L1 and NCAM adhesion molecules as signaling coreceptors in neuronal migration and process outgrowth.

Neural cell adhesion molecules (CAMs) of the immunoglobulin superfamily engage in multiple neuronal interactions that influence cell migration, axonal and dendritic projection, and synaptic targeting. Their downstream signal transduction events specify whether a cell moves or projects axons and dendrites to targets in the brain. Many of the diverse functions of CAMs are brought about through homophilic and heterophilic interactions with other cell surface receptors. An emerging concept is that CAMs act as coreceptors to assist in intracellular signal transduction, and to provide cytoskeletal l…

The effect of a GABAA agonist muscimol on acoustic injury of the mouse cochlea.

Gamma-aminobutyric acid (GABA) is one of major inhibitory neurotransmitter in the central nervous system and constitutes the cochlear effere … ed. Muscimol significantly decreased the ABR threshold shift and inhibited swelling of the afferent dendrites induced by acoustic overexposure. In addition, bicuculline inhibited the effects of muscimol. These findings suggest that activation of GABA(A) receptors reduces acoustic injury of the cochlea.

Fasciculation and elongation protein zeta-1 (FEZ1) participates in the polarization of hippocampal neuron by controlling the mitochondrial motility.

The fasciculation and elongation protein zeta-1 (FEZ1), a mammalian orthologue of Caenorhabditis elegans UNC-76 protein, is a 45-kDa protein … is considerably elongated by the RNAi treatment. The transport of mitochondria from soma to axon or dendrites is essential for the neuronal differentiation. Therefore, our results strongly suggest that FEZ1 participates in the establishment of neuronal polarity by controlling the mitochondrial motility along axon.

On the effects of psychostimulants, antidepressants, and the antiparkinsonian drug levodopa on dopamine neurons.

The dopaminergic system constitutes the principal target of many psychostimulants, antidepressant, and antiparkinsonian drugs. The effects caused by these compounds are partly associated with an increased dopamine (DA) levels within the terminal areas of DA neurons and in the ventral midbrain. Therefore, several substances of abuse, antidepressants, and endogenous compounds (levodopa and trace amines [TAs]) regulate the activity of DA cells by activating D2 autoreceptors located on the terminals, soma, and dendrites. Considering our past and recent experimental studies on this issue, here we w…

Brain plasticity in paediatric neurology.

Plasticity includes the brain's capacity to be shaped or moulded by experience, the capacity to learn and remember, and the ability to reorganize and recover after injury. Mechanisms for plasticity include activity-dependent refinement of neuronal connections and synaptic plasticity as a substrate for learning and memory. The molecular mechanisms for these processes utilize signalling cascades that relay messages from synaptic receptors to the nucleus and the cytoskeleton to control the structure of axons and dendrites. Several paediatric neurological disorders such as neurofibromatosis-1, Fra…

Correlation of factor XIIIa+ dermal dendrocytes with paracoccidioidomycosis skin lesions.

We demonstrated and quantified by immunohistochemistry the factor XIIIa+ dermal dendrocytes (FXIIIa+ DD) in paracoccidioidomycosis skin lesions. Sixty-one biopsies were classified according to the tissue response in well-organized granulomas (group 1), poorly organized granulomas (group 2) and samples showing both kinds of granuloma (group 3). Ten biopsies from normal skin were used as controls. In order to verify the internalization of Paracoccidioides brasiliensis antigens by FXIIIa+ DD, we performed a double immunostaining technique. FXIIIa+ DD were hypertrophied with prominent dendrites an…

Stimulation of mouse melanocyte proliferation by Piper nigrum fruit extract and its main alkaloid, piperine.

During a herbal screening programme to find potential repigmenting agents for the treatment of vitiligo, Piper nigrum L. fruit (black pepper … igrum extract and piperine induced morphological alterations in melan-a cells, with more and longer dendrites observed. The augmentation of growth by piperine was effectively inhibited by RO-31-8220, a selective protein kinase C (PKC) inhibitor, suggesting that PKC signalling is involved in its activity. This is the first full report on such an activity of …

Axonal calcium entry during fast 'sodium' action potentials in rat cerebellar Purkinje neurones.

1. Using laser-scanning confocal microscopy, fast Ca2+ transients were recorded in individual not yet myelinated axons of Purkinje neurones in cerebellar slices from young rats. Axonal Ca2+ transients could be detected during a single action potential and had progressively larger amplitudes when the number of action potentials was increased. 2. Under voltage-clamp conditions, axonal Ca2+ transients were as large as those observed in dendrites and in the cell body. Axonal Ca2+ transients were completely blocked by 100 nM of the neurotoxin omega-agatoxin IVA, indicating that they were caused by …

Immunohistochemical method for the diagnosis of olfactory disturbance.

To confirm the diagnosis and determine the cause of the olfactory disturbance, we used an immunohistochemical method to examine biopsy specimens of the olfactory mucosa from a patient who complained of anosmia after head injury. Neuron-specific enolase immunoreactivity was found in the olfactory vesicles and dendrites of the receptor cells in the olfactory epithelium. S-100 protein immunoreactivity was found in the ductal cells of Bowman's gland in the olfactory epithelium and in the acinar cells of Bowman's gland in the lamina propria. This suggests that the olfactory receptor cells and Bowma…

Friend or Foe: The Protective and Pathological Roles of Inducible Bronchus-Associated Lymphoid Tissue in Pulmonary Diseases.

