Paracoccidoides brasiliensis 30 kDa adhesin: identification as a 14-3-3 protein, cloning and subcellular localization in infection models.

Paracoccidoides brasiliensis adhesion to lung epithelial cells is considered an essential event for the establishment of infection and different proteins participate in this process. One of these proteins is a 30 kDa adhesin, pI 4.9 that was described as a laminin ligand in previous studies, and it was more highly expressed in more virulent P. brasiliensis isolates. This protein may contribute to the virulence of this important fungal pathogen. Using Edman degradation and mass spectrometry analysis, this 30 kDa adhesin was identified as a 14-3-3 protein. These proteins are a conserved group of

Vector-Borne Zoonotic Pathogens in Eurasian Moose (Alces alces alces).

Climate change, with warmer temperatures and altered precipitation patterns, has affected the distribution of vectors and vector-borne diseases. In the northern hemisphere, vectors are spreading north, and with them, pathogens of zoonotic and animal health impact. Eurasian moose (Alces alces alces) are physiologically and anatomically adapted for cold climate, and are rarely considered ideal hosts of vectors, apart from deer keds (Lipoptena cervi). To investigate the presence of vector-borne pathogens, spleen samples from 615 moose were collected in southern Sweden from 2008 to 2015. The sampl

Integrated multi-omic analyses in Biomphalaria-Schistosoma dialogue reveal the immunobiological significance of FREP-SmPoMuc interaction.

The fresh water snail Biomphalaria glabrata is one of the vectors of the trematode pathogen Schistosoma mansoni, which is one of the agents responsible of human schistosomiasis. In this host-parasite interaction, co-evolutionary dynamic results into an infectivity mosaic known as compatibility polymorphism. Integrative approaches including large scale molecular approaches have been conducted in recent years to improve our understanding of the mechanisms underlying compatibility. This review presents the combination of integrated Multi-Omic approaches leading to the discovery of two repertoires

Effect of Histoplasma capsulatum glucans on host innate immunity.

β-1,3-Glucan is important for infective forms (mycelial phase) of Histoplasma capsulatum and shares many features allotted to pathogen-associated molecular patterns. These cell wall carbohydrates interact with phagocytes by binding to Toll and lectin-like receptors, present on cell surfaces of macrophages, neutrophils, and dendritic cells. This review focuses on recent findings of the major H. capsulatum and host carbohydrate-driven interactions that account for internalization of fungal infective forms into phagocytes, and its subsequent avoidance of intracellular elimination. The yeast phase

Extracellular Proteins of Mycoplasma synoviae.

Mycoplasma synoviae is a Gram positive bacteria lacking of cell wall that affects chickens and turkeys causing infection in the upper respiratory tract and in some cases arthritis, with economical impact to broiler breeders. Treatment and prevention of avian synovitis depend on knowledge of the infectious process. Secreted or surface-exposed proteins play a critical role in disease because they often mediate interactions between host and pathogen. In the present work, we sought to identify possible M. synoviae secreted proteins by cultivating the bacteria in a modified protein-free Frey medium

Immunobiology of experimental cutaneous leishmaniasis.

The study of the murine model of infection with Leishmania major is providing important insights into the understanding of the complex interactions between the host and intracellular pathogens. Using this model system, basic research is actively leading to the identification of host factors promoting or circumventing the development of immunity to L. major. Here, Geneviève Milon, Giuseppe Del Giudice and Jacques A. Louis review recent results related to the characterization of immunological host factors determining resistance and susceptibility to this parasite, and try to identify areas where

Environmental and physiological factors influencing the survival of resistant Salmonella under infection-related gut conditions.

SUMMARYThe human gastrointestinal tract is a dynamic and complex ecosystem that plays a pivotal role in maintaining host health and regulating microbial homeostasis. This environment significantly impacts the survival and evolution of antimicrobial-resistant organisms. Various innate factors, including pH, temperature, osmotic stress, oxygen levels, host secretions, immune response, bile salts, and nutrient gradients, influence microbial behavior, interspecies interactions, and the spread of resistance genes to ensure successful colonization. The potential of the bacteria to avoid or overcome

Identification of host cell proteins that interact with the M protein of Pigeon paramyxovirus type 1.

