Paclitaxel drug-coated balloon angioplasty for de novo coronary lesions in an expanded real world clinical setting: the multicenter ALLIANCE registry.

Despite limited clinical evidence, the use of drug-coated balloons (DCB) in percutaneous coronary intervention for de novo coronary lesions is expanding and increasing. ALLIANCE is a prospective, multicenter, all-comer registry designed to evaluate the efficacy and safety of the paclitaxel-DCB in an expanded real-world clinical setting. A calculated sample size of 1500 patients was expected to provide 95% power to detect non-inferiority against a performance goal of 7.5% target lesion failure (TLF) at 1 year with a 3.0% absolute non-inferiority margin. Cases in which DCB was performed after su

Spent Coffee Ground-Derived Carbon Dots as Redox-Modulating Biomaterials for Astrocytic Differentiation of Human Neural Progenitor Cells.

Food waste-derived carbon dots (CDs) from spent coffee grounds or spinach were synthesized via hydrothermal and solvothermal methods and systematically characterized for their photoluminescence and surface chemical properties. Biological evaluations using human neural progenitor cells (hNPCs) demonstrated efficient cellular uptake and high biocompatibility across all synthesized CDs. Notably, coffee-derived blue-emitting CDs (B-CDs) exhibited robust concentration-dependent antioxidant activity and reduced intracellular reactive oxygen species (ROS) levels. In addition, B-CDs were associated wi

Mechanically robust zwitterionic hydrogel coating reinforced by microgels for antibacterial and anticoagulant applications.

Nosocomial infections and thrombosis associated with implantable medical devices have emerged as increasingly critical clinical challenges. Antifouling hydrogels, owing to their biocompatibility and highly hydrophilic surfaces, have garnered significant attention as a promising strategy to mitigate these complications. However, conventional hydrogels often suffer from poor mechanical strength due to their high-water content and the absence of efficient energy dissipation mechanisms, leading to weak adhesion to underlying substrates and potential detachment under physiological conditions. Peptide dendritic polymerization-enabled stable and tailorable nanoparticles as crosslinkers for fabricating multifunctional hydrogels with enhanced wound healing efficacy.

Developing biomedical nanomaterials that seamlessly integrate structural stability, precise modifiability, and inherent biocompatibility remains a significant challenge. Peptides are ideal candidate building blocks for constructing biocompatible nanomaterials. However, traditional peptide assembly often relies on weak non-covalent interactions, limiting their stability and functional diversity. Drawing on dendrimer design principles, we design peptide monomers capable of undergoing covalent dendritic polymerization via efficient click chemistry to generate peptide nanoparticles. These nanopart

Harmonizing the adhesion-release dilemma: a bio-based injectable hypertonic hydrogel for bladder perfusion therapy.

The high incidence of bladder diseases significantly impairs patients' quality of life, and there is an urgent need for improved local treatment strategies for superficial bladder cancer. However, conventional intravesical instillation suffers from a short drug residence time (less than 2 hours) due to urine washout and lacks adequate wet adhesion to the bladder wall. Moreover, it remains challenging to achieve controlled drug release triggered by the pathological microenvironment (e.g., weak acidity and enzyme overexpression). While existing hydrogel materials often rely on strong covalent bo

The Effect of Multifunctional Wet Adhesive Hydrogels on the Repair of Full-Thickness Oral Mucosal Defects.

Patients' quality of life is severely compromised by full-thickness oral mucosal abnormalities, and traditional treatment approaches are severely hampered by the dynamic wet environment and tissue heterogeneity. Hydrogels have demonstrated remarkable efficacy in promoting skin wound repair; however, their ability to adhere effectively and sustain functionality within the oral cavity remains inadequate. This work used hyaluronic acid (HA), 3-aminophenylboronic acid (3-APBA), tannic acid (TA), and deferoxamine (DFO) as major components to create a multifunctional hydrogel called HPTA@DFO (abbrev

Photocrosslinkable injectable glycol chitosan/gelatin biohybrid thermogel as a localized delivery platform for bone regeneration.

Injectable thermogels provide a minimally invasive strategy for localized biomaterial delivery, but insufficient mechanical stability and limited cell-adhesive properties often hinder regenerative performance. Here, we developed a photocrosslinkable injectable biohybrid thermogel by blending methacrylated glycol chitosan (MGC) with gelatin methacryloyl (GM). The MGC/GM precursor exhibited a flowable temperature window for syringe injection, followed by thermo-induced gelation at physiological temperature and UV-triggered covalent crosslinking for in situ mechanical stabilization. The photocros

Orange Peel Extract-Mediated Green Synthesis of PEG-Modified ZnO Nanoparticles: Structural Characterization, Biocompatibility, and Anticancer Efficacy.

OBJECTIVES: Engineered nanomaterials could potentially interact with biomolecules and intracellular processes, as many biological activities take place at the nanoscale level. Conventional anticancer therapies have several limitations, which warrant the introduction of alternative cancer cell killing agents. METHODS: In the present study, we have synthesized ZnO nanoparticles using orange peel extract and encapsulated them inside polyethylene glycol (PEG), a biocompatible polymer (PEG-ZnO-OP). Different characterization techniques were performed to ensure the proper synthesis of the nanoparti

A zwitterionic catalytic coating for in situ nitric oxide generation enables synergistic antithrombotic and anti-inflammatory effects for blood-contacting devices.