Inducible bronchus-associated lymphoid tissue (iBALT) is a tertiary lymphoid structure that resembles secondary lymphoid organs. iBALT is induced in the lung in response to Ag exposure. In some cases, such as infection with Mycobacterium tuberculosis, the formation of iBALT structure is indicative of an effective protective immune response. However, with persistent exposure to Ags during chronic inflammation, allergy, or autoimmune diseases, iBALT may be associated with exacerbation of inflammation. iBALT is characterized by well-organized T and B areas enmeshed with conventional dendritic cel

Facet-selective molecular regulation of Zn metal anodes for durable aqueous zinc-ion batteries.

Aqueous zinc-ion batteries (AZIBs) are attractive for grid-scale energy storage, yet Zn anodes suffer from dendrites, side reactions and restricted current windows. Herein, capecitabine (CAP) is employed as a electrolyte additive for Zn-deposition regulation and interfacial stabilization. The N/O-containing groups in CAP partially reconstruct the Zn2+ solvation and water hydrogen-bond network, reducing water activity. Meanwhile, facet-selective adsorption of CAP on Zn(100), forms an adsorption-mediated interfacial layer that suppresses parasitic reactions and promotes compact Zn(100)-textured …

Kinetics of Stacking-Order Evolution during Heterogeneous Ice Formation.

The selection of stacking order in a broad range of close-packed polymorphic materials remains a challenging enigma. Using in situ cryogenic transmission electron microscopy, we uncover the atomistic mechanisms governing the vapor deposition growth of ice. We find that the heterogeneous ice nucleation and growth undergo recrystallization accompanied by bifurcation, reflecting a coherent epitaxial transition from a cubic-ice embryonic core to hexagonal-ice prismatic dendrites, with intermediate stacking-disordered layers serving as a dynamic fluctuating bridge. Supported by molecular dynamics s…

The Diagnostic Value of Cerebrospinal Fluid Neurogranin in Neurodegenerative Diseases.

Synaptic pathology is crucial in neurodegenerative diseases (NDs), and numerous studies show a correlation between synaptic proteins and the rate of cognitive decline in Alzheimer's disease, Parkinson's disease, dementia, and Creutzfeldt-Jacob's disease. Due to the fact that altered synaptic function is considered a core feature of the pathophysiology of neurodegenerative disorders, synaptic proteins, such as neurogranin, may serve as a biomarker of these diseases. Neurogranin is a postsynaptic protein located in the cell bodies and dendrites of neurons, foremost in the cerebral cortex, hippoc…

The multiple faces of the oxytocin and vasopressin systems in the brain.

Classically, hypothalamic neuroendocrine cells that synthesise oxytocin and vasopressin were categorised in two major cell types: the magnoc … ted. It is now clear that oxytocin and vasopressin in the brain are released extrasynaptically from dendrites and from varicosities in distant axons. The peptides act principally to modulate information transfer through conventional synapses (such as glutamate synapses) by actions at respective receptors that may be preferentially localised to synaptic regi …

Pyramidal neuron growth and increased hippocampal volume during labor and birth in autism.

We report that the apical dendrites of CA3 hippocampal pyramidal neurons are increased during labor and birth in the valproate model of autism but not in control animals. Using the iDISCO clearing method, we show that hippocampal, especially CA3 region, and neocortical volumes are increased and that the cerebral volume distribution shifts from normal to lognormal in valproate-treated animals. Maternal administration during labor and birth of the NKCC1 chloride transporter antagonist bumetanide, which reduces [Cl-]i levels and attenuates the severity of autism, abolished the neocortical and hip…

Presynaptic Expression of LRIT3 Transsynaptically Organizes the Postsynaptic Glutamate Signaling Complex Containing TRPM1.

Throughout the CNS, interactions between pre- and postsynaptic adhesion molecules establish normal synaptic structure and function. Leucine-rich repeat (LRR) domain-containing proteins are a large family that has a diversity of ligands, and their absence can cause disease. At the first retinal synapse, the absence of LRIT3 expression leads to the disassembly of the postsynaptic glutamate signaling complex (signalplex) expressed on depolarizing bipolar cell (DBC) dendrites. The prevalent view is that assembly of the signalplex results from direct postsynaptic protein:protein interactions. In co…

Pre-symptomatic alterations in subcellular betaCGRP distribution in motor neurons precede astrogliosis in ALS mice.

In our study we investigated the pathology-related expression patterns of the two calcitonin gene-related peptide (CGRP) isoforms in spinal cord motor neurons of SOD1(G93A) mice, an animal model of the human motor neuron disease, amyotrophic lateral sclerosis (ALS). We found that alphaCGRP and betaCGRP gene expression and alphaCGRP immunoreactivity remained unaltered throughout disease, and alphaCGRP gene deficiency had no effect on disease progression. In contrast, betaCGRP immunoreactivity appeared at atypical sites in degenerating motor neuron cell bodies, axons, and dendrites already in th…
