Autologous bone marrow stromal cells loaded onto porous hydroxyapatite ceramic accelerate bone repair in critical-size defects of sheep long bones.

The ability of marrow-derived osteoprogenitor cells to promote repair of critical-size tibial gaps upon autologous transplantation on a hydroxyapatite ceramic (HAC) carrier was tested in a sheep model. Conditions for in vitro expansion of sheep bone marrow stromal cells (BMSC) were established and the osteogenic potential of the expanded cells was validated. Ectopic implantation of sheep BMSC in immunocompromised mice led to extensive bone formation. When used to repair tibial gaps in sheep, cell-loaded implants (n = 2) conducted a far more extensive bone formation than did cell-free HAC cylin…

Efficacy of an outer membrane protein of Pasteurella haemolytica A2, A7 or A9-enriched vaccine against intratracheal challenge exposure in sheep.

The outer membrane proteins (OMP) were extracted from the P. haemolytica A2, A7 and A9 to determine their potential as immunogens and their capability for cross-protection. Sixty lambs of approximately 9 months old were divided into four main groups. Animals in Group 1 were vaccinated with 2ml vaccine containing 100microg/ml of the outer membrane proteins of P. haemolytica A2. Animals in Group 2 were similarly vaccinated with the OMPs of P. haemolytica A7 while Group 3 with OMPs of P. haemolytica A9. Animals in Group 4 were unvaccinated control. During the course of the study, serum was collec

Impairment of gas exchange due to alveolar oedema during xylazine sedation in sheep; absence of a free radical mediated inflammatory mechanism.

We studied the mechanism of impairment of gas exchange following sedation with the alpha2 adrenoreceptor agonist, xylazine, in Suffolk cross-bred sheep spontaneously breathing room air. Xylazine caused a significant fall in PaO2 from a mean (pre-xylazine) of 97.9 mm Hg (6.7 mm Hg SEM) to a mean of 38.1 mm Hg (3.2 mm Hg SEM) one minute after injection with a transient increase in PaCO2 from a mean (pre-xylazine) of 32.6 mm Hg (1.9 mm Hg SEM) to a mean of 40.2 mm Hg (3.0 mm Hg SEM). There was no significant fall in mean arterial pressure or in white cell count. There was no significant change in…

Human thymus lymphoid tissue (HTL) antigen, complement receptor and rosette formation with sheep erythrocytes of the lymphocytes from primary immunodeficiency diseases.

The presence of human thymus lymphoid tissue (HTL) antigen, which we have proposed as a marker of thymus-derived lymphocytes (T cells) in human beings, complement receptor and rosette formation with sheep erythrocytes were tested on the lymphocytes from various immunodeficiency diseases. It is postulated that both T and B cells are reduced in severe combined immunodeficiency, B but not T cells are defective in infantile X-linked agammaglobulinaemia, and both T and part of B cell populations (IgA producers) are impaired in ataxia telangiectasia. The HTL antigen-positive cells (HTLC) were reduce…

Establishment and in vitro validation of a sheep-specific, low-cost TLR7/8 agonist R848-mediated X/Y sperm separation system.

Sex control is crucial for maximizing the economic benefits of sheep husbandry: male lambs increase farm income by 30-40% due to superior growth and feed efficiency, while female lambs are essential for fine-wool production and population expansion. However, the only commercialized method (flow cytometry) has critical sheep-specific limitations, including 25-30% flow channel blockage, sperm DNA oxidative damage, and prohibitive cost for large-scale breeding. TLR7/8 agonist R848 has been validated for sperm separation in mice, cattle and goats, but no sheep-specific optimized system has been es…

RFRP-3 suppresses ovarian granulosa cell function in Tan sheep via dual signaling pathways: PI3K/AKT/mTOR and MAPK/ERK1/2.

The inhibitory role of RFRP-3 in the hypothalamic-pituitary axis is established, but its direct intra-ovarian actions remain poorly defined. This study reveals, for the first time, the expression of RFRP-3 and its receptor GPR147 in ovarian granulosa cells of Tan sheep. We demonstrate that RFRP-3 acts locally to suppress steroidogenesis by downregulating key steroidogenic enzymes (STAR, CYP19A1, HSD17B2). Furthermore, it inhibits proliferation and promotes apoptosis by modulating cell-cycle regulators (PCNA, CCND1, CDK4, CDK1, CCNB) and apoptosis-related proteins (Bcl-2, Bax, p53). A pivotal a…

Integrative transcriptomics and metabolomics analyses reveal changes in meat quality and muscle lipid metabolism in sheep supplemented with rumen-protected glucose.