The use of extracorporeal life support and indwelling blood-contacting devices in critically ill patients is frequently complicated by device-associated thrombosis, a persistent clinical challenge not fully addressed by current antithrombogenic coatings. This limitation is largely attributable to the profound systemic inflammatory state inherent to critical illness, which drives a hypercoagulable milieu through mechanisms of immunothrombosis that conventional coatings fail to mitigate. To address this dual pathophysiological challenge, we designed a novel composite coating comprising a zwitter

Versatile Nano-Crosslinker Enhanced Injectable Hydrogel Toward Rapid Hemostasis and Efficient Trauma Repair.

Uncontrolled traumatic hemorrhage, often complicated by infection and poor healing, accounts for over 30% of trauma-related deaths worldwide. Injectable hydrogels with robust multi-bond crosslinked networks, integrating fluidity and in situ stability, are promising for efficient hemostasis, but their crosslinkers' relatively single structures limit the multifunctionality required for effective trauma management in resource-scarce environments. Herein, a new class of versatile ε-polylysine grafted manganese dioxide (EPL-g-MnO2) nano-crosslinkers is synthesized to construct an injectable hydroge

Decoration of Porous TiO2 Implants With Metal-Organic Composites Enables Synergistic Therapy for Osteoporotic Fracture Healing.

Osteoporotic fracture healing is trapped in a vicious cycle where multiple pathological factors, including persistent oxidative stress, inadequate vascular supply, and an imbalance bone remodeling process that favors resorption, combine to form a hostile niche for regeneration. The intricate interaction of these pathologies highlights the need to transform titanium implants from inert devices into active platforms for bone regeneration. In this study, a multifunctional coating where titanium dioxide nanotubes (TNT) are decorated with metal-organic networks (TNT@EZCA) is developed for coordinat

Temporal trends in device selection and clinical outcomes after femoropopliteal endovascular therapy: a multicenter real-world study.

Drug-coated balloons (DCBs) are increasingly used in femoropopliteal (FP) endovascular therapy (EVT) to limit permanent scaffold implantation. Although this paradigm shift has substantially altered treatment strategies, its effects on real-world clinical outcomes remain unclear. Temporal trends in device selection and clinical outcomes after FP EVT were evaluated in contemporary practice. This multicenter, retrospective study included patients who underwent EVT for de novo FP artery disease at six institutions between July 2017 and December 2023. Patients were stratified into three equal, chro

Nerve conduits with multiple physical and biochemical guiding factors for peripheral nerve defects.

Peripheral nerve injury is a common complication in patients with trauma, and severe cases can result in sensory and motor function loss, affecting the quality of life. Although the peripheral nervous system has spontaneous regenerative ability, bridging long-distance nerve gaps remains a clinical challenge. Therefore, we innovatively constructed a multi-guided nerve conduit to address this issue. Using gelatin methacryloyl as the raw material, directional micropores were created through casting, demolding, and directional freezing to achieve topographical guidance. A concentration gradient of

Development of a Laminin-511 Inspired 3D Zwitterionic Hydrogel for Human Pluripotent Stem Cell Culture.

Human pluripotent stem cells (hPSCs) hold enormous potential for regenerative medicine and disease modeling applications. These cells, however, are commonly cultured on Matrigel, a 2D coating of mouse origin and significant complexity, hindering their clinical translation. These issues have motivated the development of (a) 2D natural alternatives, which can mimic the extracellular matrix niche, but result in cytoskeletal and epigenetic changes, insufficient cell-cell and cell-matrix interactions, lineage biases upon differentiation, and challenges with scale-up due to surface area constraints

Laser patterning of ECM-derived biomaterials to direct degradation, site-specific resorption, controlled vascularization and functional repair of large nerve defects.

Controlled degradation of extracellular matrix-derived biomaterials in a site-specific and temporal sequence might facilitate early vascularization and improve tissue regeneration. In this study, we developed a tailored laser patterning treatment that successfully addresses this challenge. We show that application of a focused diode-pumped solid-state laser (532 nm) for 30 s duration leads to local heating and reduction of collagen fibril integrity in localized laser-patterned areas of a collagen biomaterial. When implanted in vivo, these thermally degraded regions then become susceptible to f

Injectable polysaccharide-based composite hydrogel promotes regeneration of critical-sized bone defects.

Critical-sized bone defects (CSDs) fail to undergo spontaneous regeneration. Conventional treatment methods, including bone grafts, as well as therapies based on growth factors and cytokines often face serious limitations, including limited availability, immune rejection, high cost, and safety concerns. Although tissue engineering using scaffolds has emerged as a promising alternative, many scaffold-based approaches still rely on the incorporation of exogenous growth factors or cytokines to achieve adequate osteoinductive performance, adding complexity, cost, and potential safety concerns to t

Silver nanoparticle-antimicrobial peptide supramolecular nanocomposite coating improves implant osseointegration under mechanical loading.

BACKGROUND: Long-term implant success depends on preventing infection and achieving initial stability, yet many antimicrobial coatings impair osseointegration, and most animal models overlook the impact of mechanical loading. To overcome the limitations of static in vivo models that do not reflect the mechanical environment of dental implants, this study developed a loaded animal model that evaluates the osteogenic properties of an Ag-GL coating under clinically relevant biomechanical forces. METHODS: Silver nanoparticles (AgNPs) were synthesized with a redox reaction, while the antimicrobial
