Cell adhesion molecules in neural plasticity and pathology: similar mechanisms, distinct organizations?

Brain plasticity and the mechanisms controlling plasticity are central to learning and memory as well as the recovery of function after brain injury. While it is clear that neurotrophic factors are one of the molecular classes that continue to regulate brain plasticity in the adult central nervous system (CNS), less appreciated but equally profound is the role of cell adhesion molecules (CAMs) in plasticity mechanisms such as long term potentiation, preservation of neurons and regeneration. Ironically, however, CAMs can also reorganize the extra-cellular space and cause disturbances that drive

Pregnancy outcome and neurodevelopment of children exposed in utero to psychoactive drugs: the Motherisk experience.

This paper presents an overview of the Motherisk Program data on pregnancy outcome and neurodevelopment of children exposed in utero to selected psychoactive drugs. First, the use of cocaine during pregnancy has been associated with increased risk of spontaneous abortions, abruptio placenta, premature labor, and stillbirth. Twenty-three adopted children exposed in utero to cocaine demonstrated an 8-fold increase in risk for microcephaly compared with controls. Global intelligence quotients (IQ) did not differ between the 2 groups, but the cocaine-exposed children achieved significantly lower s

Rifaximin, a non-absorbable rifamycin, for the treatment of hepatic encephalopathy. A double-blind, randomised trial.

The aim of this study was to evaluate the efficacy and tolerability of rifaximin, a non-absorbable intestinal antibiotic, in comparison to neomycin in the short- and long-term treatment of hepatic encephalopathy (HE). Forty-nine patients with a definite diagnosis of cirrhosis were included in this double-blind, randomised, controlled trial. Patients were randomly assigned to one of the following treatments: (1) rifaximin 400 mg three times daily; (2) neomycin 1 g three times daily. Both drugs were administrated orally as tablets during 14 consecutive days each month, for a period of six months

Neuroligand-evoked calcium-dependent release of excitatory amino acids from cultured astrocytes.

The release of excitatory amino acids (EAAs) from neuron-free cultures of neocortical astrocytes was monitored using HPLC. The neuroligand bradykinin caused a dose-dependent receptor-mediated increase in release of the EAAs glutamate and aspartate from type 1 astrocyte cell cultures obtained from rat cerebral cortex. Removal of calcium from the extracellular fluid prevented the bradykinin-induced release of EAAs from astrocytes. The addition of the calcium ionophore ionomycin caused a calcium-dependent release of EAAs. Inhibitors of the glutamate transporters p-chloromercuriphenylsulfonic acid

Long-term immunity to measles, mumps, and rubella after allogeneic bone marrow transplantation.

A total of 124 patients who had survived at least 2 years after allogeneic bone marrow transplantation (BMT) were studied. Serum was collected at least once yearly. IgG antibodies were determined by enzyme-linked immunosorbent assay for measles and mumps. Rubella antibodies were analyzed by radial hemolysis. Antibody levels were interpreted as representing immunity, seronegativity, or seropositivity, but with uncertain immunity. The median follow-up of the patients was 6.5 years (range, 2 to 13.5 years). The calculated probabilities of being immune to measles at 3, 5, and 7 years from BMT were

Molecularly nanoengineered dendritic cells for tumor-specific antigen capture and sono-activatable cytokine-mediated immunotherapy.

Engineering artificial dendritic cells (DCs) holds great promise for cancer immunotherapy, but achieving spatiotemporally controlled multifunctionality within the immunosuppressive tumor microenvironment remains a challenge. Here, we report a chemically engineered, ultrasound (US)-activatable artificial DCs nanosystem (nDC-M) that mimics the DC-mediated T-cell immunity through a rationally designed molecular assembly. The nDC-M integrates three key functional modules including a semiconducting polymer nanoparticle and interleukin-2 (IL-2) crosslinked by hypoxia-cleavable fragment for on-demand

Robust Demodulation of eLoran Tri-State PPM via Multi-Scale Spatiotemporal Modeling.

Enhanced Loran (eLoran) provides a terrestrial complement to Global Navigation Satellite Systems (GNSSs), but tri-state pulse position modulation (PPM) demodulation remains difficult under composite interference. This study proposes a label-independent Nominal-Center (NC) sampling strategy and a Multi-Scale Spatiotemporal Network (MSST-Net). NC extracts each pulse around its predicted nominal timing anchor, thereby avoiding target-dependent actual-center alignment while preserving the displacement that encodes the PPM state. MSST-Net combines multi-scale one-dimensional convolutions, Squeeze-a

TL-CRANN: a transfer-learning-based hybrid CNN-RNN model with attention mechanism for cross-domain IoT attack detection.

INTRODUCTION: The rapid and pervasive growth of Internet of Things (IoT) devices has significantly expanded the attack surface of current network environments, making them vulnerable to advanced and evolving cyber threats. However, current intrusion detection systems (IDS) usually suffer from weak cross-domain generalization, high false positive rates, and poor performance against novel attacks. METHODS: To overcome these issues, this paper proposes TL-CRANN, a transfer-learning-based hybrid deep learning framework utilizing a convolutional neural network (CNN), bidirectional long short-term

Corrected Version: Sodium Benzoate, A Metabolite of Cinnamon and A Food Additive, Improves Cognitive Functions in Mice After Controlled Cortical Impact Injury.

Traumatic brain injury (TBI) is a major health concern, sometimes leading to long-term neurological disability, especially in children, young adults, and war veterans. Although the research investigators and clinicians have applied different treatment strategies or neurosurgical procedures to solve this health issue, we are still in need of effective therapy to halt the pathogenesis of brain injury. Earlier we have reported that sodium benzoate (NaB), a metabolite of cinnamon and a Food and Drug Administration-approved drug against urea cycle disorders and glycine encephalopathy, protects neur

Load-Decomposition Learning Enhances Ground Reaction Force Estimation Using IMUs.

