Application of high-density DNA resequencing microarray for detection and characterization of botulinum neurotoxin-producing clostridia.

BACKGROUND: Clostridium botulinum and related clostridia express extremely potent toxins known as botulinum neurotoxins (BoNTs) that cause severe, potentially lethal intoxications in humans. These BoNT-producing bacteria are categorized in seven major toxinotypes (A through G) and several subtypes. The high diversity in nucleotide sequence and genetic organization of the gene cluster encoding the BoNT components poses a great challenge for the screening and characterization of BoNT-producing strains. METHODOLOGY/PRINCIPAL FINDINGS: In the present study, we designed and evaluated the performan

Development of molecular-based methods for determination of high histamine producing bacteria in fish.

Histamine (or scombroid) fish poisoning is a significant cause of food borne disease in the United States. In this study, we describe the development of a molecular-based technique which uses digoxigenin (DIG) labeled DNA probes for the detection of gram negative bacteria producing high amounts of histamine (>1000 ppm). A cocktail of PCR amplification fragments corresponding to a 709 bp fragment of the histidine decarboxylase (hdc) gene of four high producing bacteria (Morganella morganii, Enterobacter aerogenes, Raoultella planticola and Photobacterium damselae) was DIG-labeled and screened a

Microscopic and molecular studies of the diversity of free-living protozoa in meat-cutting plants.

The diversity of free-living protozoa in five meat-cutting plants was determined. Light microscopy after enrichment culturing was combined with sequencing of PCR-amplified, denaturing gradient gel electrophoresis (DGGE)-separated 18S rRNA gene fragments, which was used as a fast screening method. The general results of the survey showed that a protozoan community of amoebae, ciliates, and flagellates was present in all of the plants. Protozoa were detected mainly in floor drains, in standing water on the floor, on soiled bars of cutting tables, on plastic pallets, and in out-of-use hot water k

Longitudinal study of Salmonella infection in Italian farrow-to-finish swine herds.

A longitudinal study of Salmonella enterica infection was carried out in five Italian farrow-to-finish swine herds previously known to be infected by Salmonella. Five litters were randomly selected from each herd and in each litter six piglets were randomly selected and individually identified. Thus, the study included 30 pigs from each farm. At weaning, individual blood samples were collected for serological examination from all selected piglets and on the same day from all sows in the farrowing unit. Piglets were bled again at approximately 60, 90, 150, 210 and 270 days of life whereas the l

Detection of classical enterotoxins and identification of enterotoxin genes in Staphylococcus aureus from milk and dairy products.

Milk and dairy products are frequently contaminated with enterotoxigenic Staphylococcus aureus, which is often involved in staphylococcal food poisoning. The distribution of genes encoding staphylococcal enterotoxins (SE) in S. aureus isolated from bovine, goat, sheep and buffalo milk and dairy products was verified by the presence of the corresponding SE production. A total of 112 strains of S. aureus were tested for SE production by immuno-enzymatic (SEA-SEE) and reversed passive latex agglutination (SEA-SED) methods, while multiplex-PCR was applied for SE genes (sea, sec, sed, seg, seh, sei

Low prevalence of yadA-positive Yersinia enterocolitica in sows.

Yersinia enterocolitica 4/O:3 is commonly isolated from fattening pigs in Finland, but its prevalence in sows is relatively unknown. The current study determined the prevalence of yadA-positive Y. enterocolitica in sows and fattening pigs with polymerase chain reaction (PCR) and culture methods. The strains were characterized with pulsed-field gel electrophoresis (PFGE) using NotI DNA-restriction enzyme. Pathogenic Y. enterocolitica was detected in only 14% of sow tonsils compared with 56% of tonsils of fattening pigs. The prevalence varied between seven slaughterhouses from 0% to 30% in sows

Diversity of enterovirus sequences detected in oysters by RT-heminested PCR.

Oysters harvested in western France, from five sites associated with outbreaks of food-borne norovirus gastroenteritis between February 2000 and March 2001, were assayed for enterovirus RNA by reverse transcriptase-heminested polymerase chain reaction (RT-heminested PCR). Forty percent (21/52) of shellfish samples (pool of seven oysters) were contaminated by enteroviruses. Infectious coxsackieviruses serotype A21 were isolated from three of these positive samples. Amplicons corresponding to 65 base sequences in the 5' untranslated region of the enteroviral genome were analyzed by direct sequen

Regulatory veterinary medicine in a changing environment.

There is an increasing need for the veterinary profession to meet the new challenges brought about by increased and intensified livestock production. These challenges consist of control and eradication of diseases, the humane treatment of animals and the prevention of transmission of disease from animals or animal products to man. Examples are given of the contribution which regulatory medicine activities have made to the prevention/control/eradication of screw worm, foot-and-mouth disease, Venezuelan equine encephalomyelitis, Newcastle disease, bovine brucellosis, hog cholera, and certain oth

Chlordecone, a persistent pollutant: analytical challenges and sustainable remediation solutions.

