Stimulatory release of hepatic lipase activity from liver by epidermal growth factor.

We have found that release of hepatic lipase activity is stimulated from liver slices by epidermal growth factor (EGF) into the medium in a time-dependent manner. This novel effect of EGF was markedly decreased by various tyrosine kinase inhibitors, such as alpha-cyano-3-ethoxy-4-hydroxy-5-phenyl-thiomethyl cinnamamide, amiloride and biochanin A. The stimulation by EGF was also suppressed, however, by dibutyryl cyclic adenosine monophosphate, and 3-isobutyl-1-methylxanthine, a cyclic nucleotide dependent phosphodiesterase inhibitor. These findings show that the stimulatory release of the enzym

Localization of calpain immunoreactivity in senile plaques and in neurones undergoing neurofibrillary degeneration in Alzheimer's disease.

An antibody raised against the calcium activated neutral protease (calpain) was used to investigate the possible involvement of this enzyme in the formation of plaques and tangles in Alzheimer-type dementia (ATD) brain. Our results revealed the presence of a number of strongly stained calpain positive neurones in the normal human cerebral cortex and a loss of calpain positive cells in ATD brain. Furthermore, double staining experiments revealed that calpain immunoreactivity was present in cells undergoing tangle formation, and was also present in senile plaques. These data suggest that activat

A possible new class of octopamine receptors coupled to adenylate cyclase in the brain of the dipterous Ceratitis capitata. Pharmacological characterization and regulation of 3H-octopamine binding.

Octopamine exerts its effects in insects through interaction with at least two classes of receptors, designated octopamine-1 and octopamine-2. Octopamine-2 receptors are positively coupled to adenylate cyclase, while octopamine-1 receptors are not coupled to this enzyme system. Ceratitis capitata brain appears to have octopamine receptors as unique aminergic receptors coupled to adenylate cyclase. These receptors show some pharmacological analogies with respect to octopamine-2 receptors, however they should constitute a new class of octopamine receptors. C. capitata brain octopamine receptors

Trifluoperazine inhibition of fibrinogen receptor redistribution in surface activated platelets: correlative video-enhanced differential interference contrast light microscopic, high voltage electron microscopic and scanning electron microscopic studies.

Video-enhanced differential interference contrast (VDIC) light microscopy in conjunction with fibrinogen labelled colloidal gold was employed as a probe to follow the mobility of the fibrinogen receptor on platelets. Correlative studies by both high voltage and scanning electron microscopy confirms localization of labels relative to platelet ultrastructural and surface characteristics, respectively. Treatment of platelets with trifluoperazine prior to and after incubation with fibrinogen-gold labels results in a concentration dependent inhibition of receptor movement. The results obtained from

Vinyl chloride-induced deactivation of cytochrome P-450 and other components of the liver mixed function oxidase system: an in vivo study.

Liver microsomal enzyme activities were measured 24 hr after a 6 hr exposure to 5% vinyl chloride in male rats pretreated with phenobarbital, Aroclor 1254 or the control vehicle. Liver injury as indicated by serum transaminase elevations after vinyl chloride, was found only in phenobarbital and Aroclor 1254 animals while cytochrome P-450 contents and oxidative N-demethylation of aminoantipyrine and ethylmorphine were markedly decreased in all groups. Glucose-6-phosphatase, cytochrome b5 and NADPH and NADH cytochrome P-450 reductase were scarcely affected. This selective enzymatic deactivation

Synthesis and biological evaluation of a diazepanone-based library of liposidomycins analogs as MraY inhibitors.

New inhibitors of the bacterial tranferase MraY are described. A scaffold strategy based on the diazepanone central core of liposidomycins, natural inhibitors of MraY has been developed. It involves the introduction of key structural fragments required for biological activity on enantiopure diazepanones by reductive amination, esterification and glycosylation. Biological evaluation of these compounds on MraY enzyme revealed interesting inhibitory activity for compounds displaying three fragments on the scaffold: a palmitoyl chain, an aminoribose part and an alkyluracil moiety. The inhibitors w

Biology of the molybdenum cofactor.

