Frasier syndrome in 17-year-old girl--case report.

The authors present a case of Frasier syndrome in a 17-year-old girl with nephrotic syndrome and male pseudohermaphroditism. Due to the existing risk of developing tumors in dysgenetic gonads, the patient was admitted to the clinic for prophylactic gonadectomy The operation was then postponed as a result of rapid progression to end-stage renal failure, and the patient was placed on hemodialysis. During subsequent laparoscopy both ovaries and dysgenetic gonads were resected. Histopathological examination revealed the presence of both seminal ducts and epididymis. Early prophylactic resection of

Orthopedic and orthodontic treatment in central giant cell granuloma treated with calcitonin.

Central giant cell granuloma of the jaw is a benign lesion of unknown etiology that occurs with very low frequency. It mainly occurs in children and young adults and is more common in the mandible. The most common treatment is surgical removal; however, alternative therapies (intralesional injections of corticosteroids, interferon alpha, and calcitonin) have been used in order to avoid undesirable damage to the jaws and teeth. The lesion may cause root resorption, tooth germ displacement, and other dental problems, as well as malocclusion that must be treated orthodontically. The orthodontic,

The effect of carbamazepine on sperm counts in Wistar rats--reflecting upon its mitogenic potential.

The present study was aimed to investigate the effects of carbamazepine, an antiepileptic drug, on sperm count in rats. Male Wistar rats were treated with carbamazepine at doses of 9, 18, and 36 mg/kg for five consecutive days. Following the last exposure, on days 14 and 35, spermatozoa were collected from epididymis and counted. On day 14, carbamazepine treatment decreased the sperm number in a dose dependent pattern. On day 35, 9 mg/kg and 36 mg/kg of carbamazepine increased the sperm number in comparison with untreated rats. The results of the study suggest that carbamazepine is a germ cell

A case of T-lineage lymphoblastic lymphoma/leukemia with t(4;11)(q21;p15) that switched to myelomonocytic leukemia at relapse.

A 51-year-old Japanese woman, initially diagnosed with T-lineage (CD2+, CD7+, CD3-, CD4-, CD8-) lymphoblastic lymphoma with t(4;11)(q21;p15), relapsed with acute myelomonocytic leukemia with the identical chromosomal abnormality. Southern-blot analysis revealed clonal rearrangements of an immunoglobulin heavy chain gene (JH) and T-cell receptor genes (J delta 1, J gamma 1, C beta 1) at first presentation, but germ line configurations of these genes at relapse. Leukemias with t(4;11)(q21;p15) may involve a hematopoietic progenitor capable of multilineage differentiation.

Bilateral laparoscopic gonadectomy in a patient with complete androgen insensitivity. A case report.

BACKGROUND: Complete androgen insensitivity (CAI) is the most common cause of male pseudohermaphroditism. Affected individuals are advised to have bilateral gonadectomies to prevent the development of germ cell tumors. CASE: A 19-year-old phenotypic female presented for the evaluation of primary amenorrhea and was diagnosed with CAI. Operative videolaparoscopy was performed, and the intraabdominal gonads were resected; the patient was discharged in < 24 hours. CONCLUSION: Laparoscopic gonadectomy can be performed in individuals with CAI. This approach eliminates the need for laparotomy and r

Ovarian neoplasms in children and adolescents in Papua New Guinea.

A study of ovarian tumours in young females under the age of 20 years in Papua New Guinea between the period 1976-1986 is presented. During these 11 years a total of 101 cases was seen among 802 ovarian tumours reported in all age groups. Epithelial tumours comprised the largest number (66.3%) followed by germ cell tumours (24.8%). Ten patients (9.9%) presented with torsion. Only 3 (3%) cases of Burkitt lymphoma were recorded (3%); the series included 1 case of primary non-gestational choriocarcinoma of the ovary in an 11-year-old premenarchal girl.

Surgically preventable site of local recurrence of testicular carcinoma.

