Deficits in skilled motor and auditory learning in a rat model of Rett syndrome.

BACKGROUND: Rett syndrome is an X-linked neurodevelopmental disorder caused by a mutation in the gene MECP2. Individuals with Rett syndrome display developmental regression at an early age, and develop a range of motor, auditory, cognitive, and social impairments. Several studies have successfully modeled some aspects of dysfunction and Rett syndrome-like phenotypes in transgenic mouse and rat models bearing mutations in the MECP2 gene. Here, we sought to extend these findings and characterize skilled learning, a more complex behavior known to be altered in Rett syndrome. METHODS: We evaluate

Clinical and neuroimaging characteristics of Chinese dementia with Lewy bodies.

Dementia with Lewy bodies (DLB) is the second most common subtype of degenerative dementia. To our knowledge, available information about the clinical features of DLB in China remains limited. Our study therefore aimed to address this issue. Thirty-seven Chinese patients with probable DLB were recruited for this study. All subjects underwent neuropsychological assessment by trained neurologists, as well as undergoing MRI, 11C-PIB PET scans for Aβ deposition and 18F-FDG PET scans for regional cerebral glucose metabolism. Our results showed that the gender ratio of patients was 16:21 (F:M). The

General Anesthesia for a Patient With Pelizaeus-Merzbacher Disease.

We report the successful management of general anesthesia for a patient with Pelizaeus-Merzbacher disease (PMD). PMD is one of a group of progressive, degenerative disorders of the cerebral white matter. The typical clinical manifestations of PMD include psychomotor retardation, nystagmus, abnormal muscle tone, seizures, and cognitive impairment. General anesthesia for a patient with PMD may be difficult mainly because of seizures and airway complications related to poor pharyngeal muscle control. In addition, the possibility of exacerbation of spasticity should be considered. A 20-year-old ma

Individual and environmental contributions to treatment outcomes following a neuroplasticity-principled speech treatment (LSVT LOUD) in children with dysarthria secondary to cerebral palsy: a case study review.

This study describes the use of a neuroplasticity-principled speech treatment approach (LSVT(®)LOUD) with children who have dysarthria secondary to cerebral palsy. To date, the authors have treated 25 children with mild-to-severe dysarthria, a continuum of gross and fine motor functions, and variable cognitive abilities. From this data set, two case studies are presented that represent as weak or strong responders to LSVT LOUD. These case studies demonstrate how individual and environmental features may impact immediate and lasting responses to treatment. Principles that drive activity-depende

Reversing cocaine-induced synaptic potentiation provides enduring protection from relapse.

Cocaine addiction remains without an effective pharmacotherapy and is characterized by an inability of addicts to inhibit relapse to drug use. Vulnerability to relapse arises from an enduring impairment in cognitive control of motivated behavior, manifested in part by dysregulated synaptic potentiation and extracellular glutamate homeostasis in the projection from the prefrontal cortex to the nucleus accumbens. Here we show in rats trained to self-administer cocaine that the enduring cocaine-induced changes in synaptic potentiation and glutamate homeostasis are mechanistically linked through g

Dyslexia and music. From timing deficits to musical intervention.

The underlying causes of the language and literacy difficulties experienced by dyslexic children are not yet fully understood, but current theories suggest that timing deficits may be a key factor. Dyslexic children have been found to exhibit timing difficulties in the domains of language, music, perception and cognition, as well as motor control. The author has previously suggested that group music lessons, based on singing and rhythm games, might provide a valuable multisensory support tool for dyslexic children by encouraging the development of important auditory and motor timing skills and

Sex Differences in Prefrontal Oxygenation and Neuromuscular Fatigue to Incremental Cycling and Cognitive Testing Lead to Similar Cognitive Performance.

PURPOSE: Cognitive-motor dual-task performance is prevalent in many sports and appears to be impaired during high-intensity exercise, in association with central fatigue and cerebral deoxygenation. However, the extent to which these responses differ between males and females remains unknown. METHODS: Forty-eight well-trained adults (16 females) performed an incremental cycling test with a sustained-attention Mackworth task. The protocol consisted of 3-min ramp stages (starting at 1 W·kg -1, +0.4 W·kg -1 per stage) until an "extremely strong" perceived exertion was achieved (Borg CR100, task-f

Biomarker-Conditioned Vision Transformers with Deformable Biomarker Attention for Alzheimer's Disease Classification.

