Imageomics defines granular morphological changes of human skin with age and reveals a rejuvenating effect of xenografting.

Rejuvenating aging human skin is a major therapeutic goal, but objective, quantitative measures of intrinsic aging are limited. We performed a cross-sectional histological study of UV-protected buttock and abdominal skin in adults spanning multiple decades of life to identify features that reliably track age. Epidermal thickness measured between rete ridges was unchanged, but rete ridge size declined linearly with age: ridges became shorter and thinner in both sites, though rete ridge number decreased only in the abdomen. Consistent with these structural changes, proliferative cells (Ki67+) pe

Combined blockade of VEGFR-3 and Itga-9 inhibits corneal lymphangiogenesis and valvulogenesis in vivo and promotes high-risk transplant survival.

PURPOSE: Corneal transplantation is the last hope for vision restoration for corneal blindness. The rejection rate of high-risk grafts can be 50-90%, irrespective of current treatment modalities. As VEGFR-3 (vascular endothelial growth factor receptor-3) is a central regulator of lymphatic vessel formation (lymphangiogenesis, LG), integrin alpha9beta1 (Itga-9) is essential for lymphatic maturation by luminal valve formation (valvulogenesis, VG). This study is to investigate the combined blockade of VEGFR-3 and Itga-9 on high-risk corneal transplant rejection. MATERIALS AND METHODS: High-risk

Plasma proteomic profiles of patients with atopic dermatitis with moderate-to-severe pruritus treated with a single dose of nemolizumab.

Nemolizumab demonstrates efficacy against pruritus and eczema by inhibiting IL-31 signaling in patients with atopic dermatitis. However, its effect on the systemic immune response at the molecular level remains unknown. In this study, we aimed to elucidate it by investigating plasma proteins and pathways modulated by nemolizumab under concomitant topical treatment. Plasma protein profiling was conducted for 25 Japanese patients with atopic dermatitis who received a single 60 mg dose of nemolizumab at baseline and 1, 2, 4, and 8 weeks after administration using the SomaScan 7k assay. Proteome d

Inflammatory Bowel Disease: Mechanisms and Therapeutic Advances.

Inflammatory bowel disease (IBD), which includes ulcerative colitis and Crohn's disease, represents a chronic, immune-mediated pathology in which genetic susceptibility, environmental exposures, and microbial perturbations converge to destabilize intestinal homeostasis. Despite a steady global rise in incidence affecting more than 10 million people worldwide, current single-pathway biologic and small-molecule therapies still leave 30-40% of patients without durable remission, with progression to stricturing, fistulizing, or transmural complications. Yet the mechanistic integration of innate an

Platelet Casein Kinase 2α is a pivotal player in arterial thrombotic occlusion and post-ischemic myocardial remodeling.

AIMS: Platelets play a major role in thrombo-inflammatory cardiovascular diseases such as myocardial infarction. Although platelet function is crucially determined by kinases, the impact of Casein Kinase 2α (CK2α) on platelet activation during arterial thrombosis and myocardial remodeling following ischemia and reperfusion (I/R) injury is not known. METHODS AND RESULTS: Using platelet-specific deletion of Csnk2a1 in mice, the evaluation of the CK2α-dependent platelet phosphoproteome revealed a diminished phosphorylation of the IP3 receptor type-1 in Csnk2a1-deficient mice. This finding was ac

Exploring the role of differential splicing of Pekin ducks resistant to DHAV-3 based on full-length transcriptome.

Duck Hepatitis A Virus type 3 (DHAV-3) causes high mortality in Pekin ducks, and the role of post-transcriptional regulation in disease resistance remains unclear. Here, we performed Oxford Nanopore full-length transcriptome sequencing on liver samples from resistant and susceptible ducks at 0 h and 24 h after DHAV-3 infection. Only one differentially expressed gene (DEG) and four differentially expressed transcripts (DETs) were identified pre-infection (0 h), whereas post-infection (24 h) we detected 2,539 DEGs and 2,909 DETs, indicating strong group-specific expression divergence. Functional

Matrix Viscoelasticity Regulates the Stemness and Multilineage differentiation of Primary Neural Progenitor-Stem Cells in 3D.

