Human pathologies associated with defective RNA transport and localization in the nervous system.

RNA localization is emerging as an important process to restrict certain proteins to specific subcellular domains and thus spatially control the expression of genes within cells. It is used, for instance, to compartmentalize the developing embryo during early embryogenesis. The localization of RNA also plays important roles later during development, such as in asymmetric cell divisions, cell migration and the outgrowth and pathfinding of axons and dendrites. In differentiated cells, it serves to subdivide the cell into functionally distinct compartments. For example, in mature neurons it is be…

Human eastern equine encephalitis: immunohistochemistry and ultrastructure.

The brain of a 7-year-old boy who died of eastern equine encephalitis (EEE) was examined by immunohistochemical and ultrastructural techniques to detect the presence and distribution of viral antigen. A mouse polyclonal antibody was most effective for demonstrating the presence of antigen previously unreported in this disease in humans. Antigen was localized to the perikaryon and dendrites of neurons; little was detected in glial cells. Cell death by apoptosis was conspicuous, but it was primarily identified in glial and inflammatory cells. Neuronal death was most commonly marked by cytoplasmi…

Misrouting of tyrosinase with a truncated cytoplasmic tail as a result of the murine platinum (cp) mutation.

Mice homozygous for the platinum (cp) allele at the albino locus manifest severe oculocutaneous albinism despite the presence in vitro of tyrosinase activity at 25% wild-type levels. We demonstrate that the cp allele results from an A-->T substitution, changing a lysine residue at position 489 to a termination codon, with truncation of tyrosinase's cytoplasmic tail. In choroidal melanocytes of neonatal mutant mice, tyrosinase activity could be detected in the trans Golgi network, but was absent from melanosomes. Instead, it was detected in vesicles in the cell periphery and dendrites, and on t…

Hereditary spastic paraparesis with dementia, amyotrophy and peripheral neuropathy. A neuropathological study.

Hereditary, probably autosomal recessive, spastic paraparesis in two siblings was associated with dementia of frontal lobe type, amyotrophy and peripheral sensory and motor polyneuropathy. Neuropathological findings correlate with neurological deficits, although neuron loss in the caudate and putamen, substantia nigra, and loss of Purkinje cells were clinically silent. Loss of neurons occurred in all cortical layers of the prefrontal lobe and superior temporal gyrus. Immunohistochemical studies showed reduced parvalbumin immunoreactivity in dendrites, and reduced numbers of calbindin D28k-immu…

[A comparative study of the quantitative distribution and ultrastructure of Langerhans cells in the epidermis of patients with chronic benign dermatoses and mycosis fungoides].

Using histoenzymatic assay for ATPase activity and electron microscopy, Langerhans' cells (LC) in biopsy specimens taken from skin lesions of 20 patients with chronic benign dermatoses (CBD) and of 28 patients suffering from mycosis fungoides (MF) were identified. In the early lesions of MF the reduction of the number of cell dendrites, disappearance of normal arrangement and the reduced absolute quantity of LC were observed on comparison with patients with CBD. Ultrastructurally, the destroyed forms of LC were predominantly observed in MF lesions, while in cases of CBD most of the LC were fun…

Myelination and glial ensheathment of Purkinje cells in cerebellar cultures are not inhibited by antibodies to the neural cell adhesion molecule, N-CAM.

Mouse cerebellar cultures were exposed to anti-N-CAM antibodies throughout their in vitro development. Some cultures were stripped of myelinating oligodendrocytes and functionally competent astrocytes by treatment with cytosine arabinoside (Ara C), while others were left untreated and were potentially capable of forming myelin around axons and astrocytic sheaths around Purkinje cell somata and dendrites. As expected, the antibodies inhibited axonal fasciculation in the Ara C treated cultures. However, the same antibodies had no discernible effect on formation of myelin or astrocytic sheaths in…

Brain cell alterations suggesting premature aging induced by dietary deficiency of vitamin B6 and/or copper.

Weanling albino rats were fed semisynthetic diets deficient or sufficient in vitamin B6 or copper, or both, for 2 or 3 months. Brains were examined by light and electron microscopy after Golgi impregnation or conventional tissue processing for electron microscopy. Golgi impregnation revealed that some pyramidal cells of the cerebral cortex, particularly in layers III and V, showed partial to nearly complete dendritic loss. This occurred in all deficient groups but was most typical of deficiency of vitamin B6. Swelling in dendrites or perikarya was more typical of copper deficiency. Ultrastruct…

Fetal alcohol syndrome and mental retardation: spine distribution of pyramidal cells in prenatal alcohol-exposed rat cerebral cortex; a Golgi study.

Among the various possible effects of the fetal alcohol-syndrome (FAS), mental retardation can be considered the most deleterious. In animal studies, prenatal alcohol exposure has been shown to result in increased neonatal mortality, retarded cerebellar development and a significant decrease in neonatal brain weight. In a Golgi study on Wistar rats that were prenatally exposed to alcohol the spine distribution in proximal apical dendrites of layer V pyramidal cells of the parietal cortex was examined. As compared with controls, a distinct spine abnormality could be demonstrated at 12 days and …

mTORC1-dependent protein synthesis underlying rapid antidepressant effect requires GABABR signaling.

