Haematological toxicity associated with antineoplastic drugs: a pharmacovigilance analysis based on the FDA adverse event reporting system database.

BACKGROUND: Antineoplastic drugs are effective for malignant tumours, but they frequently cause severe adverse drug reactions (ADRs), which seriously affect treatment continuity and patient prognosis. Post - marketing real - world safety monitoring is therefore critical. METHODS: We conducted a retrospective pharmacovigilance study using FAERS (FDA Adverse Event Reporting System) data covering the period from 2004 Q1 to 2025 Q4. Safety signals were detected via disproportionate analysis, and associations were identified through LASSO and multivariable logistic regression, followed by an analy

Between Denial and Delay: Barriers to Accessing Nonpharmacological Interventions for Seniors With Mild-to-Moderate Dementia in Canada.

Dementia is a growing public health concern in Canada, yet timely access to nonpharmacological interventions (NPIs) for persons with dementia (PWDs) remains poorly understood. This critical ethnographic study explored factors influencing access to NPIs among private-dwelling seniors with mild-to-moderate dementia and their family/friend care partners (CPs) in Alberta, Canada. Guided by McLeroy et al.'s Ecological Perspective and Habermas' Critical Social Theory, we conducted 53 semi-structured interviews with eight PWDs and 21 CPs across 20 families. Reconstructive, thematic analysis revealed

Appetite for change - the need to revisit malnutrition screening and assessment in oncology.

Malnutrition is highly prevalent among oncology patients, with large-scale studies reporting involuntary weight loss in 31-87%, depending on tumour site and disease stage. A combination of nutrition-impact symptoms, reduced oral intake and systemic inflammation lead to poor tolerance to treatment, diminished quality of life and reduced survival. Systemic inflammation is a hallmark of cancer-associated malnutrition and contributes to loss of lean mass and abnormal body composition phenotypes (sarcopenia, cachexia and low muscle density) which may coexist with overweight and obesity. Malnutritio

Kinetic and biocatalytic evaluation of dodecyl nicotinate hydrolysis in aqueous buffers and biological media.

OBJECTIVE: This study evaluated the hydrolytic behavior of dodecyl nicotinate under chemical and biological conditions to determine its suitability as a controlled-release prodrug of niacin. SIGNIFICANCE: Niacin therapy is limited by a short half-life and dose-dependent flushing, which compromise adherence despite its lipid-modifying benefits. Prodrug design provides a strategy to enhance stability, prolong release, and facilitate enzymatic activation. Dodecyl nicotinate, a lipophilic C12 ester of niacin, is expected to balance membrane permeability with enzymatic lability. While shorter (oct

Immunomodulatory Empty/Hollow Nanoparticles as Potential Therapeutic Strategies for Septic Shock.

Septic shock is a life-threatening manifestation of sepsis characterized by dysregulated immune responses, excessive inflammation, oxidative stress, and progressive multi-organ dysfunction. Despite advances in antimicrobial therapy and supportive care, mortality remains high, highlighting the need for therapeutic strategies that target immune dysregulation in addition to infection control. The review evaluates the potential of hollow nanoparticles as immunomodulatory therapies for septic shock, focusing on lipid-based, polymeric, protein-based, biomimetic, inorganic, carbon-based, and hybrid n

Manual wheelchair training for people with chronic and progressive conditions: priority recommendations for a qualitative multi-method study.

PURPOSE: Manual wheelchair training is essential for safe and independent wheelchair use; however, provision remains inconsistent and lacks alignment with the needs of people who commence wheelchair use later in life secondary to a chronic or progressive condition. Existing recommendations call for coordinated service delivery, clearer accountability and safe, contextually relevant training, but offer limited guidance on how recommendations should be prioritised or implemented. This study aimed to prioritise recommendations and provide practical guidance for manual wheelchair training for peop

Feasibility of metabolite-mediated bioleaching on composite-mineral waste from FRP strengthening: experimental insights under realistic debonding.

The present study investigates the end-of-life management of fiber-reinforced polymer (FRP) strengthening systems, which are widely adopted to enhance structural safety, by exploring metabolite-mediated bioleaching as a sustainable treatment strategy for composite-mineral waste. In particular, the study focuses on residues resulting from debonding phenomena, which reflect the actual condition of FRP systems removed from existing structures. To this aim, single shear-lap tests were performed on concrete and masonry specimens strengthened with externally bonded carbon FRP strips, with the object

Context-dependent regulation of IL-6 in exercise and colorectal cancer cachexia.

