Potential role of high mobility group box protein 1 and intermittent PTH (1-34) in periodontal tissue repair following orthodontic tooth movement in rats.

OBJECTIVES: Recent studies indicate that high mobility group box protein 1 (HMGB1) can be released by necrotic and damaged cells and functions as an alarmin that is recognized by the innate immune system. Little is known about the role of HMGB1 within the periodontal ligament (PDL). Therefore, we examined HMGB1 expression by PDL cells in vitro and compared the findings to an in vivo model of orthodontically induced tooth root resorption. In addition, we addressed the question of whether a potentially anabolic intermittent administration of parathyroid hormone (iPTH) would modulate the expressi

Sinus grafting with mineralized allograft and staged implant placement.

Advanced atrophy of the partially edentulous maxilla necessitates bone augmentation as a prerequisite for successful implant osseointegration. This case shows that sinus grafting at the left first molar (free-end gap) by means of particulated mineralized solvent-preserved cancellous bone and sealing of the recipient site with a bioabsorbable xenogenic collagenous membrane provides optimum conditions for a staged implant placement. Four months after sinus membrane elevation, a trephine biopsy was recovered from the augmented floor and a microtextured implant placed into the augmented area. The

Clinical, radiographic, and histologic evaluation of localized ridge augmentation using a mandibular bone block.

This clinical case report presents a new technique for reconstructing the vertical and horizontal dimensions of the alveolar ridge using a mandibular bone block at sites planned for single implants. Augmentation was eased with the use of a mandibular bone block, which was shaped after placement with a round bur to passively fit the dimensions of the defect, at the recipient site. After treatment, approximately 7 mm of horizontal and 3 mm of vertical augmentation were gained. Histologic observation showed that the grafted bone was well integrated with the original bone. The volume of tissue gai

Aspirin, acetaminophen, and ibuprofen: their effects on orthodontic tooth movement.

INTRODUCTION: Orthodontic patients often take analgesics for pain during treatment. But various analgesics have different capacities to inhibit prostaglandins, and these differences might affect tooth movement. The purposes of this study were to determine by direct measurement the effects that acetylsalicylic acid, ibuprofen, and acetaminophen have on orthodontic tooth movement in rats and to evaluate histologically the differences in bone resorption in the pressure area in rats treated with these analgesics. METHODS: Thirty-six adult male Wistar rats were divided in 4 groups of 9 each. Ortho

Treatment of gingival recession using a collagen membrane with or without the use of demineralized freeze-dried bone allograft for space maintenance.

BACKGROUND: Studies utilizing collagen membranes for guided tissue regeneration (GTR)-based root coverage procedures have reported promising results. However, creating and maintaining space underneath the membrane remains a challenge. Therefore, the purpose of this clinical trial was to determine whether the addition of bone graft (i.e., demineralized freeze-dried bone allograft [DFDBA]) significantly affects the outcome of collagen membrane GTR-based root coverage procedures. METHODS: Twenty patients participated. One Miller's Class I or II recession defect per patient was treated with a col

Dominant lung involvement and heterozygous IFIH1 variants.

BACKGROUND: Heterozygous gain-of-function (GoF) variants in IFIH1 are associated with rare autoinflammatory diseases characterized by heightened interferons-stimulated gene (ISG) signatures. These include Aicardi-Goutières syndrome, presenting with slowly progressive spastic paraparesis of the lower limbs, intracerebral calcifications, and neuroinflammation, and Singleton-Merten syndrome, defined by tooth loss, bone demineralization, and vascular calcification. RESEARCH QUESTION: Do rare IFIH1 GoF variants contribute to an interstitial lung disease phenotype, thereby expanding the spectrum of…

Early-onset neuroinflammation drives neurodegeneration caused by lysosomal PI(3,5)P2 insufficiency.

Phosphatidylinositol 3,5-bisphosphate [PI(3,5)P2] is a lysosomal signaling lipid whose deficiency, caused by mutations in the PIKfyve complex subunits FIG4 or VAC14, underlies a spectrum of fatal neurologic diseases including Charcot-Marie-Tooth type 4J (CMT4J) and amyotrophic lateral sclerosis (ALS). To map the molecular consequences of PI(3,5)P2 insufficiency in the brain, we performed quantitative proteomic and transcriptomic analyses of three mouse lines bearing distinct loss-of-function mutations in Fig4 or Vac14, examining the brain at the presymptomatic and end stages. Strikingly, profo

WTAP Contributes to Periodontitis Pathogenesis by Promoting PDLSC Senescence and Impairing Osteogenic Differentiation via m6A-Dependent Regulation of TP53BP1.

BACKGROUND: Periodontitis, a chronic inflammatory disease, represents the primary cause of tooth loss in Chinese adults. Wilms tumor 1-associating protein (WTAP) is a key component of the N6-methyladenosine (m6A) methyltransferase complex, and has an unclear role in periodontitis pathogenesis, particularly concerning its regulatory functions in periodontal ligament stem cells (PDLSCs). METHODS: The target gene was identified through the GES260558 dataset and Genecards database. Gene expression was measured using reverse transcription-quantitative PCR (RT-qPCR) and western blot. Periodontitis-…

Direct and indirect regulation of SIX1+EYA transcriptional activity by PA2G4, MCRS1, and SOBP.

Branchio-oto-renal (BOR) syndrome is an autosomal dominant condition characterized by variable malformations including hearing loss and renal dysfunction. Variants in SIX1 or its activating co-factor EYA1 are causative in about 50% of patients. Some patients carrying BOR variants also present with craniosynostosis, indicating that cranial skeletal dysmorphologies could be an under-diagnosed feature. To date, most studies on the role of SIX1 have focused on its role in the cranial placode-based development of the inner ear, whereas its role in the neural crest cells of the mandibular arch, whic
