Menin inhibitors in acute myeloid leukaemia: Mechanisms, clinical pharmacology, and emerging therapeutic strategies.

Acute myeloid leukaemia (AML) remains associated with poor outcomes in relapsed or refractory disease and in molecularly defined high-risk subgroups. AML with KMT2A rearrangements (KMT2Ar) or NPM1 mutations (NPM1m) shares a dependence on the menin-KMT2A interaction, which sustains HOXA/MEIS1-driven transcription required for leukaemic maintenance. This dependence has provided the rationale for developing menin inhibitors as a targeted approach in selected AML subsets. This review summarises the mechanistic basis, clinical pharmacology, and current clinical role of menin inhibitors in AML. Revu

NMDA receptor subunit-dependent analysis of radiprodil inhibition in neonatal male rat substantia nigra dopaminergic neurons.

BACKGROUND AND PURPOSE: Radiprodil is a GluN2B subunit selective NMDA receptor negative allosteric modulator which is currently under clinical investigation as a possible anti-epileptic drug for paediatric use. Our goal was to investigate the inhibition of dopaminergic neuron NMDA responses by radiprodil. EXPERIMENTAL APPROACH: Whole-cell and outside-out patch-clamp recordings from substantia nigra dopaminergic neurons in acute mid-brain slices from neonatal male rats were used to investigate radiprodil action. Radiprodil action was analysed using a 'hypercube' model with explicit receptor Gl

Copper (I) nicotinate complex potentiates chemotherapy-induced apoptosis in HCC-1806 triple negative breast cancer cells: An in vitro study.

Triple negative breast cancer (TNBC) remains a clinically challenging subtype because it lacks established molecular targets and often depends on cytotoxic chemotherapy. Although doxorubicin (DOX) and mTOR inhibitors such as Torin1 (TOR) exhibit antitumor activity, their clinical use may be constrained by toxicity and resistance. Copper (I) Nicotinate Complex (CNC) has emerged as promising candidates due to their superoxide dismutase (SOD) -mimicking and cytoprotective properties. This study aimed to evaluate the potential effect of CNC combined with DOX or TOR to enhance cytotoxicity against

Pharmacogenomic Synthetic Lethal Screens Reveal Hidden Vulnerabilities and New Therapeutic Approaches for Treatment of NF1-Associated Tumors.

Neurofibromatosis type I (NF1) is a common cancer predisposition syndrome caused by heterozygous loss-of-function mutations in the tumor-suppressor gene NF1. Individuals with NF1 develop benign tumors of the peripheral nervous system (neurofibromas), originating from the Schwann cell (SC) lineage after somatic loss of the wild-type NF1 allele, some of which progress further to malignant peripheral nerve sheath tumors (MPNST). There is only one FDA-approved targeted therapy for symptomatic plexiform neurofibromas and none approved for MPNSTs. The genetic basis of NF1 syndrome makes associated t

Beyond poly(ADP-ribose) polymerase inhibition: emerging therapeutic strategies for germline BRCA1/2-mutated and PALB2-mutated breast cancers.

PURPOSE OF REVIEW: Germline or somatic biallelic inactivation of BRCA1, BRCA2 and PALB2 genes define a clinically and biologically distinct subset of breast cancers characterized by homologous recombination deficiency (HRD) and sensitivity to PARP inhibitors. Despite regulatory approvals of olaparib and talazoparib for breast cancers in patients harboring germline BRCA1 and BRCA2 pathogenic/likely pathogenic variants, therapeutic progress in this setting has been limited by therapeutic resistance and treatment-related toxicity. This review studied emerging therapeutic strategies aimed at exten

Reverse Transcriptase Connection and RNase H Domain Variation in HIV-1 Subtype C Among Individuals with Virologic Failure in Botswana.

Background: Classical reverse transcriptase (RT) drug resistance mutations (DRMs) are primary determinants of antiretroviral treatment failure, but polymorphisms within the RT connection and RNase H domains may also influence RT inhibitor susceptibility. These regions remain poorly characterised in HIV-1 subtype C (HIV-1C), the predominant subtype in sub-Saharan Africa. This study investigated RT connection and RNase H polymorphisms in treatment-experienced people with HIV (PWH) experiencing virologic failure (VF) in Botswana. Methods: Seventeen plasma samples from the Botswana National HIV Dr

Staphylococcal proliferation on skin models to investigate novel anti-infective treatments against dysbiosis.

Inflammatory skin conditions like Acne inversa are characterized by dysbiosis, an imbalance of commensal and pathogenic bacteria, posing challenges for specific treatments. Consequently, we investigated how biofilm formation, low-nutrition skin environments, and air interfaces influence susceptibility to anti-infective treatments in mixed bacterial cultures. To achieve this in a cost-effective and reproducible manner, we developed a simplified substrate made of gelatin, hyaluronic acid, chondroitin sulphate, and alginate (=Gel-Alg). This in vitro model simulates biofilm cultivation on skin sur

Harnessing Repurposed Drugs to Enhance Temozolomide Efficacy in Glioblastoma.

BACKGROUND: Glioblastoma (GB) is the most aggressive primary malignant brain tumor in adults and remains associated with poor survival despite surgical resection followed by radiotherapy and temozolomide (TMZ) chemotherapy. Intrinsic and acquired resistance to TMZ, including MGMT-dependent DNA repair and activation of pro-survival pathways, could decrease treatment efficacy. Drug repurposing offers an attractive strategy to identify agents that may enhance TMZ activity because these drugs already have known pharmacokinetic and safety profiles. This narrative review summarizes the available evi

Preclinical Prediction of Resistance Mutations and Proposal of Sequential Treatment Strategies for ALK-positive Lung Cancer Using Next-generation ALK Inhibitors.

BACKGROUND: Anaplastic lymphoma kinase (ALK) gene rearrangements occur in approximately 5% of non-small cell lung cancers (NSCLCs). Although ALK tyrosine kinase inhibitors provide substantial clinical benefits, acquired resistance-conferring mutations frequently emerge, leading to disease progression. Preclinical prediction of these mutations might help guide the development of more effective sequential treatment strategies prior to clinical application. OBJECTIVE: To predict the emergence of resistance mutations to the investigational ALK inhibitors zotizalkib (TPX-0131), gilteritinib (ASP22

In vitro and In vivo evaluation of phage-antibiotic synergy for the treatment of urinary tract infections.

The therapeutic challenges caused by multidrug-resistant (MDR) and extensively drug-resistant (XDR) bacteria have necessitated the development of alternative treatment strategies. Phage-antibiotic synergy (PAS) has recently emerged to replace or possibly supplement antibiotics. We isolated the lytic phage NTNC80A from hospital sewage in Chandigarh, India, and it belongs to the class Caudoviricetes of viruses. We tested multiple strategies to investigate the synergistic effects of phage and antibiotic combinations against highly drug-resistant uropathogenic Escherichia coli isolates. The maximu

Enhancing nanomedicine efficacy in KPC pancreatic tumors through ketotifen-mediated tumor microenvironment remodeling.

The tumor microenvironment (TME) in pancreatic ductal adenocarcinoma (PDAC) poses major barriers to the efficacy of anticancer therapies, particularly through features such as desmoplasia, elevated stiffness, and impaired perfusion that restrict drug delivery. Strategies that modulate these physical abnormalities hold promise for improving therapeutic outcomes, especially for nanomedicines. In this study, we investigated the potential of the antihistamine ketotifen to remodel the TME and enhance intratumoral delivery and efficacy of liposomal therapeutics, PEGylated liposomal doxorubicin (Doxi
