β-Glucan-rich Hericium erinaceus hot-water extract ameliorates acute colitis by suppressing inflammation, preserving the epithelial barrier, and modulating gut microbiota ecosystem.

Inflammatory bowel disease (IBD) is a chronic disorder marked by mucosal barrier disruption, immune imbalance, and gut dysbiosis. Polysaccharide-enriched extracts of Hericium erinaceus (HE) have shown anti-colitic effects, but the role of its β-glucan-rich water extract (HE-HW) remains unclear. By investigating a commercially relevant β-glucan-rich extract rather than isolated polysaccharides, this work addresses a key translational gap in HE research. HE-HW, rich in 1,3:1,6-β-glucan (286.4 mg/g), was evaluated using cells and mouse colitis models. In RAW 264.7 macrophages, HE-HW (1-10 μg/mL)

Epi-Berberine Alleviates Ulcerative Colitis, Protects the Intestinal Mucosal Barrier, and Inhibits Colonic Inflammation via Glutaminyl-Peptide Cyclotransferase in a Mouse Model of DSS-Induced Colitis.

The global incidence of ulcerative colitis (UC), a chronic inflammatory bowel disease, is rising. Epi-berberine (EPI), an alkaloid from Coptidis Rhizoma, shows potential for UC treatment. This study aimed to assess EPI's effect on UC and explore the mechanism underlying its UC-alleviating effect. The therapeutic effects of EPI on UC were validated in dextran sulfate sodium (DSS)-induced colitis mice and lipopolysaccharide (LPS)-induced Caco-2 inflammatory models and further investigated the underlying mechanism. In vivo, high-dose EPI (200 mg/kg) attenuated weight loss, reduced disease activit

A sulfated polysaccharide from Laminaria japonica alleviated colitis via remodeling gut microbiota and promoting butyrate-mediated barrier repair.

Ulcerative colitis (UC) is a chronic inflammatory bowel disease characterized by dysregulated immunity, gut barrier dysfunction, and microbial dysbiosis. Polysaccharides from Laminaria japonica have shown anti-inflammatory properties and microbial-modulating potential, but their precise protective mechanisms, particularly through an integrated "microbiota-metabolite-barrier-inflammation" axis, remain unclear. In this study, a sulfated polysaccharide (ALP) was extracted from L. japonica and its protective effects were evaluated in a DSS-induced colitis mice model. ALP administration significant

Activating PXR by Curcumol Ameliorates DSS-Induced Ulcerative Colitis via Transcriptional Repression of STING Signaling.

Pregnane X receptor (PXR) deficiency is implicated in ulcerative colitis (UC) pathogenesis, although its mechanistic role remains incompletely defined. Curcumol (CUR), a natural compound derived from the medicinal and culinary plant Curcuma longa L., possesses broad pharmacological activities (e.g., anti-inflammatory, antifibrotic, anticancer) as reported in the literature. However, its therapeutic potential for UC remains unreported. This study aimed to investigate whether CUR has pharmacological activity that improves UC in mice and its mechanism of action. Through our research, we demonstra

Optimization of corneal preservation media with novel antifungal agents.

The performance of ophthalmic surgeries, particularly penetrating keratoplasty, is associated with an increased risk of ocular infections, especially fungal keratitis. In addition to infections such as post-surgical occlusions, microbial contamination of the donor’s tissue, predominantly fungal in nature, can also lead to the disposal of said tissue. The genus Fusarium, along with several species from the Candida genus, emerge as the primary fungal agents, displaying notable biofilm-forming abilities on surfaces. To address these issues, the addition of antifungals to preservation media vials

Colon-targeted mucin/cyclosporine A co-loaded microspheres synergistically ameliorate ulcerative colitis via immunosuppression and mucosal barrier repair.

Ulcerative colitis (UC) is a chronic inflammatory disease of the colon and rectum, associated with epithelial barrier defects and dysregulated immune responses. Here, we developed Eudragit S100-coated mucin/CyA co-loaded calcium alginate microspheres (E-CMAMs) for synergistic UC therapy via immunosuppression and mucosal barrier repair. E-CMAMs were prepared by a high-voltage electrostatic method and an emulsification-solvent evaporation method. Their characteristics were analyzed by inverted optical microscope, SEM, FTIR, and DSC. E-CMAMs exhibited excellent pH-responsive behavior, as they rem

Larimichthys crocea isinglass polysaccharide attenuates DSS-induced colitis via gut microbiota regulation and NF-κB signaling inhibition.

This study aimed to investigate the therapeutic efficacy of Larimichthys Crocea Isinglass polysaccharide (CIP) in ameliorating DSS-induced colitis. CIP, a complex polysaccharide composed of six monosaccharides with a molecular weight of 3.97 kDa. Specifically, CIP treatment mitigated weight loss and histopathological damage in the colon, increased colon length, and by inhibiting the TLR4/MyD88/NF-κB pathway significantly downregulated the secretion levels of pro-inflammatory cytokines TNF-α, IL-6, and IL-1β, while simultaneously upregulating the anti-inflammatory cytokine IL-10 and inhibition

Microenvironment-induced programmable nanotherapeutics restore mitochondrial dysfunction for the amelioration of non-alcoholic fatty liver disease.

Nonalcoholic fatty liver disease (NAFLD) is a metabolic liver disorder with severe complications. Mitochondrial dysfunction due to over-opening of the mitochondrial permeability transition pore (mPTP) in liver cells plays a central role in the development and progression of NAFLD. Restoring mitochondrial function is a promising strategy for NAFLD therapy. Herein, we designed and developed a microenvironment-induced programmable nanotherapeutic to restore mitochondrial function and ameliorate NAFLD. cyclosporine (Cyclosporine capsules) A (CsA), as a highly effective inhibitors of the opening of
