high density, the level of parasitaemia is limited at a sublethal level by differentiation to the non-replicative "stumpy" form and by the host immune response. Here, we derive continuous time equations to model the time-course, cell types and level of trypanosome parasitaemia, and compare the best fits with experimental data. The best fits that were obtained favour a model in which both density-dependent trypanosome differentiation and host immune respons

CD5+ B lymphocytes are the main source of antibodies reactive with non-parasite antigens in Trypanosoma congolense-infected cattle.

Mice infected with African trypanosomes produce exceptionally large amounts of serum IgM, a major part of which binds to non-trypanosome antigens such as trinitrophenol and single-strand DNA. In this paper, we describe that in cattle infected with Trypanosoma congolense and T. vivax, similar antibodies are found, although they bind mainly to protein antigens, such as beta-galactosidase, ovalbumin and ferritin. The parasite non-specific IgM antibodies appear around the same time as the parasite-specific antibodies, but their origin and function are not clear. We tested the hypothesis that CD5+

Selective inhibition of trypanosomal glyceraldehyde-3-phosphate dehydrogenase by protein structure-based design: toward new drugs for the treatment of sleeping sickness.

Within the framework of a project aimed at rational design of drugs against diseases caused by trypanosomes and related hemoflagellate parasites, selective inhibitors of trypanosomal glycolysis were designed, synthesized, and tested. The design was based upon the crystallographically determined structures of the NAD:glyceraldehyde-3-phosphate dehydrogenase complexes of humans and Trypanosoma brucei, the causative agent of sleeping sickness. After one design cycle, using the adenosine part of the NAD cofactor as a lead, the following encouraging results were obtained: (1) a 2-methyl substitutio

Modulation of the phenotype and function of bovine afferent lymph cells during infection with Trypanosoma congolense.

Alterations in the phenotype and function of cells isolated from bovine afferent lymph were studied following tsetse-transmitted Trypanosoma congolense infection. Little alteration was observed in the output of the CD2+ T cells in the lymph, and within this population the CD4:CD8 ratio remained relatively constant. By contrast, a marked decrease was observed in the output of gamma delta T cells over the first 7 days following infection. The number of B cells increased between 2 and 6 days post-infection, and thereafter returned to pre-infection values. Little change was observed within the aff

Organization of two invariant surface glycoproteins in the surface coat of Trypanosoma brucei.

The surface coat of Trypanosoma brucei, formed by about 10(7) molecules of the membrane-form variant surface glycoprotein (mfVSG) per cell, is generally considered to constitute a barrier against the access of antibodies directed to invariant surface proteins. The recent characterization of two invariant surface glycoproteins (ISGs) with apparent molecular masses of 65 and 75 kDa (ISG65 and ISG75; 70,000 and 50,000 molecules per cell, respectively), which are both predicted to be composed of large extracellular domains, single transmembrane alpha-helices, and small intracellular domains, enabl

Independent regulation of B cell responses to surface and subsurface epitopes of African trypanosome variable surface glycoproteins.

Regulation of B cell responses to the trypanosome surface Ag was examined in H-2k compatible "responder" B10.BR and "nonresponder" C3H mice after infection with two variant clones of Trypanosoma brucei rhodesiense. Development of a selective RIA for independent detection of antibody binding to surface (exposed) and subsurface (buried) epitopes of the trypanosome variable surface glycoprotein (VSG) molecule permitted sensitive quantitation and kinetic characterization of immune responses to these epitopes. The infected B10.BR mice responded to both exposed and buried VSG epitopes of clone LouTa

Mechanisms of self-cure from Trypanosoma congolense infection in mice.

The mechanism(s) of resistance to African trypanosomiasis caused by Trypanosoma congolense was investigated by using the Dinderesso/80/CRTA/3 isolate to which C57B1/6 are resistant (low parasitemia and self-cure) and BALB/c sensitive (high parasitemia and death). The resistance of C57B1/6 is similar to that found in some natural hosts of African trypanosomes such as certain indigenous West African cattle and wild Bovidae. The antibody response to epitopes exposed on the variant surface glycoprotein of a clone obtained from the Dinderesso/80/CRTA/3 isolate was measured by a complement-mediated …

Activity of tunicamycin against Trypanosoma brucei in vitro and in vivo.

Tunicamycin, a specific inhibitor of glycoprotein biosynthesis, was evaluated for activity and chemotherapeutic potential against Trypanosoma brucei in vitro and in vivo. The inhibition of parasite incorporation of two radiolabeled macromolecular precursors, [methyl-3H]thymidine and D-[2-3H]mannose, was measured in short-term (4-h) microcultures. Mannose incorporation was inhibited over the concentration range of 0.01 to 10.0 microgram/ml, reaching 60% at the latter level. Thymidine incorporation was not affected. In vivo experiments indicated that a single dose of 2.0 mg of tunicamycin per kg

[Multidrug-resistant organisms (MDROs) in healthcare associated infections (H.A.Is): one threat above].

The use of antimicrobials has meant for humanity one of the biggest progresses in our recent History and has enhanced an improvement in livi … of attention. In this sense Paul Ehlrich (1908), father of modern chemotherapy, observed during the trypanosomiasis treatment that the pathogen emerged as a resistant variant after the use of an arsenical compound. Neuschlosz S. also describes in 1919 the resistance to quinine. But it was in the 20th century, that with the development of the specific investigatio …

Spiroplasma in Glossina fuscipes fuscipes: a potential manipulator of male reproduction.

Glossina fuscipes fuscipes (Gff) is a riverine species of the Glossina genus (tsetse flies). It is one of the principal vector species responsible for transmitting human African trypanosomiasis across a broad swath of sub-Saharan Africa. Consequently, it remains a primary target for vector-control interventions in endemic regions. As with other Glossina species, G. fuscipes houses a highly streamlined microbiota, composed of maternally transmitted endosymbionts mandatory for survival, which also significantly impacts the vectorial capacity. Among them, the obligate Wigglesworthia provides esse…

APOL1 Nephropathy: A Population Genetics and Evolutionary Medicine Detective Story.

Common DNA sequence variants rarely have a high-risk association with a common disease. When such associations do occur, evolutionary forces must be sought, such as in the association of apolipoprotein L1 (APOL1) gene risk variants with nondiabetic kidney diseases in populations of African ancestry. The variants originated in West Africa and provided pathogenic resistance in the heterozygous state that led to high allele frequencies owing to an adaptive evolutionary selective sweep. However, the homozygous state is disadvantageous and is associated with a markedly increased risk of a spectrum

We Remember… Elders' Memories and Perceptions of Sleeping Sickness Control Interventions in West Nile, Uganda.

The traditional role of African elders and their connection with the community make them important stakeholders in community-based disease control programmes. We explored elders' memories related to interventions against sleeping sickness to assess whether or not past interventions created any trauma which might hamper future control operations. Using a qualitative research framework, we conducted and analysed twenty-four in-depth interviews with Lugbara elders from north-western Uganda. Participants were selected from the villages inside and outside known historical sleeping sickness foci. El