Neural progenitor-stem cells (NPSCs) reside in mechanically dynamic brain microenvironments and give rise to neurons, astrocytes, and oligodendrocytes. Although recent studies have shown that matrix viscoelasticity can influence neural maturation and neurogenic differentiation, its role in primary NPSCs beyond neurogenesis remains less defined. How matrix viscoelasticity or stress relaxation regulates primary NPSC stemness, neuronal and glial differentiation, and the associated matrix-cell mechanotransduction pathways are not well understood. Here, we use alginate hydrogels with independently

Platelet transfusion refractoriness in Glanzmann thrombasthenia: a case report with literature review.

INTRODUCTION: Glanzmann thrombasthenia (GT) is a rare inherited platelet function disorder characterized by a deficiency or dysfunction of the αIIbβ3 integrin, leading to impaired platelet aggregation. Platelet transfusion refractoriness (PTR) represents a therapeutic challenge, often resulting from alloimmunization against HLA antigens or the αIIbβ3 complex. METHODS: We report the case of a patient with type I GT, who presented with recurrent mucosal hemorrhages and chronic anemia requiring repeated transfusions. RESULTS: By the age of 8 years, the patient developed transfusion refractorine

Hypoxia drives ANGPTL4 expression in tumor-associated macrophages and promotes intrahepatic cholangiocarcinoma intrahepatic metastasis.

Intrahepatic metastasis (IM) is a critical predictor of poor prognosis in intrahepatic cholangiocarcinoma (iCCA), yet the tumor microenvironmental mechanisms driving this process remain poorly understood. By integrating single-cell RNA sequencing data from 15 iCCA samples with a public transcriptomic cohort of 255 patients (OEP001105), we characterized the immune and malignant epithelial landscapes associated with IM. We identified ANGPTL4-positive tumor-associated macrophages and hypoxia-associated malignant epithelial cells that were enriched in the IM group. Mechanistically, hypoxia induced

Fenofibrate attenuates hyperhomocysteinemia-potentiated thrombosis by restoring platelet fatty acid β-oxidation.

Hyperhomocysteinemia (HHcy) is an independent risk factor for thrombotic cardiovascular events. We previously demonstrated that homocysteine (Hcy) amplifies platelet activation by promoting membrane remodeling and enhancing signaling through surface platforms such as integrin αIIbβ3 and G protein-coupled receptors. However, the mechanisms by which Hcy remodels platelet membrane lipid metabolism remain poorly understood. Here, using an integrated proteomic and lipidomic approach, we showed that Hcy disrupted platelet lipid homeostasis by impairing fatty acid β-oxidation (FAO), a metabolic pathw

Mechanobiology and Molecular Regulation of Periodontal Tissue Remodeling During Orthodontic Tooth Movement: From Periodontal Phenotype to Precision Orthodontics-A Narrative Review.

Orthodontic tooth movement has traditionally been planned according to biomechanical principles; however, growing evidence indicates that treatment outcomes are strongly influenced by individual biological responsiveness. This narrative review summarizes current knowledge on the mechanobiological and molecular mechanisms regulating periodontal tissue remodeling during orthodontic tooth movement and discusses their implications for personalized orthodontic treatment. The literature was critically analyzed with particular emphasis on mechanotransduction pathways, including Piezo1, TRPV4, integri

DAB2IP: unifying cardiovascular pathogenesis and cardiovascular brain crosstalk.

Cardiovascular diseases and brain disorders collectively represent a major global health burden. Not only do they frequently occur as comorbidities, but their pathological mechanisms are also intricately linked through a complex network of cardiovascular-brain crosstalk. Cardiovascular diseases (CVDs) and neurological disorders together impose high global health burdens. The DAB2IP gene, initially characterized as a tumor suppressor, serves as a multifunctional regulator and performs roles in both the cardiovascular and neurological systems. In atherosclerosis (AS), DAB2IP suppresses the infla

Beyond Hemostasis: Platelet Biomarkers and Vascular-Immune Crosstalk in Dengue.

