INTRODUCTION: In recent years, the scientific community, together with regulatory authorities, institutions and stakeholders, joined in the common objective of changing the regulatory toxicology paradigm, moving from classical toxicological animal testing towards an exposure-driven risk assessment, based on mechanistic information derived from alternative methods, globally known as new approach methodologies (NAMs). The transition to the next generation risk assessment (NGRA) requires the collective effort of all involved actors. One approach addressing the needs of the NGRA is the adverse out

Copper-induced transmission of antimicrobial resistance has been well documented in livestock farming environments, but the in vivo mechanisms driving fecal resistome development remain unclear. Here, 120 mg/kg CuSO4 and copper-peptide were supplemented to piglets, and the fecal resistome development was first analyzed by metagenomic sequencing. In this study, dietary CuSO4 drove abundant and diverse ARGs and MRGs. Following CuSO4 deprivation, ARGs and copper resistance exhibited a persistent promotion, whereas most MRGs rapidly declined. The resistance development was characterized by abundan

Unveiling the enigmatic processes underlying the rapid disappearance of arthropod corpses in nature is not only fascinating but also essential for understanding their ecological fate. Functional traits retained in freshly dead arthropods may influence their consumption by invertebrate scavengers. To validate this idea, we conducted a field trapping survey using four types of baits (arthropod corpses, yak carrion, yak dung, and horse dung) to investigate the attraction of arthropod corpses to necrophagous beetles in a Tibetan alpine meadow, complemented by a laboratory experiment to further exa

The gut microbiota has received considerable attention in the field of colorectal cancer (CRC) research in recent years. In this review, we have explored the multifaceted relationship between the gut microbiota and CRC progression and treatment. The composition, distribution, and normal physiological functions of the gut microbiota have been summarized, along with the association between gut dysbiosis and CRC based on the body of evidence from animal experiments and clinical studies. In addition, we have discussed the mechanisms through which specific microbial configurations or microbiota-der

New approach methodologies (NAMs) have long contributed to pharmaceutical development with growing attention now focused on their use at regulatory decision points to reduce reliance on laboratory animal models in drug safety and efficacy assessment. Yet validated NAMs remain limited to specific endpoints, and uncertainty persists regarding human translatability and health authority acceptance. In the absence of globally harmonized frameworks, validated-method databases, and clearly defined contexts of use, companies vary in how they identify, evaluate, govern, and implement NAMs. This article

This research provides novel insights into the diversity of DNA extracted from samples collected from the Turin Shroud in 1978, revealing its biological complexity through rigorous DNA and metagenomic analyses. Our findings highlight its preservation conditions and environmental interactions, offering valuable perspectives into the identified genetic variants, which originated from multiple biological sources. We identified several human mitochondrial DNA (mtDNA) lineages, including K1a1b1a, which matches the 1978 official collector's mitogenome, H2a2 (i.e., the lineage of the mtDNA reference

The use of arthropods in a medicolegal framework is well-known and widely adopted in forensic death investigations. Typically, the life cycle and community composition of carrion arthropods can be used to establish a timeframe of events occurring after the death of an individual. However, because of their unique life strategies (eg the precise detection, location, and exploitation of ephemeral resources) and high dispersal abilities as adults, carrion arthropods can reveal much more information than meets the eye. In recent years, carrion arthropods have revealed important biological and chemi

This study evaluated the effects of dietary supplementation with Bacillus-based probiotics on growth performance, nutrient digestibility, rumen fermentation, and microbial functional capacity in weaned yaks. Twenty animals were randomly assigned to a basal diet (control group, CON) or the same diet supplemented with Bacillus subtilis and Bacillus licheniformis (probiotic group, PRO) for 90 days. Probiotic supplementation increased average daily gain (P < 0.05) and tended to increase dry matter intake (P = 0.059). In addition, neutral detergent fibre and acid detergent fibre digestibility were

Animals must allocate limited energetic resources across competing defense programs in response to infection. Here, we show that the conserved nuclear hormone receptor NHR-68 integrates fatty acid metabolism with the neural control of molecular and behavioral immunity in Caenorhabditis elegans. Acting in parallel with NHR-10, NHR-68 controls genes involved in polyunsaturated fatty acid (PUFA) metabolism. Loss of NHR-68 disrupts linoleic acid (LA) homeostasis, impairing pathogen avoidance behavior. Supplementation with LA restores avoidance, and fat-3 inhibition, which elevates LA, enhances pat

Plant-derived exosomes (PDEs) are gaining attention owing to their key implications in cross-kingdom communication, facilitating bioactive entities among plants and animals. PDEs are tiny nanoscale vesicles generally comprised of RNAs, proteins, and secondary metabolites and are involved in the regulation of physiological processes (immune modulation, cell regeneration, and stress response). An important feature of PDEs is to enable cross-kingdom regulation in skin wound repair. This is because PDEs can modulate several signaling pathways (PI3K-Akt, TGF-β, and mitogen-activated protein kinase)