Alzheimer's disease (AD) classification from structural magnetic resonance imaging (MRI) remains challenging, particularly when distinguishing mild cognitive impairment (MCI) from both cognitively normal (CN) ageing and established AD. Multimodal approaches that combine imaging with clinical information are promising, but most confine the influence of clinical variables to the classifier head, so biomarkers cannot shape spatial feature extraction inside the imaging encoder. We propose a bimodal deep learning framework operating on two input sources: 3D T1-weighted structural MRI and tabular cl

The AI Colleague: Reimagining Interprofessional Collaboration and Education in Medicine.

AI is no longer confined to optional decision support; it is becoming a routine presence in clinical workflows, shaping diagnostic hypotheses, triage priorities, risk estimates, and documentation. Yet most educational responses still treat AI as a tool operated by an individual clinician. This framing underestimates how AI reshapes the actual unit of practice: the interprofessional team. We propose the concept of AI-expanded interprofessional collaboration (AI-IPC), in which AI systems function as consequential participants in team cognition-not as moral agents, but as sources of recommendatio

Design and development of a convolutional neural network based on human cognitive attention mechanism for automatic classification of leukemia.

Cancer occurs when healthy cells in the body grow abnormally and out of control. Leukemia is a type of cancer that affects White Blood Cells (WBCs) and can cause a lethal infection and early death. Identification and classification of different types of leukemia are performed manually and automatically. The doctors analyze blood samples under a microscope and consider any changes in the number and structure of WBCs as a sign of cancer in the manual method. It is a time-consuming, inaccuracy-prone process that depends on the expertise and skill of the physician and the type of laboratory equipm

EEG microstate dynamics and metal metabolism in Parkinson's disease dementia: An exploratory study with machine learning-assisted sensitivity analysis.

BACKGROUND: Electroencephalogram (EEG) microstates effectively characterise cognitive-related brain networks, and metal homeostasis is crucial for maintaining cognitive function. However, the relationship between these factors in Parkinson's disease (PD) with dementia (PDD) remains unclear. We investigated this association and evaluated the performance of these combined features in classifying PDD. METHODS: We analysed linear and nonlinear features of EEG microstates in 60 patients with PDD, 56 patients with PD without dementia (PDND), and 60 healthy controls (HC). We measured indicators of m

Revealing hidden knowledge in amnestic mice.

Alzheimer's disease (AD) is a form of dementia in which memory and cognitive decline is thought to arise from underlying neurodegeneration. These cognitive impairments, however, are transient when they first appear and can fluctuate across disease progression. Here, we investigate the neural mechanisms underlying fluctuations of performance in amnestic mice. We trained APP/PS1+ mice on an auditory go/no-go task that dissociated learning of task contingencies (knowledge) from its more variable expression under reinforcement (performance). APP/PS1+ exhibited significant performance deficits comp

Predicting biological activity and design of 5-HT6 antagonists through assessment of ANN-QSAR models in the context of Alzheimer's disease.

CONTEXT: Alzheimer's disease (AD) is the leading cause of dementia around the world, totaling about 55 million cases, with an estimated growth to 74.7 million cases in 2030, which makes its treatment widely desired. Several studies and strategies are being developed considering the main theories regarding its origin since it is not yet fully understood. Among these strategies, the 5-HT6 receptor antagonism emerges as an auspicious and viable symptomatic treatment approach for AD. The 5-HT6 receptor belongs to the G protein-coupled receptor (GPCR) family and is closely implicated in memory loss

A Rapid Response Mobile Application Improves First-Year Resident Clinical Performance During Simulated Care Events: A Randomized Controlled Trial.

BACKGROUND: Rapid response teams (RRTs) are critical to the timely and appropriate management of acutely decompensating patients. In the academic setting, the vital role of RRT leader is often filled by a junior resident physician who may lack the necessary medical knowledge and experience. Cognitive aids help improve guideline adherence and may support resident performance as they transition into leadership roles. OBJECTIVE: This study evaluated the impact of a rapid response mobile application on intern performance during simulated rapid response events. DESIGN: This randomized controlled