The transition element molybdenum (Mo) is an essential micronutrient for plants where it is needed as a catalytically active metal during enzyme catalysis. Four plant enzymes depend on molybdenum: nitrate reductase, sulphite oxidase, xanthine dehydrogenase, and aldehyde oxidase. However, in order to gain biological activity and fulfil its function in enzymes, molybdenum has to be complexed by a pterin compound thus forming the molybdenum cofactor. In this article, the path of molybdenum from its uptake into the cell, via formation of the molybdenum cofactor and its storage, to the final modifi

Microvillar enzyme assays in amniotic fluid and fetal tissues at different stages of development.

A systematic study of microvillar enzyme activities in the amniotic fluid in correlation with their values in different fetal tissues during development has been undertaken. Microvillar enzymes appeared in the amniotic fluid at the time of disappearance of the anal membrane, 12-13 weeks, and declined from the 18th week until the 24th week. The study of fetal tissues and fluids has shown that gamma-glutamyltranspeptidase is mainly of liver origin. The significant decrease of the activities of these amniotic fluid enzymes has been the basis of prenatal diagnosis of cystic fibrosis. These assays

Plasma lecithin: cholesterol acyltransferase in copper-deficient rats.

Some groups of people with high risk of ischemic heart disease have low lecithin:cholesterol acyltransferase activity in plasma and vice versa. Because we hypothesized a relationship between inadequate copper nutriture and the risk of ischemic heart disease, we measured plasma lecithin: cholesterol acyltransferase in copper deficient male Sprague-Dawley rats. Deficiency was verified by the presence of anemia, hypercholesterolemia and low copper concentrations in kidney and skeletal muscle. Three experiments showed a significant decrease (22-32% reduction) in enzyme activity in deficiency. Copp

Induction of tyrosine hydroxylase by cyclic AMP and glucocorticoids in a rat pheochromocytoma cell line: effect of the inducing agents alone or in combination on the enzyme levels and rate of synthesis of tyrosine hydroxylase.

The enzymatic activity of tyrosine hydroxylase (EC 1.14.16.2) increases in rat pheochromocytoma PC18 cells exposed to either elevated levels of cyclic AMP or glucocorticoids. The cyclic AMP-mediated increase in activity is elicited by cyclic AMP analogs or by compounds which activate adenylate cyclase or inhibit phosphodiesterase. The glucocorticoid-mediated increase is elicited only by glucocorticoid steroid hormones; nonglucocorticoid steroid hormones have no effect on tyrosine hydroxylase. In PC18 cells exposed simultaneously to both cyclic AMP-elevating agents and glucocorticoids, the incr

Inhibition effects and binding behavior of alginate oligosaccharides on tyrosinase and elastase: multi-spectroscopic approaches and zebrafish evaluation.

Alginate oligosaccharides (AOSs) are natural marine oligosaccharides with diverse physiological functions, which have broad application prospects in both the cosmeceutical and food industries. This study systematically investigated the inhibitory mechanisms of AOSs against tyrosinase and elastase, two key enzymes associated with skin aging, by combining multi-spectroscopic approaches with zebrafish in vivo evaluation. Results demonstrated that AOSs acted as a reversible mixed-type inhibitor for both enzymes, exhibiting moderate inhibitory activities with IC50 values of 7.37 ± 0.15 mM for tyros

Rational Thermostabilization of the Fe(II)/α-Ketoglutarate-Dependent Halogenase BesD While Preserving Halogenation Selectivity.

Fe(II)/α-ketoglutarate (αKG)-dependent halogenases that catalyze site-selective C-H halogenation of free substrates without carrier proteins are attractive biocatalysts for diversifying pharmaceuticals and agrochemicals. However, their application remains limited by the narrow diversity of natural halogenases, poor stability, and restricted substrate scope. Protein stabilization is a common strategy to enhance mutational tolerance during enzyme engineering; however, Fe(II)/αKG-dependent halogenases are structurally closely related to hydroxylases, and consensus-based stabilizing mutations risk

Pancreatic Lipase in Eutectogels as Emerging Materials: Exploring Their Properties and Potential Applications in Biosensing.

Eutectogels are advanced gel-based systems that integrate deep eutectic solvents (DES) into polymer networks. In this study, we report the first detailed characterization of an enzyme-containing eutectogel, representing a significant step toward advanced biosensing and biocatalytic applications. Specifically, we have incorporated pancreatic lipase, one of the main target enzymes in the treatment of obesity, in eutectogels via UV-induced radical polymerization of suitable precursors in appropriate DESs. Prior to immobilization, the enzyme was solubilized in selected DESs and its activity and co

Thiomandelic acid, a broad spectrum inhibitor of zinc beta-lactamases: kinetic and spectroscopic studies.

