Primary selective IgM deficiency: an ignored immunodeficiency.

Immunoglobulin M (IgM) provides the initial response to foreign antigen and plays a regulatory role in subsequent immune response development, accelerating the production of high-affinity IgG. Though selective IgM deficiency was described more than 45 years ago in children with fulminant meningococcal septicemia, it has been largely an ignored primary immunodeficiency. It appears to be more common than originally realized. Selective IgM deficiency is observed in both children and adults with no gender bias. The most common clinical manifestation of selective IgM deficiency is infections with e

How innate immune mechanisms contribute to antibody-enhanced viral infections.

Preexisting antibodies may enhance viral infections. In dengue, nonneutralizing antibodies raised by natural infection with one of four dengue viruses (DENVs) may enhance infection with a different virus by a process we term "intrinsic antibody-dependent enhancement" (iADE). In addition, nonprotective antibodies raised by formalin-inactivated respiratory syncytial virus (RSV) and measles virus vaccines have led to enhanced disease during breakthrough infections. Infections under iADE conditions not only facilitate the process of viral entry into monocytes and macrophages but also modify innate

IL-10 is required for optimal CD8 T cell memory following Listeria monocytogenes infection.

IL-10 is an important immunoregulatory cytokine that plays a central role in maintaining a balance between protective immunity against infection and limiting proinflammatory responses to self or cross-reactive Ags. We examined the full effects of IL-10 deficiency on the establishment and quality of T cell memory using murine listeriosis as a model system. IL-10(-/-) mice had reduced bacterial loads and a shorter duration of primary infection than did wild-type mice. However, the number of Ag-specific T cells in secondary lymphoid and nonlymphoid organs was diminished in IL-10(-/-) mice, compar

Glucose and electrolyte supplementation of drinking water improve the immune responses of poults with inanition.

Enteric disorders predispose poultry to malnutrition. The objectives of this paper were 1) to simulate the inanition of poult enteritis mortality syndrome by restricting feed intake and 2) to develop a drinking water supplement that supports the immune functions of poults with inanition. Poults were restricted to 14 g of feed/d for 7 d beginning at 14 d of age then fed ad libitum until 36 d (recovery). The control was fed ad libitum. During the feed-restriction period, duplicate groups of 6 poults received 1 of 5 drinking water treatments: 1) restricted feed, unsupplemented water; 2) restricte

Pregnancy in patients with idiopathic thrombocytopenic purpura: assessing the risks for the infant at delivery.

The objective of this review is to estimate the risk of severe thrombocytopenia at birth and minor or major morbidity for the infant of a pregnancy complicated by immune thrombocytopenic purpura. English language publications on immune thrombocytopenic purpura in pregnancy from January 1980 to December 1990 were used. Manuscripts considered for infant risk assessment fulfilled the following criteria: 10 or more entrants, reported fetal platelet samples (cordocentesis, scalp sampling or cord sample at birth), and provided all infant platelet data. All other data not fulfilling these criteria ar

The sympathetic nerve--an integrative interface between two supersystems: the brain and the immune system.

The brain and the immune system are the two major adaptive systems of the body. During an immune response the brain and the immune system "talk to each other" and this process is essential for maintaining homeostasis. Two major pathway systems are involved in this cross-talk: the hypothalamic-pituitary-adrenal (HPA) axis and the sympathetic nervous system (SNS). This overview focuses on the role of SNS in neuroimmune interactions, an area that has received much less attention than the role of HPA axis. Evidence accumulated over the last 20 years suggests that norepinephrine (NE) fulfills the c

Exploring the relationship between metabolism and immune microenvironment in breast cancer bone metastasis based on metabolic pathways.

BACKGROUND: Bone metastasis is a significant contributor to mortality in patients with advanced breast cancer. Its progression is deeply intertwined with tumor metabolic reprogramming and the remodeling of the immune microenvironment. However, the dynamic interplay between metabolic pathways and immune regulation remains incompletely elucidated. METHODS: In this study, leveraging RNA-seq data and clinical information from breast cancer bone metastasis (BCBM) patients sourced from the GEO database, integrated bioinformatic analyses were employed to determine the activity of metabolic pathways

Dietary β-1,3/1,6-Glucan from Baker's Yeast Supports Upper Respiratory Mucosal Immune Health in Healthy Adults: Evidence from a Randomized, Double-Blind, Placebo-Controlled Trial.

Background/Objective: Dietary polysaccharides are increasingly recognized as functional nutritional components that support human health through modulation of immune function. However, clinical evidence linking their intake to site-specific upper respiratory mucosal immune health in humans remains limited. This study investigated whether dietary β-1,3/1,6-glucan (SC-BG) from baker's yeast may support upper respiratory mucosal immune health in healthy adults. Methods: Following in vitro assays on human dendritic cells (DCs), a randomized, double-blind, placebo-controlled parallel-group trial wa

Bifidobacterium lactis XLTG11 Reduces Eczema and Infections in Infants: A Randomized Trial.

BACKGROUND: Early-life gut microbiota profoundly influences immune system maturation and disease susceptibility. Perturbations in microbial development have been linked to rising rates of allergic and infectious diseases in children. Probiotic interventions offer a promising strategy to restore microbial-immune homeostasis; however, evidence from rigorously designed, strain-specific randomized trials integrating clinical and microbiome outcomes remains limited. OBJECTIVE: To evaluate the efficacy of Bifidobacterium animalis subsp. lactis XLTG11 in reducing the incidence of eczema and respirat

Alveolar echinococcosis drives functional reprogramming of hepatic CD8+ T cells.

