Protective and pathogenic: The Janus-faced role of cGAS-STING in the brain.

Innate immune signaling preserves brain integrity during infection and sterile injury but can also drive neurodegeneration and cognitive decline. Over the past 2 decades, work has shown that cytosolic DNA sensing via the cGAS-STING pathway can precondition the brain against ischemic injury while impairing post-stroke recovery. Our interest in this pathway arose from the discovery that tilorone, an antiviral immunomodulator, activated a hypoxia-inducible factor (HIF) reporter. Although pre-stroke tilorone treatment was strongly neuroprotective, mechanistic studies revealed that type I interfero

Rapid Manufactured CAR-T Cells enriched for CD45RA-negative T cells Exhibit Superior Persistence Compared to Conventional CAR-T Therapy.

CD19 CAR-T has achieved high remission rates in relapsed/refractory B-cell acute lymphoblastic leukemia (B-ALL). However, rapid generation with durable in vivo persistence remains a major challenge. CD45RA-negative T cells exhibit sustained antileukemic activity and preserved immune memory. Here, we developed a rapid protocol (InstanCAR-T) enriched for CD45RA-negative T cells, and systematically compared it with conventional CAR-T across in vitro functional assays, murine B-ALL xenograft models, and a Phase I clinical trial. InstanCAR-T shortened ex vivo culture to 3 days while enriching CD45R

Do subjective and objective baseline sleep disturbances predict post-traumatic stress disorder treatment response? A secondary analysis of a randomized controlled trial.

Background: Sleep disturbances are highly prevalent in individuals with post-traumatic stress disorder (PTSD) and might interfere with trauma-focused treatments by disrupting memory consolidation, extinction and safety learning. However, evidence on the impact of sleep deficits on PTSD treatments remains inconclusive. Objective: This study conducted a secondary analysis of a randomized controlled trial comparing cognitive processing therapy (CPT) and dialectical behaviour therapy for PTSD (DBT-PTSD) in women with PTSD. We examined whether subjective and objective baseline sleep disturbances pre

Fatigue resistant elastocaloric effect in TiNi via texture-precipitate synergy.

Shape memory alloys with both a large elastocaloric effect and exceptional fatigue resistance are key candidates for next-generation solid-state cooling technologies. Here, we report a textured TiNi alloy containing engineered Ti₄Ni₂O precipitates, fabricated via controlled directional solidification, that achieves a large adiabatic temperature change of -15.9 K after 10 million compressive cycles. Microstructural analyses reveal columnar B2 grains aligned with the solidification direction and a uniform dispersion of high-density Ti₄Ni₂O precipitates. A strong crystallographic texture is obser

Fractional-Order Spiking Bayesian Neural Model for Cognitive Computations.

Cognition under uncertainty can be formalized through Bayesian inference, but biologically plausible neural implementations remain a challenge. This study develops a Bayesian neural model for lifespan prediction that integrates fractional-order dynamics into classical Leaky Integrate-and-Fire and Izhikevich neuron models. The inclusion of fractional derivative introduces long-term memory, and thus enhancing both biological plausibility and representational capacity of the Bayesian neural model. Experimental results demonstrate that fractional-order neuron models consistently provide closer ali

Igniting cold tumors: reprogramming the tumor immune microenvironment with dual-payload ADCs in combination with PD-L1&4-1BB bispecific antibodies.

BACKGROUND: The immunologically "cold" microenvironment of triple-negative breast cancer presents a major obstacle to treatment, conferring resistance to current immunotherapies. Targeting the stimulator of interferon genes (STING) pathway has emerged as a promising strategy to enhance immunotherapy efficacy. METHODS: In this preclinical study, we leveraged a novel integrated approach by employing a B7-H3-targeted dual-payload antibody-drug conjugate (ADC) to induce innate immune responses and reprogram the tumor microenvironment. RESULTS: Our preliminary results indicated that the dual-payl

Aluminum Chloride-Driven Neurodegeneration: Interlinking Redox Imbalance, Inflammation Cascade, and Excitotoxic Damage Across Molecular and Behavioral Domains.

A common environmental toxin, aluminum chloride (AlCl₃), has been closely linked to the development of neurological and neurodevelopmental diseases. This review incorporates recent research showing how long-term exposure to AlCl₃ impairs the brain's cellular, molecular, and behavioral processes. AlCl₃ causes significant oxidative stress, which is typified by the production of excessive amounts of reactive oxygen species, mitochondrial malfunction, and the activation of apoptotic pathways. Neuronal susceptibility is further exacerbated by parallel neuroinflammatory responses, such as increased

Cannabinoids in Alzheimer's disease: animal-human evidence and clinical pharmacology challenges.

