Streptococcus equi subsp. zooepidemicus (SEZ) has a wide host spectrum, including humans and domestic animals. The SEZ-caused swine streptococcicosis outbreak has occurred in several countries, and the swine-isolated strains usually have specific S. zooepidemicus M-like (szm) gene types. In this study, we found that the production of this specific szm gene (SzM protein) was an effective vaccine candidate. It could provide better protection with a 7-day interval immune procedure than the traditional vaccine strain ST171 and attenuate the strain ΔsezV against swine-isolated hypervirulent SEZ inf

VlsE (variable major protein-like sequence, expressed) is an outer surface protein of the Lyme disease pathogen (Borreliella species) responsible for its within-host antigenic variation and a key diagnostic biomarker of Lyme disease. However, the high sequence variability of VlsE poses a challenge to the development of consistent VlsE-based diagnostics and therapeutics. In addition, the standard diagnostic protocols detect immunoglobins elicited by the Lyme pathogen, not the presence of the pathogen or its derived antigens. Here, we described the development of recombinant monoclonal antibodie

Mandibular reconstruction with vascularized bone grafts is complicated by donor-host nonunion, and current virtual surgical planning produces a geometric plan rather than optimizing for bone-union propensity at the donor-host interface. We present OsteoOpt++, an image-to-decision planning loop for patient-specific mandibular reconstruction. A pre-operative computed tomography (CT) is converted into a personalized digital twin through template-to-patient registration and CT-derived updates of the muscle and temporomandibular-joint parameters. Bayesian optimization with an expected-improvement-p

Profound lymphopenia has been observed during many acute viral infections, and our laboratory has previously documented a type I IFN-dependent loss of CD8 T cells immediately preceding the development of the antiviral T cell response. Most memory (CD44(high)) and some naive (CD44(low)) CD8 T cells are susceptible to IFN-induced attrition, and we show in this study that the IFN-induced attrition of CD8(+)CD44(high) T cells is associated with elevated activation of caspase-3 and caspase-8. We questioned whether TCR engagement by Ag would render CD8 T cells resistant to attrition. We tested wheth

The interaction between the host immune response and infected hepatocytes plays a central role in the pathogenesis of hepatitis C virus (HCV). The lack of a suitable animal or in vitro model has hindered our understanding of the host T-cell/HCV interaction. Our aim was to develop an in vitro model to study the mechanisms of HCV-specific T-cell-mediated antiviral and cytolytic function. The HCV replicon was HLA typed and lymphocytes were obtained from an HLA class I-matched subject. CD8(+) T cells were expanded with 2 HCV-specific/HLA-restricted peptides for NS3. Lymphocyte preparations were co

Cryptococcus neoformans is an environmentally acquired fungal pathogen that causes over 140,000 deaths per year. Cryptococcal infection occurs when infectious particles are deposited into the lung, where they encounter host phagocytic cells. C. neoformans may be engulfed by these phagocytes, an important step of infection that leads to outcomes ranging from termination of infection to cryptococcal dissemination. To study this critical process, we screened approximately 4,700 cryptococcal gene deletion mutants for altered uptake, using primary mouse and human phagocytic cells. Among the hits of

As one of the world's megadiverse countries, Australian biodiversity is vital for global biodiversity. Nematodes belonging to the genus Anisakis (family Anisakidae) are an important part of this biodiversity due to their ability to be repeatedly transmitted among their intermediate hosts before reaching the top of the food pyramid. Therefore, they have a significant impact on the community structures of various ecosystems. In addition, globally, they are known to be of medical and veterinary significance. The aim of this article is to provide an update on the current knowledge about these impo

SUMMARYColorectal cancer (CRC) is a significant global health concern that is growing in prevalence, especially in younger populations. The gut microbiome is an increasingly recognized factor in the development and progression of numerous diseases, including CRC. This review explores the current research on the causal relationship between the microbiome and CRC, including the strengths and limitations of the current models for studying this complex interaction. We then delve into key microbial metabolites and their effects on host signaling pathways in the context of CRC and highlight specific

Many phages encode proteins that specifically inhibit host RNA polymerase activity, thereby sabotaging and, in some cases, hijacking the host transcription machinery to serve their needs. Traditional methods for identifying new phage proteins that inhibit bacterial transcription are labor intensive and require access to live phages. To overcome these limitations, we develop a highly efficient pipeline for AlphaFold 3-guided discovery of phage proteins that inhibit bacterial transcription initiation. Using this pipeline, three phage proteins are identified and characterized. Structural and bioc

Fusarium pseudograminearum, the dominant causal agent of Fusarium crown rot (FCR), secretes numerous effectors to modulate host immunity during infection, but the functions of most remain unknown. In this study, we identified FpSP5, an effector from F. pseudograminearum, as a pathogen-associated molecular pattern (PAMP) that triggers plant immunity. FpSP5 induces cell death, oxidative burst, and PTI in Nicotiana benthamiana, conferring resistance against multiple pathogens. Furthermore, its perception specifically depends on the coreceptor BAK1 in N. benthamiana. Interestingly, FpSP5 is not on