Accurate estimation of ground reaction forces (GRFs) is fundamental to gait analysis and supports functional gait assessment, surgical planning, and rehabilitation. We propose a physics-learning hybrid framework that combines linked-segment dynamics with a bidirectional long short-term memory network to estimate vertical GRFs (vGRFs) from seven inertial measurement units (IMUs). The learning module produces an unconstrained allocation variable that decomposes the physics-derived net force into limb-specific vGRFs. The framework was systematically evaluated against machine learning-based baseli

Machine Learning-Based Estimation of Knee Joint Mechanics from Kinematic and Neuromuscular Inputs: A Proof-of-Concept Using the CAMS-Knee Datasets.

This study explores the feasibility of estimating tibiofemoral joint contact forces using deep learning models trained on in vivo biomechanical data. Leveraging the comprehensive CAMS-Knee datasets, we developed and evaluated two machine learning network architectures, a bidirectional Long Short-Term-Memory Network with a Multilayer Perceptron (biLSTM-MLP) and a Temporal Convolutional Network (TCN) model, to predict medial and lateral knee contact forces (KCFs) across various activities of daily living. Using a leave-one-subject-out validation approach, the biLSTM-MLP model achieved root mean

Progress on exercise therapy in type 2 diabetes mellitus with cognitive impairment.

Patients with type 2 diabetes mellitus (T2DM) face a significantly elevated risk of developing cognitive impairment (CI), which has been recognized as an independent risk factor for dementia. Current glucose-lowering medications are limited by poor central nervous system penetration, delayed intervention, and single-target approaches, highlighting an urgent need for safe and effective complementary strategies. Exercise therapy, leveraging its advantage in "metabolic-neural bidirectional regulation," demonstrates considerable potential in ameliorating T2DM-related CI. This article systematicall

Kras G12C- and G12D-driven lung cancers differ in oncogenic potency, immunogenicity, and relapse after Kras inhibition in mouse models.

The development of allele-specific KRAS inhibitors underscores the importance of understanding the distinct tumor biology associated with common KRAS mutations, G12D and G12C, in genetically engineered mouse models (GEMMs) of non-small cell lung cancer (NSCLC) and patient samples. Lung tumors driven by the most common KRAS mutation, G12C, show delayed onset and slower progression compared with those driven by KRAS G12D in patients and mice. G12C tumors display lower proliferation and increased immune cell engagement, the latter of which is consistent with observations in patient tumors. Allele

Smart Biosensing Nanomaterials for Alzheimer's Disease: Advances in Design and Drug Delivery Strategies to Overcome the Blood-Brain Barrier.

Alzheimer's disease (AD) is a progressive neurodegenerative disorder marked by persistent memory impairment and complex molecular and cellular pathological changes in the brain. Current treatments, including acetylcholinesterase inhibitors and memantine, only help with symptoms for a short time and do not stop the disease from getting worse. This is mainly because these drugs do not reach the brain well and are quickly removed from the body. The blood-brain barrier (BBB) restricts the entry of most drugs into the central nervous system; therefore, new methods of drug delivery are needed. Nanot

Advances and challenges of precision epigenetic therapy in treating genomic imprinting diseases.

Genomic imprinting disorders (IDs) arise from the disruption of parent-of-origin-specific gene expression, a process governed by heritable epigenetic marks. Conventional therapies are largely symptomatic and fail to correct the underlying defect in dynamic gene regulation. Epigenome editing, using programmable DNA-targeting tools to rewrite epigenetic information, has emerged as a powerful therapeutic strategy that directly addresses this molecular pathology. This review synthesizes the transformative progress of the past decade, arguing that epigenome editing is poised to revolutionize the tr

Research progress on 40 Hz sensory stimulation for the treatment of Alzheimer's disease.

Alzheimer's disease (AD) is a prevalent neurodegenerative disorder characterized by β-amyloid (Aβ) deposition, tau protein hyperphosphorylation, and synaptic dysfunction. In recent years, 40 Hz sensory stimulation-including visual, auditory, and multimodal modalities-has emerged as a novel, non-invasive intervention demonstrating potential efficacy in both animal models and preliminary clinical studies. Preclinical evidence indicates that such stimulation can markedly reduce cerebral Aβ burden (by approximately 37%-53%), inhibit tau protein phosphorylation, enhance neuronal network synchrony a

Identification and validation of a prognostic risk-scoring model in drug-resistant ALL and evaluation of immune micro-environment.

ALL is the commonest childhood hematological malignancy; modern multi-agent chemotherapy cures about 80% of patients, yet roughly 20% experience primary or secondary chemotherapy resistance that sharply reduces long-term survival. The molecular circuits driving this resistance remain poorly defined, and validated biomarkers for early risk stratification are lacking. Growing data implicate reprogramming of the immune microenvironment as a critical determinant of treatment response and relapse risk. We therefore interrogated transcriptomic profiles from GEO and TARGET datasets to identify genes

Biological implications and therapeutic potential of phosphodiesterase inhibitors: A review.

Phosphodiesterase (PDE) inhibitors regulate cyclic adenosine monophosphate and cyclic guanosine monophosphate pathways, which influence neurodevelopment, cardiovascular function, and immune responses. Multiple PDE families exist, classified as dual-substrate (PDE1, PDE2, PDE3, PDE10, PDE11) or non-dual-substrate (PDE4, PDE5, PDE6, PDE7, PDE8, PDE9), each with distinct biological roles. This review summarizes the therapeutic applications of PDE inhibitors, evaluates evidence across different disease domains, and highlights challenges and future research priorities.
