Mesenchymal stromal cells support the viability and differentiation of follicular lymphoma-infiltrating follicular helper T-cells.

The biology of follicular lymphoma (FL) is largely dictated by the immune-effector and stromal cells that comprise its tumor microenvironment. FL-infiltrating T-cell populations that are thought to be fundamental to FL biology are follicular helper T-cells (TFH), follicular regulatory T-cells (TFR), a recently described population that regulates TFH activity, and regulatory T-cells (Treg). These T-cell populations have dynamic interactions with mesenchymal stromal cells (MSCs) in the tumor microenvironment. Whereas MSCs have been shown to support FL B-cell and Treg viability, their effects on …

Myeloma bone disease: Pathophysiology and management.

Multiple myeloma bone disease is marked by severe dysfunction of both bone formation and resorption and serves as a model for understanding the regulation of osteoblasts (OBL) and osteoclasts (OCL) in cancer. Myeloma bone lesions are purely osteolytic and are associated with severe and debilitating bone pain, pathologic fractures, hypercalcemia, and spinal cord compression, as well as increased mortality. Interactions within the bone marrow microenvironment in myeloma are responsible for the abnormal bone remodeling in myeloma bone disease. Myeloma cells drive bone destruction that increases t

Lenalidomide for the treatment of B-cell malignancies.

Lenalidomide is a novel anticancer agent that has made a major impact in the treatment of patients with B-cell malignancies. A more potent analog of thalidomide, lenalidomide was developed to enhance immunomodulatory properties with improved safety profile. Its antitumor activity seems mediated through modulation of both the cytokine and cellular tumor cell microenvironment. Preclinical as well as clinical observations demonstrate that lenalidomide downregulates production of various critical prosurvival cytokines in the tumor microenvironment while concurrently promoting activation of T- and …

CLDN18.2 and 4-1BB bispecific antibody givastomig exerts antitumor activity through CLDN18.2-expressing tumor-directed T-cell activation.

BACKGROUND: Claudin18.2 (CLDN18.2) is a tight junction protein that has been identified as a clinically proven target in gastric cancer. Stimulation of 4-1BB with agonistic antibodies is also a promising strategy for immunotherapy and 4-1BB+ T cells were reported to be present within the tumor microenvironment of patients with gastric cancer. However, hepatotoxicity-mediated by 4-1BB activation was observed in clinical trials of agonistic anti-4-1BB monoclonal antibodies. METHODS: To specifically activate the 4-1BB+ T cells in tumor and avoid the on-target liver toxicity, we developed a novel…

Evaluation of pain related behaviors and disease related outcomes in an immunocompetent mouse model of prostate cancer induced bone pain.

Cancer-induced bone pain (CIBP) is the most common and devastating symptom of bone metastatic cancer that substantially disrupts patients' quality of life. Currently, there are few effective analgesic treatments for CIBP other than opioids which come with severe side effects. In order to better understand the factors and mechanisms responsible for CIBP it is essential to have clinically relevant animal models that mirror pain-related symptoms and disease progression observed in patients with bone metastatic cancer. In the current study, we characterize a syngeneic mouse model of prostate cance

Osteotropic peptide-mediated bone targeting for photothermal treatment of bone tumors.

The treatment of bone tumors is a challenging problem due to the inefficient delivery of therapeutics to bone and the bone microenvironment-associated tumor resistance to chemo- and radiotherapy. Here, we developed a bone-targeted nanoparticle, aspartate octapeptide-modified dendritic platinum-copper alloy nanoparticle (Asp-DPCN), for photothermal therapy (PTT) of bone tumors. Asp-DPCN showed much higher affinity toward hydroxyapatite and bone fragments than the non-targeted DPCN in vitro. Furthermore, Asp-DPCN accumulated more efficiently around bone tumors in vivo, and resulted in a higher t

Metabolic reprogramming in colorectal cancer: Mechanisms and therapeutic insights.

Metabolic reprogramming is an important factor contributing to tumor medication resistance and recurrence in cancer cells. There is ample proof that metabolic remodeling is vital in colorectal cancer's genesis, progression, and resistance to treatment. The molecular processes behind changes in metabolism in colorectal cancer are methodically examined in this review, including its interactions with the tumor microenvironment and key signaling pathways, and its potential clinical translational value. Investigating the metabolic characteristics of colorectal cancer and identifying novel metabolic…

Inflammatory 68 Ga FAPI-46 Uptake Mimicking a Sister Mary Joseph's Nodule in a Case of Gastric Signet Ring Cell Carcinoma.

