Food-borne pathogens are ongoing problems, and new pathogens are emerging. The impact of fungi, however, is largely underestimated. Recently, commercial yogurts contaminated with Mucor circinelloides were sold, and >200 consumers became ill with nausea, vomiting, and diarrhea. Mucoralean fungi cause the fatal fungal infection mucormycosis, whose incidence has been continuously increasing. In this study, we isolated an M. circinelloides strain from a yogurt container, and multilocus sequence typing identified the strain as Mucor circinelloides f. circinelloides. M. circinelloides f. circinelloi

Metazoan organisms employ an array of overlapping innate and adaptive immune mechanisms to defend against infection and prevent disease. Z-nucleic acid-binding protein (ZBP)1 (also denoted DAI or DLM1) has emerged as a key nucleic acid (NA) sensor within mammalian cells responding to stress, infection or inflammation. Once engaged and activated by Z-nucleic acid, ZBP1 recruits receptor-interacting protein (RIP) kinase (RIPK)3 through RIP homotypic interaction motif (RHIM)-mediated interactions to promote regulated cell death and inflammatory signaling, with necroptosis representing a particula

Ca2+ signaling via the store-operated Ca2+ entry (SOCE) mediated by STIM1 and STIM2 proteins and the ORAI1 Ca2+ channel is important in saliva fluid secretion and has been associated with Sjogren's disease (SjD). However, there are no studies addressing STIM1/2 dysfunction in salivary glands or SjD in animal models. We report that mice lacking Stim1 and Stim2 [Stim1/2K14Cre(+)] in salivary glands exhibited reduced Ca2+ levels and hyposalivate. SOCE was functionally required for the activation of the Ca2+ activated Cl- channel ANO1. Ageing Stim1/2K14Cre(+) mice showed no evidence of lymphocytic

Pregnancy-associated malaria (PAM) caused by Plasmodium falciparum can result in detrimental outcomes for both mother and infant, including low infant birth weight, preterm birth, maternal anemia, spontaneous abortion, and maternal and/or infant mortality. Maternal anemia is a particularly complex outcome, as the body must both maintain erythropoiesis and tolerance of the growing fetus, while directing a Th1 response against the parasite. Underlying the pathogenesis of PAM is the expression of variant surface antigens (VSAPAM) on the surface of infected red blood cells (iRBC) that mediate sequ

To complement the development of new or repurposed drugs for improving the treatment outcomes of drug-susceptible and drug-resistant tuberculosis, current insight also focuses on the use of host-directed therapy. Metformin, a drug often used in the management of type 2 diabetes mellitus, has attracted attention by virtue of its favourable activity as an adjunctive agent against tuberculosis, discovered through laboratory and clinical studies. To definitively establish its role as a host-directed therapeutic in tuberculosis, more preclinical and clinical research is still required to better del

Leishmaniasis is an important parasitic disease found in the tropics and subtropics. Cutaneous and visceral leishmaniasis affect an estimated 1.5 million people worldwide. Despite its human health relevance, relatively little is known about the cell death pathways that control Leishmania replication in the host. Necroptosis is a recently identified form of cell death with potent antiviral effects. Receptor interacting protein kinase 1 (RIPK1) is a critical kinase that mediates necroptosis downstream of death receptors and TLRs. Heme, a product of hemoglobin catabolism during certain intracellu

Necrosis has long been considered as a passive event resulting from a cell extrinsic stimulus, such as pathogen infection. Recent advances have refined this view and it is now well established that necrosis is tightly regulated at the cell level. Regulated necrosis can occur in the context of host-pathogen interactions, and can either participate in the control of infection or favor it. Here, we review the two main pathways implicated so far in bacteria-associated regulated necrosis: caspase 1-dependent pyroptosis and RIPK1/RIPK3-dependent necroptosis. We present how these pathways are modulat

PURPOSE OF REVIEW: Many cellular processes are controlled via either stable or transient protein-protein interactions (PPIs). Protein complexes are 'molecular machines' in which multiple interactive partners carry out various cellular functions. Given that almost a third of the proteome consists of membrane proteins and that more than 50% of currently available drugs are targeted toward them, investigation of membrane protein complexes has taken center stage over the past years. Thus, gaining an in-depth understanding of PPI networks will give us more insight into the functional relationship a

Acamprosate is a new psychotropic drug used in the treatment of alcohol (ethanol)-dependence. Recent studies suggest that acamprosate inhibits neuronal hyperexcitability by antagonising excitatory amino acids. It is available as a 333 mg enteric-coated tablet, with a recommended dosage of 1.3 g/day for patients with a bodyweight < 60 kg and 2 g/day for patients with a bodyweight > or = 60 kg. Treatment with higher dose strength tablets 2 x 500 mg twice daily is bioequivalent to treatment with the 2 x 333 mg 3 times daily dosage regimen. Acamprosate is absorbed via the paracellular route in the

Prolonged confinement is a consequence of amoeboid cancer-cell invasion. In this issue of Developmental Cell, Zhang et al.1 show that sustained mechanical stress activates PIEZO1-STAT1-dependent MLKL expression, leading to MLKL-dependent membrane-lytic death in cells without detectable RIPK3 and canonical MLKL phosphorylation, broadening current models of necroptotic competence.