Chlordecone, a persistent organochlorine insecticide, was extensively applied in the French West Indies between 1972 and 1993. Decades after its ban, it continues to contaminate soils and waters, cycling through food webs and resulting in chronic exposure of both ecosystems and human populations. Its environmental persistence, strong affinity for organic matter, and adverse effects on human health have made it a pressing public health concern. The region now exhibits the highest incidence of prostate cancer globally, reflecting complex interactions between environmental exposure and genetic su

Analysis of soil and water pollution produced by improper medical waste disposal and its impacts.

Many countries struggle with the systematic separation and disposal of medical waste, which poses significant risks of disease transmission and cross-contamination. This study synthesizes literature from 2017 to 2023 to examine the environmental consequences of improper healthcare waste management, specifically regarding soil and water ecosystems. The review establishes a direct scientific link between medical waste dumping sites and the presence of heavy metal concentrations that exceed safety thresholds in surrounding agricultural lands. A key finding of this research is the identification o

Rational Design of a Dual Mutant Degrading Enzyme with Improved Catalytic Efficiency and Acid Tolerance for Four Aflatoxins.

Aflatoxins are major contaminants in raw food materials and their products. Developing enzymes that efficiently degrade multiple aflatoxins under acidic conditions has great practical value. In this study, the aflatoxin-degrading enzyme ADPPIII from Aspergillus terreus was systematically engineered. Substrate tunnel optimization obtained mutant F375L, which showed about twice the degradation activity for four aflatoxins versus the wild-type. Surface charge neutralization generated dual mutant F375L-K570Q with significantly improved acid tolerance. At pH 5.0, its degradation rates of aflatoxins

A comprehensive review on microbial degradation of polyethylene plastics: ecotoxicity, deterioration process, recurrent intermediates and key enzymes involved.

Plastic contamination is a universal threat that affects the flora and fauna in the environment. This review focuses on biodegradation of polyethylene (PE). PE is a widely used plastic in food packaging, agricultural materials (mulch films) and disposable single-use plastic bags owing to its durability and cost-effectiveness. Being a hydrophobic, recalcitrant and persistent polymer, PE contaminates air, soil and water. The ecotoxicity, sources, fate and transport of PE and detection techniques like micro-Fourier Transform Infrared Spectroscopy (micro-FTIR) are discussed. Considering the enviro

Effects of iron additives on the removal of antibiotics and antibiotic resistance genes in anaerobic fermentation of food waste.

The presence of antibiotics and antibiotic resistance genes (ARGs) in food waste (FW) during anaerobic fermentation poses significant environmental and health risks. This study elucidated the potential of iron additives, specifically 500-nm and 50-nm zero-valent iron (ZVI) and magnetite, in mitigating these contaminants. These findings revealed that 500-nm magnetite significantly reduced tetracyclines by 81.04%, while 500-nm ZVI effectively reduced cefotaxime by 89.90%. Furthermore, both 500-nm and 50-nm ZVI were observed to decrease different types and abundance of heavy metal resistance and

Total mercury, methylmercury, phosphate, and sulfate inputs to a bog ecosystem from herring gull (Larus smithsoniansus) guano.

Avian biovector transport is an important mechanism for the movement of contaminants and nutrients to remote locations, usually bird colonies, through excretion, molting and decomposition of carcasses. Methylmercury (MeHg) is a bioaccumulative neurotoxin and endocrine disruptor which is present in many remote ecosystems. We collected guano samples biweekly from a herring gull (Larus smithsoniansus) colony over two summers and analyzed MeHg, total mercury (THg), water-extractable phosphate (PO43-) and sulfate (SO42-). Concentrations of THg in guano declined through the summer months while %MeHg

Mercury methylation in rice paddy and accumulation in rice plant: A review.

The bioavailability and toxicity of mercury (Hg) are dependent on its chemical speciation, in which methylmercury (MeHg) is the most toxic compound. Inorganic Hg can be transformed into MeHg in anaerobic conditions. Subsequent accumulation and biomagnification in the food chain pose a potential threat to human health. Previous studies have confirmed that paddy soil is an important site for MeHg production, and rice fields are an important source of MeHg in terrestrial ecosystems. Rice (Oryza sativa L.) is recently confirmed as a potential bioaccumulator plant of MeHg. Understanding the behavio

Cadmium contamination of agricultural soils and crops resulting from sphalerite weathering.

The biogeochemistry and bioavailability of cadmium, released during sphalerite weathering in soils, were investigated under contrasting agricultural scenarios to assess health risks associated with sphalerite dust transport to productive soils from mining. Laboratory experiments (365 d) on temperate and sub-tropical soils amended with sphalerite (<63 μm, 0.92 wt.% Cd) showed continuous, slow dissolution (0.6-1.2% y(-1)). Wheat grown in spiked temperate soil accumulated ≈38% (29 μmol kg(-1)) of the liberated Cd, exceeding food safety limits. In contrast, rice grown in flooded sub-tropical soil

Soil-related bacterial and fungal infections.

A variety of classic and emerging soil-related bacterial and fungal pathogens cause serious human disease that frequently presents in primary care settings. Typically, the growth of these microorganisms is favored by particular soil characteristics and may involve complex life cycles including amoebae or animal hosts. Specific evolved virulence factors or the ability to grow in diverse, sometimes harsh, microenvironments may promote pathogenesis. Infection may occur by direct inoculation or ingestion, ingestion of contaminated food, or inhalation. This narrative review describes the usual pres
