Immunotherapeutic Concepts to Target Acute Myeloid Leukemia: Focusing on the Role of Monoclonal Antibodies, Hypomethylating Agents and the Leukemic Microenvironment.

Intensive chemotherapeutic protocols and allogeneic stem cell transplantation continue to represent the mainstay of acute myeloid leukemia (AML) treatment. Although this approach leads to remissions in the majority of patients, long-term disease control remains unsatisfactory as mirrored by overall survival rates of approximately 30%. The reason for this poor outcome is, in part, due to various toxicities associated with traditional AML therapy and the limited ability of most patients to tolerate such treatment. More effective and less toxic therapies therefore represent an unmet need in the m

sGC-cGMP signaling: target for anticancer therapy.

The biologic endogenous production of cGMP was reported in the 1960s and followed by the demonstration of guanylyl cyclase activity and the isoforms of soluble and membrane-bound guanylyl cyclases. During the same period, cGMP specific phosphodiesterases also was discovered. Murad's lab established link between the endothelium derived relaxation factor (EDRF) and elevated cGMP concentration in the vascular system. October 12, 1998, the Nobel Assembly awarded the Nobel Prize in Medicine or Physiology to scientists Robert Furchgott, Louis Ignarro, and Ferid Murad for their discoveries concerning

Immune Microenvironment in Sporadic Early-Onset versus Average-Onset Colorectal Cancer.

The incidence of left-sided colon and rectal cancer in young people are increasing worldwide, but its causes are poorly understood. It is not clear if the tumor microenvironment is dependent on age of onset, and little is known about the composition of tumor-infiltrating T cells in early-onset colorectal cancer (EOCRC). To address this, we investigated T-cell subsets and performed gene expression immune profiling in sporadic EOCRC tumors and matched average-onset colorectal cancer (AOCRC) tumors. Left-sided colon and rectal tumors from 40 cases were analyzed; 20 EOCRC (<45 years) patients were…

Antigen-experienced NK-T cells: integrating new insights and exploring therapeutic potential in cancer immunotherapy.

Conventional CD8+ T cells can acquire natural killer (NK) cell receptors upon persistent antigen exposure, forming antigen-experienced NK-T (AENK-T) cells. Their dual-state plasticity within the tumor immune microenvironment determines cancer immunotherapy outcomes. This narrative review synthesizes recent transcriptomic, epigenomic, and functional data to define the trajectory, regulation, and therapeutic relevance of AENK-T cells. The analysis establishes a "temporal hierarchy" differentiating AENK-T cells from innate-like T cells: activating NK receptors (NKG2C, NKG2D) emerge early, while i

Integrating machine learning of bulk and single cell RNA data to characterize immune mechanism of m6A related mitophagy genes in malignant gastric cancer.

BACKGROUND AND OBJECTIVE: Gastric cancer is a heterogeneous and complicated epithelial cancers. Chronic H. pylori and EBV infection, as well as intestinal microbiota exposure make gastric cancer encountered a complex tumor immune microenvironment. Mitophagy and m6A are deeply involved in immune microenvironment in the development of tumors. METHODS: We used integrating machine learning of bulk and single cell RNA sequencing to explore the immune mechanisms of m6A related mitophagy genes (MRMGs) in gastric cancer. RT-qPCR and immunochemistry were used to verify gene expression. RESULTS: Progn

Comparative Proteomic Analysis of the Secretome of Control and BRAF/MEK Inhibitor-Resistant Melanoma Cells.

Treatment based on BRAF/MEK kinase inhibitors is one of the most commonly used methods in advanced melanoma therapy, but patients often develop resistance to treatment. Treatment-resistant cells can affect other cancer cells and the tumor microenvironment through the factors that they secrete. Therefore, this study aimed to examine the protein composition of the secretome of cells resistant to vemurafenib (a BRAF inhibitor) and cobimetinib (a MEK inhibitor) and to compare it with that of nonresistant cells. Proteomic analysis, followed by gene ontology (GO) analysis, identified many difference…

Bone Metastasis in Estrogen Receptor-Positive Breast Cancer: Molecular Insights and Therapeutic Advances.

Estrogen receptor-positive (ER+) breast cancer represents the most prevalent molecular subtype of breast cancer, characterized by hormone-dependent growth, relatively indolent progression, and a pronounced tendency to metastasize to bone. While endocrine therapies remain the cornerstone of treatment, a significant proportion of ER+ tumors eventually develop resistance, culminating in distant metastases-most frequently to the bone. Bone metastasis substantially compromises patient survival and quality of life, highlighting the critical need to elucidate its molecular underpinnings. Recent multi

Machine Learning-Assisted Nanoplasmonic Barcode Array-Coupled Microfluidic Platforms for Exosome Profiling and Organotropic Metastasis Analysis.