Pathogenic bacteria are remarkably inventive when it comes to manipulating their hosts. In this issue of Cell Host & Microbe, McCaffrey et al. identify a previously uncharacterized bacterial effector from Legionella pneumophila that modifies not host proteins but messenger ribonucleic acid (mRNA) to dampen the host inflammatory response.

Host-microbial co-metabolites are small bioactive molecules generated through obligatory sequential or complementary enzymatic transformations by both gut microbiota and host tissues, including secondary bile acids, trimethylamine N-oxide (TMAO), indoxyl sulfate, p-cresyl sulfate, phenylacetylglutamine, and hippurate. The dysregulation of this co-metabolic axis, often through gut microbial dysbiosis, contributes to chronic low-grade inflammation and has been implicated in inflammatory bowel disease, metabolic disorders, cardiovascular disease, kidney disease, neurological disorders, and cancer

Dermatophytosis, a prevalent superficial fungal infection caused by fungi such as Trichophyton rubrum, Trichophyton mentagrophytes, and Microsporum canis, develops through the dynamic interaction between fungal virulence factors - including adhesion, secretion of keratinolytic proteases, and hyphal invasion - and the host's multifaceted immune response, which involves antimicrobial peptides, pattern recognition receptors, and cellular immunity. Recent evidence identifies non-coding RNAs (ncRNAs), including miRNAs, lncRNAs, circRNAs, and tRFs, as key regulators of this host-fungal crosstalk. Th

The human gut microbiome exhibits profound interindividual variability, challenging the efficacy of generalized interventions and underscoring the need for precision approaches to microbiome-associated diseases. Such strategies require comprehensive, individualized assessment of microbial ecosystems and their functional responses, an objective that remains difficult to achieve in vivo. In vitro gut models offer a promising alternative, maintaining personalized microbial communities in controlled environments and enabling detailed investigation of microbiome dynamics. Ulcerative colitis (UC), a chronic inflammatory bowel disorder characterized by progressive colonic inflammation, presents escalating global incidence rates and heightened risk of colorectal carcinogenesis. Despite emerging evidence implicating lactylation - a novel post-translational modification - in diverse biological pathways, its mechanistic involvement in UC pathogenesis remains poorly elucidated. Through integrative analysis of GEO datasets, we systematically identified lactylation-associated signatures in UC using differential expression profiling, weighted gene co-expression network a

Store-operated calcium entry (SOCE), mediated by the endoplasmic reticulum (ER) Ca2+ sensors stromal interaction molecule (STIM) proteins and the plasma membrane (PM) Orai channels, is essential for calcium signaling and a wide range of physiological processes. Precise regulation of SOCE is critical for maintaining tissue homeostasis, whereas its dysregulation contributes to diverse pathological conditions, particularly organ fibrosis. In this review, we outline the molecular basis of SOCE and discuss how its dysregulation is implicated in human disease. We further emphasize the pivotal role o

INTRODUCTION: Overgrowth and colonization by non-typeable Haemophilus influenzae (NTHi) is a common feature of increasing disease severity, treatment resistance and increased susceptibility to disease exacerbations across chronic airways diseases (CADs). Whether NTHi is a driver of respiratory disease or reflects that the damaged airway has become a permissive environment for growth remains to be proven.
AREAS COVERED: In this review, we discuss the potential roles of hypermutation, biofilm formation and intracellular living in allowing NTHi to adapt to living in the lungs of individuals with

Viral infectious diseases pose serious threats to global public health. Host non-coding RNAs (ncRNAs), including miRNAs, lncRNAs, circRNAs, and vtRNAs, act as master regulators of host antiviral defense and viral replication. Current studies on ncRNA-virus interactions are generally fragmented, and most reviews focus on a single virus or a single class of ncRNAs, without combining ncRNA expression dynamics with their biological functions for systematic collation. Herein, we establish a novel four-category classification framework based on ncRNA expression patterns and functional characteristic

Social interactions across species rely on neural representations of self- and other-models that guide sequential behavioral exchanges. In this review, we propose an integrated framework in which these internal models interact with endocrine systems to regulate mating behavior and mother-infant bonding. External sensory cues and internal bodily states are combined in the central nervous system to generate predictions about a partner's actions and to adjust one's own behavior accordingly. Sex hormones organize and activate sex-specific neural circuits that stabilize male- and female-type behavi

Wound closure by suture and staple demands surgical skills and carries risks of traumatic complications, whereas current tissue adhesives exhibit insufficient wet adhesion strength, unsatisfactory in vivo biodegradability, and durability. Inspired by the robust underwater adhesion of mussels, whose underwater adhesion derives from the synergy of hydrophobic and adhesive matrix, a tough bioadhesive (PLD) was developed by coassembly of lipoic acid (LA) and a dopamine-functionalized lipoic acid derivative (LAD). Mimicking the mussel's adhesion mechanism, PLD expelled interfacial water via hydroph