Resistance to beta-lactam antibiotics mediated by metallo-beta-lactamases is an increasingly worrying clinical problem. Candidate inhibitors include mercaptocarboxylic acids, and we report studies of a simple such compound, thiomandelic acid. A series of 35 analogues were synthesized and examined as metallo-beta-lactamase inhibitors. The K(i) values (Bacillus cereus enzyme) are 0.09 microm for R-thiomandelic acid and 1.28 microm for the S-isomer. Structure-activity relationships show that the thiol is essential for activity and the carboxylate increases potency; the affinity is greatest when t

Antioxidant defences in the microaerophilic protozoan Trichomonas vaginalis: comparison of metronidazole-resistant and sensitive strains.

The sensitivity of the microaerophilic protozoan Trichomonas vaginalis to oxygen and products of its reduction, and the antioxidant defences employed by this organism, were investigated. Studies revealed that this amitochondrial flagellate is sensitive to oxygen tensions above those experienced in situ in the vagina (i.e. > 60 microM) and that metronidazole-resistant strains (CDC 85 and IR78) were more sensitive to elevated oxygen levels than a metronidazole-sensitive isolate (1910). In the presence of radical scavengers, inactivation of organisms at 60 microM oxygen was significantly lessened

Impacts of corn stover management and fertilizer application on soil nutrient availability and enzymatic activity.

Corn stover is a global resource used in many industrial sectors including bioenergy, fuel, and livestock operations. However, stover removal can negatively impact soil nutrient availability, especially nitrogen (N) and phosphorus (P), biological activity, and soil health. We evaluated the effects of corn stover management combined with N and P fertilization on soil quality, using soil chemical (nitrate, ammonium and Bray-1 P) and biological parameters (β-glucosidase, alkaline phosphatase, arylsulfatase activities and fluorescein diacetate hydrolysis-FDA). The experiment was performed on a Mol

Strain-differences and inducibility of microsomal oxidative enzymes in Drosophila melanogaster flies.

Some basic characteristics of the enzyme system involved in the oxidative metabolism of xenobiotic compounds were investigated in Drosophila melanogaster flies. Attention was focussed on (1) the normal levels of these enzymes and their activities in whole flies, in different parts of the fly's body and in different sexes, (2) the changes in levels and activities of the enzymes elicited by pretreatment of the flies with known enzyme inducers and (3) differences between strains. Four commonly used wild-type (WT) strains, three insecticide resistant strains (IR) and one white-eyed mutant strain w

Treatment of atypical hemolytic uremic syndrome and C3 glomerulopathy in mice by hepatic expression of factor D.

Atypical hemolytic uremic syndrome (aHUS) and C3 glomerulopathy (C3G) are diseases driven by dysregulation of the alternative pathway (AP) complement. Current treatments of these diseases involve the use of frequent and often large doses of monoclonal antibodies, peptide inhibitors, or chemical inhibitors of complement proteins, with various limitations. In this study, we describe ectopic expression of factor D (FD) in the liver as a potential novel therapy for these indications. Unlike most plasma complement proteins, which are synthesized in the liver, FD, a serine protease in the AP, is syn

Paeonol alleviates atherosclerosis by inhibiting CD8+ T-cell activation via targeting the SYK/NFATc1 signaling pathway in ApoE-/- mice.

ETHNOPHARMACOLOGICAL RELEVANCE: Moutan Cortex, a traditional Chinese medicine, has been used to treat cardiovascular diseases. Paeonol (Pae), a key bioactive compound, is responsible for its anti-atherosclerotic effects. Although CD8+ T-cell activation drives atherosclerosis, Pae's role in this process remains unclear. AIM OF THE STUDY: We investigated whether Pae inhibits atherosclerosis by targeting the spleen tyrosine kinase (SYK)/nuclear factor of activated T-cells c1 (NFATc1) pathway, thereby reducing CD8+ T-cell activation and attenuating vascular endothelial cell injury. MATERIALS AND