Bacteria belonging to the relapsing fever-causing genus Borrelia, are important causes of zoonotic veterinary and human infections. The genus contains both species with a specific avian association and species with a suggested wider host range, including avian hosts. Despite this, existing studies exploring the Borrelia-avian interaction are relatively few. In this short communication, we report on the in vitro survival of eight strains of relapsing fever from seven species in chicken serum and its correlation to bacterial binding of serum components reactive to a polyclonal anti-complement re

Precise spatial positioning of cellular constituents is critical for bacterial replication, interactions, and behavior. Rod-shaped bacteria may concentrate molecules and machinery at the cell poles, often by leveraging polar scaffolding proteins to recruit other factors. These typically small proteins can self-assemble into higher-order structures that provide platforms to direct fundamental processes like chromosome segregation, division plane selection, and differentiation. Beyond fundamental cellular processes, polarity is implicated in a wide range of bacterial behaviors, including environ

The genus Taenia belongs to the class Cestoda and comprises a diverse group of species of medical, veterinary, and ecological importance. These tapeworms exhibit complex life cycles involving carnivorous definitive hosts and herbivorous or omnivorous intermediate hosts. Taenia solium, T. saginata, and T. asiatica are particularly important due to their impact on human health, notably through taeniasis and cysticercosis. Mitochondrial genomes (mitogenomes) have emerged as powerful tools for taxonomy, diagnostics, and evolutionary studies of these parasites. Complete mitogenome sequences are ava

The gut microbiome harbors immense metabolic capacity, much of which is still being uncovered. Recent discoveries have highlighted the ability of gut microbes to metabolize inositol-derived compounds, including free inositols and their phosphorylated derivatives, inositol phosphates. While these compounds are abundant in plant-based diets, our understanding of the specific bacterial taxa involved and their metabolic pathways has only recently begun to emerge. Intriguingly, inositols and inositol phosphates have been associated with a range of health benefits. Although some effects have been at

Adaptive immunity, performed by T and B lymphocytes, seeks total virus elimination through specific recognition of viral antigens. It has been shown that innate or adaptive immune response regulation variations are associated with an excessive immune response, leading to tissue damage with an increased risk of complications and death. This article is a novel contribution focused on models that represent pathogenic interactions with humans. In our case, the objective was to build and analyze a mathematical model for SARS-CoV-2 infection in the human host, including elements of respiratory cell

Herein, a novel synthetic strategy for macrocycles based on a three-dimensional carborane skeleton via a cyclization reaction was developed. Further substrate scope experiments confirm that this strategy possesses excellent reaction compatibility. In addition, the cavity size of the macrocycle can be regulated by the bonding angle, in which the size of the macrocycle cavity formed by para-carborane (180°) is larger than that formed by meta-carborane (120°). More importantly, when a pyridine group is introduced into this type of macrocycle, nanotubes can be constructed through the coordination

Although many host factors important for severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection have been reported, the mechanisms by which the virus interacts with host cells remain elusive. Here, we identified tripartite motif containing (TRIM) 28, TRIM33, euchromatic histone lysine methyltransferase (EHMT) 1, and EHMT2 as proviral factors involved in SARS-CoV-2 infection by CRISPR-Cas9 screening. Our result suggested that TRIM28 may play a role in viral particle formation and that TRIM33, EHMT1, and EHMT2 may be involved in viral transcription and replication. UNC0642, a com

This study examined the correlation between intestinal protozoans and the bacterial microbiome in faecal samples collected from 463 patients in New Zealand who were diagnosed with gastroenteritis. In comparison to traditional microscopic diagnosis methods, Multiplexed-tandem PCR proved to be more effective in detecting intestinal parasites. Among the identified protozoans, Blastocystis sp. and Dientamoeba fragilis were the most prevalent. Notably, D. fragilis was significantly associated with an increase in the alpha-diversity of host prokaryotic microbes. Although the exact role of Blastocyst

Due to the recurrent pandemic outbreaks that occurred during the last century, Influenza A viruses are considered a serious potential danger to human health. Among the innate immune pathways in eukaryotes, RNA interference plays a significant role in the interaction between viruses and host cells. RNA interference is addressed by small dsRNA molecules produced by the host itself (miRNAs, i.e. "micro-RNAs") but can be triggered also by the administration of exogenous short RNAs (siRNAs, "short interfering RNAs"). In this work, artificial siRNA pools targeting NP and PB genomic regions of the In

Sepsis is a syndrome characterized by life-threatening organ dysfunction caused by the dysregulated host response to an infection. Sepsis, especially septic shock and multiple organ dysfunction is a medical emergency associated with high morbidity, high mortality, and prolonged after-effects. Over the past 20 years, regulatory T cells (Tregs) have been a key topic of focus in all stages of sepsis research. Tregs play a controversial role in sepsis based on their heterogeneous characteristics, complex organ/tissue-specific patterns in the host, the multi-dimensional heterogeneous syndrome of se