Elucidating the mechanisms of organotropic metastasis requires the in situ profiling of exosomes within the tumor microenvironment. However, conventional assays are normally based on the isolation of exosomes from biological fluids, which disrupts their native spatial context. Moreover, these assays frequently exhibit limited analytical sensitivity, and the types of exosome phenotypes detected simultaneously are inherently restricted by fluorescent spectral overlap. To address the issue, we present an integrated microfluidic platform coupling a biomimetic tumor-stroma coculture system with a n…

Targeting the ac4C 'Writer' NAT10 enhances pancreatic cancer immunotherapy via dual modulation of CD8+ T cells and tumor cells.

Pancreatic cancer (PC) is highly lethal because of its immunosuppressive tumor microenvironment and resistance to immunotherapy. This study explored the role of NAT10-mediated N4-acetylcytidine (ac4C) RNA modification in pancreatic cancer progression and immune evasion. NAT10 (N-acetyltransferase 10) is overexpressed in pancreatic cancer tissues and correlates with poor prognosis. Mechanistically, NAT10 stabilizes ETS2 mRNA through ac4C acetylation, forming a positive feedback loop that upregulates NAT10 and PD-L1, thereby suppressing CD8 + T cell infiltration and promoting immune evasion. In …

Disrupting Siglec-mediated interactions to develop immunotherapies for cancer treatment.

INTRODUCTION: Recent advances in cancer immunotherapy have improved patient outcomes, even in advanced stages of the disease. However, the effectiveness of current cancer immunotherapies remains limited to a small subset of patients because of resistance and an immunosuppressive tumor microenvironment. AREAS COVERED: Research performed during the last years have identified immunosuppressive interactions between sialic acid-containing glycans and sialic acid-binding immunoglobulin-like lectin (Siglec) receptors as a potential new, targetable pathway to overcome resistance to immunotherapy. In …

Piezo1 Activation Improves NSCLC Liver Metastasis Immunotherapy by Overriding Matrix Stiffness-Mediated Bimodal PD-L1/CXCL10 Regulation.

Immunotherapy efficacy in NSCLC is significantly reduced upon liver metastasis due to profound alterations in the tumor microenvironment, characterized by the absence of cyclic mechanical stretch and increased extracellular matrix (ECM) stiffness. However, the mechanisms underlying the synergistic regulation of these two mechanical cues on the immunotherapy response in NSCLC cells remain poorly understood. In this study, it is demonstrated that both mechanical and biochemical activation of the mechanosensitive ion channel Piezo 1 induces nuclear translocation of YAP, thereby promoting an immun…

Treatment resistance in pancreatic and biliary tract cancer: molecular and clinical pharmacology perspectives.

INTRODUCTION: Treatment resistance poses a significant obstacle in oncology, especially in biliary tract cancer (BTC) and pancreatic cancer (PC). Current therapeutic options include chemotherapy, targeted therapy, and immunotherapy. Resistance to these treatments may arise due to diverse molecular mechanisms, such as genetic and epigenetic modifications, altered drug metabolism and efflux, and changes in the tumor microenvironment. Identifying and overcoming these mechanisms is a major focus of research: strategies being explored include combination therapies, modulation of the tumor microenvi…

Lect2 inhibited EOC cells' invasive phenotype and suppressed EOC's transcoelomic metastasis by targeting c-Met signaling. In addition, Lect2 downregulation induced the accumulation and activation of myeloid-derived suppressor cells (MDSCs). This fo …

Identification of a mechanogenetic link between substrate stiffness and chemotherapeutic response in breast cancer.

Mechanical feedback from the tumor microenvironment regulates an array of processes underlying cancer biology. For example, increased stiffness of mammary extracellular matrix (ECM) drives malignancy and alters the phenotypes of breast cancer cells. Despite this link, the role of substrate stiffness in chemotherapeutic response in breast cancer remains unclear. This is complicated by routine culture and adaptation of cancer cell lines to unnaturally rigid plastic or glass substrates, leading to profound changes in their growth, metastatic potential and, as we show here, chemotherapeutic respon…

IL-17A-producing CD30(+) Vδ1 T cells drive inflammation-induced cancer progression.

Although it has been suspected that inflammation is associated with increased tumor metastasis, the exact type of immune response required to initiate cancer progression and metastasis remains unknown. In this study, by using an in vivo tumor progression model in which low tumorigenic cancer cells acquire malignant metastatic phenotype after exposure to inflammation, we found that IL-17A is a critical cue for escalating cancer cell malignancy. We further demonstrated that the length of exposure to an inflammatory microenvironment could be associated with acquiring greater tumorigenicity and th