BACKGROUND: Alveolar echinococcosis (AE), caused by the larval stage of Echinococcus multilocularis, exhibits infiltrative, tumor-like behavior in the liver and persists within its tolerogenic immune environment. Although T cells are central to host defense, the stage-specific remodeling of their lineage states during AE remains unclear. METHODS: A secondary AE infection model was established by portal vein injection of approximately 1,000 viable protoscoleces in C57BL/6 mice. Liver tissues collected at 3 days (3 dpi) and 3 months (3 mpi) post-infection were analyzed using single-cell RNA seq

Impact of Immune System Activation and Vascular Impairment on Male and Female Sexual Dysfunction.

INTRODUCTION: Male and female sexual dysfunction (SD) is considered a multifactorial condition. Numerous studies have shown the involvement of inflammatory processes in this pathological condition. Sexual intercourse requires healthy and functioning vessels to supply the pelvic region in both males and females, generating penile erection and clitoral and vaginal lubrication, respectively. Cardiovascular diseases and associated risk factors may contribute negatively to pelvic blood flow, possibly through immune system activation. AIM: The study aimed to address the correlation between vascular

The History of Macrophage Activation Syndrome in Autoimmune Diseases.

In 1979, it became recognized in the literature that what we call hemophagocytic lymphohistiocytosis (HLH) was a nonmalignant disease of histiocytes. Subsequently a familial form and a secondary form of HLH were differentiated. When HLH is secondary to an autoimmune disease, rheumatologists refer to this entity as macrophage activation syndrome (MAS) to differentiate it from HLH itself. Although the first cases of MAS likely appeared in the literature in the 1970s, it was not until 1985 that the term activated macrophages was used to describe patients with systemic juvenile idiopathic arthriti

Survey of the Italian Registry of Renal Biopsies. Frequency of the renal diseases for 7 consecutive years. The Italian Group of Renal Immunopathology.

PURPOSE: This report describes data collected by the Italian Registry of Renal Biopsies (IRRB) (E mail http://www.teseo.it/nephrology/irrb), which includes 15461 renal biopsies (RB) performed over a period of 7 years (1987-1993). METHODS: From 1987 to 1988, the Registry recorded only the histological diagnosis of 3686 RB, whereas beginning in 1989, additional records (sex, age, clinical symptoms and renal function) were obtained from 11,775 patients undergoing RB. This is the first report on such a large number of RB containing data from 96% of Italian renal units which routinely perform RB.

A predictive model for identifying secondary underlying diseases of hemophagocytic lymphohistiocytosis.

BACKGROUND: Secondary hemophagocytic lymphohistiocytosis (HLH) is a rare, life-threatening disease of immune hyperactivation that arises in the context of infectious, inflammatory, or neoplastic triggers. The aim of this study was to establish a predictive model for the timely differential diagnosis of the original disease resulting in HLH by validating clinical and laboratory findings to further improve the efficacy of therapeutics for HLH. METHODS: We retrospectively enrolled 175 secondary HLH patients in this study, including 92 patients with hematologic disease and 83 patients with rheuma

Genetic line and major histocompatibility complex effects on primary and secondary antibody responses to T-dependent and T-independent antigens.

The effects of MHC and nonMHC (background) genetics on the kinetics of primary and secondary antibody responses to T-cell-dependent (SRBC) and T-cell-independent [Brucella abortus (BA)] antigens were investigated. Eight genetic groups were represented, with four homozygous MHC haplotypes [B1-IrGATlow (IrGAT = immune response to GAT locus); B1-IrGAThigh; B19-Ir-GATlow; B19-IrGAThigh] on two genetic backgrounds, the S1 and G lines. Birds were injected simultaneously with BA and SRBC at 4 and 7 wk of age, and blood samples were taken weekly from 4 to 10 wk of age for measurement of total agglutin

Safety and immunogenicity of a first-generation modified vaccinia virus Ankara-vectored SARS-CoV-2 vaccine candidate with mRNA-based booster vaccination: A phase 1 trial.

The rapid development of effective COVID-19 vaccines was enabled by the parallel assessment of multiple candidates early in the pandemic, including candidates based on the Modified Vaccinia virus Ankara (MVA) vector platform. This Phase 1 trial (NCT04569383) evaluated the MVA-based vaccine candidate MVA-SARS-2-S, expressing the full-length native SARS-CoV-2 spike glycoprotein (Wuhan-HU-1). Between October 2020 and May 2021, 30 healthy, SARS-CoV-2-naive adults received two doses of either 107 or 108 international units (IU) of MVA-SARS-2-S 28 days apart, with 6-month follow-up. Prospective Evaluation of Antibody Response post COVID-19 vaccination in older persons ≧ 60 years old (PEARL 60): A longitudinal 15-months study in a tertiary centre in Malaysia.

Older adults exhibit heterogeneous immune responses to COVID-19 vaccination, yet the relative contributions of age, comorbidity, vaccine platform, and infection history to antibody durability remain incompletely defined. Understanding these determinants is essential to inform booster strategies in ageing populations. We conducted a longitudinal observational study of 300 participants (250 aged ≥60 years and 50 younger controls) followed for up to 15 months. Anti-spike (anti-S) antibody responses were assessed at four event-anchored timepoints: ≤ 3 months post-primary vaccination (TP1), ~ 3 mon