We report on a patient with a mixed germ cell testicular carcinoma in whom recurrence developed at the site of retained spermatic cord elements subsequent to retroperitoneal lymph node dissection. This site for recurrence can be prevented by total removal of the spermatic cord. Recurrence at this site often involves the peritoneum thus significantly reducing the survival rate. It is our policy to attach a long length of nonabsorbable suture marker material to the most proximal aspect of the spermatic cord when performing a radical inguinal orchiectomy. Such a marker facilitates the removal of

Computed tomography characterization of anterior mediastinal tumors.

The computed tomography manifestations of 39 patients with histologically proved anterior mediastinal tumors were studied by categorizing the tumors by content as solid homogeneous, cystic homogeneous, and heterogeneous. Among 24 solid tumors, thymomas were easily distinguished by their lack of invasiveness from malignant lymphomas and germ-cell tumors. Among 11 cystic tumors, pericardial and thymic cysts were differentiated from cystic teratomas and bronchogenic cysts by their lack of recognizable capsules with or without calcification. The differential diagnosis of a variety of tumors of the

Gestational and nongestational trophoblastic neoplasia: a historic and pathobiologic survey.

A peculiar fascination has existed with trophoblastic neoplasia as an entity from the earliest cognitive periods. An intensive level of controversy surrounded the question of histogenesis which was finally settled in the closing years of the 19th century as a neoplasm of fetally derived structures. It was appreciated that similar appearing tumors occurred in extrauterine sites and that males were also affected. The totipotential germ cell was suspected as the progenitor of these neoplasms. The purpose of this report is to review some of the milestone developments in our knowledge of gestationa

Cdkn1c orchestrates a molecular network that regulates euploidy of male mouse germline stem cells.

Karyotype instability in the germline leads to infertility. Unlike the female germline, the male germline continuously produces fertile sperm throughout life. Here, we present a molecular network responsible for maintaining karyotype stability in the male mouse germline. Loss of the cyclin-dependent kinase inhibitor Cdkn1c in undifferentiated spermatogonia induced degeneration of spermatogenesis prior to entry into the differentiating spermatogonia stage. In vitro analysis of mouse spermatogonial stem cells revealed that CDKN1C localized to spindle microtubules during metaphase, and that disru

The Plasmodium transmission-blocking symbiont, Microsporidia MB, is vertically transmitted through Anopheles arabiensis germline stem cells.

Microsporidia MB is a promising candidate for developing a symbiont-based strategy for malaria control because it disrupts the capacity of An. arabiensis to transmit the Plasmodium parasite. The symbiont is predominantly localized in the reproductive organs and is transmitted vertically from mother to offspring and horizontally (sexually) during mating. Due to the contribution of both transmission routes, Microsporidia MB has the potential to spread through target vector populations and become established at high prevalence. Stable and efficient vertical transmission of Microsporidia MB is imp

Per- and Poly-fluoroalkyl Substances (PFAS) Exposure and Risk of Kidney, Liver, and Testicular Cancers: A Systematic Review and Meta-Analysis.

INTRODUCTION: Per- and poly-fluoroalkyl substances (PFASs) are a large, complex group of synthetic chemicals humans can be exposed to from occupational or environmental sources. In this systematic review and meta-analysis, we examined the association between PFAS exposure, particularly Perfluorooctanoic Acid (PFOA), and Perfluorooctane Sulfonic Acid (PFOS), and risk of kidney, liver, and testicular cancer. METHODS: We systematically searched PubMed to identify cohort and case-control studies reported after the Monograph of the International Agency for Research on Cancer and the Toxicological

Suppression of trinucleotide repeat expansion in spermatogenic cells in Huntington's disease.

Trinucleotide repeats (TNRs) are dispersed throughout the human genome. About 20 loci are related to human diseases, such as Huntington's disease (HD). A larger TNR instability is predominantly observed in the paternal germ cells in some TNR disorders. Suppressing the expansion during spermatogenesis can provide a unique opportunity to end the vicious cycle of genetic anticipation. Here, using an in vitro differentiation method to derive advanced spermatogenic cells, we investigated the efficacy of two therapeutic agents, araC (cytarabine) and aspirin, on stabilizing TNRs in spermatogenic cell…

[Methylation pattern of imprinted genes H19 and IGF2r in spermatogenic cells cultured in vitro].