Cannabinoids have emerged as potential modulators of pathological processes in Alzheimer's disease (AD), including neuroinflammation, synaptic dysfunction, and protein aggregation. Cannabidiol (CBD) and Δ9-tetrahydrocannabinol (THC), the main phytocannabinoids from Cannabis sativa, interact with the endocannabinoid system and may influence neuronal and glial signaling pathways relevant to AD pathology. This mini review summarizes evidence from transgenic animal models and clinical studies evaluating CBD, THC, and their combination in AD. Preclinical studies show that CBD and THC reduce β-amylo

Food-based multisensory stimulation ameliorates cognitive impairment after mild traumatic brain injury in male rats by modulating intestinal and brain inflammation.

Mild traumatic brain injury (mTBI) often leads to cognitive impairment (CI), with neuroinflammation and gut microbiota dysbiosis playing pivotal roles in its pathogenesis. This study aimed to investigate whether food-based multisensory stimulation could ameliorate cognitive deficits in mTBI rats via modulation of the gut-brain axis. Using a rat model of mTBI, we demonstrated that food-based multisensory stimulation significantly improved spatial and recognition memory, as evidenced by performance in the Morris water maze and novel object recognition tests, and reduced serum biomarkers of neuro

Towards an Integrative Framework of Self-Regulation: A Scoping Review on the Interplay Between Emotion Regulation, Executive Functions and Decision-Making in Clinical Populations.

BACKGROUND/OBJECTIVE: Emotion regulation (ER) is closely linked to decision-making (DM). Although executive functions (EF) are often suggested as a key mechanism underlying this relationship, evidence from different clinical conditions remains fragmented. This scoping review aims to map the literature on the relationship between ER and DM in clinical populations, with a specific focus on the interaction between EF and ER in shaping adaptive DM processes, particularly in populations characterised by cognitive impairment and emotional dysregulation. METHODS: A search of electronic databases was

Innovative multi-epitope vaccine development for rheumatoid arthritis via immunoinformatics.

Rheumatoid arthritis (RA) is a chronic autoimmune disorder marked by persistent joint inflammation, pain, and progressive disability, affecting millions worldwide. Current therapies, including anti-inflammatory and immunosuppressive agents, provide symptomatic relief but fail to offer long-term disease control or immune tolerance, highlighting the need for innovative therapeutic strategies. Here, we present a novel multi-epitope vaccine designed using immunoinformatics to target key RA-associated molecules-TNF-α, IL-23R, PTPN22, PADI2, and PADI4-critical drivers of inflammation and autoimmunit

GPR68, a proton-sensing GPCR, as a therapeutic target in depression: structural, signalling, and mechanistic perspectives.

Brain acid signalling refers to the process by which brain tissue utilizes protons (H+) as signalling molecules to sense changes in the extracellular microenvironmental pH via specific receptors such as proton-sensing GPCRs. This signalling initiates downstream pathways that regulate neural activity and maintain brain homeostasis. As a prototypical proton-sensing GPCR, GPR68 plays a pivotal role in maintaining physiological homeostasis, regulating tissue functions, and contributing to pathological processes, owing to its high sensitivity to extracellular pH changes. GPR68 is expressed in neuro

The brain angiotensin converting enzyme 2 and angiotensin-(1-7) as potential therapeutic targets for the central nervous system diseases.

The renin-angiotensin system (RAS) serves as a key regulator of body fluid balance and cardiovascular homeostasis, whose dysregulation is involved in the pathogenesis of various central nervous system (CNS) diseases. Beyond their canonical functions such as vascular regulation, the constituents of the RAS, particularly angiotensin (Ang) II, exert inflammatory and oxidative injury on neurons in the brain. Meanwhile, the non-classical pathway angiotensin-converting enzyme (ACE)2/Ang-(1-7) axis shows beneficial effects against CNS diseases. Activation of cerebral ACE2/Ang-(1-7) exerts regulatory

Repeated mRNA vaccination and breakthrough infection reveal durable dominance but diminishing recall of vaccine-induced CD8⁺ T-cell clones.

Successive mRNA vaccinations preserve severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2)-specific T-cell immunity, but how individual CD8⁺ T-cell clones behave across repeated booster doses and breakthrough infection (BTI) remains poorly defined. We longitudinally tracked bulk CD8⁺ T-cell receptor beta (TCRβ) repertoires and spike-specific tetramer⁺ CD8⁺ T cells in adult participants across the third and fourth vaccine doses and subsequent BTI. Each booster continued to recruit previously unexpanded "new" responder clonotypes, but both the number and summed frequency of newly engaged

Gut microbiota dysbiosis is associated with the synaptic deficits and cognitive impairment induced by LCM mixtures within a proposed mechanistic framework consistent with the AOP concept.

Liquid crystal monomers (LCMs), as emerging environmental contaminants, have poorly understood cognitive effects under realistic mixture exposure scenarios. This study integrated ecological and human exposure data to characterize LCM mixtures and assessed their neurotoxicity using in vitro and in vivo models. We found that long-term exposure to LCM mixtures at environmental levels more strongly inhibited viability in hippocampal HT22 cells and microglial BV2 cells than individual compounds, and was associated with memory deficits in mice accompanied by neuroinflammation and reduced hippocampal