Administration of N-methyl-D-aspartate receptors (NMDAR) antagonists initiates a rapid anti-depressant response requiring mammalian Target of Rapamycin Complex 1 (mTORC1) kinase; however the molecular mechanism is unknown. We have determined that upon NMDAR blockade, dendritic γ-amino-butyric acid B receptors (GABABR) facilitate dendritic calcium entry. The GABABR-mediated increase in calcium signal requires the availability of dendritic L-type calcium channels. Moreover, GABABR can activate mTOR and increase mTOR dependent expression of BDNF under the same NMDAR blocked conditions. In vivo, b

Presentation of an epithelioid cell histiocytoma on the ventral tongue.

The epithelioid cell histiocytoma (ECH) is a polypoidal benign tumor of superficial connective tissue that is often diagnosed as a pyogenic granuloma. ECHs are speculated to originate from dermal dendritic subunits and are composed of 2 primary cell populations, ie, CD34+ primitive fibroblastic dendrocytes and factor XIIIa+ histiocytes. Although dendritic subunits are distributed throughout most collagenous tissues inclusive of oral mucosa, to date, all reported cases of ECH have been cutaneous lesions. ECHs' putative pathogenesis entails activation of CD34+ "sentinel" reserve dendrocytes, fol

Cytokeratin-positive malignant tumors with reticulum cell morphology: a subtype of fibroblastic reticulum cell neoplasm?

Cytokeratin-positive interstitial reticulum cells (CIRCs) have been described as a subset of fibroblastic reticulum cells (FBRCs) normally found in lymph nodes, the spleen, and tonsils. Although tumors derived form other reticulum (dendritic) cells, specifically follicular dendritic cells, interdigitating dendritic cells, and cytokeratin-negative FBRCs, have been well documented and are now accepted, this is not the case for tumors of CIRCs. A possible reason for this failure is the difficulty in distinguishing them from other tumors, particularly carcinoma. We report three cases of cytokerati

Expression of proliferating cell nuclear antigen (PCNA): proliferative phase functions and malignant transformation of melanocytes.

The UV-dependent G2-phase functions of melanocytes include dendricity, the expression of melanocyte stimulating hormone (MSH) receptors and neural differentiation. The present report studied highly dendritic melanocytes in epidermis overlying tumours, seborrhoeic keratosis, basal cell carcinoma and melanomas. The expression of the proliferative protein PCNA was studied by immunohistochemistry, as this indicates cells in S/G2-phase. In the non-neoplastic dendritic melanocytes, PCNA is retained in the cytoplasm, resulting in the arrest of the cells in the S/G2-phase for prolonged periods, as ind

Control of Intestinal Type 3 Immunity by the Unfolded Protein Response Sensor Inositol-Requiring Enzyme 1α in Antigen-Presenting Cells.

BACKGROUND & AIMS: The unfolded protein response is an essential cellular program maintaining intestinal homeostasis by acting on several cell types. However, whether the unfolded protein response sensor inositol-requiring enzyme 1α acts on dendritic cells and other antigen-presenting cells for shaping intestinal immunity remains poorly defined. METHODS: Here, we dissect the role of the endoribonuclease domain of the unfolded protein response sensor inositol-requiring enzyme 1α in intestinal CD11c+ antigen-presenting cells, using inositol-requiring enzyme 1α endoribonuclease reporter mice and

The murine ortholog of Kaufman oculocerebrofacial syndrome protein Ube3b regulates synapse number by ubiquitinating Ppp3cc.

Kaufman oculocerebrofacial syndrome (KOS) is a severe autosomal recessive disorder characterized by intellectual disability, developmental delays, microcephaly, and characteristic dysmorphisms. Biallelic mutations of UBE3B, encoding for a ubiquitin ligase E3B are causative for KOS. In this report, we characterize neuronal functions of its murine ortholog Ube3b and show that Ube3b regulates dendritic branching in a cell-autonomous manner. Moreover, Ube3b knockout (KO) neurons exhibit increased density and aberrant morphology of dendritic spines, altered synaptic physiology, and changes in hippo

WIP is a negative regulator of neuronal maturation and synaptic activity.

Wiskott-Aldrich syndrome protein (WASP) -interacting protein (WIP) is an actin-binding protein involved in the regulation of actin polymerization in cells, such as fibroblasts and lymphocytes. Despite its recognized function in non-neuronal cells, the role of WIP in the central nervous system has not been examined previously. We used WIP-deficient mice to examine WIP function both in vivo and in vitro. We report here that WIP(-)(/-) hippocampal neurons exhibit enlargement of somas as well as overgrowth of neuritic and dendritic branches that are more evident in early developmental stages. Dend