The effects of dietary rumen-protected glucose (RPG) supplementation on Dumengsa sheep growth performance, meat quality, and transcriptomic and metabolomic profiling are reported. Twelve sheep were randomly assigned to a control (basal diet, n = 6) or RPG (basal diet +1.0% RPG, n = 6) group for 100 d. RPG increased serum malondialdehyde (P = 0.015) and cholesterol (P = 0.046) concentrations, enhanced intramuscular fat content (P = 0.016), and tended to produce lower meat lightness (P = 0.072), redness (P = 0.053), and hue angle (P = 0.072) values. In total, 319 differentially expressed genes a…

GMS-YOLO11n: A Sheep Detection Model for Challenging Fixed-View Farm Conditions Integrating Spatially Gated Structural Enhancement and Multi-Scale Attention.

Accurate sheep detection supports counting, tracking, behavior analysis, and health monitoring in intelligent livestock farming. However, occlusion, scale variation, nighttime low light, and background interference can cause missed detections and inaccurate localization. This study proposes GMS-YOLO11n, an improved YOLO11n-based detector for fixed-view sheep monitoring. A spatially gated bottleneck convolution module was introduced at key backbone downsampling stages to strengthen local structural cues, while a multi-scale attention fusion module combined deep semantic information with shallow…

Differences in meat and wool quality and gastrointestinal/hypothalamic functions in Dexin fine-wool meat sheep with divergent residual feed intake.

Residual feed intake (RFI) is an important indicator for measuring feed efficiency and can be used to screen sheep populations with low feed consumption and excellent growth performance. Its mechanism is closely related to the hypothalamus, which regulates animal feeding behavior, as well as the rumen and duodenum, which are responsible for the main digestive and absorptive functions. In theory, the hypothalamus-gastrointestinal axis is one of the key pathways affecting feed efficiency in ruminants. However, existing research has focused on the gastrointestinal microbiota and lacks systematic …

Noninvasive assessment of fetal metabolic acidemia by automated analysis of cardiac function using color tissue Doppler imaging in a sheep model.

Reliable noninvasive methods for accurately diagnosing fetal metabolic acidemia are still lacking. Our aim was to evaluate an ultrasound-based noninvasive method for assessing metabolic acidemia by an automated analysis of cardiac motion using color tissue Doppler imaging (cTDI) of the fetal heart in an experimental sheep model. Ten pregnant sheep and their fetuses were instrumented at 119-120 days gestation under general anesthesia. After a four-day recovery period, fetal arterial blood pressure, blood gases and heart rate were measured invasively. Cine-loops of cTDI images of the fetal heart…

Molecular Prevalence, Genotypic Distribution and Zoonotic Potential of Enterocytozoon bieneusi in Hu Sheep and Goats in Southern Zhejiang Province, Eastern China.

Enterocytozoon bieneusi is a major zoonotic pathogen causing human microsporidiosis, with small ruminants serving as crucial reservoir hosts. Hu sheep and local goats are pillar livestock breeds in Zhejiang Province, yet no parallel comparative epidemiological data on E. bieneusi exist for sympatric small ruminants in southern Zhejiang, and the infection status in Hu sheep remains entirely uncharacterized. Here, a total of 297 fecal samples (159 Hu sheep, 138 goats) were collected from four administrative regions in southern Zhejiang from 2021 to 2023 and screened via nested PCR targeting the …

Generation of Genetically Engineered Embryos Using Gene Editing and Somatic Cell Nuclear Transfer for Production of Sheep Models of Human Disease.

Large animal models are valuable tools for investigating human disease. Sheep, pigs, and goats often better recapitulate the anatomy and physiology of human organs and the complexity of human disease, thereby enhancing their clinical relevance compared to rodents. CRISPR-Cas9 and somatic cell nuclear transfer (SCNT) enable the generation of large animal models with greater precision, versatility, and genetic uniformity. The primary benefit of this approach, compared with zygote microinjection, is the ability to confirm in vitro whether the desired genetic modification and potential off-target …

Nutrient Utilization and Requirements in Sheep and Goats Raised Under Different Systems and Fed Low Nutritional Novel Feeds for Meat Production.

Sheep and goats are essential in many parts of the world, providing meat, milk, and fiber while also aiding rural and low-income communities. The cost of feeding is a major factor in the overall expenses of animal production. Therefore, it is crucial to improve nutrient utilization and accurately assess nutrient needs. Sheep and goats in different parts of the world are raised under different systems influenced by variations in environmental conditions. In tropical areas, sheep and goats often consume low-quality diets that do not meet their production requirements. These substandard diets, wh…