Colorectal cancer (CRC) is a common malignancy of the digestive system and remains a major cause of cancer-related morbidity and mortality worldwide. Its development and progression are influenced by genetic susceptibility, lifestyle factors, metabolic dysregulation, and chronic inflammatory states. Cancer cachexia is a multifactorial wasting syndrome characterized by weight loss, skeletal muscle atrophy, metabolic disturbances, and functional decline, which substantially impairs quality of life and survival outcomes in patients with CRC. In this context, interleukin-6 (IL-6) has emerged as a

[Multisystem interactive mechanisms of cancer-related fatigue: challenges of modern medical interventions and prospects for integration with TCM].

Cancer-related fatigue(CRF) is one of the most distressing symptoms in patients with malignant tumors, characterized by persistent physical and mental exhaustion that is not alleviated by rest, significantly impairing patients' quality of life. The pathogenesis of CRF involves complex multi-system interactions, which can be summarized as neuro-immune dysregulation and skeletal muscle and mitochondrial dysfunction. Currently, modern medicine faces significant challenges in the clinical management of CRF, including unclear mechanisms of pharmacological interventions, insufficient evidence-based

Vagus nerve-driven microbiota homeostasis: a promising integrative add-on therapy for neurodevelopmental disorders.

Over the past two decades, the conceptualization of neurodevelopmental disorders (NDDs) has undergone a profound transformation, shifting from a primarily brain-centric framework toward a systems-level perspective that integrates peripheral physiological processes. Among these, the gut microbiota has emerged as a critical determinant of neurodevelopmental trajectories. Through its involvement in immune modulation, metabolic signaling, and neural communication, the microbiota-gut-brain axis (MGBA) exerts a pervasive influence on brain maturation and function. A growing body of evidence indicate

New strategies for valve replacement and interventional options in adults with congenital heart disease: new insights from Euro-ACHD 2025.

Recent advances in congenital heart disease management reflect a paradigm shift towards less invasive, physiology-preserving, and durable interventions. Transcatheter pulmonary valve implantation has become an established therapy for right ventricular outflow tract dysfunction, supported by multiple valve systems tailored to increasingly complex anatomies. These devices have demonstrated high procedural success and sustained hemodynamic performance, although infective endocarditis and arrhythmic complications remain clinical concerns. In parallel, transcatheter edge-to-edge repair for atrioven

Esophageal Exclusion as a Damage-Control Strategy for Acute Esophageal Necrosis Secondary to Ruptured Stanford Type B Aortic Dissection: A Case Report.

Acute esophageal necrosis (AEN) is a rare and life-threatening condition most commonly associated with systemic hypoperfusion and critical illness. Aortic-related AEN is exceptionally uncommon and may result from direct mediastinal compression and vascular compromise rather than generalized hemodynamic insufficiency. We report the case of a 77-year-old man presenting with a Stanford type B aortic dissection associated with aneurysmal degeneration complicated by rupture. The patient presented with progressive dysphagia, back pain, and hemoptysis. Computed tomography angiography demonstrated a l

An atypical presentation of Pembrolizumab-induced bullous pemphigoid in a patient with uterine serous carcinoma.

BACKGROUND: Immune checkpoint inhibitor-induced bullous pemphigoid (ICI-BP) is a rare but recognized complication of immune checkpoint inhibitor therapy. Compared to conventional BP, ICI-BP can present atypically, leading to delayed diagnosis and treatment, which can devastate quality of life for cancer patients. Literature on the presentation, diagnosis, and management of ICI-BP in patients with gynecologic cancer is scarce. CASE PRESENTATION: We report a case of ICI-BP presenting as erythema multiforme (EM)-like lesions in a woman in her early 60s with stage IVB uterine serous carcinoma. Up

Specific aspects of kidney care for Africans living in Europe.

Chronic kidney disease (CKD) disproportionately affects people of African ancestry worldwide including those living in Europe and the United States (US), with overrepresentation among those with advanced CKD and kidney failure. This excess burden reflects the convergence of genetic susceptibility, including Apolipoprotein-L1 (APOL1) risk variants and sickle-cell-related nephropathy, with early-life factors such as low birth weight, high rates of hypertension and diabetes, and recurrent acute kidney injury from infections or environmental exposures. Social and structural drivers including pover
