Scaffold-free tenogenic macro-tissues engineered by multi-layered bioassembly of rat tendon fibroblast spheroids.

Tissue-engineered tendon constructs that replicate the structural and functional properties of natural tendons are crucial in regenerative medicine to improve treatment outcomes after tendon injury. This study aimed to engineer 3-dimensional biomimetic tendon macro-tissues through bioassembly of cell spheroids. Rat tendon fibroblasts seeded at different cell numbers (1 × 104, 5 × 104, 1 × 105, 2 × 105, 3 × 105) were analysed for spheroid formation and development for 28 d. The spheroid diameters decreased over time with a reduction in cell density while synthesising their own collagen fibres. …

Umbilical cord-derived mitochondrial transplantation restores tendon structure and function in a preclinical model of achilles tendinopathy.

BACKGROUND: Achilles tendinopathy is a degenerative musculoskeletal disorder for which disease-modifying therapies remain limited, largely due to the inability of current interventions to directly restore cellular bioenergetic function. Emerging evidence has identified mitochondrial dysfunction as a central contributor to tendon degeneration, highlighting mitochondria as a potential therapeutic target. Here, we evaluated umbilical cord-derived mitochondria (UC-MT) as a dose-defined, cell-free therapeutic strategy for tendinopathy. METHODS: UC-MT were isolated from human umbilical cord-derived

Rice bodies, extensor digitorum longus tendon tenosynovitis and sarcoidosis - multimodal ultrasound approach. A case report.

We present the case of a young woman in whom the association between sarcoidosis, extensor digitorum longus tendon tenosynovitis, and the presence of numerous rice bodies within the tendon sheath revealed a new clinical manifestation of sarcoidosis, in the absence of pulmonary or lymph nodes involvement. The most important aspect of this case is the assessment of tenosynovitis using multimodal ultrasound (grey scale, color/power Doppler, strain elastography, contrast enhanced ultrasound) which demonstrated specific findings that may facilitate further recognition of this clinical condition.

Filamented light (FLight) biofabrication of mini-tendon models show tunable matrix confinement and nuclear morphology.

One hallmark of healthy tendon tissue is the high confinement of tenocytes between tightly packed, highly aligned collagen fibers. During tendinopathy, this organization becomes dysregulated, leading to cells with round-shaped morphology and collagen fibers which exhibit crimping and misalignment. The elongated nuclei in healthy tendons are linked to matrix homeostasis through distinct mechanotransduction pathways, and it is believed that the loss of nuclear confinement could upregulate genes associated with abnormal matrix remodeling. Replicating the cell and nuclear morphology of healthy and…

Transosseous Quadriceps Tendon Repair With Knotless All-Suture Anchors Is Biomechanically Superior to Traditional Transosseous Fixation.

BACKGROUND: Reruptures and functional deficits can occur with conventional transosseous quadriceps tendon repair. Previous work has demonstrated the biomechanical superiority of adjustable transosseous metal cortical button fixation over conventional repair. Knotless all-suture anchor (ASA) buttons may provide a similar improvement but have not yet been investigated. PURPOSE: To biomechanically compare adjustable transosseous cortical fixation with knotless ASAs to traditional transosseous repair. STUDY DESIGN: Controlled laboratory study. METHODS: Eight matched pairs of male cadaveric knee

Pulmonary Embolism Following Quadriceps Tendon Repair With Tourniquet Use Despite Adequate Thromboprophylaxis: A Case Report.

Venous thromboembolism (VTE) is a recognized complication following orthopaedic surgery of the lower limb, but its incidence after quadriceps tendon repair remains poorly described. We report the case of a 53-year-old man (BMI 42 kg/m²) with morbid obesity who developed acute pulmonary embolism (PE) following quadriceps tendon repair performed with tourniquet assistance, despite receiving extended perioperative thromboprophylaxis with tinzaparin. This case highlights the interplay between tourniquet use, patient-specific risk factors (obesity, delayed surgery, and reduced mobility), and the ad

Increasing ground contact time reduces Achilles tendon forces during rearfoot strike running outdoors.

OBJECTIVES: Strategies to reduce Achilles tendon forces during running may be beneficial for injury prevention. Increasing ground contact time could reduce Achilles tendon forces during running but may also elicit changes in cadence that could offset these reductions. The purpose of this study was to determine if changing ground contact time altered Achilles tendon forces during running, with both a fixed and a free cadence. DESIGN: This was a cross-sectional study of healthy rearfoot strike runners. METHODS: Thirty rearfoot strike runners ran on a concrete sidewalk at 3.0 m/s with preferred