Marine heatwaves are increasing in frequency and intensity worldwide and represent a growing threat to coastal ecosystems and the animals inhabiting them. The vulnerability of marine species to these events depends on several factors, including upper thermal tolerance, the thermal sensitivity of physiological performance, and the capacity for rapid physiological acclimation. We examined upper thermal tolerance and short-term acclimation capacity in 10 mobile macroinvertebrate species inhabiting shallow coastal ecosystems in the Northeast Atlantic. Individuals were exposed either to ambient the

BACKGROUND: Animal venoms may alter postmortem carcass conditions and thereby influence carrion-associated insects. However, it remains unclear whether lethal envenomization affects adult Diptera as a broad assemblage-level response or as taxon-specific changes during decomposition.
METHODS: Fifteen rabbits were assigned to control, snake-envenomized, or scorpion-envenomized treatments (with five carcasses each). Carcasses were exposed under semi-natural conditions, until complete dryness, over 11 days postmortem. Adult flies were collected daily for analyzing abundance at total, family, and

Footpad dermatitis (FPD) is a common welfare and production problem in intensively reared poultry, yet the mechanisms responsible for its chronic progression remain poorly understood. To investigate the biological changes associated with chronic FPD, healthy and FPD-affected ducks (Health and FPD groups) were compared using histopathology, transcriptomics, proteomics, microbiome profiling, and metabolomics, with 3-8 biological replicates per group depending on the analytical platform. Histological examination demonstrated marked extracellular matrix (ECM) remodeling, characterized by collagen

The International Committee on Taxonomy of Viruses (ICTV) is considering a proposal submitted in July 2025 to create an optional additional taxonomic rank below the level of species. This was designed to increase the precision of taxonomic assignment where clinical or other regulatory distinctions need to be made between viruses assigned to the same species. To understand current classification practice and to gauge opinion on the proposal to formalize a below-species taxon, current ICTV study group members (n=746) representing over 100 Study Groups were invited to complete a survey, from whom

Animal tissues have diverse architectures and cell behaviors across the epithelial-mesenchymal spectrum. Cell adhesion mediated by classical cadherins is foundational. Cadherins nucleate complexes of dozens of proteins connecting junctions to the cytoskeleton and signaling downstream. Many junctional proteins are well-studied in epithelia, but less is known about roles during mesenchymal migration. The nascent myotubes of the pupal Drosophila testis provide an excellent model for N-cadherin mediated mesenchymal migration. We combined a proximity proteomics dataset of adherens junction proteins

Ectoparasitoses are caused by organisms that parasitize their host's skin. They affect a considerable proportion of the global population, with a disproportionately greater burden in disadvantaged individuals and communities, although any human is susceptible. They are more prevalent in tropical climates. They can have a major impact on infected individuals' quality of life and on affected populations, and they can result in severe complications. Despite their contribution to the global burden of disease, ectoparasitoses remain largely neglected. Making their characteristics visible and unders

Mucosal microbiota contribute broadly to host defense and immune homeostasis, while the lung and gut microbiota form a particularly important bidirectional ecological and immunological network that shapes pulmonary host defense, inflammatory injury, and tissue repair. In pneumonia, loss of colonization resistance and altered microbial metabolite production may weaken innate and adaptive immunity; respiratory infection, antibiotics, and critical-care exposures can, in turn, remodel both microbial communities. In acute lung injury (ALI) and acute respiratory distress syndrome (ARDS), intestinal

Coccidioidomycosis (Valley fever) is acquired through inhalation of spores produced by fungi in the genus Coccidioides. Coccidioides is commonly detected in, and cultivated from, the lung tissue of native rodents and the soils within their burrows. Coccidioidomycosis is acquired by exposure to environmentally produced spores and is not spread between hosts. Thus, determining the location of rodent burrows with soils harboring Coccidioides will be critical for understanding coccidioidomycosis incidence and modelling how Coccidioides distributions will be affected by global change. Coccidioides

Dirofilaria immitis remains a significant canine pathogen in Brazil despite the long-standing availability of chemoprophylactic products. Higher infection rates in low-income areas, and the persistence of cases in populations with veterinary access, suggest gaps in preventive governance. A cross-sectional survey was conducted in two high-challenge areas of metropolitan Rio de Janeiro, Brazil, from June 2023 to January 2024. Dogs older than 1 year and not on heartworm prophylaxis during the previous 6 months were enrolled. Blood samples were tested for D. immitis antigen, and owner questionnair
