Electromagnetic modulation of biological processes: influence of culture media and significance of methodology in the Ca-uptake by embryonal chick tibia in vitro.

The present studies are aimed at establishing molecular correlations in the interaction of very low frequency electromagnetic fields with biological systems. Ca-uptake by chick embryo tibia rudiment in short-term culture was a useful model. Tibiae of 8- to 10-day-old chick embryos were incubated 60 min in simplified culture media in the presence of 45Ca at 37.5 +/- 0.5 degrees C either inside or outside pulsating electromagnetic fields. Radioactivity count in the medium was the most accurate method for determining Ca-uptake by the rudiment. The effect of the fields on the Ca-uptake depended ma

Deficient guanine nucleotide regulatory unit activity in cultured fibroblast membranes from patients with pseudohypoparathyroidism type I. a cause of impaired synthesis of 3',5'-cyclic AMP by intact and broken cells.

Deficient activity of the guanine nucleotide regulatory protein (G unit), an integral component of the membrane-bound adenylate cyclase complex, has been implicated as the biochemical lesion in many patients with pseudohypoparathyroidism (PHP) type I. In addition to renal resistance to parathyroid hormone in this disorder, there is decreased responsiveness of diverse tissues to hormones that act via 3',5'-cyclic AMP (cAMP). To assess whether a deficiency of G units could account for impaired adenylate cyclase activity, we studied cAMP production in intact cultured fibroblasts and fibroblast pl

Regulation of platelet adenylate cyclase by adenosine.

The stimulatory and inhibitory effects of adenosine on the adenylate cyclases of human and pig platelets were studied. Stimulation occurred at lower concentrations than did inhibition, and the stimulatory effect was prevented by methylxanthines. Stimulation by adenosine was immediate in onset and was reversible, under conditions when cyclic AMP formation was linear with respect to time and protein concentration. The stimulatory and inhibitory effects could be distinguished further by the use of various analogues of adenosine and could be prevented by adenosine deaminase. The data suggest that

Nutrition in growth and development of oral tissues.

The development of teeth, salivary glands, and other oral tissues can be seriously compromised by a deficiency of a number of essential nutrients. Because nutritional stress during critical development periods may decrease the ultimate resistance of the oral tissues to challenge, the prevention of disease and maintenance of oral health are intimately dependent on the nutrient supply during development. Appropriate dietary counseling, particularly with reference to critical developmental times and intake of essential nutrients, can insure optimal oral tissue development.

Activation of protein kinase in thyroid slices by thyroid-stimulating hormone.

Protein kinase activity in homogenates of control thyroid slices and those incubated with thyroid-stimulating hormone (TSH) and prostaglandin EI was assayed and correlated with changes in cyclic adenosine 3':5'-monophosphate (cAMP) concentrations and binding of [3H]cAMP. Both TSH and prostaglandin E1 (25 mug/ml) increased protein kinase activity and the activity ratio (expressed as activity - cAMP to activity plus cAMP). It is unlikely that such activation reflects effects of the increased cAMP liberated at the time of homogenization. Hormone-induced activation of protein kinase persisted even

Differences in coupling of energy to glycine and phenylalanine transport in aerobically grown Escherichia coli.

Differences exist in the coupling of energy to transport of glycine and phenylalanine in aerobically grown cells of Escherichia coli. Energy derived from respiration, although involved in both uptake systems, is not employed identically as shown by kinetic effects of cyanide and anoxia and by temperature dependencies. Additional evidence for aerobic differences was provided by the effects of azide which greatly decreased initial rates of uptake of glycine but not phenylalanine. The effect on glycine uptake was not due to uncoupling of oxidative phosphorylation or to a decrease in respiration r

Subclinical Leaflet Thrombosis, Transcatheter Heart Valve Characteristics and Structural Valve Deterioration: Rationale and Design of the POPular PET TAVI Study.

BACKGROUND: Subclinical leaflet thrombosis is common after transcatheter aortic valve implantation (TAVI) and may contribute to structural valve deterioration over time, this may be influenced by transcatheter heart valve (THV) design and other valve-related factors. METHODS: The 'Effect of Subclinical Leaflet Thrombosis and Prosthesis Type on Transcatheter Aortic Valve Degeneration (POPular PET TAVI)' study is an international, multicenter, cross-sectional cohort study. Patients (n=180) are enrolled at five years after TAVI for a detailed assessment of THV structure and function using a mult

A comparative assessment of radiochemical purity and yield of [18F]PSMA-1007 production using two different automated synthesis platforms: a head-to-head comparison.