Pigeon paramyxovirus type 1 (PPMV-1) belongs to the avian paramyxovirus type 1 group of viruses, which can cause tremors, torticollis, and respiratory signs in domestic and wild pigeons. The M protein of PPMV-1 is a multifunctional structural protein. It not only helps in the assembly, budding, and positioning of the virus but also inhibits the host's immune response and promotes replication of the virus in the host. In this study, the GST pull-down method was used to screen host proteins that interact with PPMV-1 M protein, and then mass spectrometry (MS) was used to analyse the screened host

Demonstration of transplacental transmission of a human isolate of Anaplasma phagocytophilum in an experimentally infected sheep.

Anaplasma phagocytophilum, first identified as a pathogen of sheep in Europe, more recently has been recognized as an emerging tick-borne pathogen of humans in the U.S. and Europe. Transmission of A. phagocytophilum is reported to be by ticks, primarily of the genus Ixodes. While mechanical and transplacental transmission of the type genus organism, A. marginale, occur in addition to tick transmission, these modes of transmission have not been considered for A. phagocytophilum. Recently, we developed a sheep model for studying host-tick-pathogen interactions of the human NY-18 A. phagocytophil

Host Species and Meteorological Influences on the Prevalence of Babesia microti (Aconoidasida: Piroplasmida) in Wild Rodents: Insights From a Meta-Analysis in Japan.

Rodents play a central role as reservoirs of zoonotic pathogens, yet the ecological and meteorological factors affecting pathogen prevalence remain unclear. In this study, a meta-analysis was used to evaluate the influence of host species and meteorological factors on the prevalence of Babesia microti in two common Japanese field mice, Apodemus speciosus and A. argenteus. The data were collected from seven published studies conducted across Japan. To examine the effects of total annual precipitation, average annual temperature, and host species, generalized linear mixed models were employed. T

Lymphocytes in host defense against Pneumocystis carinii.

Host defense against Pneumocystis carinii depends on complex interactions between host immune cells and mediators. In immunocompetent hosts, the immune system provides efficient and effective defense against P. carinii. Clinical and experimental investigations confirm that lymphocytes control and coordinate this defense. During states of immunosuppression, however, lymphocyte function is impaired and clinical P. carinii pneumonia results. Lymphocytes participate in host defense by regulating other immune cells (CD4+ and CD8+ T cells), by producing antibodies against pathogens (B cells), and by

Discovery of the Role of Tick Salivary Glands in Enhancement of Virus Transmission-Beginning of an Exciting Story.

There is increasing evidence that arthropod-borne pathogens exploit saliva of their vectors during the transmission process to vertebrate hosts. Extensive research of the composition of tick saliva and its role in blood-feeding and transmission of pathogens started in the late 1980s and led to a number of discoveries on the composition and function of salivary molecules, some of which are associated with pathogen transmission. The study by Jones et al. published in 1989 can be ranked among the pioneer works in this field as it demonstrated for the first time the role of tick salivary glands in

Enzyme activity and expression of catalases in response to oxidative stress in Sporothrix schenckii.

Sporothrix schenckii is a dimorphic fungus, pathogenic to humans and animals, which is usually infective in the yeast form. Reactive oxygen species (ROS) play an important role in the host's defense, damaging the pathogen's DNA, proteins, and lipids. To prevent oxidative damage, the ROS are detoxified by pathogen-derived antioxidant enzymes such as catalases (CATs). In this work, we analyzed the activity and expression level of three S. schenckii genes, designated as CAT1, CAT2, and CAT3, that putatively encoded for three isoforms of monofunctional CAT with a predicted molecular weight of 57.6

Spirochete Flagella and Motility.

Spirochetes can be distinguished from other flagellated bacteria by their long, thin, spiral (or wavy) cell bodies and endoflagella that reside within the periplasmic space, designated as periplasmic flagella (PFs). Some members of the spirochetes are pathogenic, including the causative agents of syphilis, Lyme disease, swine dysentery, and leptospirosis. Furthermore, their unique morphologies have attracted attention of structural biologists; however, the underlying physics of viscoelasticity-dependent spirochetal motility is a longstanding mystery. Elucidating the molecular basis of spiroche

Potential pharmacological strategies targeting the Niemann-Pick C1 receptor and Ebola virus glycoprotein interaction.

Niemann-Pick C1 (NPC1) receptor is an intracellular protein located in late endosomes and lysosomes whose main function is to regulate intracellular cholesterol trafficking. Besides being postulated as necessary for the infection of highly pathogenic viruses in which the integrity of cholesterol transport is required, this protein also allows the entry of the Ebola virus (EBOV) into the host cells acting as an intracellular receptor. EBOV glycoprotein (EBOV-GP) interaction with NPC1 at the endosomal membrane triggers the release of the viral material into the host cell, starting the infective