Fibroblast activation protein (FAP) is a type II transmembrane protease expressed in cancer-associated fibroblasts (CAF) that constitute a part of the tumor stroma and influence the tumor microenvironment. Although FAP is not expected to be physiologically expressed in noncancer-associated cells, it can be observed in wound healing, arthritis, atherosclerotic plaques, and fibrosis. In this case, we would like to present early-stage 68 Ga FAPI-46 PET/CT inflammation findings at the trocar entry site mimicking the Sister Mary Joseph's nodule in a patient diagnosed with gastric signet ring cell c…

Protocol for isolating patient-derived ascites cells and extracellular vesicles from gastric cancer peritoneal metastases.

Peritoneal carcinomatosis (PC) cells and extracellular vesicles (EVs) from gastric cancer ascites are valuable for studying tumor-stroma interactions. Here, we present a protocol for isolating PC cells and EVs from patient ascites. We describe steps for PC cell culture, iodixanol-based EV purification, electron microscopy, nanoparticle tracking analysis, flow cytometry, and proteomic profiling. This protocol also includes TD-139 loading into exosomes for functional assays to evaluate their role in modulating the tumor microenvironment. For complete details on the use and execution of this prot…

Regulatory Effects of Histone Deacetylase Inhibitors on Myeloid-Derived Suppressor Cells.

Histone deacetylase inhibitors (HDACIs) are antitumor drugs that are being developed for use in clinical settings. HDACIs enhance histone or nonhistone acetylation and promote gene transcription via epigenetic regulation. Importantly, these drugs have cytotoxic or cytostatic properties and can directly inhibit tumor cells. However, how HDACIs regulate immunocytes in the tumor microenvironment, such as myeloid-derived suppressor cells (MDSCs), has yet to be elucidated. In this review, we summarize the effects of different HDACIs on the immunosuppressive function and expansion of MDSCs based on …

Clinical and Biological Prognostic Factors in Follicular Lymphoma.

Follicular lymphoma comprises approximately 20-30% of all cases of B-cell lymphomas. Median survival has improved significantly in the modern era. Prognostic factors include histologic grade, cytogenetics, molecular mutations, the tumor microenvironment, and tumor burden. Clinical prognostic indices are available and increasingly incorporate genetic information. Prognostic factors also arise during the course of treatment. Early progression within 24 months of initial chemoimmunotherapy is an adverse prognostic marker of inferior survival. Other high-risk populations include those with double …

Bioinformatic analysis of serpina1 expression in papillary thyroid carcinoma and its potential association with Hashimoto's thyroiditis.

PURPOSE: Previous studies have suggested that SERPINA1 may promote a better prognosis in papillary thyroid carcinoma (PTC) along with Hashim … linked to certain clinical pathological features of PTC and the infiltration of immune cells in the tumor microenvironment. Single-cell transcriptome analysis reveals that SERPINA1 is primarily expressed in thyrocytes and myeloid cells. In thyrocytes, SERPINA1 is associated with complement-related proteins (e.g., C3, CD55). In poorly differentiated thyrocytes, it is lin …

SPP1 in Immunosuppressive Signaling in Lung Adenocarcinoma: Bulk and Single-Cell Transcriptomic Analysis.

BACKGROUND: Secreted phosphoprotein 1 (SPP1) has been implicated in various tumors and is known to correlate with immune infiltration and poor prognosis. However, its functional role as an immune communication mediator within the lung adenocarcinoma (LUAD) tumor microenvironment (TME) remains poorly defined. This study aimed to investigate the prognostic value of SPP1 and its association with inferred intercellular communication patterns in LUAD. METHODS: We integrated bulk RNA-seq datasets (GSE7670, GSE43458, TCGA-LUAD) and single-cell RNA-seq data (GSE123902) to identify differentially expr…

Single-cell analysis reveals an endothelial TP53-CXCL14 axis in breast cancer progression.

BACKGROUND: The TME of BRCA plays a core role in disease progression and treatment resistance. As a pleiotropic chemokine, CXCL14 exhibits context-dependent roles in cancer. While its potential prognostic value in BRCA has been noted, its precise role and regulatory mechanisms remain elusive. METHOD: The expression of CXCL14, its prognostic significance, and its association with the immune microenvironment were systematically analyzed using public databases, such as TCGA, GEO, and cBioPortal. In vitro experiments were performed to generate HUVEC cells with stable TP53 silencing or overexpress

PRKX-mediated stabilization of PD-L1 characterizes an immunosuppressive gastric cancer subtype.

BACKGROUND: Gastric cancer (GC) derives limited benefit from immunotherapy, with clinical responses observed in only a minority of patients. Increasing evidence suggests that heterogeneity within the tumor immune microenvironment (TME) is a critical determinant of immunotherapeutic efficacy, highlighting the need for precise immune stratification and the identification of molecular biomarkers that shape the TME. METHODS: We integrated single-cell transcriptomic data from our cohort and public datasets to characterize immune microenvironment heterogeneity in GC. Functional experiments were per