BACKGROUND: Glutamate carboxypeptidase II (GCPII), also known as prostate-specific membrane antigen (PSMA) is overexpressed in 90-100% of prostate cancer cells. The radiopharmaceutical [18F]PSMA-1007, recognised as a PET tracer for prostate cancer imaging, is based on PSMA inhibitor [Glu-CO-Lys(2Nal-Amb-Glu-Glu-PyTMA)] bound to the radioisotope Fluorine-18. [18F]Fluoride was obtained via the 18O(p,n)18F reaction using a cyclotron for medical use, while synthesis of [18F]PSMA-1007 was performed with two different platforms: FASTlab2 and NEPTIS® Perform. Both modules enabled synthesis through nu

Fluorine determination in biological tissues and its impact oxidative biomarkers.

This study proposes a strategy for the determination of total fluorine in biological tissues using microwave-induced combustion (MIC) for sample preparation, followed by determination by ion chromatography (IC), after evaluating different drying strategies. An ion-selective electrode (ISE) was also used to compare the results. The method was applied to liver, kidney, and brain samples from Winstar rats exposed to increasing concentrations of fluorine in drinking water. MIC parameters were evaluated through independent recovery tests, with the use of 150 mmol L-1 (NH4)2CO3 as an absorbent solut

A Continuous Culture Millifluidic Device for the Study of Escherichia coli under Low and Fluctuating Nutrient Conditions.

Microbial habitats in nature are often characterized by low concentrations of mixed nutrients, spatial heterogeneity, and temporal fluctuations. However, traditional laboratory culturing methods fail to replicate these conditions. Batch cultures cannot sustain growth in low-nutrient environments, while chemostats maintain steady-state growth with a single limiting nutrient but are challenging to implement when the goal is to maintain defined low concentrations of nutrient mixtures or to introduce rapid fluctuations. Microfluidic systems generate dynamic environments but yield insufficient biom

Ester Bond-Anchored TMP Moieties for Durable and Rechargeable Anti-biofouling PVDF Membranes.

Global water scarcity and pollution have elevated membrane separation to an indispensable technology for sustainable water purification. Poly(vinylidene fluoride) (PVDF), prized for its exceptional chemical and mechanical stability, faces a long-standing performance trade-off, as its inherent hydrophobicity causes severe biofouling, while chlorine regeneration for biofilm removal degrades surface-grafted antibacterial functional layers. Herein, we report a high-performance chlorine-rechargeable N-halamine antibacterial membrane (denoted as PPVDF-O-TP-Cl, a PEG-blended PVDF grafted with chlorin

Drying free strategies for sustainable fluorine-18 radiochemistry.

Azeotropic removal of water remains a significant limitation in fluorine-18 radiochemistry, often leading to longer synthesis times, variable yields, increased solvent use and operational complexity, and, indirectly, increased resource demand due to decay- and loss-driven activity requirements and incompatibility with base- or heat-sensitive precursors. This review critically evaluates drying-free strategies that mitigate or obviate this step while sustaining high radiochemical performance and compliance with good manufacturing practice. Methods are organized into controlled hydrous fluorinati

Metabolic Glycoengineering Enables Fluorine-18 Radiolabeling of T Lymphocytes via Dual-Bioorthogonal Chemistry.

The ability to track therapeutic cells is critical for advancing adoptive cell therapy. Positron emission tomography (PET) offers sensitive, quantitative imaging, but improved strategies for cell labeling remain needed. Here, we report a metabolic glycoengineering approach that installs azide groups onto the Jurkat T lymphocyte surface using the canonical tetraacetylated N-azidoacetylmannosamine (Ac4ManNAz). Azide-bearing cells were functionalized via strain-promoted azide-alkyne cycloaddition (SPAAC-based ligation) with a dual-clickable trancyclooctene (TCO)-bearing dibenzocyclooctyne (DBCO)