OBJECTIVE: To compare the methylation patterns of imprinting control region (ICR) of H19 gene and differentially methylated region (DMR) of IGF2r gene in mature sperms derived from epididymis of Kunming mice and in vitro cultured haploid spermatids. METHODS: The H19 ICR and IGF2r DMR2 were detected by bisulfite sequencing PCR (BSP). The results were compared with standard sequence derived from GenBank using a DNAman software. RESULTS: 96.67% (15 CpG sites) of H19 ICR was found to be methylated, and 94.29% IGF2r DMR2 was found to be unmethylated in mature sperms. By contrast, 69.33% of H19 IC

Congenital cerebral primitive neuroectodermal tumor with astrocytic differentiation and extracranial metastases.

A cerebral primitive neuroectodermal tumor with astrocytic differentiation and extracranial metastases in a 28-day-old infant is reported. The infant presented with a progressively enlarged head, cutaneous lesions in the neck, and enlarged cervical lymph nodes. A computed tomography brain scan demonstrated a giant thalamic tumor with subarachnoid dissemination and hydrocephalus. Biopsy material from the cervical lesions showed a picture of glioma with anaplastic astrocytes. The patient received a ventriculoperitoneal shunt operation and palliative chemotherapy, but died at 3 months of age. Aut

Hyaline-cell pleomorphic adenoma. Diagnosis by fine needle aspiration biopsy.

The cytologic, histologic, immunohistochemical and ultrastructural characteristics are presented of a case of hyaline-cell pleomorphic adenoma of the soft palate diagnosed by aspiration biopsy. Hyaline cells are commonly found in pleomorphic adenomas of minor salivary gland origin. The lack of cohesiveness of the hyaline cells makes them readily amenable to sampling by fine needle aspiration biopsy. When identified in a neoplasm, these cells are characteristic, and probably diagnostic, of a mixed tumor.

Rosmarinic acid alleviates colitis and repairs mucus barrier damage in a gut microbiota-dependent manner.

Rosmarinic acid (RA) significantly alleviates DSS-induced colitis by repairing the damaged colonic mucus barrier and reversing gut microbiota dysbiosis. However, it remains unclear whether the gut microbiota is required for this protective effect. To address this, pseudo-germ-free mouse models and fecal microbiota transplantation (FMT) were used to evaluate whether the gut microbiota is required for RA to restore mucus barrier integrity and alleviate colitis. The study showed that transplanting fecal microbiota from RA-pretreated donor mice into pseudo-germ-free recipients with colitis markedl

Generation of hiPSCs (JUCGRMi008-A) from a β-propeller protein-associated neurodegeneration patient with WDR45 mutation.

The WDR45 gene encodes WIPI4, a protein that plays a crucial role in autophagy. Mutations in WDR45 lead to excessive iron deposition in the central nervous system, resulting in β-propeller protein-associated neurodegeneration (BPAN) which is characterized by progressive dystonia and Parkinsonism. In this study, we established an induced pluripotent stem cell (iPSC) line from a BPAN patient with WDR45 mutation. The iPSC line exhibited typical morphology, a normal karyotype, expression of markers of the undifferentiated hPSC state, and the capacity to differentiate into all three germ layers. Th

Generation of an induced pluripotent stem cell line (NCHi023-A) from a 41-year-old male with nemaline myopathy carrying autosomal dominant ACTA1 c.809-10C>A mutation.

Nemaline myopathy is a rare genetic condition characterized by weakened muscles due to thread-like rods, called nemaline bodies, in muscle fibers. This condition varies in time of onset and severity. Although there is no cure, therapies and treatments are available to reduce symptoms. This iPSC line, NCHi023-A, was reprogrammed using Sendai virus from skin fibroblasts from a male patient with nemaline myopathy carrying a pathogenic heterozygous ACTA1 mutation. Characterization of this line was successful, with validation of cell identity, normal morphology and karyotype, positive expression of