Dengue is an arboviral disease with major global public health impact. Its pathophysiology involves complex interactions between the virus and the host, including the immune response and the vascular endothelium. In this context, platelets play roles beyond hemostasis, acting as key immunomodulatory cells involved in inflammation and the regulation of vascular permeability. Therefore, this review summarizes and discusses the so called classic and non-classic platelet-related and endothelial biomarkers described in dengue virus infection. Classical biomarkers include P-selectin (CD62P), PF4/CXC

CALR-dependent endoplasmic reticulum N-glycoprotein maturation promotes Japanese encephalitis virus entry and replication.

The endoplasmic reticulum (ER) chaperone machinery participates in multiple stages of viral infection, from entry through assembly and release. However, how ER-resident quality-control mechanisms shape cellular permissiveness to Japanese encephalitis virus (JEV) remains unclear. Here, we identify the ER-resident chaperone calreticulin (CALR) as a key host factor that promotes cellular permissiveness to JEV infection. CRISPR/Cas9-mediated knockout of CALR markedly reduces JEV attachment, internalization, and productive replication. Mechanistically, CALR depletion disrupts N-linked glycosylation

ITGA4 drives pathogenesis of flavivirus and inhibition of it protects mice against Japanese encephalitis virus infection.

Multiple studies have revealed a potential role for integrin α4 (ITGA4) in the pathogenesis of SARS-CoV-2/HIV/SIV infection, but its role in flavivirus infection remains elusive. Here, we used Japanese encephalitis virus (JEV) as a model, aiming to dissect the regulatory role of ITGA4 in flavivirus infection. We found that JEV infection activates vitamin A metabolism, which in turn induces the upregulation and activation of ITGA4. ITGA4 triggers cytoskeletal reorganization enhancing early viral replication in monocytes via the ITGA4-HSP70-pFAK-Rac1 signaling axis, and reinforces interactions b

The immunophenotype and proviral landscape of HIV-infected CD4 T cells during antiretroviral therapy.

In individuals on effective antiretroviral therapy (ART), integrated HIV proviruses persist within CD4 T cells, forming a viral reservoir that rebounds if treatment is stopped. Identifying and targeting these rare, infected cells is critical for advancing therapies, but methods to study reservoir cells are limited, and their unique properties remain largely unknown. We applied DAb-seq, a high-throughput method that combines single-cell DNA and surface protein sequencing, to profile ~527,000 CD4 T cells from the blood of six individuals on ART. Infected cells were distributed across all CD4 T c

Engineering In Vitro Models of Skeletal Muscle With Neuromuscular Junctions Using Hierarchical Micro-Nano Biomaterials: Cooperative Effect of Adhesion Ligand Nanoclustering and Surface Anisotropy.

The development of high-fidelity in vitro contractile skeletal muscle containing neuromuscular junctions (NMJs) is an unmet challenge, largely because current systems cannot accurately reproduce hierarchical micro- to nano-scale biointerfacial cues of the native extracellular matrix. This limitation impairs tissue regeneration and limits the clinical relevance of existing models for drug screening and studies of disease pathophysiology. Herein, we present hierarchical, anisotropic biomaterials that induced early maturation of myotubes and NMJ development during co-culture with motor neurons. W

Anticancer Peptides: Design Principles, Translational Bottlenecks, and Emerging Opportunities.

Anticancer peptides (ACPs) are frequently discussed as a single therapeutic class, yet the term encompasses molecules that perform markedly different jobs: direct tumor-cell killing, intracellular target modulation, tumor homing and penetration, or selective delivery of a separate payload. This functional diversity creates opportunity, but it also obscures why many potent peptides fail during translation. In this review, we organize ACPs according to their intended pharmacologic role and examine the molecular and product-development principles that determine whether an active sequence can beco

Regulation of Adult Zebrafish Retinal Regeneration by Lamβ1b-Chain-Containing Laminins.

Retinal degenerative diseases are a major cause of blindness in humans that often result in permanent and progressive loss of vision. Unlike humans, zebrafish possess the remarkable ability to regenerate lost retinal neurons through Müller glia (MG) reprogramming and asymmetric cell division to produce multipotent retinal progenitor cells (RPCs). While most studies on the molecular mechanisms underlying this regeneration process have focused on intracellular mechanisms, the role of the microenvironment surrounding retinal cells, the extracellular matrix (ECM), has been understudied. Laminins a
